1Electroacupuncture in the repair of spinal cord injury:inhibiting the Notch signaling pathway and promoting neural stem cell proliferation显示文摘Electroacupuncture for the treatment of spinal cord injury has a good clinical curative effect, but the underlying mechanism is unclear. In our experiments, the spinal cord of adult Sprague-Dawley rats was clamped for 60 seconds. Dazhui(GV14) and Mingmen(GV4) acupoints of rats were subjected to electroacupuncture. Enzyme-linked immunosorbent assay revealed that the expression of serum inflammatory factors was apparently downregulated in rat models of spinal cord injury after electroacupuncture. Hematoxylin-eosin staining and immunohistochemistry results demonstrated that electroacupuncture contributed to the proliferation of neural stem cells in rat injured spinal cord, and suppressed their differentiation into astrocytes. Real-time quantitative PCR and western blot assays showed that electroacupuncture inhibited activation of the Notch signaling pathway induced by spinal cord injury. 4Exploratory clinical study of chidamide,an oral subtype-selective histone deacetylase inhibitor,in combination with exemestane in hormone receptor-positive advanced breast cancer显示文摘Objective: The recurrence or progression under endocrine therapy in hormone receptor-positive(HR+)advanced breast cancer(ABC) remained a critical clinical challenge.Chidamide is an oral subtype-selective histone deacetylase(HDAC) inhibitor with multiple functions in tumor growth inhibition and microenvironment modulation via epigenetic reprogramming.The purpose of this study was to evaluate the safety,pharmacokinetics(PK),and preliminary efficacy of chidamide in combination with exemestane in HR+ ABC patients.Methods: Eligible patients were postmenopausal women with HR+ ABC recurrent or progressed to at least one endocrine therapy.Blood samples were obtained in the run-in period and the first day of combination treatment for PK analysis.In combination treatment,patients were given exemestane 25mg daily and chidamide 30mg twice a week(BIW) until progression of disease or intolerable toxicities.A treatment cycle was defined as 4 weeks.Safety,PK parameters,and preliminary efficacy were evaluated.Results: A total of 20 patients were enrolled between July and December,2015.The median number of treatments cycle was 5.2(20.8 weeks) with 2 patients still on treatment at the data cut-off date of October,2017.The treatment-related adverse events(AE) ≥ grade 3 in more than 2 patients were neutropenia(35%),thrombocytopenia(30%),and leucopenia(20%).The plasma exposure of exemestane was consistent in the presence or absence of chidamide.A slight increase in chidamide exposure was noted in the presence of exemestane,probably due to the inter-and intra-patient variations.The best response in 16 evaluable patients was assessed by Response Evaluation Criteria in Solid Tumors(RECIST),including 4 patients with partial response,10 patients with stable disease.The median progression-free survival(PFS) was 7.6 months.Conclusions: The combination of chidamide with exemestane was generally well tolerated with promising preliminary efficacy in HR+ ABC patients.The overall results from this study encourage further pivotal trial in this patient population.Qingyuan Zhang Tao Wang Cuizhi Geng Yue Zhang Jinwen Zhang Zhiqiang Ning Zefei Jiang 2018Chinese Journal of Cancer Research2018,30,6:11

6Modified citrus pectin inhibited bladder tumor growth through downregulation of galectin-3显示文摘Modiled citrus pectin (MCP) is a carbohydrate enriched complex, which has been implicated in cancer treatment and prevention. However, the effects of MCP on urinary bladder cancer (UBC) are unknown. In this study, MCP was first tested in T24 and J82 human UBC cells and showed the inhibition of cell viability by the sulforhodamine B (SRB) assay. The MCP-treated UBC cells exhibited G2/M phase arrest with the decrease of Cyclin B1 and phosphorylated Cdc2. Caspase-3 was also activated, leading to the cleavage of Caspase-3 and PARP. We further explored the possible molecular mechanisms upon MCP treatment in UBC cells. Reduction of galectin-3 was observed and followed with the inactivation of Akt signaling pathway. Of note, galectin-3 knockdown by RNA interference recapitulated the MCP-mediated anti-proliferation, cell cycle arrest and apoptosis. Moreover, oral administration of MCP to the T24 xenograft-bearing nude mice inhibited the tumor growth significantly (P<0.05). Quantification analysis of immunohistochemistry staining for Ki67 and cleaved Caspase-3 confirmed the decrease of proliferation index (P<0.05) and the increase of apoptosis index (P<0.01) in 700 mg/kg MCP-fed UBC xenografts. Using the information from TCGA database, we revealed that the overexpression of galectin-3 was associated with high tumor grade with lymph node metastasis, poor overall survival in UBC patients. 7Unpalatable Weed Stellera chamaejasme L. Provides Biotic Refuge for Neighboring Species and Conserves Plant Diversity in Overgrazing Alpine Meadows on the Tibetan Plateau in China显示文摘Steller chamaejasme L.(S. chamaejasme for short) is one of the most noxious unpalatable weeds in China, which has been frequently reported its negative interaction(i.e. competition and allelopathy) with other herbaceous species in grasslands. This study compared species diversity, biomass and sexual reproduction of herbaceous plants in meadows with S. chamaejasme and in open meadows without S. chamaejasme in overgrazing meadows on the Tibetan Plateau in China to determine whether positive facilitation exist between S. chamaejasme and other herbaceous species under livestock's overgrazing. The results showed that there are more herbaceous species in meadows with S. chamaejasme than those in open meadows(35s and30s, respectively). Diversity index and above-ground biomass were also significantly higher in meadows with S. chamaejasme. There were 39%(11/28) of all species with sexual reproduction found in meadows with S. chamaejasme, which was 7 times more than those in open meadows. Our study showed that S. chamaejasme could provide biotic refuge for neighboring plants and preserve plant diversity from livestock's overgrazing in alpine meadows on the Tibetan Plateau. 10Protective effects of sirtuin 3 on titanium particle-induced osteogenic inhibition by regulating the NLRP3 inflammasome via the GSK-3β/β-catenin signalling pathway显示文摘Periprosthetic osteolysis(PPO)remains the key factor in implant failure and subsequent revision surgery and is mainly triggered by wear particles.Previous studies have shown that inhibition of osteoblastic differentiation is the most widespread incident affecting the interface of trabecular and loosening prostheses.Additionally,the NLRP3 inflammasome is activated by prosthetic particles.Sirtuin3,an NAD+-dependent deacetylase of mitochondria,regulates the function of mitochondria in diverse activities.However,whether SIRT3 can mitigate wear debris-induced osteolysis by inhibiting the NLRP3 inflammasome and enhancing osteogenesis has not been previously reported.Therefore,we investigated the role of SIRT3 during the process of titanium(Ti)particle-induced osteolysis.We revealed that upregulated SIRT3 dramatically attenuated Ti particle-induced osteogenic inhibition through suppression of the NLRP3 inflammasome and improvement of osteogenesis in vivo and in vitro.Moreover,we found that SIRT3 interference in the process of Ti particle-induced osteolysis relied on the GSK-3β/β-catenin signalling pathway.Collectively,these findings indicated that SIRT3 may serve as a rational new treatment against debris-induced PPO by deacetylase-dependent inflammasome attenuation.Kai Zheng Jiaxiang Bai Ning Li Meng Li Houyi Sun Weicheng Zhang Gaoran Ge Xiaolong Liang Huaqiang Tao Yi Xue Yuefeng Hao Chen Zhu Yaozeng Xu Dechun Geng 2021Bioactive Materials2021,6,10:5
11Effects of Waxy Maize Relay Intercropping and Residue Retention on Rhizosphere Microbial Communities and Vegetable Yield in a Continuous Cropping System显示文摘Intercropping and residue retention contribute to high yield and quality of crops. However, their coupled effects on rhizospheric microbial communities under a continuous vegetable cropping system have not been adequately addressed. The objective of the present study was to assess the effects on soil microbial community and yields of waxy maize(Zea mays L.) intercropped with or without residue retention in a continuous broccoli(Brassica oleracea L.) cropping system, i.e., relay intercropping of broccoli and waxy maize(B/M-B), relay intercropping of broccoli and waxy maize with residue retention(B/MR-B), and broccoli monoculture(B-B). The biomass yields of spring and autumn vegetables in B/MR-B were 16.3%–32.5% and 30.1%–46.1% higher than those of B-B,respectively. Autumn vegetable economic yields of B/MR-B were 28.2%–40.3% higher than B-B. The average well color development followed the order: B/MR-B > B/M-B > B-B. The Shannon index, Simpson index, and Mc Intosh index were higher in B/MR-B than under monoculture. A principal component analysis showed that microbial communities of B/MR-B soils differed from those of B/M-B and B-B soils. Carbon(C) sources utilized by the rhizosphere microorganisms were mainly carbohydrates, carboxylic acids, amino acids, and polymers; however, the C sources for the soil microbial community differed between intercropping and monoculture. The communities from B/MR-B preferred amino acids and polymers. Available nitrogen(N), potassium(K), and phosphorus(P) had an obvious impact on soil microbial community. Additionally, the C source utilization by microorganisms was significantly affected by p H and available K and P. 12Sustained high level transgene expression in mammalian cells mediated by the optimized piggyBac transposon system显示文摘Sustained,high level transgene expression in mammalian cells is desired in many cases for studying gene functions.Traditionally,stable transgene expression has been accomplished by using retroviral or lentiviral vectors.However,such viral vector-mediated transgene expression is often at low levels and can be reduced over time due to low copy numbers and/or chromatin remodeling repression.The piggyBac transposon has emerged as a promising nonviral vector system for efficient gene transfer into mammalian cells.Despite its inherent advantages over lentiviral and retroviral systems,piggyBac system has not been widely used,at least in part due to their limited manipulation flexibilities.Here,we seek to optimize piggyBac-mediated transgene expression and generate a more efficient,user-friendly piggyBac system.By engineering a panel of versatile piggyBac vectors and constructing recombinant adenoviruses expressing piggyBac transposase(PBase),we demonstrate that adenovirusmediated PBase expression significantly enhances the integration efficiency and expression level of transgenes in mesenchymal stem cells and osteosarcoma cells,compared to that obtained from co-transfection of the CMV-PBase plasmid.We further determine the drug selection timeline to achieve optimal stable transgene expression.Moreover,we demonstrate that the transgene copy number of piggyBac-mediated integration is approximately 10 times higher than that mediated by retroviral vectors.Using the engineered tandem expression vector,we show that three transgenes can be simultaneously expressed in a single vector with high efficiency.Thus,these results strongly suggest that the optimized piggyBac system is a valuable tool for making stable cell lines with sustained,high transgene expression.Xiang Chen Jing Cui Zhengjian Yan Hongmei Zhang Xian Chen Ning Wang Palak Shah Fang Deng Chen Zhao Nisha Geng Melissa Li Sahitya K.Denduluri Rex C.Haydon Hue H.Luu Russell R.Reid Tong-Chuan He 2015Genes & Diseases2015,2,1:5
13Genetic Diversity, Antimicrobial Resistance, and Virulence Genes of Aeromonas Isolates from Clinical Patients, Tap Water Systems, and Food显示文摘Objective This study aimed to evaluate the genetic diversity,virulence,and antimicrobial resistance of Aeromonas isolates from clinical patients,tap water systems,and food.Methods Ninety Aeromonas isolates were obtained from Ma’anshan,Anhui province,China,and subjected to multi-locus sequence typing(MLST)with six housekeeping genes.Their taxonomy was investigated using concatenated gyr B-cpn60 sequences,while their resistance to 12 antibiotics was evaluated.Ten putative virulence factors and several resistance genes were identified by PCR and sequencing.Results The 90 Aeromonas isolates were divided into 84 sequence types,80 of which were novel,indicating high genetic diversity.The Aeromonas isolates were classified into eight different species.PCR assays identified virulence genes in the isolates,with the enterotoxin and hemolysin genes act,aer A,alt,and ast found in 47(52.2%),13(14.4%),22(24.4%),and 12(13.3%)of the isolates,respectively.The majority of the isolates(≥90%)were susceptible to aztreonam,imipenem,cefepime,chloramphenicol,gentamicin,tetracycline,and ciprofloxacin.However,several resistance genes were detected in the isolates,as well as a new mcr-3 variant.Conclusions Sequence type,virulence properties,and antibiotic resistance vary in Aeromonas isolates from clinical patients,tap water systems,and food.MENG Shuang WANG Yong Lu LIU Chen Geng YANG Jing YUAN Min BAI Xiang Ning JIN Dong LIANG Jun Rong CUI Zhi Gang LI Juan 2020Biomedical and Environmental Sciences2020,33,6:5
14Threshold effects of tourism agglomeration on the green innovation efficiency of China's tourism industry显示文摘Industrial agglomeration is an essential and effective way to integrate resources and elements. Its impact on the green innovation efficiency of the tourism industry is a process of continuous development and dynamic change. This paper, based on the provincial panel data from 2006 to 2015, uses super SBM model to measure the green innovation efficiency of China's tourism industry.The threshold regression model is employed to conduct empirical tests of the nonlinear threshold effect of agglomeration on the green innovation efficiency of tourism industry. The inter-provincial differences of various threshold effects and their possible causes are analyzed. Results indicate that the green innovation efficiency of China's tourism industry is generally growing, while regional disparity is significant with a gradient decrease along the eastern-central-western regions. There is an obvious positive nonlinear relationship between tourism agglomeration and green innovation efficiency. It is also found that with the increase of agglomeration, its influence is at a high level. As the level of agglomeration crosses the first threshold, its impact is at a low level, and when it crosses the second threshold, the impact of tourism agglomeration is at an intermediate level. 15Agricultural Land Suitability of Production Space in the Taihang Mountains, China显示文摘The contradiction between the shortage of land for agriculture due to rapid expansion of industrialization and urbanization and increasing population pressure is projected to impose great threats to future food security.Agricultural land suitability evaluation is an effective approach to improve the utilization of land resources for crop production and thus enhance the capacity of food provision.In this study, we evaluated the land suitability for agriculture of the production space in the Taihang Mountains by three steps: establishing indicator system, determining weights for indicators, and constructing a fuzzy matter-element model to assess the grades of suitability.Results showed that the land suitability had a significant linear correlation with potential crop yields, indicating our evaluation was effective to predict crop production.The spatial pattern of land suitability for crop production demonstrated that land with higher suitability was generally located in piedmont plains and basins, while land with lower suitability was mostly situated in mountainous areas.The area of highly, moderately, marginally suitable and unsuitable land for agriculture was 32.13%, 28.58%, 37.49% and 1.80% of the production space, respectively.However, the correlation degree analysis indicated that the requirements of these four suitability grades were currently not satisfied but could be potentially fulfilled.In terms of indicator weights, soil properties were much more important than topography and location conditions to influence the grades of suitability.Among all indicators, slope, soil organic matter, soil texture and soil depth were the most influential factors, so slope farming prevention and organic fertilization were most likely to improve land suitability for agriculture.Compared the outputs of our land suitability evaluation model with the distribution of the existing croplands, we found that about 66.52% of marginally suitable and 54.55% of unsuitable land for agriculture were currently used for croplands.Therefore, de-farming policy should be implemented in areas of these two suitability grades.In contrast, cropland expansion was encouraged in the land that was highly or moderately suitable for agriculture.Our evaluation of agricultural land suitability is beneficial for future land use planning and decision-making in the Taihang Mountains.GENG Shoubao SHI Peili ZONG Ning ZHU Wanrui 2019Chinese Geographical Science2019,29,6:4
16First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 18Numerical simulation of flow focusing pattern and morphological changes in two-phase flow inside nozzle显示文摘The flow focusing nozzle is a new type of nozzle that performs effective atomization of the discrete phase by means of high-speed motion of the continuous phase.The flow pattern and its morphological changes have a significant effect on the atomization, but the influence of different parameters on the morphological change of the flow pattern remains unclear.The flow focusing pattern and morphological changes in the two-phase flow inside the nozzle were simulated numerically, based on the volume of fluid method.The results demonstrate that the ratio of the nozzle-to-capillary distance and capillary diameter, the gas–liquid velocity ratio, and capillary diameter have significant effects on the flow pattern.When the ratio of the nozzle-to-capillary distance H and capillary diameter D increases, or the capillary diameter D increases, the flow pattern tends to transform into a laminar form; however, when the gas–liquid velocity ratio V increases, the flow pattern tends to transform into a turbulence form.Furthermore, we define the cone-shaped expansion rate, cone-shaped focusing rate,and cone angle in order to study the morphological changes in the cone shape inside the nozzle.The results indicate that the morphological change of the cone shape and flow pattern transformation is interrelated.When the cone shape tends to be unstable, the flow pattern changes towards flow blurring, whereas, a stable cone indicates that the flow tends to exhibit a droplet pattern.Jin Zhao Zhi Ning Ming Lü Geng Wang 2019Chinese Journal of Chemical Engineering2019,27,1:2
19Temperature dependence of densities of Sb and Bi melts显示文摘The densities of Sb and Bi melts were investigated by an improved Archimedean method. The results show that the density of the Sb melt decreases linearly with increasing temperature,but the density of the Bi melt firstly increases and then decreases as the temperature increases. There is a maximum density value of 10.002 g/cm3 at 310 ℃ ,about 39℃ above the melting point. The temperature depend-ence of the Sb melt is well fitted with the expression ρ =6.8590-5.8105×10-4T,and that of the Bi melt is fitted with ρ =10.3312-1.18×10-3T. 20Highly chemoselective lipase from Candida sp.99-125 catalyzed ring-opening polymerization for direct synthesis of thiol-terminated poly(e-caprolactone)显示文摘Lipase from Candida sp.99-125 catalyzed ring-opening polymerization of e-caprolactone in the presence of 6-mercapto-1-hexanol was presented as a new metal-free approach for direct synthesis of welldefined thiol-terminated poly(e-caprolactone).Remarkably,high chemoselectivity of lipase from Candida sp.99-125 toward hydroxyl and thiol was exhibited and quantitative thiol fidelity over 90% was achieved.The tedious protecting/deprotecting steps for thiol and metal residue were avoided.The polymerizations with around 70% monomer conversion were conducted in bulk and toluene at relative low temperature of 40 8C.Number–average molecular weight of resulted polymers ranged from 3000 to4700 Da by changing the feed ratio between monomer and initiator.The structures of obtained thiolterminated poly(e-caprolactone) were demonstrated by combining NMR and SEC analyses.Ning Zhu Zi-Long Zhang Wei He Xiao-Cheng Geng Zheng Fang Xin Li Zhen-Jiang Li Kai Guo 2015Chinese Chemical Letters2015,26,3:2
