| 5 | Mixture Toxicities of Three Pesticides Having Different Time-Toxicity Profiles显示文摘In the environment,chemical pollutants always exist as mixtures.To evaluate mixture toxicity,we should ex-amine the concentration ratios and the concentration levels of mixture components as well as the interactive time between the components and testing organism.Selecting three pesticides,metribuzin(MET),dodine(DOD)and ri-domil(RID),as mixture components,Vibrio qinghaiensis sp.-Q67 as testing organism,and 96-well microplate as exposure experiment carrier,the luminescence inhibition toxicities of the pesticides and their binary mixtures at six exposure time points(0.25,2,4,8,12,and 16 h)were determined.The results showed that the three pesticides have different time-toxicity profiles.The toxicity of MET increases over time,and those of DOD and RID show a slight decrease or increase in the first 4 h,respectively,and then remain unchanged to 16 h.The toxicities of the binary mixtures consisting of the three pesticides depend on not only the mixture composition but also the exposure time.The time-toxicity profiles of all rays in MET-DOD mixture system are U-shaped curves,those in MET-RID system are basically monotonically increased and those in DOD-RID system are basically monotonically decreased.How-ever,our findings indicate that for the test substances,mixture toxicity can be predicted by the extended concentra-tion addition model. | Mo Yu Shushen Liu Mengchao Wang Fu Chen Hanxiao Tang | 2014 | Chinese Journal of Chemistry2014,32,6: | 2 |
| 6 | Cold-induced retrotransposition of fish LINEs显示文摘Classes of retrotransposons constitute a large portion of metazoan genome. There have been cases reported that genomic abundance of retrotransposons is correlated with the severity of low environmental temperatures. However, the molecular mechanisms underlying such correlation are unknown. We show here by cell transfection assays that retrotransposition(RTP) of a long interspersed nuclear element(LINE) from an Antarctic notothenioid fish Dissostichus mawsoni(dmLl) could be activated by low temperature exposure, causing increased dmL1 copies in the host cell genome. The cold-induced dmL1 propagation was demonstrated to be mediated by the mitogen-activated protein kinases(MAPK)/p38 signaling pathway, which is activated by accumulation of reactive oxygen species(ROS) in cold-stressed conditions. Surprisingly, dmL1 transfected cells showed an increase in the number of viable cells after prolonged cold exposures than non-transfected cells. Features of cold inducibility of dmL1 were recapitulated in LINEs of zebrafish origin both in cultured cell lines and tissues, suggesting existence of a common cold-induced LINE amplification in fishes. | 8 | Experimental study on enhancement of the metastatic potential of portal vein tumor thrombus-originated hepatocellular carcinoma cells using portal vein serum显示文摘Objective: Portal vein metastasis of hepatocellular carcinoma(HCC) results in a poor prognosis and seriously affects the survival rate of patients. The mechanism underlying the formation of portal vein tumor thrombus(PVTT) is complex and is not yet fully understood. This study was conducted to investigate the impact of portal vein blood on the proliferation, metastasis, invasion and apoptosis of PVTT cells and to explore its possible mechanisms, which was expected to lay a foundation for ascertaining the mechanism underlying the portal vein metastasis of HCC.Methods: Peripheral blood and portal vein blood were collected from patients with HCC, and the sera from these two sources were used to culture the PVTT-originated HCC cell line CSQT-2. The cells were collected after 24 h, and flow cytometry was performed to detect cell proliferation, cell cycle stages and apoptosis. Transwell migration and invasion assays were applied to detect the metastasis and invasion of the cells in each group. The changes in the expression of MMP-2 and MMP-9 in cells were detected via Western blotting. The contents of IL-12, IFN-γ, IL-1β, IL-2 and TNF-α in the two groups of sera were quantified using corresponding kits. Results: Compared with the group of cells cultured with peripheral serum, the cells cultured with portal vein serum showed significantly lower apoptosis(P<0.01), significantly enhanced cell metastasis and invasion(P<0.01), whereas cell proliferation and the stages of the cell cycle did not differ significantly(P>0.05). A significantly increased expression level of MMP-2 has been observed in tumor cells treated portal vein serum. In addition, compared with peripheral serum, the content of IL-12 was significantly decreased in portal vein serum(P<0.05), while the contents of IFN-γ, IL-1β, IL-2, and TNF-α did not differ significantly(P>0.05). Conclusions: Portal vein serum from HCC patients could inhibit the apoptosis of PVTT-originated HCC cells and promote cell metastasis and invasion. | 9 | The Mitochondrial Genome of Pseudocalotes microlepis (Squamata: Agamidae) and its Phylogenetic Position in Agamids显示文摘We describe the complete mitochondrial genome of a small-scaled forest agamid(Pseudocalotes microlepis), which is 17 873 bp in size, containing 13 PCGs, 2 r RNAs, 22 t RNAs and non-coding regions. The mitogenome has a typical gene order among squamates. 13 PCGs include 2 start codons(ATG and ATA), 3 stop codons(TAG, TAA and AGG), and an incomplete stop codon(T-). Codon usage analyses showed that CUA-Leu1 and CGG-Arg are the most frequently and rarely used codon, respectively. All 22 t RNAs were predicted to form canonical cloverleaf secondary structures, except for two t RNAs(t RNACys and t RNASer(AGY)) lacking the dihydorouridine(DHU) arm. The large noncoding region(control region) is 2 687 bp long(28.3% T, 18.2% C, 42.3% A, and 11.2% G), with four different types of repeating sequences. The phylogenetic tree resulting from BEAST analyses based on concatenated 2 r RNAs and 13 PCGs in sequence revealed that the newly sequenced P. microlepis, where the genus Acanthosaura were aggregated. | 11 | Lineage diversification and niche evolution in the Reeves’Butterfly Lizard Leiolepis reevesii(Agamidae)显示文摘We used mitochondrial cytochrome b and ND4 genes and 9 microsatellite loci to determine genetic diversity,population structure,evolutionary history,and migration patterns within the Reeves’butterfly lizard Leiolepis reevesii(Agamidae).Considering molecular-based phylogeographical lineages,we then performed niche equivalency and similarity tests between divergent lineages.Phylogenetic analyses based on mitochondrial DNA(mtDNA)data revealed 2 lineages(A and B)diverging≈0.84 million years ago and,respectively,restricted to the northern and southern portions of the Wuzhishan and Yinggeling mountain ranges.Lineage B contains individuals from southern Hainan;Lineage A includes individuals from all other localities and can be further divided into 3 clusters according to microsatellite data.The null hypothesis that the 2 lineages shared identical niches was rejected in all niche equivalency tests,indicating niche shifts during genetic divergence.Similarity tests provided evidence of niche conservatism,suggesting that the 2 lineages share more characteristics of their niche spaces than randomly expected.The niche similarity and equivalency tests indicated a complex niche pattern in which both lineages share a main portion of their ecological spaces.The climatic niche of Lineage B represented a marginal and specialized fraction of the entire ecological space of the climatic niche of Lineage A,with warmer conditions.Isolation caused by orogenesis and subsequent niche divergence,together with local adaptation,may have led to genetic differentiation and further lineage sorting in L.reevesii. | Xiaming ZHU Lei HUA Mengchao FANG Yu DU Chixian LIN Longhui LIN Xiang JI | 2021 | Integrative Zoology2021,16,3: | 1 |
| 12 | A straightforward spectral emissivity estimating method based on constructing random rough surfaces显示文摘Spectral emissivity is an essential and sensitive parameter to characterize the radiative capacity of the solid surface in scientific and engineering applications,which would be non-negligibly affected by surface morphology.However,there is a lack of assessment of the effect of roughness on emissivity and a straightforward method for estimating the emissivity of rough surfaces.This paper established an estimating method based on constructing random rough surfaces to predict rough surface(Geometric region)emissivity for metal solids.Based on this method,the emissivity of ideal gray and non-gray body surfaces was calculated and analyzed.The calculated and measured spectral emissivities of GH3044,K465,DD6,and TC4 alloys with different roughness were compared.The results show that the emissivity increases with the roughness degree,and the enhancement effect weakens with the increase of roughness or emissivity due to the existing limit(emissivityε=1.0).At the same time,the roughness would not change the overall spectral distribution characteristics but may attenuate the local features of the spectral emissivity.The estimated results are in good agreement with the experimental data for the above alloys'rough surfaces.This study provides a new reliable approach to obtaining the spectral emissivity of rough surfaces.This approach is especially beneficial for measuring objects in extreme environments where emissivity is difficult to obtain.Meanwhile,this study promotes an understanding of surface morphology's effect mechanism on emissivity. | Zezhan Zhang Mengchao Chen Lichuan Zhang Hongzu Li Hairui Huang Zilong Zhang Peifeng Yu Yi Niu Shan Gao Chao Wang Jing Jiang | 2023 | Light(Science & Applications)2023,12,12: | 0 |

| 15 | Macroecological Patterns of Climatic Niche Breadth Variation in Lacertid Lizards显示文摘Measuring climatic niche position and breadth may help to determine where species can occur over space and time. Using GIS-based and phylogenetic comparative methods, we investigated global patterns of variation in climatic niche breadth in lacertid lizards to test the following three hypotheses about climatic niche widths. First, does a species' temperature or precipitation niche breadth relate to its temperature or precipitation niche position(the mean value of annual mean temperature or annual precipitation across sampled localities in the range of each species)? Second, are there trade-offs between a species' temperature niche breadth and precipitation niche breadth? Third, does a species' temperature or precipitation niche breadth relate to altitude or latitude? We expect that:(1) species distributed in cold regions are specialized for low-temperature environments(i.e. narrow niche breadth center around low temperatures);(2) a negative relationship between species niche breadth on temperature and precipitation axes according to the tradeoff hypothesis(i.e. species that tolerate a broad range of precipitation regimes cannot also tolerate a broad range of temperatures);(3) precipitation niche breadth decreases with altitude or latitude, whereas temperature climatic niche breadth increases with altitude or latitude. Based on the analytical results we found that:(1) temperature niche breadth and position are negatively related, while precipitation niche breadth and position are positively related;(2) there is no trade-off between temperature and precipitation niche breadths; and (3) temperature niche breadth and latitude/altitude are positively related, but precipitation niche breadth and latitude/altitude are not significantly related.
