| 1 | QTG-Seq Accelerates QTL Fine Mapping through QTL Partitioning and Whole-Genome Sequencing of Bulked Segregant Samples显示文摘Deciphering the genetic mechanisms underlying agronomic traits is of great importance for crop improvement. Most of these traits are controlled by multiple quantitative trait loci (QTLs), and identifying the underlying genes by conventional QTL fine-mapping is time-consuming and labor-intensive. Here, we devised a new method, named quantitative trait gene sequencing (QTG-seq), to accelerate QTL fine-mapping. QTGseq combines QTL partitioning to convert a quantitative trait into a near-qualitative trait, sequencing of bulked segregant pools from a large segregating population, and the use of a robust new algorithm for identifying candidate genes. Using QTG-seq, we fine-mapped a plant-height QTL in maize (Zea mays L.), qPH7, to a 300-kb genomic interval and verified that a gene encoding an NF-YC transcription factor was the functional gene. Functional analysis suggested that qPH7-encoding protein might influence plant height by interacting with a CO-like protein and an AP2 domain-containing protein. Selection footprint ana卜 ysis indicated that qPH7 was subject to strong selection during maize improvement. | 2 | How does the motor relearning program improve neurological function of brain ischemia monkeys?显示文摘The motor relearning program can significantly improve various functional disturbance induced by ischemic cerebrovascular diseases. However, its mechanism of action remains poorly understood. In injured brain tissues, glial fibrillary acidic protein and neurofilament protein changes can reflect the condition of injured neurons and astrocytes, while vascular endothelial growth factor and basic fibroblast growth factor changes can indicate angiogenesis. In the present study, we induced ischemic brain injury in the rhesus macaque by electrocoagulation of the M1 segment of the right middle cerebral artery. The motor relearning program was conducted for 60 days from the third day after model establishment. Immunohistochemistry and single-photon emission CT showed that the numbers of glial fibrillary acidic protein-, neurofilament protein-, vascular endothelial growth factor- and basic fibroblast growth factor-positive cells were significantly increased in the infarcted side compared with the contralateral hemisphere following the motor relearning program. Moreover, cerebral blood flow in the infarcted side was significantly improved. The clinical rating scale for stroke was used to assess neurological function changes in the rhesus macaque following the motor relearning program. Results showed that motor function was improved, and problems with consciousness, self-care ability and balance function were significantly ameliorated. | 3 | Stability influence factors analysis and construction of a deep beam anchorage structure in roadway roof显示文摘Deep beam anchorage structures based on spatial distribution analysis of the cable prestressed field have been proposed for roadway roof support. Stability and other factors that influence deep beam structures are studied in this paper using mechanical calculations, numerical analysis and field measurements. A mechanical model of deep beam structure subjected to multiple loading is established, including analysis of roof support in the return airway of S1203 working face in the Yuwu coal mine, China. The expression of maximum shear stress in the deep beam structure is deduced according to the stress superposition criterion. It is found that the primary factors affecting deep beam structure stability are deep beam thickness, cable pre-tension and cable spacing. The variation of maximum shear stress distribution and prestressed field diffusion effects according to various factors are analyzed using Matlabòand FLAC3D^(TM) software, and practical support parameters of the S1203 return airway roof are determined.According to the observations of rock pressure, there is no evidence of roof separation, and the maximum values of roof subsidence and convergence of wall rock are 72 and 48 mm, respectively. | 6 | A Model-Embedded Trend Test with Incorporating Hardy-Weinberg Equilibrium Information显示文摘The genetic models are greatly important in the analysis of genetic epidemiologic studies and many of the studies are conducted using the trend test under the additive model. However, for many complex diseases and traits, the underlying genetic model for a genetic locus is usually uncertain. So a robust test free of genetic model is appropriate. In this paper, the authors propose a model-embedded trend test by incorporating Hardy-Weinberg equilibrium information and obtain the explicit formula to calculate its statistical significance. Extensive simulation studies show the proposed test is more robust than the existing procedures. | 8 | FBXL19-AS1 promotes cell proliferation and inhibits cell apoptosis via miR-876-5p/FOXM1 axis in breast cancer显示文摘As the most common cancer and one of the leading causes of cancer-associated mortality,breast cancer continues to need more key molecules to regulate its progression.F-box and leucine-rich repeat protein 19 antisense RNA 1(known as FBXL19-AS1)is a long non-coding RNA(lncRNA)which has been reported as an oncogene in several types of human cancers.However,the specific downstream targets of FBXL19-AS1 remain unknown.In this study,we set out to find more reliable downstream molecules of FBXL19-AS1 in breast cancer.FBXL19-AS1 was expressed at a high level in breast cancer cells.Loss-of-function experiments revealed that silencing FBXL19-AS1 could impair cell proliferation and induce cell apoptosis in breast cancer.In addition,the location of FBXL19-AS1 in the cytoplasm was detected by fluorescent in situ hybridization assay,while FBXL19-AS1 regulated the expression of Forkhead box M1(FOXM1)by directly absorbing miR-876-5p.Through rescue assays,it was observed that FOXM1 overexpression recovered the inhibited tumor growth caused by FBXL19-AS1 downregulation.We affirmed the function of FBXL19-AS1 in breast cancer and described the mechanism of the FBXL19-AS1/miR-876-5p/FOXM1 axis.The current work presents the molecular mechanism which underlies FBXL19-AS1 in breast cancer and suggests a comprehensive,feasible FBXL19-AS1-mediated therapeutic approach for treating breast cancer. | Guolei Dong Teng Pan Dongdong Zhou Chunyan Li Jingjing Liu Jin Zhang | 2019 | Acta Biochimica et Biophysica Sinica2019,51,11: | 1 |

The pressure distribution is calculated by the air-bearing model at the head/disk interface with respect to the helium content and the pressure obtained is then input to the molecular dynamics model to understand the lubricant transfer mechanism. Finally, the effects of pressure at the boundary condition and disk velocity on lubricant transfer are discussed in relation to the helium fraction within the air-helium gas mixtures. Results show there is a decrease in the pressure difference with an increase in the helium percentage, which leads to a decrease in the volume of the lubricant transferred. | 15 | Expert consensus on odontogenic maxillary sinusitis multi-disciplinary treatment显示文摘Odontogenic maxillary sinusitis (OMS) is a subtype of maxillary sinusitis (MS). It is actually inflammation of the maxillary sinus that secondary to adjacent infectious maxillary dental lesion. Due to the lack of unique clinical features, OMS is difficult to distinguish from other types of rhinosinusitis. Besides, the characteristic infectious pathogeny of OMS makes it is resistant to conventional therapies of rhinosinusitis. Its current diagnosis and treatment are thus facing great difficulties. The multi-disciplinary cooperation between otolaryngologists and dentists is absolutely urgent to settle these questions and to acquire standardized diagnostic and treatment regimen for OMS. However, this disease has actually received little attention and has been underrepresented by relatively low publication volume and quality. Based on systematically reviewed literature and practical experiences of expert members, our consensus focuses on characteristics, symptoms, classification and diagnosis of OMS, and further put forward multidisciplinary treatment decisions for OMS, as well as the common treatment complications and relative managements. | 16 | Grain refinement and strength enhancement in Mg wrought alloys: A review显示文摘Low absolute strength becomes one major obstacle for the wider applications of low/no rare-earth(RE) containing Mg alloys. This review firstly demonstrates the importance of grain refinement in improving strength of Mg alloys by comprehensively comparing with other strategy, e.g., precipitation strengthening. Dynamic recrystallization(DRX) plays a crucial role in refining grain size of Mg wrought alloys.Therefore, secondly, the DRX models, grain nucleation mechanisms and the related grain refinement abilities in Mg alloys are summarized,including phase boundary, twin boundary and general boundary induced recrystallization. Thirdly, the newly developed low-RE containing Mg alloy, e.g., Mg-Ce, Mg-Nd and Mg-Sm based alloys, and the RE-free Mg alloys, e.g., Mg-Al, Mg-Zn, Mg-Sn and Mg-Ca based alloy,are reviewed, with the focus on enhancing the mechanical properties mainly via the grain refinement strategy. At the last section, the perspectives and outstanding issues concerning high-performance Mg wrought alloys are also proposed. | 18 | Genetic dissection of N use efficiency using maize inbred lines and testcrosses显示文摘Although the use of heterosis in maize breeding has increased crop productivity,the genetic causes underlying heterosis for nitrogen(N) use efficiency(NUE) have been insufficiently investigated.In this study,five N-response traits and five low-N-tolerance traits were investigated using two inbred line populations(ILs) consisting of recombinant inbred lines(RIL) and advanced backcross(ABL) populations,derived from crossing Ye478 with Wu312.Both populations were crossed with P178 to construct two testcross populations.IL populations,their testcross populations,and the midparent heterosis(MPH)for NUE were investigated.Kernel weight,kernel number,and kernel number per row were sensitive to N level and ILs showed higher N response than did the testcross populations.Based on a highdensity linkage map,138 quantitative trait loci(QTL) were mapped,each explaining 5.6%–38.8% of genetic variation.There were 52,34 and 52 QTL for IL populations,MPH,and testcross populations,respectively.The finding that 7.6% of QTL were common to the ILs and their testcross populations and that 11.7% were common to the MPH and testcross population indicated that heterosis for NUE traits was regulated by non-additive and non-dominant loci.A QTL on chromosome 5 explained 27% of genetic variation in all of the traits and Gln1-3 was identified as a candidate gene for this QTL.Genome-wide prediction of NUE traits in the testcross populations showed 14%–51% accuracy.Our results may be useful for clarifying the genetic basis of heterosis for NUE traits and the candidate gene may be used for genetic improvement of maize NUE. | Xiaoyang Liu Kunhui He Farhan Ali Dongdong Li Hongguang Cai Hongwei Zhang Lixing Yuan Wenxin Liu Guohua Mi Fanjun Chen Qingchun Pan | 2023 | The Crop Journal2023,11,4: | 0 |
