1Immune mechanisms of Concanavalin A model of autoimmune hepatitis显示文摘As a chronic inflammatory disease of the liver,the pathogenic mechanisms of autoimmune hepatitis (AIH) have not yet been elucidated,with prognosis and diagnosis remaining unsatisfied.Currently the only viable treatments of AIH are immunosuppressant application and liver transplantation.It is considered that lack of good animal AIH models is the main reason for the shortage of a simple and efficient cure.The Concanavalin A (Con A) model is a typical and well established model for investigating T-cell and macrophage dependent liver injury in mice,which closely mimics the pathogenesis mechanisms and pathological changes of patients,and is regarded as the best experimental model for AIH research so far.In this paper we eluci-dated the pathogenic mechanisms of AIH and the evolution of relative animal models.We go on to further focus on Con A-induced liver injury from the point of immunological mechanisms and the change of cytokine levels.Finally,we manifested the clinical significance of the AIH animal models and the challenges they would meet during their future development.Hai-Xia Wang Man Liu Shun-Yan Weng Jing-Jing Li Chao Xie Hong-Lin He Wen Guan Yun-Sheng Yuan Jin Gao 2012World Journal of Gastroenterology2012,18,2:52

3Atomically dispersed Au_1 catalyst towards efficient electrochemical synthesis of ammonia显示文摘Tremendous efforts have been devoted to explore energy-efficient strategies of ammonia synthesis to replace Haber–Bosch process which accounts for 1.4% of the annual energy consumption. In this study,atomically dispersed Au_1 catalyst is synthesized and applied in electrochemical synthesis of ammonia under ambient conditions. A high NH_4^+ Faradaic efficiency of 11.1% achieved by our Au_1 catalyst surpasses most of reported catalysts under comparable conditions. Benefiting from efficient atom utilization, an NH_4^+ yield rate of 1,305 lg h^(-1) mg_(Au)^(-1) has been reached, which is roughly 22.5 times as high as that by supported Au nanoparticles. We also demonstrate that by employing our Au_1 catalyst, NH4+can be electrochemically produced directly from N_2 and H_2 with an energy utilization rate of 4.02 mmol kJ^(-1). 5Investigation of quality of life in patients with lung cancer by the EORTC QLQ-C30(V3.0) Chinese version显示文摘Objective To explore quality of life(QOL) and its influencing factors in patients with lung cancer.Methods A QOL questionnaire(European Organization for Research and Treatment of Cancer [EORTC] Quality of Life Questionnaire-Core 30 [QLQ-C30] Chinese version) was used with 138 patients with lung cancer participating in the study. A statistical description of the general characteristics of the patients with lung cancer was performed. The patients' QLQ-C30 scores were compared with the reference value for each dimension. To analyze the influence of QOL in different genders, age groups, and cancer stages, ttests and rank sum tests were used to compare the differences in QOL using a 5% significance level. Results The QLQ-C30 function scores in PF(physical functioning), EF(emotional functioning), SF(social functioning), and GH(global functioning), and symptom scales in NV(nausea and vomiting), DY(dyspnea), SL(insomnia), and FI(financial difficulties) were significantly different(P < 0.05) in comparison to the reference values. Female patients were worse than males(P < 0.05) in EF, NV, and DI(diarrhea). The later the stage of lung cancer, the worse the quality of life became; the functional scales in RF(role functioning), EF, CF, SF, and GH, and symptom scales in PA(pain), AP(appetite), and SL differences were statistically significant(P < 0.05). 6Experimental investigation of a 500 W traveling-wave thermoacoustic electricity generator显示文摘In this paper, an experimental investigation of a traveling-wave thermoacoustic electricity generator, which consists of a traveling-wave thermoacoustic heat engine and a linear alternator driven by that engine, is presented. Using the results of previous theoretical and experimental research, we designed and fabricated a traveling-wave thermoacoustic heat engine and a linear alternator. 7Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 8Perfluorooctane sulfonate(PFOS) and related fluorochemicals in chicken egg in China显示文摘The ubiquitous occurrence of perfluorinated compounds(PFCs) in environmental samples has drawn much attention.Recent human exposure studies found relatively high perfluorooctane sulfonate(PFOS) concentrations in blood samples from several cities in China when compared with other countries.The objectives of the present study were:(1) to measure PFC concentrations and compositions in chicken egg samples from local markets in China;and(2) to conduct a preliminary human health risk assessment of egg consumption.Eight pooled egg samples from eight locations were analyzed for 11 PFCs.The results showed that close to 100% of the PFOS in the egg was distributed in egg yolk and PFOS was not detected in egg white(<0.08 ng/g wet weight,w/w).Of the perfluoroalkylsulfonates,only PFOS was detected in all egg samples,while of the perfluoroalkylcarboxylates,perfluoroundecanoic acid(PFUnDA) was detected in all samples,followed by perfluorooctanoate(PFOA)(75% occurrence) and perfluorodecanoic acid(PFDA)(50% occurrence).PFOS concentrations in egg ranged from 45.0 to 86.9 ng/g w/w.The results suggested that current concentrations of PFOS in domestic chicken eggs are unlikely to cause immediate harm to Chinese populations.WANG Yuan YEUNG Leo Wai Yin YAMASHITA Nobuyoshi TANIYASU Sachi SO Man Ka Margaret B. 11Dental implant treatment for renal failure patients on dialysis:a clinical guideline显示文摘Chronic kidney disease(CKD) is a worldwide public health problem that is growing in prevalence and is associated with severe complications. During the progression of the disease, a majority of CKD patients suffer oral complications. Dental implants are currently the most reliable and successful treatment for missing teeth. However, due to complications of CKD such as infections,bone lesions, bleeding risks, and altered drug metabolism, dental implant treatment for renal failure patients on dialysis is more challenging. In this review, we have summarized the characteristics of CKD and previous publications regarding dental treatments for renal failure patients. 12Metformin promotes microglial cells to facilitate myelin debris clearance and accelerate nerve repairment after spinal cord injury显示文摘Spinal cord injury(SCI)is one kind of severe trauma for central nervous system.Myelin debris clearance and axon regeneration are essential for nerve regeneration after SCI.Metformin,a glucose-lowering drug,has been demonstrated to promote the locomotor functional recovery after SCI.In this study,we investigated the role and molecular mechanism of metformin on myelin preservation in a rat SCI model.SCI was induced in rats by compression at T9 level using a vascular clip.We showed that administration of metformin(50 mg·kg^(-1)·d^(-1),ip)for 28 days significantly improved locomotor function in SCI rats.Metformin also ameliorated SCI-induced neuronal apoptosis and promoted axon regeneration in the spinal cord.Using co-immunofluorescence of IBa-1 and MBP,and luxol fasting blue(LFB)staining,we demonstrated that metformin promoted the transformation of M1 to M2 phenotype polarization of microglial cells,then greatly facilitated myelin debris clearance and protected the myelin in SCI rats.Furthermore,metformin ameliorated SCI-induced blockade of autophagic flux in the spinal cord,and enhanced the fusion of autophagosome and lysosome by inhibiting the AMPK-mTOR signaling pathway.Moreover,metformin significantly attenuated inflammatory responses in the spinal cord.In LPS-treated BV2 cells,pretreatment with metformin(2 mM)significantly enhanced autophagy level,suppressed inflammation and cell apoptosis.The protective effects were blocked in the presence of an autophagy inhibitor 3-methyladenine(3-MA,5 mM),suggesting that the effect of metformin on autophagy in microglial cells is essential for the myelin preservation during nerve recovery.This study reveals a novel therapeutic effect of metformin in SCI recovery by regulating the activation of microglial cells and enhancing its autophagy level.Yan-qing Wu Jun Xiong Zi-li He Yuan Yuan Bei-ni Wang Jing-yu Xu Man Wu Su-su Zhang Shu-fang Cai Jia-xin Zhao Ke Xu Hong-yu Zhang Jian Xiao 2022Acta Pharmacologica Sinica2022,43,6:8
13MicroRNA396-mediated alteration in plant development and salinity stress response in creeping bentgrass显示文摘The conserved microRNA396(miR396)is involved in plant growth,development,and abiotic stress response in multiple plant species through regulating its targets,Growth Regulating Factor(GRF)transcription factor genes.However,the role of miR396 has not yet been characterized in perennial monocot species.In addition,the molecular mechanism of miR396-mediated abiotic stress response remains unclear.To elucidate the role of miR396 in perennial monocot species,we generated transgenic creeping bentgrass(Agrostis stolonifera)overexpressing Osa-miR396c,a rice miRNA396 gene.Transgenic plants exhibited altered development,including less shoot and root biomass,shorter internodes,smaller leaf area,fewer leaf veins,and epidermis cells per unit area than those of WT controls.In addition,transgenics showed enhanced salt tolerance associated with improved water retention,increased chlorophyll content,cell membrane integrity,and Na^(+)exclusion during high salinity exposure.Four potential targets of miR396 were identified in creeping bentgrass and up-regulated in response to salt stress.RNA-seq analysis indicates that miR396-mediated salt stress tolerance requires the coordination of stress-related functional proteins(antioxidant enzymes and Na+/H+antiporter)and regulatory proteins(transcription factors and protein kinases).This study establishes a miR396-associated molecular pathway to connect the upstream regulatory and downstream functional elements,and provides insight into the miRNA-mediated regulatory networks.Shuangrong Yuan Junming Zhao Zhigang Li Qian Hu Ning Yuan Man Zhou Xiaoxia Xia Rooksie Noorai Christopher Saski Shigui Li Hong Luo 2019Horticulture Research2019,6,1:8

15Highly efficient,genotype-independent transformation and gene editing in watermelon(Citrullus lanatus)using a chimeric ClGRF4-GIF1 gene显示文摘Efficient genetic transformation has the potential to advance research and breeding in watermelon(Citrullus lanatus),but regeneration from tissue culture remains challenging.Previous work showed that expressing a fusion of two interacting transcription factors,GROWTH-REGULATING FACTOR4(GRF4)and GRF-INTERACTING FACTOR1(GIF1),improved regeneration in wheat(Triticum aestivum).By overexpressing a chimeric fusion of Cl GRF4 and Cl GIF1,we achieved highly efficient transformation in watermelon.Mutating the mi396 micro RNA target site in Cl GRF further boosted the transformation efficiency up to 67.27%in a genotype-independent manner.Cl GRF4-GIF1 can also be combined with clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)genome editing tools to achieve highly efficient gene editing in watermelon,which we used to successfully create diploid seedless watermelon.This research thus puts forward a powerful transformation tool for future watermelon research and breeding.Qin Feng Ling Xiao Yizhen He Man Liu Jiafa Wang Shujuan Tian Xian Zhang Li Yuan 2021Journal of Integrative Plant Biology2021,63,12:8
16Aspirin inhibits the proliferation of hepatoma cells through controlling GLUT 1-mediated glucose metabolism显示文摘Aspirin can efficiently inhibit liver cancer growth,but the mechanism is poorly,understood.In this study,we-report that aspirin modulates glucose uptake through downregulating glucose transporter 1 (GLUT1),leading to the inhibition of hepatoma cell proliferation.Our data showed that aspirin significantly decreased the levels of reactive oxygen species (ROS)and glucose consumption in hepatoma cells.Interestingly,we identified that GLUT1 and HIF1α could be decreased by aspirin.Mechanically,we demonstrated that the -1008/-780 region was the regulatory element of transcriptional factor NF-κB in GLUT1 promoter by luciferase report gene assays.PDTC,an inhibitor of NF-KB,could suppress the expression of GLUT1 in HepG2 and H7402 cells,: followed by affecting the levels of ROS and glucose consumption.CoCl2-activated HIF1α expression could slightly rescue the GLUT1 expression inhibited by aspirin or PDTC,suggesting that aspirin depressed GLUT1 through targeting NF-κB or NF-κB/HIFla signaling.Moreover,we found that GLUT1 was highly expressed in clinical HCC tissues relating to their paired adjacent normal tissues.Importantly,we observed that high level of GLUT1 was significantly correlated with the poor relapse-free survival of HCC patients by analysis of public data.Functionally,overexpression of GLUT1 blocked the PDTC-induced or aspirin-induced inhibition of glucose metabolism in HepG2 cells.Conversely,aspirin failed to work when GLUT1 was stably knocked down in the cells. 17SLOW WALKER3,Encoding a Putative DEAD-box RNA Helicase,is Essential for Female Gametogenesis in Arabidopsis显示文摘RNA helicases are adenosine tri-phosphatases that unwind the secondary structures of RNAs and are required in almost any aspect of RNA metabolism.They are highly conserved from prokaryotic to eukaryotic organisms.However,their precise roles in plant physiology and development remain to be clarified.Here we report that the mutation in the gene SLOW WALKER3(SWA3) results in the slow and retarded progression of mitosis during megagametogenesis in Arabidopsis.SWA3 is a putative RNA helicase of the DEAD-box subfamily.Mutant megagametophyte development is arrested at fouror eight-nucleate stages,furthermore,one of the synergids in about half of the mutant embryo sacs displays abnormal polarity,with its nucleus locating at the chalazal end,instead of the micropylar end in the wild-type.Transmission of the mutation through female gametophytes is severely reduced in swa3.However,a small portion of mutant embryo sacs are able to develop into mature and functional female gametophytes when pollination was postponed.The SWA3 in Arabidopsis is a homolog of Dbp8 in yeast.Dbp8 interacts with Efs2 and is essential for biogenesis of 18S rRNA in yeast.Our data suggest that SWA3 may form a complex with AtEfs2 and take roles in ribosomal biogenesis as RNA helicase during megagametogenesis in Arabidopsis.Man Liu Dong-Qiao Shi Li Yuan Jie Liu Wei-Cai Yang 2010Journal of Integrative Plant Biology2010,52,9:6
18IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE.Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu 2021Cellular & Molecular Immunology2021,18,7:6
19Electrolytic groundwater circulation well for trichloroethylene degradation in a simulated aquifer显示文摘An in situ groundwater remediation process, termed EGCW, was developed in this study by integrating in-well groundwater electrolysis into groundwater circulation well. Groundwater circulation carries electrolytically generated O2 and H2 into the impacted aquifer for in situ biodegradation of contaminants. In a two-dimensional tank filled with field sandy sediments,simulated trichloroethylene(TCE)-contaminated groundwater was circulated between an injection well with electrodes inside and a pumping well. Results from a 50-day EGCW experiment show that in-well electrolysis oxygenated most region near the injection well, and 10 mg/L TCE was mainly biodegraded aerobically to about 2.7 mg/L(73% removal) by the indigenous microbes. Aerobic TCE degradation was enhanced by the pulsed addition of acetate. Together with the proofs of stable carbon isotope fractionation(enrichment factor:-0.57‰–-1.53‰) and microbial community variation after EGCW treatment, aerobic cometabolism was proposed to be the most likely mechanism for TCE degradation. It is interesting to find that the intrinsic organic carbon in aquifer matrix could fuel the aerobic TCE degradation, particularly at low TCE concentrations. 20GAMETOPHYTIC FACTOR 1,Involved in Pre-mRNA Splicing,Is Essential for Megagametogenesis and Embryogenesis in Arabidopsis显示文摘RNA biogenesis is essential and vital for accurate expression of genes. It is obvious that cells cannot continue normal metabolism when RNA splicing is interfered with. sgt13018 is such a mutant,with partial loss of function of GAMETOPHYTIC FACTOR 1(GFA1) ;a gene likely involved in RNA biogenesis in Arabidopsis. The mutant is featured in the phenotype of diminished female gametophyte development at stage FG5 and is associated with the arrest of early embryo development in Arabidopsis. Bioinformatics data showed that homologs of gene GFA1 in yeast and human encode putative U5 snRNP-specific proteins required for pre-mRNA splicing. Furthermore,the result of yeast two-hybrid assay indicated that GFA1 physically interacted with AtBrr2 and AtPrp8,the putative U5 snRNP components,of Arabidopsis.
