| 4 | License Assisted Access-WiFi Coexistence with TXOP Backoff for LTE in Unlicensed Band显示文摘License assisted access(LAA) was proposed as a solution to the challenge of satisfying high throughput requirement in cellular network with limited licensed band. LAA uses long term evolution(LTE) carrier aggregation technique to combine licensed and unlicensed bands, specifically 5 GHz Wi Fi band, in order to achieve high data rates. However, LAA has several critical concerns for services provision on 5 GHz unlicensed band. In this article, an overview of the techniques concerning the coexistence of LTE and Wi Fi on the same unlicensed band is presented. We first present several general approaches including power control of LTE evolved Node B(e NB), carrier selection, LTE duty cycling, self clear to send(CTS) and request to send/CTS(RTS/CTS), and listen before talk(LBT). Then, we focus on LBT techniques for LAA-Wi Fi coexistence and discuss how LBT based coexistence solutions can dynamically respond to the variations in network load and ensure fair coexistence in a distributed manner. Based on the LBT mechanism, we also present a fairness-aware LAA-WiF i coexisting scheme with TXOP(transmitted in a single transmission opportunity) backoff for LAA e NB. | 7 | SUMOylation-triggered ALIX activation modulates extracellular vesicles circTLCD4-RWDD3 to promote lymphatic metastasis of non-small cell lung cancer显示文摘Lymph node(LN)metastasis is one of the predominant metastatic routes of non-small cell lung cancer(NSCLC)and is considered as a leading cause for the unsatisfactory prognosis of patients.Although lymphangiogenesis is well-recognized as a crucial process in mediating LN metastasis,the regulatory mechanism involving lymphangiogenesis and LN metastasis in NSCLC remains unclear.In this study,we employed high-throughput sequencing to identify a novel circular RNA(circRNA),circTLCD4-RWDD3,which was significantly upregulated in extracellular vesicles(EVs)from LN metastatic NSCLC and was positively associated with deteriorated OS and DFS of patients with NSCLC from multicenter clinical cohort.Downregulating the expression of EV-packaged circTLCD4-RWDD3 inhibited lymphangiogenesis and LN metastasis of NSCLC both in vitro and in vivo.Mechanically,circTLCD4-RWDD3 physically interacted with hnRNPA2B1 and mediated the SUMO2 modification at K108 residue of hnRNPA2B1 by upregulating UBC9.Subsequently,circTLCD4-RWDD3-induced SUMOylated hnRNPA2B1 was recognized by the SUMO interaction motif(SIM)of ALIX and activated ALIX to recruit ESCRT-III,thereby facilitating the sorting of circTLCD4-RWDD3 into NSCLC cell-derived EVs.Moreover,EV-packaged circTLCD4-RWDD3 was internalized by lymphatic endothelial cells to activate the transcription of PROX1,resulting in the lymphangiogenesis and LN metastasis of NSCLC.Importantly,blocking EV-mediated transmission of circTLCD4-RWDD3 via mutating SIM in ALIX or K108 residue of hnRNPA2B1 inhibited the lymphangiogenesis and LN metastasis of NSCLC in vivo.Our findings reveal a precise mechanism underlying SUMOylated hnRNPA2B1-induced EV packaging of circTLCD4-RWDD3 in facilitating LN metastasis of NSCLC,suggesting that EV-packaged circTLCD4-RWDD3 could be a potential therapeutic target against LN metastatic NSCLC. | Xiayao Diao Chao Guo Hanhao Zheng Ke Zhao Yuming Luo Mingjie An Yan Lin Jiancheng Chen Yuanlong Li Yuting Li Xuehan Gao Jiaqi Zhang Mengxin Zhou Wenliang Bai Lei Liu Guige Wang Lanjun Zhang Xiaotian He Rusi Zhang Zhihua Li Changhao Chen Shanqing Li | 2023 | Signal Transduction and Targeted Therapy2023,8,12: | 0 |
| 8 | Experimental Study on the Influencing Factors of Motion Responses on an Air-Floating Caisson with Multiple Compartments显示文摘The structure of an air-floating caisson is suitable for the major structure of caisson-type artificial islands.Thus,it has been rapidly developed and widely used in the exploration and development of oil and gas fields in shallow sea and intertidal zones.Air-floating transportation technology is one of the key technologies employed in this structure.In this paper,the factors influencing the dynamic response characteristics of air-floating caisson with multi-compartments(AFCMC)were studied using model tests.The length and the height of each air-floating structure in the model were 1.0 and 0.1 m,respectively.In addition,the 1:100 models with 6,8,and 10 compartments under regular waves were tested in the wave flume,respectively.In the experiments,the respective water depths were set at 0.2,0.3,and 0.4 m,and the corresponding drafts were 0.05,0.06,and 0.07 m.Results show that with the increase of draft,the heave natural period increased and the maximum amplitude of the heave motion decreased.Meanwhile,the pitch motion decreased at 6 and 8 compartments and increased at 10 compartments.As the water depth increased,the maximum amplitude and amplitude change of heave and pitch motions first increased and then decreased.However,several amplitudes close to the maximum amplitude appeared in the measured period at shallower water depth,thereby indicating the vertical movements of the structure enhanced under shallow water.The increase in the number of compartments reduced the vertical movements under 6.0 m draft,but it increased the vertical movements under 5.0 and 7.0 m draft.Thus,increasing the number of compartments has a limited capacity to improve the motion performance of the structure. | LIU Xianqing ZHANG Puyang ZHAO Mingjie DING Hongyan LE Conghuan LV Nan LUO Sheng | 2022 | Journal of Ocean University of China2022,21,1: | 0 |
| 9 | Berberine alleviates myocardial diastolic dysfunction by modulating Drp1-mediated mitochondrial fission and Ca^(2+)homeostasis in a murine model of HFpEF显示文摘Heart failure with preserved ejection fraction(HFpEF)displays normal or near-normal left ventricular ejection fraction,diastolic dysfunction,cardiac hypertrophy,and poor exercise capacity.Berberine,an isoquinoline alkaloid,possesses cardiovascular benefits.Adult male mice were assigned to chow or high-fat diet with L-NAME(“two-hit”model)for 15 weeks.Diastolic function was assessed using echocardiography and noninvasive Doppler technique.Myocardial morphology,mitochondrial ultrastructure,and cardiomyocyte mechanical properties were evaluated.Proteomics analysis,autophagic flux,and intracellular Ca^(2+)were also assessed in chow and HFpEF mice.The results show exercise intolerance and cardiac diastolic dysfunction in“two-hit”-induced HFpEF model,in which unfavorable geometric changes such as increased cell size,interstitial fibrosis,and mitochondrial swelling occurred in the myocardium.Diastolic dysfunction was indicated by the elevated E value,mitral E/A ratio,and E/e’ratio,decreased e’value and maximal velocity of re-lengthening(-dL/dt),and prolonged re-lengthening in HFpEF mice.The effects of these processes were alleviated by berberine.Moreover,berberine ameliorated autophagic flux,alleviated Drp1 mitochondrial localization,mitochondrial Ca^(2+)overload and fragmentation,and promoted intracellular Ca^(2+)reuptake into sarcoplasmic reticulum by regulating phospholamban and SERCA2a.Finally,berberine alleviated diastolic dysfunction in“two-hit”diet-induced HFpEF model possibly because of the promotion of autophagic flux,inhibition of mitochondrial fragmentation,and cytosolic Ca^(2+)overload. | Miyesaier Abudureyimu Mingjie Yang Xiang Wang Xuanming Luo Junbo Ge Hu Peng Yingmei Zhang Jun Ren | 2023 | Frontiers of Medicine2023,17,6: | 0 |

| 13 | Anti-apoptotic protein BCL-XL as a therapeutic vulnerability in gastric cancer显示文摘Background: New therapeutic targets are needed to improve the outcomes for gastric cancer(GC) patients with advanced disease. Evasion of programmed cell death(apoptosis) is a hallmark of cancer cells and direct induction of apoptosis by targeting the pro-survival BCL2 family proteins represents a promising therapeutic strategy for cancer treatment. Therefore, understanding the molecular mechanisms underpinning cancer cell survival could provide a molecular basis for potential therapeutic interventions. Method: Here we explored the role of BCL2L1 and the encoded anti-apoptotic BCL-XL in GC. Using Droplet Digital PCR(ddPCR) technology to investigate the DNA amplification of BCL2L1 in GC samples and GC cell lines, the sensitivity of GC cell lines to selective BCL-XL inhibitors A1155463 and A1331852, pan-inhibitor ABT-263, and VHL-based PROTAC-BCL-XL was analyzed using(CellTiter-Glo) CTG assay in vitro. Western Blot(WB) was used to detect the protein expression of BCL2 family members in GC cell lines and the manner in which PROTAC-BCL-XL kills GC cells. Coimmunoprecipitation(Co-IP) was used to investigate the mechanism of A1331852 and ABT-263 kills GC cell lines. DDPCR, WB, and real-time PCR(RTPCR) were used to investigate the correlation between DNA, RNA, protein levels, and drug activity. Results: The functional assay showed that a subset of GC cell lines relies on BCL-XL for survival. In gastric cancer cell lines, BCL-XL inhibitors A1155463 and A1331852 are more sensitive than the pan BCL2 family inhibitor ABT-263, indicating that ABT-263 is not an optimal inhibitor of BCL-XL. VHL-based PROTAC-BCL-XL DT2216 appears to be active in GC cells. DT2216 induces apoptosis of gastric cancer cells in a time-and dose-dependent manner through the proteasome pathway. Statistical analysis showed that the BCL-XL protein level predicts the response of GC cells to BCL-XL targeting therapy and BCL2L1 gene CNVs do not reliably predict BCL-XL expression.Conclusion: We identified BCL-XL as a promising therapeutic target in a subset of GC cases with high levels of BCL-XL protein expression. Functionally, we demonstrated that both selective BCL-XL inhibitors and VHL-based PROTAC BCL-XL can potently kill GC cells that are reliant on BCL-XL for survival. However, we found that BCL2L1 copy number variations(CNVs) cannot reliably predict BCL-XL expression, but the BCL-XL protein level serves as a useful biomarker for predicting the sensitivity of GC cells to BCL-XL-targeting compounds. | 14 | An HGF-dependent positive feedback loop between bladder cancer cells and fibroblasts mediates lymphangiogenesis and lymphatic metastasis显示文摘Background Cancer-associated fibroblasts (CAFs) play a vital role in facilitating tumor progression through extensive reciprocal interplay with cancer cells. Tumor-derived extracellular vesicles (EVs) are the critical mediators involved in the crosstalk between cancer cells and stromal cells, contributing to the metastasis of cancers. Yet, the biological mechanisms of tumor-derived EVs in triggering CAFs phenotype to stimulate the lymph node (LN) metastasis of bladder cancer (BCa) are largely unknown. Here, we aimed to explore the effects and molecular mechanisms of tumor-derived EV-mediated CAFs phenotype in regulating BCa LN metastasis. Methods The high-throughput sequencing was utilized to identify the crucial long non-coding RNA (lncRNA) associated with CAF enrichment in BCa. The functional role of the transition of fibroblasts to CAFs induced by LINC00665-mediated EVs was investigated through the in vitro and in vivo assays. Chromatin isolation by RNA purification assays, fluorescence resonance energy transfer assays, cytokine profiling and patient-derived xenograft (PDX) model were performed to explore the underlying mechanism of LINC00665 in the LN metastasis of BCa. Results We found that CAFs are widely enriched in the tumor microenvironment of BCa, which correlated with BCa lymphangiogenesis and LN metastasis. We then identified a CAF-associated long non-coding RNA, LINC00665, which acted as a crucial mediator of CAF infiltration in BCa. Clinically, LINC00665 was associated with LN metastasis and poor prognosis in patients with BCa. Mechanistically, LINC00665 transcriptionally upregulated RAB27B expression and induced H3K4me3 modification on the promoter of RAB27B through the recruitment of hnRNPL. Moreover, RAB27B-induced EVs secretion endowed fibroblasts with the CAF phenotype, which reciprocally induced LINC00665 overexpression to form a RAB27B-HGF-c-Myc positive feedback loop, enhancing the lymphangiogenesis and LN metastasis of BCa. Importantly, we demonstrated that blocking EV-transmitted LINC00665 or HGF broke this loop and impaired BCa lymphangiogenesis in a PDX model. | 16 | Role of PIVKA-II in screening for malignancies at a hepatobiliary andpancreatic disease center: A large-scale real-world study显示文摘Background and aims:Protein induced by vitamin K absence or antagonist II(PIVKA-II)is a well-accepted biomarker for diagnosing hepatocellular carcinoma(HCC).Although nonspecific increase of PIVKA-II has been reported,real-world evidence remains scarce.Based on real-world data,this study aimed to comprehensively describe the use of PIVKA-II at a hepatobiliary and pancreatic disease center and to assess its utility for the initial screening of HCC or other hepatobiliary–pancreatic malignancies.Methods:This real-world retrospective study is based on the PIVKA-II results of 16,215 individuals and other relevant laboratory test(alpha-fetoprotein[AFP],carbohydrate antigen 19-9[CA19-9],liver function,blood coagulation indicators,hepatitis B virus,and hepatitis C virus).However,only the first PIVKA-II results of 7809 eligible individuals were included.Between-group comparisons,correlation analysis,and receiver operating characteristic curve analysis were performed.Results:PIVKA-II results were abnormal in patients with HCC(55.9%),biliary carcinoma(BC,13.4%),gastroin-testinal and pancreatic cancer(6.3%),and benign diseases(23.5%)as well as in healthy individuals(0.92%).The area under the curve of PIVKA-II for detecting malignancies was 0.7754(0.7620–0.7688),whereas that for detecting HCC was 0.7509(0.7357–0.7662).Stratifying the PIVKA-II values or combining PIVKA-II with AFP or CA19-9 helped improve the diagnostic performance of PIVKA-II for HCC.PIVKA-II values were significantly positively correlated with AST in patients with HCC and with bilirubin in patients with BC.Conclusions:This study determined the role of PIVKA-II in malignancy screening at hepatobiliary and pancreatic disease centers.It was also noted that the diagnostic efficacy of PIVKA-II for HCC improved after combining PIVKA-II with AFP or stratifying its value. | Chenghao Ge Mingjie Luo Kaiyuan Guo Dong Zhu Ning Han Tengjiao Wang Xiuying Zhao | 2022 | iLIVER2022,1,4: | 0 |
| 17 | A test method for analyzing the deformation of landslide model显示文摘In order to study the overall deformation of geotechnical model conveniently,the worksite of landslide bridge foundation reinforced by the front and rear row anti-slide piles in Chenglan railway was taken as an example.On the basis of shaking tabe test of a 1/40 reduced scale model,the landslide deformation caused by vibration waves was monitored through burying self-made phosphor bronze strips in soil.Combined with the horizontal and vertical coordinates of the bending strain points on the phosphor bronze strips,the digital matrix was converted by applying Renka Cline random matrix generation method,and the two-dimensional contour plots were drawn based on it.The results showed that the two-dimensional contour plots reflected the basic law of landslide deformation reasonably,and it revealed the evolution process of landslide deformation and failure.The research conclusions were consistent with the test phenomenon,which met the basic requirements of overall deformation analysis of landslide model.This proposed method can monitor multiple cross sections and was practical for model test. | Da Lei Mingjie Chen Zhihao Xu Bin Luo Guanlu Jiang Kai Fan Qihao Zhou | 2021 | Earthquake Research Advances2021,1,3: | 0 |
