1BCRP基因在非小细胞肺癌组织和非癌肺组织中的表达及意义显示文摘目的:进一步研究从人类乳腺癌细胞株 (MCF-7/AdrVp)中克隆出的、对蒽环类抗癌药耐药的 BCRP基因在非癌肺组织及非小细胞肺癌组织中的表达。方法:从术后即刻获得的正常肺组织和有活力的非小细胞肺癌组织新鲜标本中及时提取细胞总 RNA,通过 RT-PCR及 PCR法获得并扩增 BCRP基因的 cDNA产物,再经凝胶电泳分离 BCRP基因 cDNA带,然后再通过转膜技术把这些 BCRP基因 cDNA产物转移到杂交膜上,用 Southern blot杂交技术检测 BCRP基因的表达程度。结果:细胞总 RNA分别从 8个只有肺癌组织标本和 12对同时获取的肺癌组织及非癌肺组织标本中成功提取,然后用变性凝胶电泳鉴定提取 RNA的质量。其中 4例标本因术中缺血时间太长,或有坏死炎症或术前放化疗等原因,所提 RNA有降解而放弃进一步的检测。通过 RT-PCR及 PCR方法从 16例可用标本中获得并扩增 BCRP基因的 cDNA产物,再通过转膜及 Southern blot杂交技术,最终发现 BCRP基因在正常肺组织中均有不同程度的表达( 10/10),而在非小细胞肺癌组织标本中只有一半病人的标本有不同程度的表达 (8/16)。结论: BCRP基因可能是一个在非癌肺组织中常规表达的基因,而在部分病人的肺癌组织中可能此基因有缺失或被抑制。因此。毛友生 Austin Doyle Weidong Yang Yuetong Wei Mark J.Krasna Douglas D.Ross 2001癌症2001,20,3:9
2Direct laser writing of volumetric gradient index lenses and waveguides显示文摘Direct laser writing(DLW)has been shown to render 3D polymeric optical components,including lenses,beam expanders,and mirrors,with submicrometer precision.However,these printed structures are limited to the refractive index and dispersive properties of the photopolymer.Here,we present the subsurface controllable refractive index via beam exposure(SCRIBE)method,a lithographic approach that enables the tuning of the refractive index over a range of greater than 0.3 by performing DLW inside photoresist-filled nanoporous silicon and silica scaffolds.Adjusting the laser exposure during printing enables 3D submicron control of the polymer infilling and thus the refractive index and chromatic dispersion.Combining SCRIBE’s unprecedented index range and 3D writing accuracy has realized the world’s smallest(15μm diameter)spherical Luneburg lens operating at visible wavelengths.SCRIBE’s ability to tune the chromatic dispersion alongside the refractive index was leveraged to render achromatic doublets in a single printing step,eliminating the need for multiple photoresins and writing sequences.SCRIBE also has the potential to form multicomponent optics by cascading optical elements within a scaffold.As a demonstration,stacked focusing structures that generate photonic nanojets were fabricated inside porous silicon.Finally,an all-pass ring resonator was coupled to a subsurface 3D waveguide.The measured quality factor of 4600 at 1550 nm suggests the possibility of compact photonic systems with optical interconnects that traverse multiple planes.SCRIBE is uniquely suited for constructing such photonic integrated circuits due to its ability to integrate multiple optical components,including lenses and waveguides,without additional printed supports.Christian R.Ocier Corey A.Richards Daniel A.Bacon-Brown Qing Ding Raman Kumar Tanner J.Garcia Jorik van de Groep Jung-Hwan Song Austin J.Cyphersmith Andrew Rhode Andrea N.Perry Alexander J.Littlefield Jinlong Zhu Dajie Xie Haibo Gao Jonah F.Messinger Mark L.Brongersma Kimani C.Toussaint Jr Lynford L.Goddard Paul V.Braun 2020Light(Science & Applications)2020,9,1:6
3Baseline effects of lysophosphatidylcholine and nerve growth factor in a rat model of sciatic nerve regeneration after crush injury显示文摘Schwann cells play a major role in helping heal injured nerves. They help clear debris, produce neurotrophins, upregulate neurotrophin receptors, and form bands of Büngner to guide the healing nerve. But nerves do not always produce enough neurotrophins and neurotrophin receptors to repair themselves. Nerve growth factor(NGF) is an important neurotrophin for promoting nerve healing and lysophosphatidylcholine(LPC) has been shown to stimulate NGF receptors(NGFR). This study tested the administration of a single intraneural injection of LPC(1 mg/mL for single LPC injection and 10 mg/mL for multiple LPC injections) at day 0 and one(day 7), two(days 5 and 7), or three(days 5, 7, and 9) injections of NGF(160 ng/mL for single injections and 80 ng/mL for multiple injections) to determine baseline effects on crush ed sciatic nerves in rats. The rats were randomly divided into four groups: control, crush, crush-NGF, and crush-LPC-NGF. The healing of the nerves was measured weekly by monitoring gait; electrophysiological parameters: compound muscle action potential(CMAP) amplitudes; and morphological parameters: total fascicle areas, myelinated fiber counts, fiber densities, fiber packing, and mean g-ratio values at weeks 3 and 6. The crush, crush-NGF, and crush-LPC-NGF groups statistically differed from the control group for all six weeks for the electrophysiological parameters but only differed from the control group at week 3 for the morphological parameters. The crush, crush-NGF, and crush-LPC-NGF groups did not differ from each other over the course of the study. Single injections of LPC and NGF one week apart or multiple treatments of NGF at 5, 7 and 9 days post-injury did not alter the healing rate of the sciatic nerves during weeks 1-6 of the study. 9Identification of Global DNA Methylation Signatures in Glioblastoma-Derived Cancer Stem Cells显示文摘Glioblastoma(GBM)is the most common and most aggressive primary brain tumor in adults.The existence of a small population of stem-like tumor cells that efficiently propagate tumors and resist cytotoxic therapy is one proposed mechanism leading to the resilient behavior of tumor cells and poor prognosis.In this study,we performed an in-depth analysis of the DNA methylation landscape in GBMderived cancer stem cells(GSCs).Parallel comparisons of primary tumors and GSC lines derived from these tumors with normal controls(a neural stem cell(NSC)line and normal brain tissue)identified groups of hyper- and hypomethylated genes that display a trend of either increasing or decreasing methylation levels in the order of controls,primary GBMs,and their counterpart GSC lines,respectively.Interestingly,concurrent promoter hypermethylation and gene body hypomethylation were observed in a subset of genes including MGMT,AJAP1 and PTPRN2.These unique DNA methylation signatures were also found in primary GBM-derived xenograft tumors indicating that they are not tissue culture-related epigenetic changes.Integration of GSC-specific epigenetic signatures with gene expression analysis further identified candidate tumor suppressor genes that are frequently down-regulated in GBMs such as SPINT2,NEFM and PENK.Forced re-expression of SPINT2 reduced glioma cell proliferative capacity,anchorage independent growth,cell motility,and tumor sphere formation in vitro.The results from this study demonstrate that GSCs possess unique epigenetic signatures that may play important roles in the pathogenesis of GBM.Eun-Joon Lee Prakash Rath Jimei Liu Dungsung Ryu Lirong Pei Satish K.Noonepalle Austin Y.Shull Qi Feng N.Scott Litofsky Douglas C.Miller Douglas C.Anthony Mark D.Kirk John Laterra Libin Deng Hong-Bo Xin Xinguo Wang Jeong-Hyeon Choi Huidong Shi 2015Journal of Genetics and Genomics2015,42,7:1
