| 2 | Clinical characteristics and effectiveness of antiviral agents in hospitalized children with infectious mononucleosis in China:A multicenter retrospective study显示文摘Importance:The clinical characteristics of infectious mononucleosis(IM)in Chinese children have not been evaluated in multicenter studies,and the effectiveness of antiviral treatment are controversial.Objective:To investigate the clinical characteristics of Chinese children with IM and current status of antiviral therapy for affected patients.Methods:Hospitalized patients with IM were enrolled between 2018 and 2020 in five children’s hospitals in China.The clinical characteristics were compared among four age groups:<3 years,3-<6 years,6-<10 years,and≥10 years.The clinical characteristics of IM and effectiveness of antiviral therapy were compared among patients receiving acyclovir(ACV),ganciclovir(GCV),and no antiviral therapy(i.e.,non-antiviral group).Results:In total,499 patients were analyzed;most patients were 3-<6 years of age.The most common symptoms and signs included fever(100%),lymphadenopathy(98.6%),pharyngitis(86.4%),eyelid edema(76.8%),and snoring(72.9%).There were significant differences in rash,hepatomegaly,and liver dysfunction among the four age groups.Patients aged<3 years had a lower incidence of liver dysfunction and a higher incidence of rash.Among the 499 patients,50.1%were treated with GCV,26.3%were treated with ACV,and 23.6%received no antiviral therapy.Compared with the non-antiviral group,patients in the ACV and GCV groups had longer durations of fever(P<0.001).There were no significant differences in the incidences of complications among the three treatment groups.Interpretation:The incidence of IM in Chinese children peaked at 3-<6 years of age.Clinical features of IM varied according to age.Patients receiving antiviral therapy exhibited more serious clinical manifestations than did patients without antiviral therapy.The effectiveness of antiviral therapy for IM requires further analysis. | Huili Hu Huiling Deng Jing Bi Yi Xu Shuangjie Li Yue Xie Xinrong Sun Dongmeng Wang Xufang Li Wenxian Ouyang Bing Hu Yufeng Zhang He Tang Chunxiao Fang Hui Zhang Lingyun Guo Chen Wang Tianyi Wang Fengxia Yang Tao Jiang Zhengde Xie Gang Liu | 2021 | Pediatric Investigation2021,5,3: | 12 |

| 4 | Regulatory effects of antitumor agent matrine on FOXO and PI3K-AKT pathway in castration-resistant prostate cancer cells显示文摘We previously demonstrated that matrine could inhibit the proliferating, migrating, as well as invading processes of both PC-3 and DU145 cells. However, the underlying molecular mechanisms have not yet been clearly defined. In this study, using various techniques such as high throughput sequencing technology, bioinformatics, quantitative real-time PCR, and immunoblot analysis,we aimed to understand whether matrine serves as a novel regulator of FOXO and PI3K-AKT signaling pathway. DU145 and PC-3 cell lines were cultured for 24 h in vitro. Cells were treated with either matrine or control serum for 48 h, followed by extraction of total RNA. The RNA was sequenced using HiSeq 2500 high-throughput sequencing platform (Illumina). A gene library was established and quality analysis of read data carried out. Integrated database from the website DAVID was used to analyze Gene Ontology (GO), and Kyoto encyclopedia of genes and genomes (KEGG) pathway of differential genes was used for pathway analysis, screening for fold differences of more than two times. The FOXO and PI3K-AKT signaling pathways were screened, and expression levels of mRNA and core protein detected by real-time PCR and immunoblotting, respectively. High throughput sequencing and GO analysis revealed that differentially expressed genes before and after treatment played an important role in cell metabolic process, growth process, anatomical structure formation, cellular component organization, and biological regulation. KEGG signal pathway analysis revealed that FOXO and PI3K-AKT signal pathways had a significant difference between before and after matrine-treated androgen-independent prostate cancer cells PC-3 and DU145. Real-time PCR showed that matrine treatment led to a significant increase in the expression levels of FOXO1A, FOXO3A, FOXO4, and FOXO6 in DU145 and PC-3 cells (P<0.01 or P<0.05), whereas the PI3K expression levels decreased (P<0.01). Similarly, immunoblotting revealed a significant increase (P<0.05) in the expression levels of FOXO1A FOXO3A, FOXO4, and FOXO6 in both PC-3 and DU145 cells, whereas PI3K expression levels decreased (P<0.05). Matrine had a broad regulating effect on the mRNA expression profiles of both PC-3 and DU145 cells. Matrine may inhibit cell proliferation, migration, as well as invasion, and induce apoptosis in both PC-3 and DU145 cells through FOXO and PI3K-AKT signaling pathways. | 8 | Frequency-specific abnormalities in regional homogeneity among children with attention deficit hyperactivity disorder: a resting-state f MRI study显示文摘Although many functional magnetic resonance imaging(f MRI) studies have investigated the neurophysiology of attention deficit hyperactivity disorder(ADHD),the existing studies have not yielded consistent findings.This may be related to the different properties of different frequency bands. To investigate the frequency-specific regional homogeneity(Re Ho) of spontaneous neural activities in ADHD, the current study used resting-state f MRI to explore the Re Ho properties of five frequency bands, slow-5(0.01–0.027 Hz), slow-4(0.027–0.073 Hz),slow-3(0.073–0.198 Hz), slow-2(0.198–0.25 Hz) and the extra-low frequency(0–0.01 Hz), in 30 drug-naive boys with ADHD and 30 healthy controls. Compared with controls, the ADHD group showed decreased Re Ho in the default mode network(DMN) including the medial prefrontal cortex and precuneus, middle frontal gyrus and angular gyrus. ADHD patients also showed increased Re Ho in the posterior cerebellum. Significant interactions between frequency band and group were observed predominantly in the dorsolateral prefrontal and parietal cortices, orbital frontal cortex, supplementary motor area,inferior occipital gyrus, thalamus and anterior cerebellum.In particular, we found that the between-group difference in the extra-low frequency band(0–0.01 Hz) seemed to be greater than that in the other frequency bands for most brain regions. | 9 | Preparation of 5-fluorouracil-loaded chitosan nanoparticles and study of the sustained release in vitro and in vivo显示文摘The sustained-release properties of the biodegradable nano-drug delivery systems were used to improve the residence time of the chemotherapeutic agent in the body. These drug delivery systems were widely used to deliver chemotherapeutic drugs. The 5-fluorouracil loaded chitosan nanoparticles prepared in this paper have the above advantage. Here, we found that when the mass ratio of 5-fluorouracil and chitosan was 1:1, the maximum drug loading of nanoparticles was 20.13 ± 0.007%, the encapsulation efficiency was 44.28 ± 1.69%, the particle size was 283.9 ± 5.25 nm and the zeta potential was 45.3 ± 3.23 mV. The prepared nanoparticles had both burst-release and sustained-release phases in vitro release studies.In addition, the inhibitory effect of the prepared nanoparticles on gastric cancer SGC-7901 cells was similar to that of 5-fluorouracil injection, and the blank vector had no obvious inhibitory effect on SGC-7901 cells. | 10 | Tensile, creep behavior and microstructure evolution of an as-cast Ni-based K417G polycrystalline superalloy显示文摘The Ni-based K417 G superalloy is extensively applied as aeroengine components for its low cost and good mid-temperature(600–900?C) properties. Since used in as-cast state, the comprehensive understanding on its mechanical properties and microstructure evolution is necessary. In the present research,the tensile, creep behavior and microstructure evolution of the as-cast K417 G superalloy under different conditions were investigated. The results exhibit that tensile cracks tend to initiate at MC carbide and γ/γ' eutectic structure and then propagate along grain boundary. As the temperature for tensile tests increases from 21?C to 700?C, the yield strength and ultimate tensile strength of K417 G superalloy decreases slightly, while the elongation to failure decreases greatly because of the intermediate temperature embrittlement. When the temperature rises to 900?C, the yield strength and ultimate tensile strength would decrease significantly. The creep deformation mechanism varies under different testing conditions. At 760?C/645 MPa, the creep cracks initiate at MC carbides and γ/γ' eutectic structures,and propagate transgranularly. While at 900?C/315 MPa and 950?C/235 MPa, the creep cracks initiate at grain boundary and propagate intergranularly. | 11 | Study on the Anti-Liver Fibrosis Effect of Benefit Liver Granule (益肝冲剂) and Its Mechanism in Rats显示文摘Objective:To explore the effect of Benefit Liver Granule (BLG), a traditional Chinese medical preparation, in antagonizing liver fibrosis and its possible mechanism. Methods:Carbon tetrachloride was used to establish the experimental liver fibrosis model of rat. The model rats were randomly divided into 4 groups and BLG, Astragalus (As), Salvia (Sa) and normal saline (for control) respectively were given to them by gastrogavage. Changes of serum alanine transaminase (ALT), aspartate aminotransferase (AST), transforming growth factor α(TGF α), interleukine 6 (IL 6), liver content of hydroxyproline (Hyp), superoxide dismutase (SOD), malonyldialdehyde (MDA) as well as liver pathology after treatment were observed. A normal control group was also established for control.Results: Pathological examination showed that in the saline group, the structure of normal hepatic lobuli was destroyed with swelled liver cells, focal necrosis, extensive fatty degeneration, and focal inflammatory cell infiltration. Small amounts of proliferated fiber tissue were found in the intra lobular area, peri central vein area, portal area and limiting plate area, and formation of pseudolobuli was also seen. In the 3 treated groups, the serum ALT, AST, TGF α and IL 6 as well as liver content of Hyp, and MDA were lower and SOD were higher than those in the control group significantly, P <0.05 or P <0.01.Conclusion: BLG, As and Sa have the action of anti liver fibrosis, while BLG has the best effect. Herein, a series of Co3O4 with differe nt morphologies (ordered two-dimensio nal porous nano sheets, flowerlike and cuboidlike) were successfully prepared through facile hydrothermal and calcinatio n methods. Ordered two-dime nsional Co3O4 nano sheets show the best cycling stability. Detailed experimental results reveal that the superiority of the unique two-dimensional uniform porous structures is vital for Li-O2 batteries cathode catalysts. For further studying origins and evolution of MHC class I region haplotypes in Han and Jinuo populations in Yunnan Province, we investigated frequencies of five POALINs, their associations with HLA-A and -B, the three-loci POALINs haplotype frequencies and HLA/POALIN four-loci haplotype frequencies within the alpha block of MHC class I region. We found that a strong positive association between AluHG and HLA-A*02 is in Jinuo, but not in Yunnan Han. | 20 | Effect of Stacking Fault Energy on the Grain Structure Evolution of FCC Metals During Friction Stir Welding显示文摘The effect of stacking fault energy(SFE) on the grain structure evolution of face-centered cubic metals during friction stir welding was investigated by using pure aluminum,pure copper and Cu-30 Zn alloy as experiment materials.Tool 'stop action' and rapid cooling were employed to 'freeze' the microstructure of the flowing materials around the tool.Marker materials were used to show the streamline of the material flow.The microstructures of the three materials at different welding stages were contrastively studied by the electron backscatter diffraction technique.The results show that at the material flow stage,as the SFE decreases,the grain structure evolution changes from the continuous dynamic recrystallization to discontinuous dynamic recrystallization,and further to the dynamic equilibrium between the annealing twinning due to thermally activated grain boundary migration and the twin destruction during the plastic deformation.Owing to different grain structure evolution mechanisms,the grain structure at the end of the material flow is greatly different.Especially in copper,a lot of dislocations remain,which gives rise to the static recrystallization occurring during the subsequent cooling stage. | Xiaochao Liu Yufeng Sun Tomoya Nagira Kohsaku Ushioda Hidetoshi Fujii | 2020 | Acta Metallurgica Sinica(English Letters)2020,33,7: | 2 |
