Non-coding RNAs and other determinants of neuroinflammation and endothelial dysfunction:regulation of gene expression in the acute phase of ischemic stroke and possible therapeutic applications显示文摘Ischemic stroke occurs under a variety of clinical conditions and has different pathogeneses, resulting in necrosis of brain parenchyma.Stroke pathogenesis is characterized by neuroinflammation and endothelial dysfunction. Some of the main processes triggered in the early stages of ischemic damage are the rapid activation of resident inflammatory cells(microglia, astrocytes and endothelial cells),inflammatory cytokines, and translocation of intercellular nuclear factors.Inflammation in stroke includes all the processes mentioned above, and it consists of either protective or detrimental effects concerning the“polarization”of these processes. This polarization comes out from the interaction of all the molecular pathways that regulate genome expression:the epigenetic factors. In recent years, new regulation mechanisms have been cleared, and these include non-coding RNAs,adenosine receptors, and the activity of mesenchymal stem/stromal cells and microglia. We reviewed how long non-coding RNA and microRNA have emerged as an essential mediator of some neurological diseases. We also clarified that their roles in cerebral ischemic injury may provide novel targets for the treatment of ischemic stroke. To date,we do not have adequate tools to control pathophysiological processes associated with stroke. Our goal is to review the role of non-coding RNAs and innate immune cells(such as microglia and mesenchymal stem/stromal cells)and the possible therapeutic effects of their modulation in patients with acute ischemic stroke.A better understanding of the mechanisms that influence the“polarization”of the inflammatory response after the acute event seems to be the way to change the natural history of the disease. | Mario Daidone Marco Cataldi Antonio Pinto Antonino Tuttolomondo | 2021 | Neural Regeneration Research2021,16,11: | 9
Acute appendicitis: What is the gold standard of treatment?显示文摘McBurney’s procedure represented the gold-standard for acute appendicitis until 1981,but nowadays the number of laparoscopic appendectomies has progressively increased since it has been demonstrated to be a safe procedure, with excellent cosmetic results and it also allows a shorter hospitalization,a quicker and less painful postoperative recovery. The aim of this editorial was to perform a review of the literature in order to address controversial issues in the treatment of acute appendicitis. | Cesare Ruffolo Alain Fiorot Giulia Pagura Michele Antoniutti Marco Massani Ezio Caratozzolo Luca Bonariol Francesco Calia di Pinto Nicolò Bassi | 2013 | World Journal of Gastroenterology2013,19,47: | 8
Impact of age-related comorbidity on results of colorectal cancer surgery显示文摘AIM: To analyze the correlation between preexisting comorbidity and other clinicopathological features, short-term surgical outcome and long-term survival in elderly patients with colorectal cancer (CRC). METHODS: According to age, 403 patients operated on for CRC in our department were divided into group A (< 70 years old) and group B (≥ 70 years old) and analyzed statistically. RESULTS: Rectal localization prevailed in group A (31.6% vs 19.7%, P = 0.027), whereas the percentage of R0 resections was 77% in the two groups. Comorbidity rate was 46.2% and 69.1% for group A and B, respectively (P < 0.001), with a huge difference as regards cardiovascular diseases. Overall, postoperative morbidity was 16.9% and 20.8% in group A and B, respectively (P = 0.367), whereas mortality was limited to group B (4.5%, P = 0.001). In both groups, patients who suffered from postoperative complications had a higher overall comorbidity rate, with preexisting cardiovascular diseases prevailing in group B (P = 0.003). Overall 5-year survival rate was significantly betterfor group A (75.2% vs 55%, P = 0.006), whereas no signif icant difference was observed considering disease-specif ic survival (76.3% vs 76.9%, P = 0.674). CONCLUSION: In spite of an increase in postoperative mortality and a lower overall long-term survival for patients aged ≥ 70 years old, it should be considered that, even in the elderly group, a signif icant number of patients is alive 5 years after CRC resection. | Corrado Pedrazzani Guido Cerullo Giovanni De Marco Daniele Marrelli Alessandro Neri Alfonso De Stefano Enrico Pinto Franco Roviello | 2009 | World Journal of Gastroenterology2009,15,45: | 5
High prevalence of vitamin A deficiency in Crohn's disease patients according to serum retinol levels and the relative dose-response test显示文摘AIM:To assess the vitamin A status of patients with Crohn's disease(CD) by evaluating serum retinol levels and the relative dose response(RDR) test(liver retinol stores).METHODS:Vitamin A nutritional status was measured by serum retinol obtained by high performance liquid chromatography and the RDR test for evaluation of the hepatic stores.Body composition was performed by densitometry by dual-energy X-ray absorptiometry.Vitamin A dietary intake was assessed from a semiquantitative food frequency questionnaire.RESULTS:This study included 38 CD patients and 33 controls.Low serum retinol concentrations were detected in 29% of CD patients vs 15% in controls(P < 0.005).The RDR test was positive in 37% of CD patients vs 12% in controls, which indicated inadequate hepatic vitamin A stores(P < 0.005).Individuals with hypovitaminosis A had lower BMI and body fat compared with those without this deficiency. There was no association between vitamin A deficiency and its dietary intake, ileal location, presence of disease activity and prior bowel resections.CONCLUSION:Patients with CD have higher prevalence of vitamin A deficiency,as assessed by two independent methods. | Márcia Soares-Mota Tianny A Silva Luanda M Gomes Marco AS Pinto Laura MC Mendon?a Maria Lúcia F Farias Tiago Nunes Andrea Ramalho Cyrla Zaltman | 2015 | World Journal of Gastroenterology2015,21,5: | 4
Role of autophagy in cholangiocarcinoma:Pathophysiology and implications for therapy显示文摘Cholangiocarcinoma(CCA)is a malignant tumour of the biliary system that originates from the neoplastic transformation of cholangiocytes.CCA is characterized by late diagnosis and poor outcome, with surgery considered as the last option for management.Autophagy is a physiological lysosomal degradation process, essential for cellular homeostasis and ubiquitous in all eukaryotic cells.Several studies have reported a potential involvement of autophagy in cancer, but it remains unclear whether activation of this process represents a survival mechanism of cancer cells. In the present review,we examine the autophagic process and summarize the current knowledge about the involvement of autophagy in the progression of cancer. The link between autophagy and chemoresistance and the use of autophagic markers in diagnosis are also considered in detail.Preliminary evidence shows that the combination of autophagy modulators(activators or inhibitors)with conventional chemotherapeutic agents offers a possible treatment option against signalling pathways that are hyperactivated or altered in CCA.In vitro evidence suggests that combination of chemotherapy agents, such as cisplatin, under activation or inhibition of autophagic processes,in two different CCA cell lines, may improve chemosensitivity and reduce cell survival, respectively.A deeper understanding of these pathways,in both cancer and non-cancer cells, could unveil possible therapeutic targets to treat CCA patients. | Elisabetta Ninfole Claudio Pinto Antonio Benedetti Marco Marzioni Luca Maroni | 2021 | World Journal of Clinical Cases2021,9,22: | 3
An analysis of vehicular exhaust derived nanoparticles and historical Belgium fortress building interfaces显示文摘Air pollution monitoring is one of the most important features in contamination risk management. This is because many of the compounds contained within air pollution present a serious risk both for the preservation of open air cultural heritage and for human health.New particle formation is a major contributor to urban pollution, but how it occurs in cities is often puzzling. As more and more people enjoy an increased quality of life through outdoor activity, managing outdoor air quality is vital. This study presents the application of a low-cost system for monitoring the current level of road traffic passengers’exposure to particulate air contamination. The global rise in tourism also leads to apprehension about its probable destructive influence on various aspects of global preservation. One of the major risks encountered by tourists, stemming from modes of transport, are nanoparticles(NPs)(<100 nm)and ultra-fine particles(UFPs)(100-1000 nm)consisting of potentially hazardous elements(PHEs).This study examines Steen Castle,a medieval fortress located in Antwerp,Belgium.Significant NPs with PHEs,were found in the air sampled in this area. The self-made passive sampler(LSPS)described in this study, consisting of retainers specially designed for advanced microscopic analysis,is used for the first time as a simple way to characterize the surrounding atmospheric contamination caused by NPs and UFPs,without the need of other commonly employed more expensive particulate focused active samplers such as cascade impactors. This study aims to assess the result of the utilization of a low-cost,LSPS,to determine outdoor NPs and UFPs in a Belgian urban(Steen Castle)and rural area(Fort van Schoten). This work is the first to detail the usefulness of LSPS for the evaluation of Belgium’s outdoor air for NPs and UFPs,which contain PHEs. | Luis F.O.Silva Diana Pinto Alcindo Neckel Marcos L.S.Oliveira | 2020 | Geoscience Frontiers2020,11,6: | 2
Mathematical simulation of radial heat transfer in packed beds by pseudohomogeneous modeling显示文摘Uniform flow regime and constant effective thermal conductivity inside packed beds are commonly accepted in the evaluation of the fluid dynamics and heat transfer in such systems. However, several authors have confirmed the presence of an oscillatory velocity profile caused by the effective contribution of porosity profile in the fluid dynamic behavior of packed beds, which directly influences the heat transfer inside the beds. This paper describes the application of a pseudo-homogeneous mathematical model for describing heat transfer in packed beds with oscillatory profiles of velocity and porosity, using a radius-dependent model for effective thermal conductivity k r.Several temperature profiles were obtained in a packed bed system with thermal source located on the wall. The simulated temperature and effective thermal conductivity obtained from simulations were compared with experimental data and calculation from a model based on uniform k r fitting. The results indicate that the proposed mathematical modeling was capable of better representing the heat transfer in the packed bed. | Rodrigo Béttega Marcos Flávio Pinto Moreira Ronaldo Guimaraes Corrêa José Teixeira Freire | 2011 | Particuology2011,9,2: | 2
Impact of jejunostomy during esophagectomy for cancer on health related quality of life显示文摘Background: The aim of this study was to evaluate the impact of jejunostomy during esophagectomy for cancer on postoperative health-related quality of life(HRQL).Methods: We evaluate all consecutive patients who underwent esophagectomy for cancer at the surgical oncology unit of the Veneto Institute of Oncology(IOV-IRCCS) between January 2008 and March 2014. The primary outcome was HRQL, which was assessed using nine scales of EORTC C30 and OES18 questionnaires. General linear models were estimated to evaluate mean score difference(MD) of each selected scale in patients with and without jejunostomy, adjusting for clinically relevant confounders. The secondary outcomes were morbidity, hospital stay, postoperative weight loss and postoperative albumin impairment. Results: Jejunostomy was performed in 40 on 109 patients(41.3%) who participated in quality of life investigation. A clinically and statistically significantly worse eating at admission(P=0.009) became not clinically significant at 3 months after surgery(MD =9.1). Jejunostomy was associated to clinically and statistically significantly poorer emotional function(EF) at 3 months after surgery(MD =-15.6; P=0.04). Hospital stay was longer in jejunostomy group(median, 20 vs. 17 days, P=0.02).Conclusions: In our series patients who had a jejunostomy during esophagectomy had been selected for their risk for postoperative complication. However, their postoperative outcome was actually similar compared to those without jejunostomy. Nevertheless, jejunostomy was associated to clinically and statistically significantly poorer EF at 3 months after surgery. Therefore, patient candidate to esophagectomy and feeding jejunostomy should receive additional psychological support. | Marco Scarpa Francesco Cavallin Giulia Noaro Eleonora Pinto Rita Alfieri Matteo Cagol Carlo Castoro | 2014 | Chinese Journal of Cancer Research2014,26,6: | 2
Tracheobronchial Polyflex stents for the management of benign refractory hypopharyngeal strictures显示文摘AIM:To describe a modified technique for placement of a tracheobronchial self-expanding plastic stent(SEPS) in patients with benign refractory hypopharyngeal strictures in order to improve dysphagia and allow stricture remodeling.METHODS:A case series of four consecutive patients with complex hypopharyngeal strictures after combinedtherapy for laryngeal cancer, previously submitted to multiple sessions of dilation without lasting improvement,is presented. All patients underwent placement of a small diameter and unflared tracheobronchial SEPS.Main outcome measurements were improvement of dysphagia and avoiding of repeated dilation.RESULTS:The modified introducer system allowed an easy and technically successful deployment of the tracheobronchial Polyflex stent through the stricture. All four patients developed complications related to stent placement. Two patients had stent migration(one proximal and one distal),two patients developed phanryngocutaneous fistulas and all patients with stents in situ for more than 8 wk had hyperplastic tissue growth at the upper end of the stent.Stricture recurrence was observed at 4 wk follow-up after stent removal in all patients CONCLUSION:Although technically feasible, placement of a tracheobronchial SEPS is associated with a high risk of complications.Small diameter stents must be kept in place for longer than 3 mo to allow adequate time for stricture remodeling. | Rui Almeida Silva Nuno Mesquita Pedro Pimentel Nunes Elisabete Cardoso Ricardo Marcos Pinto Luís Moreira Dias | 2012 | World Journal of Gastroenterology2012,18,6: | 2
