| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Vulnerability and adaptation of an oasis social–ecological system affected by glacier change in an arid region of northwestern China显示文摘The Hexi Inland River Basin in an arid region of northwestern China was chosen as the study area for this research. The authors define the vulnerability of an oasis social-ecological system to glacier change; select 16 indicators from natural and socioeconomic systems according to exposure, sensitivity, and adaptive capacity; and construct a vulnerability-assessment indicator system aimed at an inland river basin in the arid region of Northwestern China. Vulnerability of the oasis socialecological system affected by glacier change in the study area is evaluated by Spatial Principal Component Analysis(SPCA) under the circumstance of glacier change. The key factors affecting the vulnerability are analyzed. The vulnerability of the oasis social-ecological system in the Hexi Inland River Basin affected by glacier change is of more than medium grade, accounting for about 48.0% of the total number of counties in the study area. In terms of the spatial pattern of the vulnerability, the oasis economic belt is the most vulnerable. With the rapid development of the area's society and economy, the exposure of the system to glacial changes is significantly increased; and an increase in glacial meltwater is not enough to overcome the impact of increased exposure, which is the main reason for the high vulnerability. | 3 | Leucine modulates the IPEC-J2 cell proteome associated with cell proliferation, metabolism and phagocytosis显示文摘Leucine can affect intestinal protein expressions, and improve mucosal immune function. However, little study has been conducted to determine the change of protein component by leucine treatment in intestine epithelial cells. The present study was to cover the key proteins and cell pathways that could be regulated by leucine treatment in porcine intestinal epithelial cell line(IPEC-J2) cells with the approach of proteome analysis. A total number of 3,211 proteins were identified in our approach by searching the database of Uniprot sus scrofa. Among identified proteins, there were 101 proteins expressed differently between control group and leucine group. Compared with the control group, there were 50 up-regulated proteins and 51 down-regulated proteins in leucine group. In these proteins, leucine treatment decreased the expression of some proteins including pyruvate kinase, glyceraldehyde-3-phosphate dehydrogenase,E3 ubiquitin ligase, cathepsin D, caspase 3 and caspase 6, and increased the levels of some proteins, such as some eukaryotic translation initiation factors, ubiquitin carboxyl-terminal hydrolase, DNA-related RNA polymerase II, urokinase plasminogen activator, cyclin-dependent kinase inhibitor 2 b, Mut L homolog 1,5-methylcytosine binding domain 4, polymerase d, a-tubulin, syntaxin 18, Ras homolog D, actin related protein 2/3 complex and cofilin. Via the analysis of Gene Ontology and pathways, these proteins in IPECJ2 cells were related with some physiological functions, such as protein metabolism, glycolysis, cell proliferation, apoptosis and phagocytosis. | 5 | Photosynthetic Physiological Response to Drought Stress of Populus euphratica at Different Ages in Minqin显示文摘[Objectives]This study was conducted to investigate the photosynthetic physiological characteristics of Populus euphratica trees at different forest ages in the desert area of Minqin in response to drought stress.[Methods]With P.euphratica trees of different ages in Minqin as the research object,the water characteristics and photosynthetic physiological indexes(chlorophyll,soluble sugar,POD,SOD and MDA)were compared under different conditions.[Results]On the time gradient,the soil water contents of P.euphratica of different ages decreased continuously with the extension of irrigation stop time,but the decreases were small.Under the same stress treatment,the soil water contents of P.euphratica of different ages increased with the deepening of soil layer,but the differences were not significant(P>0.05).Under normal condition,the chlorophyll contents of P.euphratica at three ages gradually increased with the increase of age.The chlorophyll contents in leaves of P.euphratica at different ages were all lower under normal condition and higher under water stress.The changes of POD and SOD activity in leaves of P.euphratica at different ages under different conditions were basically the same,showing that the enzyme activity was higher under water stress than under normal under.The MDA contents in leaves of the young and middle P.euphratica forests were higher under water stress at 7 and 21 d than under normal condition,and the differences were not significant(P>0.05);and the MDA content in leaves of the mature P.euphratica forest was higher under water stress at 21 and 35 d than under normal condition.Drought stress has a certain effect on the photosynthetic physiological characteristics of P.euphratica.In summary,under drought stress,the chlorophyll content,SOD and POD activity,and MDA molar concentration in the leaves of P.euphratica were basically higher than under normal condition,indicating that P.euphratica could resist drought environment through osmotic adjustment and showed strong drought resistance.[Conclusions]This study provides a theoretical reference for the restoration,protection and reconstruction of natural P.euphratica forests in the desert area of Minqin. | Chunxiu GUO Jianping MA Junmei MA Duoqing MAN Fei WANG Yunian ZHANG Peng ZHAO | 2020 | Agricultural Biotechnology2020,9,4: | 0 |
| 6 | High-performance ZnO humidity sensor synthesized by coprecipitation with PVP as surfactant for human respiration detection显示文摘A high-performance zinc oxide(ZnO)/polyvinylpyrrolidone(PVP)humidity sensor was prepared by simple coprecipitation method with PVP as surfactant.The coprecipitation method has low reaction temperature,little energy consumption and simple preparation,which is suitable for large-scale production.The PVP makes sample’s surface with more hydroxyl and oxygen vacancies than pure ZnO,which can absorb more water molecules and promote the decomposition of water molecules into H3O+to form effective ion conduction.When the molar ratio of PVP to ZnO is 1:1,the ZnO/PVP humidity sensor has low hysteresis(~4.2%),short response/recovery time(9/10 s),excellent stability and high sensitivity with more than 4 orders of magnitude in relative humidity(RH)range from 11%to 95%.Moreover,the ZnO/PVP humidity sensor can distinguish different respiratory states of human body,which has a potential in monitoring and prevention of respiratory diseases. | MAN Jianping GU Weiyuan HU Ziyan DOU Xiaozong ZHANG Hongyan | 2023 | Optoelectronics Letters2023,19,1: | 0 |
