1A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 3Diagnosis and treatment of infantile malignant solid tumors in Beijing,China:A multicenter 10-year retrospective study显示文摘Importance:Cancer is the main cause of death by disease in children.Children experience the highest incidence of cancer in the first year of life.However,there is no comprehensive registration system for children with tumors in China.Objective:To summarize the diagnosis and treatment of infant cancer and analyze the status of standardized diagnosis and management among several treatment centers in Beijing,China,thereby providing evidence to guide further clinical research.Methods:From January 1,2010 to December 31,2019,patients with newly diagnosed infantile malignant solid tumors were admitted to six large tertiary pediatric solid tumor diagnosis and treatment centers in Beijing.The epidemiology,clinical features,and therapeutic effects of tumors in these patients were analyzed retrospectively.All patients were followed up until March 31,2020.Results:In total,938 patients were enrolled in this study.There were 530 boys(56.5%)and 408 girls(43.5%);the median age was 6.0 months(range,0-12.0 months).The three most common tumors were retinoblastoma in 366 patients(39.0%),neuroblastoma in 266 patients(28.4%),hepatoblastoma in 133 patients(14.2%),and central nervous system tumors in 52 patients(5.5%).The estimated 5-year overall survival rate was 81.3%±1.8%,and the 5-year event-free survival rate was 71.8%±2.9%.The 5-year overall survival rates of non-rhabdomyosarcoma soft tissue sarcoma,neuroblastoma,and retinoblastoma were 100%,88%±2.2%,and 86.9%±2.1%,respectively.The 5-year event-free survival rates were 81.1%±2.7%for neuroblastoma,81.6%±9.8%for non-rhabdomyosarcoma soft tissue sarcoma,and 72.7%±14.1%for extracranial malignant germ cell tumors.Interpretation:The three most common infantile malignant solid tumors were retinoblastoma,neuroblastoma,and hepatoblastoma.Multidisciplinary combined diagnosis and treatment is needed for infantile tumors.Mei Jin Zhi Tian Yao Xie Zhaoxia Zhang Miao Li YaxiongYu Weiling Zhang Junyang Zhao Huanmin Wang Qi Zeng Long Li Ming Ge Ning Sun Xiaolun Zhang Jian Gong Wanshui Wu Rong Liu Weihong Zhao Dongsheng Huang Xiaoli Ma 2020Pediatric Investigation2020,4,3:3
4Tumor shrinkage by cyclopamine tartrate through inhibiting hedgehog signaling显示文摘The link of hedgehog (Hh) signaling activation to human cancer and synthesis of a variety of Hh signaling inhibitors raise great expectation that inhibiting Hh signaling may be effective in human cancer treatment. Cyclopamine (Cyc), an alkaloid from the Veratrum plant, is a specific natural product inhibitor of the Hh pathway that acts by targeting smoothened (SMO) protein. However, its poor solubility, acid sensitivity, and weak potency relative to other Hh antagonists prevent the clinical development of Cyc as a therapeutic agent. Here, we report properties of cyclopamine tartrate salt (CycT) and its activities in Hh signaling-mediated cancer in vitro and in vivo. Unlike Cyc, CycT is water soluble (5-10 mg/mL). The median lethal dose (LD50) of CycT was 62.5 mg/kg body weight compared to 43.5 mg/kg for Cyc, and the plasma half-life (T1/2) of CycT was not significantly different from that of Cyc. We showed that CycT had a higher inhibitory activity for Hh signaling-dependent motor neuron differentiation than did Cyc (IC50 = 50 nmol/L for CycT vs. 300 nmol/L for Cyc). We also tested the antitumor effectiveness of these Hh inhibitors using two mouse models of basal cell carcinomas (K14cre:Ptch1neo/neo and K14cre:SmoM2YFP). After topical application of CycT or Cyc daily for 21 days, we found that all CycT-treated mice had tumor shrinkage and decreased expression of Hh target genes. 6ROS-responsive capsules engineered from EGCG-Zinc networks improve therapeutic angiogenesis in mouse limb ischemia显示文摘The successful treatment of limb ischemia requires that promote angiogenesis along with microenvironment improvement.Zinc ions have been reported to stimulate angiogenesis,but application was limited to the toxicity concerns.We hypothesized that zinc based metal-EGCG capsule(EGCG/Zn Ps)can achieve sustained release Zn2+resulting in reduced toxicity and improve angiogenesis as well as the improvement of microenvironment by ROS scavenging of EGCG.The surface morphology,zeta potential,infrared absorbance peaks and zinc ion release profile of the EGCG/Zn Ps were measured.In vitro,EGCG/Zn showed significantly antioxidant,antiinflammatory and induced cell migration effect.In addition,EGCG/Zn Ps enabled the sustained release of zinc ions,which reduced cytotoxicity and enhanced the secretion of vascular endothelial growth factor(VEGF)in vitro and in vivo.In mouse models of limb ischemia,EGCG/Zn Ps promoted angiogenesis and cell proliferation in ischemic tissues.Moreover,EGCG/Zn Ps group exhibited the most significant recovery of limb ischemic score,limb temperature and blood flow than other groups.In conclusion,EGCG/Zn Ps is a safe and promising approach to combine the merit of Zn2+and EGCG,thus enabling the direct application to limb ischemia.Zuoguan Chen Jianwei Duan Yongpeng Diao Youlu Chen Xiaoyu Liang Huiyang Li Yuqing Miao Qing Gao Liang Gui Xiaoli Wang Jing Yang Yongjun Li 2021Bioactive Materials2021,6,1:3
7SRO1 regulates heavy metal mercury stress response in Arabidopsis thaliana显示文摘Heavy metals in the environment are harmful limiting factors for the normal growth and development of plants.Here,we isolated and identified an Arabidopsis thaliana T-DNA insertion mutant,named sro1-1,which showed a hyper-sensitive response to HgCl2.The SRO1protein contains a WWE domain that mediates proteinprotein interactions.Under HgCl2treatment,when compared with the wild-type plants,the growth of sro1-1 was repressed dramatically and the number of true leaves was reduced and etiolated.The electrolyte leakage rates showed that cell membrane integrity in sro1-1 was damaged more severely than in the wild type.DAB(3,5-diaminobenzidine)staining and confocal microscopy showed that Hg2?stress induced more hydrogen peroxide accumulation in sro1-1 than in the wild type.The qRT-PCR results indicated that the expression of some abiotic stress-induced genes,such as L-ascorbate peroxidase(APX1),was reduced under oxidative or Hg2?stress.Transgenic plants containing a GFP::SRO1 fusion protein showed that SRO1was localized in the nucleus of the cells.SRO1 was shown to be expressed in various tissues,and was most highly expressed in the vigorous tissues.Our results suggest thatSRO1 may play an important role in the stress response of A.thaliana to heavy metals.Xiaoli Zhang Xiaoliang Zhao Baozhu Li Jinchan Xia Yuchen Miao 2014Chinese Science Bulletin2014,59,25:2

11Effects Associated with Nanostructure Fabrication Using In Situ Liquid Cell TEM Technology显示文摘We studied silicon,carbon,and SiC xnanostructures fabricated using liquid-phase electron-beam-induced deposition technology in transmission electron microscopy systems.Nanodots obtained from fixed electron beam irradiation followed a universal size versus beam dose trend,with precursor concentrations from pure Si Cl4to 0%SiC l4in CH2Cl2,and electron beam intensity ranges of two orders of magnitude,showing good controllability of the deposition.Secondary electrons contributed to the determination of the lateral sizes of the nanostructures,while the primary beam appeared to have an effect in reducing the vertical growth rate.These results can be used to generate donut-shaped nanostructures.Using a scanning electron beam,line structures with both branched and unbranched morphologies were also obtained.The liquid-phase electron-beaminduced deposition technology is shown to be an effective tool for advanced nanostructured material generation.Xin Chen Lihui Zhou Ping Wang Hongliang Cao Xiaoli Miao Feifei Wei Xia Chen 2015Nano-Micro Letters2015,7,4:1
12Responses of the methanogenic pathway and fraction of CH4 oxidization in a flooded paddy soil to rice planting显示文摘Rice planting(RP)is significant to methane(CH4_(4))emissions from paddy fields,but its effect on the relative contribution of the acetoclastic methanogenesis to total CH4_(4) production(F_(ac))and the fraction of CH4_(4) oxidized(Fox)is poorly understood.To quantify the responses of the F_(ac) and Fox to RP,we investigated CH4_(4) fluxes,CH4_(4) production and oxidation potentials,dissolved CH4_(4) concentrations,and their stable carbon isotopes in a flooded paddy soil.The mcrA and pmoA gene copies were also determined by quantitative polymerase chain reaction(qPCR).Compared with the unplanted soil(control,CK),the seasonal CH4_(4) emissions from the planted soil were significantly enhanced,13.6 times,resulting in large decreases in the CH4_(4) concentrations in the soil solution.This indicated that much more CH4_(4) was released into the atmosphere by the RP than was stored in the soils.Acetoclastic methanogenesis became more important from the tillering stage(TS)to the ripening stage(RS)for the CK,with F_(ac) values increased from 17%-20% to 46%-55%.With RP,the F_(ac) values were enhanced by 10%-20%,and it significantly increased the copy numbers of the mcrA gene at the four rice stages(TS,booting stage(BS),grain-filling stage(GS),and RS).Furthermore,the effect of the RP on the abundance of the mcrA gene was highly concurrent with the effect on the F_(ac) values.At the TS,the Fox values at the soil-water interface were around 50%-75% for the CK,being 15%-20% lower than those of the RP in the rhizosphere.It increased to 65%-100% at the GS,but was reduced by 20%-30% after the RP.These differences might be because the copy numbers of the pmoA gene were significantly raised at the TS while lowered at the GS by the RP.This was further demonstrated by the strong correlations between the effect of the RP on the abundance of the pmoA gene and the effect on the Fox values.These findings suggest that RP markedly impacts on the abundances of the mcrA and pmoA genes,affecting the pathway of CH4_(4) production and the fraction of CH4_(4) oxidization,respectively.Yuting YANG Guangbin ZHANG Jing MA Qiong HUANG Haiyang YU Kaifu SONG Xiaoli ZHU Xi MIAO Hua XU 2021Pedosphere2021,31,6:1

18Cenozoic Ephedraceae adaptation to global cooling in northwestern China显示文摘Ephedraceae has been applied largely as a drought indicator to reconstruct Cenozoic paleoenvironment and paleoclimate. However, temperature indication of Ephedraceae has been largely ignored. Here, we provide a record of Ephedraceae percentage spanning from the Early Eocene to Middle Miocene (52-17 Myr B.P.) in the Xining Basin, northeastern Tibetan Plateau. This record is comparable to a compiled Cenozoic Ephedraceae record from five other basins in northwestern China. Both records show Ephedraceae percentages were high during the Early Eocene, and decreased gradually from the Middle Eocene to Late Oligocene, then maintained a stable level since the Late Oligocene. By comparing these two Ephedraceae records with the marine oxygen isotope record, we discuss the variation of Ephedraceae percentage in Middle Cenozoic in response to global temperature change. Ephedraceae percentage was high in the Early Paleogene, associated with subtropical or tropical vegetation types in a global greenhouse climate, and decreased in Early Oligocene, associated with global cooling, suggesting that Ephedraceae is warm-tolerant during the Paleogene. The low Ephedraceae percentages in the Late Oligocene and Miocene were uncoupled with global warming, which may imply that Ephedraceae began to adapt to a eurythermic climate in the inland desert environment of western China. 19Arabidopsis cryptochrome 1 promotes stomatal development through repression of AGB1 inhibition of SPEECHLESS DNA-binding activity显示文摘Cryptochromes are blue light photoreceptors that mediate various light responses in plants and mammals. The heterotrimeric G-protein is known to regulate various physiological processes in plants and mammals. In Arabidopsis, cryptochrome 1(CRY1) and the G-protein β subunit AGB1 act antagonistically to regulate stomatal development.The molecular mechanism by which CRY1 and AGB1 regulate this process remains unknown.Here, we show that Arabidopsis CRY1 acts partially through AGB1, and AGB1 acts through SPEECHLESS(SPCH), a master transcription factor that drives stomatal initiation and proliferation, to regulate stomatal development. We demonstrate that AGB1 physically interacts with SPCH to block the b HLH DNA-binding domain of SPCH and inhibit its DNA-binding activity. Moreover, we demonstrate that photoexcited CRY1 represses the interaction of AGB1 with SPCH to release AGB1 inhibition of SPCH DNA-binding activity, leading to the expression of SPCH-target genes promoting stomatal development. Taken together, our results suggest that the mechanism by which CRY1 promotes stomatal development involves positive regulation of the DNA-binding activity of SPCH mediated by CRY1 inhibition of the AGB1-SPCH interaction.
