| 1 | Nuclear factor-kappaB activation on the reactive oxygen species in acute necrotizing pancreatitic rats显示文摘AIM: To investigate the potential role of nuclear factor kappa-B (NF-κB) activation on the reactive oxygen species in rat acute necrotizing pancreatitis (ANP) and to assess the effect of pyrrolidine dithiocarbamate (PDTC, an inhibitor of NF-κB).METHODS: Rat ANP model was established by retrograde injection of 5% sodium taurocholate into biliopancreatic duct. Rats were randomly assigned to three groups (10rats each): Control group, ANP group and PDTC group. At the 6th h of the model, the changes of the serum amylase,nitric oxide (NO), malondialdehyde (MDA), superoxide dismutase (SOD) and pancreatic morphological damage were observed. The expressions of inducible nitric oxide (iNOS) were observed by SP immunohistochemistry. And the expressions of NF-κB p65 subunit mRNA were observed by hybridization in situ.RESULTS: Serum amylase and NO level decreased significantly in ANP group as compared with PDTC administrated group [(7 170.40±1 308.63) U/L vs(4 074.10±1 719.78) U/L,P<0.05], [(76.95±9.04) μmol/L vs (65.18±9.02) μmol/L,P<0.05] respectively. MDA in both ANP and PDTC group rose significantly over that in control group [(9.88±1.52)nmol/L, (8.60±1.41) nmol/L, vs (6.04±1.78) nmol/L,P<0.05], while there was no significant difference between them. SOD levels in both ANP and PDTC group underwent a significant decrease as compared with that in control [(3 214.59±297.74) NU/mL, (3 260.62±229.44) NU/mL,vs (3 977.80±309.09) NU/mL, P<0.05], but there was no significant difference between them. Though they were still higher than those in Control group, pancreas destruction was slighter in PDTC group, iNOS expression and NF-κB p65 subunit mRNA expression were lower in PDTC group as compared with ANP group.CONCLUSION: We conclude that correlation among NF-κB activation, serum amylase, reactive oxygen species level and tissue damage suggests a key role of NF-κB in the pathogenesis of ANP. | 9 | Recovery of rare earths and aluminum from FCC waste slag by acid leaching and selective precipitation显示文摘This paper investigated the recovery of rare earth elements(REEs) and aluminum(AI) from the waste slag discharged by FCC catalyst factory(FCC waste slag) via acid leaching and selective precipitation.Analysis methods such as ICP-AES, XRF and XRD were applied to obtain experiment data. The maximum leaching efficiency of REEs and Al was achieved at pH value of 1 and with liquid to solid ratio of 4:1,Under such conditions, 91.01%, 92.24% and 94.77% of La, Ce and Al were extracted at 20 ℃ for 2 h from the FCC waste slag, respectively. The SiO_2 content in the leaching residue was 88.3%, which can be used as an available silicon resource. The REEs can be precipitated in the form of REEs and sodium double sulfate(NaRE(SO_4)_2·xH_2 O) by adding Na_2 SO_4 to the leaching solution, while Al remained in the solution. Afterwards, the pH value of the filtrate was adjusted to 4.5, and Al was precipitated as AI(OH)_3. Finally,NaRE(SO_4)_2·xH_2 O and Al(OH)_3 were converted into RECl_3 and Al_2(SO4)_3 solution,respectively, which were recycled to manufacture zeolite. | 10 | Translation and validation of the Tibetan confusion assessment method for the intensive care unit显示文摘Background: At present, there is no available delirium translated assessment method for 3.3 million Tibetans. This study ai provide a method for delirium assessment for Tibetan patients speaking this language by validating a translation of the Confusion Assessment Method for the Intensive Care Unit (CAM-ICU). Methods: The study was conducted between July 2018 and November 2018. Patients were screened for delirium by a neurologist using the Diagnostic and Statistical Manual of Mental Disorders IV (DSM-IV). Patients were subsequently screened by two nurses using Tibetan translations of the CAM-ICU. With DSM-IV criterion as the reference standard, the sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV) were calculated to assess the validity of the CAM-ICU criterion. Interrater reliability was determined by comparing the CAM-ICU ratings of nurse 1 vs. nurse 2 using the k coefficient. Results: Ninety-six patients were assessed independently by two nurses and one neurologist. According to DSM-IV standard, 42 out of 96 (43.8%) patients developed delirium. The sensitivities of Tibetan CAM-ICU were 90.5% for nurse 1 and 92.9% for nurse 2, respectively. Their specificities were 85.2% and 90.7%, respectively. The PPV were 82.6% for nurse 1 and 88.6% for nurse 2. Their NPV were 92.0% and 94.2%, respectively. The Tibetan CAM-ICU was done with good interrater reliability between nurse 1 and nurse 2 (k=0.91, P<0.001). | 14 | Generation of obese rat model by transcription activator-like effector nucleases targeting the leptin receptor gene显示文摘The laboratory rat is a valuable mammalian model organism for basic research and drug discovery. Here we demonstrate an efficient methodology by applying transcription activator-like effector nucleases(TALENs) technology to generate Leptin receptor(Lepr) knockout rats on the Sprague Dawley(SD) genetic background. Through direct injection of in vitro transcribed m RNA of TALEN pairs into SD rat zygotes, somatic mutations were induced in two of three resulting pups. One of the founders carrying bi-allelic mutation exhibited early onset of obesity and infertility. The other founder carried a chimeric mutation which was efficiently transmitted to the progenies. Through phenotyping of the resulting three lines of rats bearing distinct mutations in the Lepr locus, we found that the strains with a frame-shifted or premature stop codon mutation led to obesity and metabolic disorders. However, no obvious defect was observed in a strain with an in-frame 57 bp deletion in the extracellular domain of Lepr. This suggests the deleted amino acids do not significantly affect Lepr structure and function. This is the first report of generating the Lepr mutant obese rat model in SD strain through a reverse genetic approach. | 19 | Concurrent optimization of structural topology and infill properties with a CBF-based level set method显示文摘In this paper,a parametric level-set-based topology optimization framework is proposed to concurrently optimize the structural topology at the macroscale and the effective infill properties at the micro/meso scale.The concurrent optimization is achieved by a computational framework combining a new parametric level set approach with mathematical programming.Within the proposed framework,both the structural boundary evolution and the effective infill property optimization can be driven by mathematical programming,which is more advantageous compared with the conventional partial differential equatiodriven level set approach.Moreover,the proposed approach will be more efficient in handling nonlinear problems with multiple constraints.Instead of using radial basis functions(RBF),in this paper,we propose to construct a new type of cardinal basis functions(CBF)for the level set function parameterization.The proposed CBF parameterization ensures an explicit impose of the lower and upper bounds of the design variables.This overcomes the intrinsic disadvantage of the conventional RBF-based parametric level set method,where the lower and upper bounds of the design variables oftentimes have to be set by trial and error;A variational distance regularization method is utilized in this research to regularize the level set function to be a desired distanceregularized shape.With the distance information embedded in the level set model,the wrapping boundary layer and the interior infill region can be naturally defined.The isotropic infill achieved via the mesoscale topology optimization is conformally fit into the wrapping boundary layer using the shape-preserving conformal mapping method,which leads to a hierarchical physical structure with optimized overall topology and effective infill properties.The proposed method is expected to provide a timely solution to the increasing demand for multiscale and multifunctional structure design. | Long JIANG Yang GUO Shikui CHEN Peng WEI Na LEI Xianfeng David GU | 2019 | Frontiers of Mechanical Engineering2019,14,2: | 3 |
| 20 | Association between Coronary Atherosclerotic Plaque Composition and Cardiovascular Disease Risk显示文摘Objective The objective of this study is to determine whether coronary atherosclerotic plaque composition is associated with cardiovascular disease(CVD) risk in Chinese adults. Methods We performed a cross-sectional analysis in 549 subjects without previous diagnosis or clinical symptoms of CVD in a community cohort of middle-aged Chinese adults. The participants underwent coronary computed tomography(CT) angiography for the evaluation of the presence and composition of coronary plaques. CVD risk was evaluated by the Framingham risk score(FRS) and the 10-year atherosclerotic cardiovascular disease(ASCVD) risk score. Results Among the 549 participants, 267(48.6%) had no coronary plaques, 201(36.6%) had noncalcified coronary plaques, and 81(14.8%) had calcified or mixed coronary plaques. The measures of CVD risk including FRS and ASCVD risk score and the likelihood of having elevated FRS significantly increased across the groups of participants without coronary plaques, with noncalcified coronary plaques, and with calcified or mixed coronary plaques. However, only calcified or mixed coronary plaques were significantly associated with an elevated ASCVD risk score [odds ratio(OR) 2.41; 95% confidence interval(CI) 1.09-5.32] compared with no coronary plaques, whereas no significant association was found for noncalcified coronary plaques and elevated ASCVD risk score(OR 1.25; 95% CI 0.71-2.21) after multivariable adjustment.
