| 1 | Expression and function of classical protein kinase C isoenzymes in gastric cancer cell line and its drugresistant sublines显示文摘AIM: To investigate the expression and function of classicalprotein kinase C (PKC) isoenzymes in inducing MDRphenotype in gastric cancer cells.METHODS: Two cell lines were used in the study: gastriccancer cell SGC7901 and its drug-resistant cell SGC7901/VCRstepwise-selected by vincristine 0.3, 0. 7 and 1.0 mg@ L-1 ,respectively. The expression of classical PKC (cPKC)isoenzymes in SGC7901 cells and SGC7901/VCR cells weredetected using immunofluorescent cytochemistry, laserconfocal scanning microscope and Wsstern blot. The effectsof anti-PKC isoenzymes antibody of adriamycinaccumulation in SGC7901/VCR cells were determined usingflow cytometric analysis.RESULTS: (1) SGC7901 cells exhibited positive staining ofPKC-α. SGC7901/VCR cells exhibited stronger staining ofPKC-α than SGC7901 cells. The higher dosage vincristineselected, the much stronger staining of PKC-α was observedon SGC7901/VCR cells. (2) Both SGC7901 and SGC7901/VCRcells exhibited positive staining of PKC-βⅠ and PKC-βⅡ withno significant difference. ( 3 ) Compared with SGC7901,SGC7901/VCR cells had decreased adriamycin accumulationand retention. Accumulation of adriamycin in SGC7901 was5.21 + 2.56 mg@ L-1, in SGC7901/VCR 0.3 was 0.85 + 0.29 mg@L-1 , in SGC7901/VCR 0.7 was 0.81 + 0.32 og@ L-1 , and inSGC7901NCR 1.0 was 0.80 + 0.33 mg @ L-1; Retention ofadriamycin in SGC 7901 was 2.51 + 1.23 mg@L-1, in SGC7901/VCR 0.3 was 0.47 + 0.14 mg@ L-1 , in SGC7901/VCR 0.7 was 0.44 + 0.15 mg@ L-1, and in SGC 7901/VCR 1.0 was 0.41 + 0.1 1mg @ L-1 . | 3 | Efficacy and safety of low-dose interleukin-2 in combination with methotrexate in patients with active rheumatoid arthritis:a randomized,double-blind,placebo-controlled phase 2 trial显示文摘Rheumatoid arthritis(RA)is an aggressive autoimmune arthritis,and current therapies remain unsatisfactory due to low remission rate and substantially adverse effects.Low-dose interleukin-2(Ld-IL2)is potentially a therapeutic approach to further improve the disease.This randomized,double-blind,placebo-controlled trial was undertaken to evaluate the efficacy and safety of Ld-IL2 in patients with active RA.Patients were randomly assigned(1:1)to receive Ld-IL2,defined as a dose of 1 million IU,or placebo in a 12-week trial with a 12-week follow-up.Three cycles of Ld-IL2 or placebo were administered subcutaneously every other day for 2 weeks(a total of 7 doses),followed by a 2-week break.All patients received a stable dose of methotrexate(MTX).The primary outcomes were the proportion of patients achieving the ACR20,DAS28-ESR<2.6,and the change from baseline in CDAI or SDAI at week 24.Secondary endpoints included other clinical responses and safety.The primary outcomes were achieved in the perprotocol population.The improvements from baseline in CDAI and SDAI were significantly greater across time points for the LdIL2+MTX group(n=17)than for the placebo+MTX group(n=23)(P=0.018 and P=0.015,respectively).More patients achieved ACR20 response in the Ld-IL2+MTX group than those in the placebo+MTX group at week 12(70.6%vs 43.5%)and at week 24(76.5%vs 56.5%)(P=0.014).In addition,low Treg and high IL-21 were associated with good responses to Ld-IL2.Ld-IL-2 treatment was well-tolerated in this study.These results suggested that Ld-IL2 was effective and safe in RA.ClinicalTrials.gov number:NCT 02467504. | Xiaoying Zhang Miao Miao Ruijun Zhang Xu Liu Xiaozhen Zhao Miao Shao Tian Liu Yuebo Jin Jiali Chen Huixin Liu Xia Zhang Yun Li Yunshan Zhou Yue Yang Ru Li Haihong Yao Yanying Liu Chun Li Yuhui Li Limin Ren Yin Su Xiaolin Sun Jing He Zhanguo Li | 2022 | Signal Transduction and Targeted Therapy2022，7，4： | 7 |

| 15 | Dissipative Particle Dynamics Simulation on Bonding Reaction Between Surface Modified Nanoparticles显示文摘A simulation study was carried out by using dissipative particle dynamics(DPD) method to explore the effects of properties of coating chains, such as length, density, rigidity of polymer chains, as well as the distance between nanoparticles on bonding reaction of coating chains grafted onto nanoparticles. The results show that bonding ratios of coated chains strongly depend on the length and density of coating chains. For nanoparticles with different coating densities, the optimum chain length for bonding reaction are varied. The rigidity of coating chains exhibits vigorous effects on bonding reaction that highly depends on chain lengths.
