| 1 | Indole alkaloid sulfonic acids from an aqueous extract of Isatis indigotica roots and their antiviral activity显示文摘Six new indole alkaloid sulfonic acids(1–6), together with two analogues(7 and 8) that were previously reported as synthetic products, were isolated from an aqueous extract of the Isatis indigotica root. Their structures including the absolute configurations were determined by spectroscopic data analysis, combined with enzyme hydrolysis and comparison of experimental circular dichroism and calculated electronic circular dichroism spectra. | 4 | Identifying changes in the wheat kernel proteome under heat stress using iTRAQ显示文摘Wheat(Triticum aestivum L.) is one of the three major global food crops. Hightemperature stress can affect its yield and quality. Studies of the effect of hightemperature stress on wheat kernel development are important because they can reveal the stability of wheat quality and lead to the genetic improvement of wheat quality traits. In this study, the isobaric tags for relative and absolute quantitation(iTRAQ)method was adopted to analyze changes in the protein expression profile of wheat cultivars under high temperature stress. The protein content of wheat grain increased under heat stress, while the SDS-sedimentation value and starch content decreased.Grain filling was deficient under high temperature stress, which reduced thousandkernel weight but did not affect wheat kernel length. The 207 differentially expressed proteins identified in Gaocheng 8901 under heat stress were associated with energy metabolism, growth and development, and stress response. Gene Ontology enrichment analysis showed that the annotated proteins that were differentially expressed in Gaocheng 8901 under heat stress were involved mainly in stimulus response, abiotic stress response, stress response, and plasma membrane. A set of 78 differentially expressed proteins were assigned to 83 KEGG signaling/metabolic pathways. KEGG pathway enrichment analysis showed that this set of proteins was significantly enriched in members of 51 pathways, and the proteins participated mainly in protein synthesis in the endoplasmic reticulum, starch and sucrose metabolism, and reaction on ribosomes. Five differentially expressed proteins were involved in protein–protein interaction networks that may greatly influence the yield and quality of wheat grain. | 5 | Development of high mechanical properties and moderate thermal conductivity cast Mg alloy with multiple RE via heat treatment显示文摘A new cast Mg–2 Gd–2 Nd–2 Y–1 Ho–1 Er–0.5 Zn–0.4 Zr(wt%) alloy was prepared by direct-chill semicontinuous casting technology. The microstructure, mechanical properties and thermal conductivity of the alloy in as-cast, solid-solution treated and especially peak-aged conditions were investigated. The as-cast alloy mainly consists of β-Mg matrix,(Mg, Zn)_3 RE phase and basal plane stacking faults. After proper solid-solution treatment, the microstructure becomes almost Mg-based single phase solid solution except just very few RE-riched particles. The as-cast and solid-solution treated alloys exhibit moderate tensile properties and thermal conductivity. It is noteworthy that the Mg alloy with 8 wt% multiple RE exhibits remarkable age-hardening response( HV = 35.7), which demonstrates that the multiple RE(RE = Gd, Nd, Y, Ho, Er) alloying instead of single Gd can effectively improve the age-hardening response.The peak-aged alloy has a relatively good combination of high strength/hardness(UTS(ultimate tensile strength) > 300 MPa; TYS(tensile yield strength) > 210 MPa; 115.3 HV), proper ductility(ε≈ 6%) and moderate thermal conductivity(52.5 W/(m K)). The relative mechanisms mainly involving aging precipitation of β¢ and β'' phases were discussed. | 12 | Key immunity characteristics of diverse stages of brucellosis in rural population from Inner Mongolia, China显示文摘Background: Brucellosis poses a serious threat to human and animal health,particularly in developing countries such as China.The Inner Mongolia Autonomous Region is one of the most severely brucellosis-endemic provinces in China.Currently,the host immune responses functioning to control Brucella infection and development remain poorly understood.The aim of this study is to further clarify the key immunity characteristics of diverse stages of brucellosis in Inner Mongolia.Methods: We collected a total of 733 blood samples from acute(n=137),chronic(n=316),inapparent(n=35),recovery(n=99),and healthy(n=146)groups from the rural community of Inner Mongolia between 2014 and 2015.The proportions of CD4^(+),CD8^(+),Th1,Th2,and Th17 T cells in peripheral blood and the expression of TLR2 and TLR4 in lymphocytes,monocytes and granulocytes were examined using flow cytometry analysis.The differences among the five groups were compared using one-way ANOVA and the Kruskal–Wallis method,respectively.Results: Our results revealed that the proportions of CD4^(+) and CD8^(+) T cells were significantly different among the acute,chronic,recovery,and healthy control groups(P<0.05),with lower proportions of CD4^(+) T cells and a higher proportion of CD8^(+) T cells in the acute,chronic,and recovery groups.The proportion of Th1 cells in the acute,chronic,and inapparent groups was higher than that in the healthy and recovery groups;however,there was no significant difference between patients and healthy individuals(P>0.05).The proportion of Th2 lymphocytes was significantly higher in the acute and healthy groups than in the inapparent group(P<0.05).The proportion of Th17 cells in the acute group was significantly higher than that in the healthy control,chronic,and inapparent groups(P<0.05).Finally,the highest expression of TLR4 in lymphocytes,monocytes and granulocytes was observed in the recovery group,and this was followed by the acute,chronic,healthy control,and inapparent groups.There was a significant difference between the recovery group and the other groups,except for the acute group(P<0.05).Moreover,a correlation in TLR4 expression was observed in lymphocytes,monocytes and granulocytes among the five groups(r>0.5),except for the inapparent group between lymphocytes and granulocytes(r=0.34).Conclusions: Two key factors(CD8^(+)T cells and TLR4)in human immune profiles may closely correlate with the progression of brucellosis.The detailed function of TLR4 in the context of a greater number of cell types or tissues in human or animal brucellosis and in larger samples should be further explored in the future. | Yongzhang Zhu Li Shi Yige Zeng Dongri Piao Yingbo Xie Juan Du Meng Gao Wei Gao Junli Tian Jun Yue Min Li XiaoKui Guo Yufeng Yao YaoXia Kang | 2022 | Infectious Diseases of Poverty2022,11,3: | 1 |
