1Artificial antigen-presenting cells plus IL-15 and IL-21 efficiently induce melanoma-specific cytotoxic CD8^+CD28^+ T lymphocyte responses显示文摘Objective:To develop a novel artificial antigen-presenting system for efficiently inducing melanoma-specific CD8+CD28+ cytotoxic T lymphocyte(CTL) responses.Methods:Cell-sized Dynabeads? M-450 Epoxy beads coated with H-2Kb:Ig-TRP2(181111K and anti-CD28 antibody were used as artificial antigen-presenting cells(aAPCs) lo induce melanoma-specific CD8*CD28’ CTL responses with the help of IL-2I and IL-I5.Dimer staining,proliferation,ELISPOT,and cytotoxicity experiments were conducted to evaluate the frequency and activity of induced CTLs.Results:Dimer staining demonstrated that the new artificial antigen-presenting system efficiently induced melanoma TRP2-specific CD8CD28' CTLs.Proliferation and ELISPOT assays indicated that the induced CTLs rapidly proliferate and produce increased IFN- y under the slimulalion of H-2K:Ig-TRP2-aAPCs,TL-15,and IL-21.In addition,cytoloxicily experiments showed lhat induced CTLs have specific killing activity of target cells.Conclusions:The new artificial antigen-presenting system including aAPCs plus IL-21 and IL-15 can induce a large number of antigen-specific CD8+CD28+ CTLs against the melanoma.Our study provides evidence for a novel adoptive immunotherapy against tumors.Xia Yu Jian He Sodaly Mongkhoune Yi Peng Yuan Xie Jing Su Su-Fang Zhou Xiao-Xun Xie Guo-Rong Luo Yuan Fang Xi Li Nuo Zhou Yong-Xiang Zhao Xiao-Ling Lu 2013Asian Pacific Journal of Tropical Medicine2013,6,6:3
2On-demand assembly of polymeric nanoparticles for longer-blood-circulation and disassembly in tumor for boosting sonodynamic therapy显示文摘Sonodynamic therapy (SDT) is one of the promising strategies for tumor therapy, but its application is usually hindered by fast clearance in blood-circulation, abnormal tumor microenvironment, and inefficient generation of reactive oxygen species. To solve these problems, we proposed an on-demand assembly-disassembly strategy, where the assembly is favorable for longer-blood-circulation and then the disassembly in tumor is favorable for boosting SDT. Hematoporphyrin monomethyl ether (HMME) as the model of organic sonosensitizers were conjugated with hyaluronic acid (HA). Then HA-HMME was mixed with catalase (CAT) and assembled into polymeric nanoparticles (CAT@HA-HMME NPs) with size of ~80 nm. CAT@HA-HMME NPs exhibit good biocompatibility and a longer blood half-time (t1/2 = 4.17 h) which is obviously longer than that (~0.82 h) of HMME molecules. After HA receptor-mediated endocytosis of cancer cells, CAT@HA-HMME NPs can be cleaved by endogenous hyaluronidase, resulting in the on-demand disassembly in tumor to release HA-HMME molecules and CAT. The CAT catalyzes the endogenous H_(2)O_(2) into O_(2) to relieve the hypoxic microenvironment, and the released HA-HMME exhibits a higher ROS generation ability, greatly boosting SDT for the inhibition of tumor growth. 3Optimized Bio-inspired Micro-pillar Dry Adhesive and Its Application for an Unmanned Aerial Vehicle Adhering on and Detaching from a Ceiling显示文摘Various bio-inspired dry adhesive materials have been investigated.In this work,lithography and silicon deep etching method were used to fabricate bio-inspired micro-pillar dry adhesive materials of which the parameters,such as side length,Height to Side length Aspect Ratio(HSAR),and inter-pillar Distance to Side length Aspect Ratio(DSAR)were comprehensively studied to obtain a dry adhesive with high adhesion.An orthogonal array method was designed and a series of fabrication experiments were conducted to identify optimum parameters,which resulted in a high normal adhesion of 2.98 N·cm^-2 and a shear adhesion of 9.59 N·cm^-2.An adhesion and desorption device was further designed on basis of the optimum dry adhesive,which enables an Unmanned Aerial Vehicle(UAV)to successfully adhere on and detach from a ceiling.This would allow an UAV to stay aloft for prolonged time,which could be invaluable to many applications,such as energy conservation and aerial detection.Qingsong He Xianrui Xu Zhiwei Yu Kai Huo Zhaoyang Wang Nuo Chen Xuean Sun Gui Yin Peile Du Yang Li Zhendong Dai 2020Journal of Bionic Engineering2020,17,1:2
4Natural astaxanthin enhanced antioxidant capacity and improved semen quality through the MAPK/Nrf2 pathway in aging layer breeder roosters显示文摘Background:Natural astaxanthin(ASTA)has strong antioxidant properties and has been widely used as a health product to improve human health.However,the effects of ASTA on the reproductive performance of aging roosters have been poorly studied.We aimed to investigate the effects of dietary ASTA on semen quality and antioxidant capacity in aging roosters and to explore the potential mechanism of semen quality change via antioxidation defense system.Methods:In the present study,9653-week-old Jinghong No.1 layer breeder roosters were fed a corn-soybean meal basal diet containing 0,25,50,or 100 mg/kg ASTA for 6 weeks.Results:Semen quality in the ASTA groups remarkably improved than that in the control group,and antioxidant activities,the abilities to scavenge hydroxyl radicals and superoxide anions,increased gradually with ASTA addition(P<0.05).In addition,the mRNA levels of antioxidant enzymes as well as the mRNA and protein levels of the mitogen-activated protein kinase(MAPK)and nuclear factor-erythroid 2-related factor 2(Nrf2)were markedly increased in the 50-100 mg/kg ASTA group(P<0.05).Conclusions:Collectively,these results demonstrate that dietary ASTA may improve semen quality by increasing antioxidant enzyme activities and the ability to scavenge hydroxyl radicals,which may be related to upregulation of the MAPK/Nrf2 pathway.Shan Gao Nuo Heng Fang Liu Yong Guo Yu Chen Liang Wang Hemin Ni Xihui Sheng Xiangguo Wang Kai Xing Longfei Xiao Xiaolong Qi 2022Journal of Animal Science and Biotechnology2022,13,2:2

13Effects of Leptocybe invasa Damage on Chemical Components of Eucalyptus显示文摘[Objective] The aim was to illustrate the relation between Leptocybe invasa and eucalyptus, so as to provide the basis for scientific control of L. invasa. [Method] The chemical components of eucalyptus were extracted by solid phase micro-extraction (SPME) technique and ethanol extraction method, and detected by gas chromatograph-mass spectrometer. The relative contents of various components were determined by area normalization. [Result] When eucalyptus was damaged by L. invasa, not only some substances disappeared, but also new substances were induced. Among 4 induced volatiles, the content of terpinolene was the highest of 37.66%. 16Beauvericin counteracted multi-drug resistant Candida albicans by blocking ABC transporters显示文摘Multi-drug resistance of pathogenic microorganisms is becoming a serious threat,particularly to immunocompromised populations.The high mortality of systematic fungal infections necessitates novel antifungal drugs and therapies.Unfortunately,with traditional drug discovery approaches,only echinocandins was approved by FDA as a new class of antifungals in the past two decades.Drug efflux is one of the major contributors to multi-drug resistance,the modulator of drug efflux pumps is considered as one of the keys to conquer multi-drug resistance.In this study,we combined structure-based virtual screening and whole-cell based mechanism study,identified a natural product,beauvericin(BEA)as a drug efflux pump modulator,which can reverse the multi-drug resistant phenotype of Candida albicans by specifically blocking the ATP-binding cassette(ABC)transporters;meantime,BEA alone has fungicidal activity in vitro by elevating intracellular calcium and reactive oxygen species(ROS).It was further demonstrated by histopathological study that BEA synergizes with a sub-therapeutic dose of ketoconazole(KTC)and could cure the murine model of disseminated candidiasis.Toxicity evaluation of BEA,including acute toxicity test,Ames test,and hERG(human ether-a-go-go-related gene)test promised that BEA can be harnessed for treatment of candidiasis,especially the candidiasis caused by ABC overexpressed multi-drug resistant C.albicans.Yaojun Tong Mei Liu Yu Zhang Xueting Liu Ren Huang Fuhang Song Huanqin Dai Biao Ren Nuo Sun Gang Pei Jiang Bian Xin-Ming Jia Guanghua Huang Xuyu Zhou Shaojie Li Buchang Zhang Takashi Fukuda Hiroshi Tomoda SatoshiOmura Richard DCannon Richard Calderone Lixin Zhang 2016Synthetic and Systems Biotechnology2016,1,3:1

18Synergistic effects of sodium hypochlorite disinfection and iron-oxidizing bacteria on early corrosion in cast iron pipes显示文摘Corrosion in drinking water distribution systems(DWDSs)may lead to pipe failures and water quality deterioration;biocorrosion is the most common type.Chlorine disinfectants are widely used in DWDSs to inhibit microorganism growth,but these also promote electrochemical corrosion to a certain extent.This study explored the independent and synergistic effects of chlorine and microorganisms on pipeline corrosion.Sodium hypochlorite(NaOCl)at different concentrations(0,0.25,0.50,and 0.75 mg/L)and iron-oxidizing bacteria(IOB)were added to the reaction system,and a biofilm annular reactor(BAR)was employed to simulate operational water supply pipes and explain the composite effects.The degree of corrosion became severe with increasing NaOCl dosage.IOB accelerated the corrosion rate at an early stage,after which the reaction system gradually stabilized.When NaOCl and IOB existed together in the BAR,both synergistic and antagonistic effects occurred during the corrosion process.The AOC content increased due to the addition of NaOCl,which is conducive to bacterial regrowth.However,biofilm on cast iron coupons was greatly influenced by the disinfectant,leading to a decrease in microbial biomass over time.More research is needed to provide guidelines for pipeline corrosion control.Weiying Li Yu Tian Jiping Chen Xinmin Wang Yu Zhou Nuo Shi 2022Frontiers of Environmental Science & Engineering2022,16,6:0
19Constructing on-demand single/multi-color transitioning fabrics with photocatalysis/photothermal-armed deficient semiconductors显示文摘On-demand color switching systems that utilize synchronized semiconductor-catalyzed reduction and photothermal-accelerated oxidation in liquid/solid are highly appealing.Herein,on-demand single/multi-color switching fabrics have been constructed by using defective SnO_(2):Sb-based color switching systems.SnO_(2):Sb nanocrystals with the suitable doping concentration accord lattices with abundant free electrons,conferring high photocatalytic and photothermal performances.A well-crafted set of dual light-responsive semiconductor-catalyzed systems with rapid color change can be attained via the homogenous mixture of SnO_(2):Sb with suitable redox dyes to produce single-color(RGB(red,green,blue))and multi-color transitioning(purple and green)systems.The illumination of these systems by 450 nm light triggers rapid photocatalytic discoloration,while irradiation by 980 nm light confers the photothermal effect that accelerates recoloration in air.Besides,the inks can be extended to rewritable fabrics by embedding the nanocrystals and redox dyes into hydroxyethyl cellulose(as the polymer matrix)and then coating on hydrophobic cotton fabrics to produce photo-switchable fabrics with excellent single/multi-color response.By exploiting the dual light interactions with the semiconductor-mediated systems,various images/letters can be remotely printed and erased on the rewritable fabrics which show promise for potential applications as information storage media and visual sensors.Importantly,the present rewritable fabric shows good stability and reversibility.The present work provides insights into the development of novel color-switching materials.Daniel K.Macharia Shamima Sarker Meng Liu Zeyulong Wen Nuo Yu Meifang Zhu Zhigang Chen 2024Nano Research2024,17,5:0
20A retrospective study on professor Gui-Qi Xuan’s experience in traditional Chinese medicine treatment on attention-deficit hyperactivity disorder: based on data mining and network pharmacology显示文摘Background:To investigate the clinical medication approach of Professor Guiqi Xuan(Prof.Xuan)in treating pediatric patients with attention-deficit hyperactivity disorder(ADHD)and the potential mechanism of the core herbal prescription.Methods:Following medical record information pretreatment,the Traditional Chinese Medicine(TCM)inheritance computing platform system V3.0 was utilized to analyze the standardized data.The associate rules were summarized to identify the core prescription for treating ADHD.The extracted core herbal prescription’s active compounds and potential targets were used to establish a protein-protein interaction network of active ingredient-disease targets.Cytoscape 3.9.1 software was used to analyze the network’s topological parameters to obtain the key active ingredients and their targets.The Bioconductor data package of R4.0.2 was used to analyze the gene ontology biological functions and Kyoto Encyclopedia of Genes and Genomes pathways of key targets.Results:Two hundred and twenty-seven entries derived from TCM record information were selected.Through data mining,it was found that 62.5%of pediatric patients had short-tempered behavior,nearly half had sleep problems,and 30%-40%had picky eating and polyphagia issues.The highest-frequency syndrome type was kidney deficiency and liver hyperactivity.Deficiency,fire,phlegm,and dyspeptic food were the main pathological factors for ADHD.Prof.Xuan’s treatment of ADHD mainly focused on replenishing kidney essence and subduing Yang(active,external,ascending,warm,bright,functional and excited pertain to Yang).The core herbal prescription for ADHD included Yuan-zhi,Yi-zhi,Gui-jia,Bai-shao,Long-chi,Ci-shi,Shi-chang-pu,Yu-jin,Fu-shen,and Huang-jing.The protein-protein interaction network showed that MAOA,ADRB2,FOS,MAOB,and SLC6A3 were the five key targets essential in treating ADHD with core herbal prescriptions.The gene ontology biological function of crucial targets mainly involved G protein-coupled amine receptor activity,catecholamine binding,and neurotransmitter transmembrane transporter activity.Analysis of Kyoto Encyclopedia of Genes and Genomes pathways showed that the dopaminergic synapse signaling and neuroactive ligand-receptor interaction pathways were significantly enriched and may be the primary routes for the main treatment of ADHD.Conclusion:Prof.Xuan’s treatment of ADHD has achieved satisfactory clinical effects by supplementing the kidney,replenishing the essence,opening the orifices,nourishing the Yin(static,internal,descending,cold,dim,organic,depressed and pertain to Yin),and subduing the Yang.The major prescription predominantly affects catecholamine binding,neuroactive ligand-receptor interaction,G protein-coupled amine receptor function,and signaling pathways for dopaminergic synapses.Our findings showed that the methodology and software used in this research could explore and analyze the mechanism behind Prof.Xuan’s clinical medication rule for treating ADHD in children.Jue Hu Dan-Fei Chen Fang-Fang Li Nuo Chen Chun-Lu Ye Ke-Pin Yu Xiao-Bo Xuan Gui-Qi Xuan Jian Chen 2023Life Research2023,6,3:0
