2The spatio-temporal variation of wintertime subtidal currents in the western Taiwan Strait显示文摘A new data set of observations by six cruises of ship-mounted acoustic doppler current profiler(SADCP) and three 40 d long bottom-mounted ADCPs(BADCPs) is employed to reveal the spatiotemporal variability of tidal and subtidal currents in the western Taiwan Strait(TWS) during winter season. The results confirm the existence of intense cotidal lines for M_2 tidal current, which is located north of 25°N. In this case, no existence of an amphidromic point can be identified. It is also revealed that the counter-wind current(CWC) can extend through the whole western TWS and even occupy the entire water column during winter monsoon relaxation. However,this CWC is observed to be thoroughly overwhelmed by the downwind China coastal current(CCC) during the two big monsoon bloom events in the winter of 2007, and the CCC consequently extends southward throughout the western TWS instead. Most importantly, the variation of the spatial extent for the CWC and the CCC in the western TWS is found to be well explained by the first two modes of the vector empirical orthogonal function(VEOF) analysis, that is, it is mainly controlled by a wind-driven quasi barotropic current as the first mode and slightly modulated by a relatively weak background current with a first-order baroclinic structure as the second mode.SHEN Junqiang QIU Yun GUO Xiaogang PAN Aijun ZHOU Xiwu 2017Acta Oceanologica Sinica2017,36,11:5
3Interface flux reconstruction method based on optimized weight essentially non-oscillatory scheme显示文摘Aimed at the computational aeroacoustics multi-scale problem of complex configurations discretized with multi-size mesh, the flux reconstruction method based on modified Weight Essentially Non-Oscillatory(WENO) scheme is proposed at the interfaces of multi-block grids.With the idea of Dispersion-Relation-Preserving(DRP) scheme, different weight coefficients are obtained by optimization, so that it is in WENO schemes with various characteristics of dispersion and dissipation. On the basis, hybrid flux vector splitting method is utilized to intelligently judge the amplitude of the gap between grid interfaces. After the simulation and analysis of 1D convection equation with different initial conditions, modified WENO scheme is proved to be able to independently distinguish the gap amplitude and generate corresponding dissipation according to the grid resolution. Using the idea of flux reconstruction at grid interfaces, modified WENO scheme with increasing dissipation is applied at grid points, while DRP scheme with low dispersion and dissipation is applied at the inner part of grids. Moreover, Gauss impulse spread and periodic point sound source flow among three cylinders with multi-scale grids are carried out. 4Site-Specifc Gene Targeting Using Transcription Activator-Like Effector(TALE)-Based Nuclease in Brassica oleracea显示文摘Site-specifc recognition modules with DNA nuclease have tremendous potential as molecular tools for genome targeting.The type III transcription activator-like effectors(TALEs)contain a DNA binding domain consisting of tandem repeats that can be engineered to bind user-defned specifc DNA sequences.We demonstrated that customized TALE-based nucleases(TALENs),constructed using a method called'unit assembly',specifcally target the endogenous FRIGIDA gene in Brassica oleracea L.var.capitata L.The results indicate that the TALENs bound to the target site and cleaved double-strand DNA in vitro and in vivo,whereas the effector binding elements have a 23 bp spacer.The T7 endonuclease I assay and sequencing data show that TALENs made double-strand breaks,which were repaired by a non-homologous end-joining pathway within the target sequence.These data show the feasibility of applying customized TALENs to target and modify the genome with deletions in those organisms that are still in lacking gene target methods to provide germplasms in breeding improvement.Zijian Sun Nianzu Li Guodong Huang Junqiang Xu Yu Pan Zhimin Wang Qinglin Tang Ming Song Xiaojia Wang 2013Journal of Integrative Plant Biology2013,55,11:3
5A GPU-based numerical model coupling hydrodynamical and morphological processes显示文摘Sediment transport simulations are important in practical engineering.In this study,a graphics processing unit(GPU)-based numerical model coupling hydrodynamical and morphological processes was developed to simulate water flow,sediment transport,and morphological changes.Aiming at accurately predicting the sediment transport and sediment scouring processes,the model resolved the realistic features of sediment transport and used a GPU-based parallel computing technique to the accelerate calculation.This model was created in the framework of a Godunov-type finite volume scheme to solve the shallow water equations(SWEs).The SWEs were discretized into algebraic equations by the finite volume method.The fluxes of mass and momentum were computed by the Harten,Lax,and van Leer Contact(HLLC) approximate Riemann solver,and the friction source terms were calculated by the proposed a splitting point-implicit method.These values were evaluated using a novel 2 D edge-based MUSCL scheme.The code was programmed using C++and CUDA,which could run on GPUs to substantially accelerate the computation.The aim of the work was to develop a GPU-based numerical model to simulate hydrodynamical and morphological processes.The novelty is the application of the GPU techniques in the numerical model,making it possible to simulate the sediment transport and bed evolution in a high-resolution but efficient manner.The model was applied to two cases to evaluate bed evolution and the effects of the morphological changes on the flood patterns with high resolution.This indicated that the GPU-based high-resolution hydro-geomorphological model was capable of reproducing morphological processes.The computational times for this test case on the GPU and CPU were298.1 and 4531.2 s,respectively,indicating that the GPU could accelerate the computation 15.2 times.Compared with the traditional CPU high-grid resolution,the proposed GPU-based high-resolution numerical model improved the reconstruction speed more than 2.0-12.83 times for different grid resolutions while remaining computationally efficient.Jingming Hou Yongde Kang Chunhong Hu Yu Tong Baozhu Pan Junqiang Xia 2020International Journal of Sediment Research2020,35,4:3
6Glassy metallic plastics显示文摘This paper reports a class of bulk metallic glass including Ce-, LaCe-, CaLi-, Yb-, and Sr-based metallic glasses, which are regarded as glassy metallic plastics because they combine some unique properties of both plastics and metallic alloys. These glassy metallic plastics have very low glass transition temperature (Tg^25oC to 150oC) and low Young’s modulus (~20 GPa to 35 GPa). Similar to glassy plastics, these metallic plastics show excellent plastic-like deformability on macro-, micro- and even nano-scale in their supercooled liquid range and can be processed, such as elongated, compressed, bent, and imprinted at low temperatures, in hot water for instance. Under ambient conditions, they display such metallic properties as high thermal and electric conductivities and excellent mechanical properties and other unique properties. 7Construction of recombinant plasmid pEGFP-C2-L539fs/47 and its expression in HEK293 cells显示文摘Objective:To reconstruct pEGFP-C2-L539fs/47,a HERG nonsense mutant in eukaryotic expression plasmid,and observe the fusion protein expressed in HEK293 cells(human embryo kidney cells).Methods:After double digestion of pcDNA3-L539fs/47 and pEGFP-C2-HERG with sbf I and Eco91 I,the small product fragment,from pcDNA3-L539fs/47,was subcloned into the big fragment of pEGFP-C2-HERG under T4 ligase.pEGFP-C2-L539fs/47 was identified by agarose gel electrophoresis and sequencing.pcDNA3-L539fs/47 and pEGFP-C2-L539fs/47 were transiently transfected into HEK293 cells by Lipofect,respectively.The expression of fusion protein in HEK293 cells was detected through immunofluorescence,laser confocal imaging scanning in vivo,Western blot and PCR.Results:Mutation region cDNA fragment(about 1 kb) and target vector fragment(about 7.2 kb) were ligated after purification and gel recovery.Agarose gel electrophoresis and sequencing successfully demonstrated eukaryotic expression plasmid pEGFP-C2-L539fs/47,constructed approximately 8.2 kb,sequencing consistent with template gene.The transfection efficiency of recombinant plasmid by fluorescence microscopy was more than60%.Western blot analysis detected pcDNA3-L539fs/47 expression of the protein size 60 KD,the expression of pEGFP-C2 fusion protein size of approximately 90 KD.The L539fs/47 gene expression in HEK293 cells was significant by PCR analysis.Confocal laser imaging showed that pEGFP-C2-L539fs/47 protein was successfully expressed in cytoplasm and cytomembrane of HEK293 cells.Conclusion:pEGFP-C2-L539fs/47 containing the HERG gene mutant was successfully constructed by double digestion method and expressed fusion protein in HEK293 cells,which laid a foundation for the further study on L539fs/47.Lue Ying Zhang Aifeng Han Wenqi Li Guoliang Zhang Junbo Gao Jie Pan Junqiang Zhang Yong Sun Chaofeng 2012Journal of Medical Colleges of PLA(China)2012,27,3:2

11Effect of statins use on the prevention of venous thromboembolism: a meta-analysis显示文摘Objective: To estimate the effect of statins use on the prevention of venous thromboembolism (VTE). Methods: We systematically searched MEDLINE (1980-June 2012), EMBASE (1980-June 2012), Google Scholar, Cochrane Library, and ISI Web of Science, manually reviewed references, and contacted experts. Case-control studies and cohort studies that compared any dose of statin with no statin or placebo are included. Data extraction and study quality evaluation were independently conducted in duplicate. Results: 12 studies including four cohort studies and eight case-control studies were identified and eligible for meta-analysis. Upon meta-analysis, statin use was associated with a statistically significant reduction in the odds of developing VTE (OR 0.91, 95% CI 0.86-0.96). Conclusion: This meta-analysis of current and available literature suggests that statins can reduce patient's risk of developing VTE. 12Serum carbohydrate antigen-125 is elevated in patients with chronic atrial fibrillation显示文摘Objective: To determine whether CA-125 is elevated in medically stable patients with chronic atrial fibrillation (AF) compared with patients without AF and to examine whether levels of CA-125 are associated with demographic and clinical variables in a sample of patients under study. Methods: 55 patients with chronic AF and 58 control patients in sinus rhythm were included into the study. Patients with acute heart failure (HF), chronic inflammatory or neoplastic disease were excluded from the study. Circulating levels of CA-125 were assessed; all patients underwent clinical examination, assessment and medical records including demographic data, history of comorbid conditions, current use of cardiac medications, and the results of cardiac tests including electrocardiograms. Results: The mean age of the study sample was 53.2±6.5 years and 48% were men. Patients with sinus rhythm were significantly more likely to have lower heart rates, smaller dimensions of left atrium, and to be treated with aspirin. Coumadin, coumadin and digoxin were more often prescribed in patients with chronic AF. The CA-125 levels were significantly higher in patients with chronic AF than in patients in sinus rhythm (48.5±7.65 U/ml and 28.43±5.48 U/ml, P<0.005). An inverse relation was found between CA-125 levels and left ventricular ejection fraction (LVEF) (r=-0.48, P<0.001). CA-125 was significantly related to the left atrium (LA) diameter, left ventricular end-diastolic diameter (LVEDD), left ventricular end-systolic diameter (LVESD) and brain natriuretic peptide (BNP). There was no significant correlation between CA-125 and age. 13Overoxidized poly(3,4-ethylenedioxythiophene)-gold nanoparticles-graphene-modified electrode for the simultaneous detection of dopamine and uric acid in the presence of ascorbic acid显示文摘An innovative,ternary nanocomposite composed of overoxidized poly(3,4-ethylenedioxythiophene)(OPEDOT),gold nanoparticles(Au NPs),and electrochemically reduced graphene oxide(ERGO)was prepared on a glassy carbon electrode(GCE)(OPEDOT-Au NPs-ERGO/GCE)through homogeneous chemical reactions and heterogeneous electrochemical methods.The morphology,composition,and structure of this nanocomposite were characterized by transmission electron microscopy,scanning electron microscopy,X-ray diffraction,and X-ray photoelectron spectroscopy.The electrochemical properties of the OPEDOT-Au NPs-ERGO/GCE were investigated by cyclic voltammetry using potassium ferricyanide and hexaammineruthenium(III)chloride redox probe systems.This modified electrode shows excellent electro-catalytic activity for dopamine(DA)and uric acid(UA)under physiological p H conditions,but inhibits the oxidation of ascorbic acid(AA).Linear voltammetric responses were obtained when DA concentrations of approximately 4.0-100μM and UA concentrations of approximately 20-100μM were used.The detection limits(S/N=3)for DA and UA were 1.0 and 5.0μM,respectively,under physiological conditions and in the presence of 1.0 m M of AA.This developed method was applied to the simultaneous detection of DA and UA in human urine,where satisfactory recoveries from 96.7%to 105.0%were observed.This work demonstrates that the developed OPEDOT-Au NPs-ERGO ternary nanocomposite,with its excellent ion-selectivity and electro-catalytic activity,is a promising candidate for the simultaneous detection of DA and UA in the presence of AA in physiological and pathological studies.Junqiang Pan Mei Liu Dandan Li Haonan Zheng Dongdong Zhang 2021Journal of Pharmaceutical Analysis2021,11,6:1

17How Well Do CMIP6 and CMIP5 Models Simulate the Climatological Seasonal Variations in Ocean Salinity?显示文摘This paper includes a comprehensive assessment of 40 models from the Coupled Model Intercomparison Project phase 5(CMIP5)and 33 models from the CMIP phase 6(CMIP6)to determine the climatological and seasonal variation of ocean salinity from the surface to 2000 m.The general pattern of the ocean salinity climatology can be simulated by both the CMIP5 and CMIP6 models from the surface to 2000-m depth.However,this study shows an increased fresh bias in the surface and subsurface salinity in the CMIP6 multimodel mean,with a global average of−0.44 g kg^(−1) for the sea surface salinity(SSS)and−0.26 g kg^(−1) for the 0-1000-m averaged salinity(S1000)compared with the CMIP5 multimodel mean(−0.25 g kg^(−1) for the SSS and−0.07 g kg^(−1) for the S1000).In terms of the seasonal variation,both CMIP6 and CMIP5 models show positive(negative)anomalies in the first(second)half of the year in the global average SSS and S1000.The model-simulated variation in SSS is consistent with the observations,but not for S1000,suggesting a substantial uncertainty in simulating and understanding the seasonal variation in subsurface salinity.The CMIP5 and CMIP6 models overestimate the magnitude of the seasonal variation of the SSS in the tropics in the region 20°S-20°N but underestimate the magnitude of the seasonal change in S1000 in the Atlantic and Indian oceans.These assessments show new features of the model errors in simulating ocean salinity and support further studies of the global hydrological cycle.Yuanxin LIU Lijing CHENG Yuying PAN Zhetao TAN John ABRAHAM Bin ZHANG Jiang ZHU Junqiang SONG 2022Advances in Atmospheric Sciences2022,39,10:0
18Construction of a HERG mutant L539fs/47-*558W pEGFP vector and the expression of the fusion protein in HEK293 cells显示文摘Objective:To construct a human ether-a-go-go-related gene(HERG)nonsense mutant L539fs/47-*558W into the autonomously fluorescent,eukaryotic expression vector pEGFP-C2,and to verify expression of the reconstruct in human embryonic kidney-293(HEK293)cells.Methods:The mutational fragment was subcloned into pEGFP-C2-HERG by double digestion of SbfⅠ,Eco91Ⅰand rejoining of T4 ligase.After verification,the recombinant pEGFP-C2-L539fs/47-*558W and pEGFP-C2-HERG were respectively transfected into HEK293 cells for 48 h by the Lipofect method to observe the expression location of the fusion protein by laser confocal imaging scanning in vivo.pcDNA3-L539fs/47-*558W and pcDNA3-HERG were transfected to observe the expression location of the HERG protein by immunofluoresceoce.The mutant protein size was determined by Western blotting.Results:The about 1 kb-sized mutation region cDNA fragment from pcDNA3-L539fs/47-*558W and the about 7.2 kb-sized target vector fragment from pcDNA3-HERG were ligated after purification and gel recovery.pEGFP-C2-L539fs/47*-558W,approximately 8.2 kb,was demonstrated successfully been constructed under agarose gel electrophoresis and further sequencing.Laser confocal imaging showed that pEGFP-C2-HERG was mainly expressed in the membrane,whereas truncated mutant-type HERG in the pEGFP-C2 vector was partially located in the cytoplasm,the others were transported to the cell membrane in living HEK293 cells.The same as the immunofluoresceoce results after transfection of pcDNA3-HERG and pcDNA3-L539fs/47-558W.Wild-type HERG-GFP fusion protein expressed 160 and 180 kDa bands.The mutant and mutant-GFP fusion proteins were 70 and 100 kDa,respectively.Conclusion:pEGFP-C2-L539fs/47-*558W was successfully constructed by double digestion method GFP had no effect on its protein expression and trafficking in HEK293 cells,which laid a foundation for the further study on L539fs/47-*558W.ZHANG Junbo Lü Ying ZHANG Aifeng SUN Chaofeng HAN Wenqi LI Guoliang GAO Jie HUO Jianhua PAN Junqiang ZHOU Xin NIU Xiaolin 2013Journal of Medical Colleges of PLA(China)2013,28,4:0
