1Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 3Dietary exposure to di-isobutyl phthalate increases urinary 5-methyl-2′-deoxycytidine level and affects reproductive function in adult male mice显示文摘Phthalates are a large family of ubiquitous environmental pollutants suspected of being endocrine disruptors. Epidemiological studies have associated phthalate metabolites with decreased reproductive parameters and linked phthalate exposure with the level of urinary 5-methyl-2′-deoxycytidine(5mdC, a product of methylated DNA). In this study, adult male mice were exposed to 450 mg di-isobutyl phthalate(DiBP)/(kg·day) via dietary exposure for 28 days. Mono-isobutyl phthalate(Mi BP, the urinary metabolite) and reproductive function parameters were determined. The levels of 5mdC and 5-hydroxymethyl-2′-deoxycytidine(5hmdC) were measured in urine to evaluate if their contents were also altered by DiBP exposure in this animal model. Results showed that DiBP exposure led to a significant increase in the urinary 5mdC level and significant decreases in sperm concentration and motility in the epididymis, accompanied with reduced testosterone levels and downregulation of the P450 cholesterol side-chain cleavage enzyme(P450scc) gene in the mice testes. Our findings indicated that exposure to DiBP increased the urinary 5mdC levels,which supported our recent epidemiological study about the associations of urinary 5mdC with phthalate exposure in the male human population. 43D-printed controllable gradient pore superwetting structures for high temperature efficient oil-water separation显示文摘Superwetting surfaces have the potential to address oil pollution in water,through their ability to separate the two.However,it remains a great challenge to fabricate stable and efficient separation structures using conventional manufacturing techniques.Furthermore,the materials traditionally used for oil-water separation are not stable at high temperature.Therefore,there is a need to develop stable,customizable structures to improve the performance of oil-water separation devices.In recent years,3D printing technology has developed rapidly,and breakthroughs have been made in the fabrication of complicated ceramic structures using this technology.Here,a ceramic material with a gradient pore structure and superhydrophobic/superoleophilic properties was prepared using 3D printing for high-efficiency oil-water separation.The gradient pore structure developed here can support a flux of up to 25434 L/m^(2)h,which is nearly 40%higher than that an analogous structure with straight pores.At 200℃,the oil-water separation performance was maintained at 97.4%.Furthermore,samples of the material exhibited outstanding mechanical properties,and chemical stability in a variety of harsh environments.This study provides an efficient,simple,and reliable method for manufacturing oil-water separation materials using 3D printing,and may have broader implications for both fundamental research and industrial applications.Zhipeng Jin Hui Mei Yuekai Yan Longkai Pan Hongxia Liu Shanshan Xiao Laifei Cheng 2021Journal of Materiomics2021,7,1:4
5Effects of Mercuric Chloride on Antioxidant System and DNA Integrity of the Crab Charybdis japonica显示文摘Mercury (Hg) is one of the commonly encountered heavy metals, which is widespread in inshore sediments of China. In order to investigate the toxicity of Hg on marine invertebrates, we studied the effects of the divalent mercuricion (Hg2+) (at two final concentrations of 0.0025 and 0.0050 mg L-1, prepared with HgCl2) on metallothionein (MT) content, DNA integrity (DNA strand breaks) and catalase (CAT) in the gills and hepatopancreas, antioxidant enzyme activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx), in the hemolymph, gills and hepatopancreas of the portunid crab Charybdis japonica for an experiment period up to 15 d. The results indicated that MT was significantly induced after 3 d, with a positive correlation with Hg2+ dose and time in the hepatopancreas and a negative correlation with Hg2+ dose and time in the gills. While CAT in the hemolymph was not detected, it increased in the hepatopancreas during the entire experiment; SOD and GPx in the three tissues were stimulated after 12 h, both attained peak value and then reduced during the experimental period. Meanwhile, DNA strand breaks were all induced significantly after 12 h. 6Effects of Heavy Metal Ions (Cu^(2+), Pb^(2+) and Cd^(2+)) on DNA Damage of the Gills, Hemocytes and Hepatopancreas of Marine Crab, Charybdis japonica显示文摘There are rising concerns about the hazardous effects of heavy metals on the environment. In this study, comet assay and DNA alkaline unwinding assay were conducted on the tissues (gills, hepatopancreas, and hemocytes) of Charybdis japonica in order to illustrate genotoxicity of three heavy metal ions (Cu2+, Pb2+, and Cd2+) on the marine crabs C. japonica. The crabs were exposed to Cu2+ (10, 50, and 100 ?g.L?1), Pb2+ (50, 250, and 500 ?g L?1) and Cd2+ (5, 25, and 50 ?g L?1), and the tissues were sampled at days 0.5, 1, 3, 6, 9, and 15. DNA alkaline unwinding assay was used for testing the DNA single strand break in gills and hepatopancreas and comet assay was employed for testing the DNA damage in hemocytes. The results showed that the DNA damage (F-value) of gills in the crabs exposed to the three heavy metals was decreased gradually during the exposure periods and there was a dose-time response relationship in certain time, suggesting that the levels of DNA single strand break in all the experimental groups increased significantly compared to the controls. Changes of F-value in hepatopancreas of the crabs exposed to the three heavy metals were similar to those in gills except that the peak values were found in the 500 ?g L?1 Pb2+ treatment group at day 3 and the 50 ?g L?1 Cd2+ treatment group at day 9. The ranks of DNA damage in gills and hepatopancreas induced by the three heavy metal ions (50 ?g L?1, day 15) were Cd2+ >Pb2+ >Cu2+ and Pb2+ >Cu2+ >Cd2+. The levels of DNA damage in gills were higher than those in hepatopancreas in the same experimental group. 7Effects of water stress at different growth stages on comprehensive fruit quality and yield in different bunches of tomatoes in greenhouses显示文摘The aim of this study was to investigate the effects of tomato quality and yield between different bunches and the differences between the two comprehensive evaluation methods on tomato quality ranking under water stress.Two degrees of water stress including mild water stress(W1)and moderate water stress(W2),and three growth stages that water stress applied including seedling stage(S1),flowering stage(S2)and fruit expanding stage(S3)were tested in this study.The yield and quality of different bunches of tomatoes under water stress during different growth stages were determined as responses,and the comprehensive fruit quality ranking and yield of the second and third bunches were evaluated.The results showed that water stress was important for the improvement of fruit quality,but fruit yield decreased during water stress.The yield of the third tomato bunch decreased from 11.69%(W1S1)to 30.60%(W2S2)compared to control(97.57 t/hm^(2)),and the effects of mild water stress on fruit yield were minimal at the early growth stage.However,the fruit quality in terms of soluble sugar(SS),total soluble solids(TSS),vitamin C(VC),and firmness(F)improved under water stress compared to control.The combined effects of water stress and its application period significantly affected SS and TSS.Water stress significantly improved the content of SS and TSS in the later growth period compared to seedling and flowering stages.Meanwhile,there was a significant difference in tomato quality between the second and third bunches of fruit,especially in the content of SS,organic acid(OA)and lycopene(L).Principal Component Analysis(PCA)and Grey Relational Analysis(GRA)were used to evaluate comprehensive fruit quality,and the best treatment in terms of the fruit quality was W1S3 for both bunches.The rank-sum ratio(RSR)method was used to evaluate fruit quality and yield,the results showed that W1S3 ranked first based on PCA and W1S1 ranked first based on GRA.Water stress enhanced tomato quality but inevitably reduced its yield during each growth stage.The application of mild water stress during the fruit expanding stage(W1S3)was considered to be the best treatment to provide satisfactory fruit quality and yield based on RSR.Shuxue Hao Hongxia Cao Hubing Wang Xiaoyan Pan 2019International Journal of Agricultural and Biological Engineering2019,12,3:3

16A genetic variant in pseudogene E2F3P1 contributes to prognosis of hepatocellular carcinoma显示文摘Certain pseudogenes may regulate their protein-coding cousins by competing for miRNAs and play an active biological role in cancer.However,few studies have focused on the association of genetic variations in pseudogenes with cancer prognosis.We selected six potentially functional single nucleotide polymorphisms(SNPs) in cancerrelated pseudogenes,and performed a case-only study to assess the association between those SNPs and the prognosis of hepatocellular carcinoma(HCC) in 331 HBV-positive HCC patients without surgical treatment.Log-rank test and Cox proportional hazard models were used for survival analysis.We found that the A allele of rs9909601 in E2F3P1was significantly associated with a better prognosis compared with the G allele[adjusted hazard ratio(HR) = 0.69,95%confidence interval(CI) = 0.56-0.86,P = 0.001].Additionally,this protective effect was more predominant for patients without chemotherapy and transcatheter hepatic arterial chemoembolization(TACE) treatment.Interestingly,we also detected a statistically significant multiplicative interaction between genotypes of rs9909601 and chemotherapy or TACE status on HCC survival(P for multiplicative interaction < 0.001).These findings indicate that rs9909601 in the pseudogene E2F3P1 may be a genetic marker for HCC prognosis in Chinese.Yun Pan Chongqi Sun Mingde Huang Yao Liu Fuzhen Qi Li Liu Juan Wen Jibin Liu Kaipeng Xie Hongxia Ma Zhibin Hu Hongbing Shen 2014The Journal of Biomedical Research2014,28,3:1

19Efficient CO_(2)Reduction to Formate on CsPbI_(3) Nanocrystals Wrapped with Reduced Graphene Oxide显示文摘Transformation of greenhouse gas(CO_(2))into valuable chemicals and fuels is a promising route to address the global issues of climate change and the energy crisis.Metal halide perovskite catalysts have shown their potential in promoting CO_(2)reduction reaction(CO_(2)RR),however,their low phase stability has limited their application perspective.Herein,we present a reduced graphene oxide(rGO)wrapped CsPbI_3 perovskite nanocrystal(NC)CO_(2)RR catalyst(CsPbI_3/rGO),demonstrating enhanced stability in the aqueous electrolyte.The CsPbI_3/rGO catalyst exhibited>92%Faradaic efficiency toward formate production at a CO_(2)RR current density of~12.7 mA cm^(-2).Comprehensive characterizations revealed the superior performance of the CsPbI_3/rGO catalyst originated from the synergistic effects between the CsPbI_3 NCs and rGO,i.e.,rGO stabilized theα-CsPbI_3 phase and tuned the charge distribution,thus lowered the energy barrier for the protonation process and the formation of~*HCOO intermediate,which resulted in high CO_(2)RR selectivity toward formate.This work shows a promising strategy to rationally design robust metal halide perovskites for achieving efficient CO_(2)RR toward valuable fuels.Minh Tam Hoang Chen Han Zhipeng Ma Xin Mao Yang Yang Sepideh Sadat Madani Paul Shaw Yongchao Yang Lingyi Peng Cui Ying Toe Jian Pan Rose Amal Aijun Du Tuquabo Tesfamichael Zhaojun Han Hongxia Wang 2023Nano-Micro Letters2023,15,10:1
20ASYMPTOTIC STABILITY OF VISCOUS SHOCK PROFILE FOR NON-CONVEX SYSTEM OF ONE-DIMENSIONAL VISCOELASTIC MATERIALS WITH BOUNDARY EFFECT显示文摘This paper is concerned with the asymptotic behavior of solution to the initial-boundary value problem on the half space R+ for a one-dimensional non-convex system of viscoelastic materials. The initial data has constant state at infinity and the velocity is imposed zero at the boundary x = 0. By virture of the boundary effect, the solution is expected to behave as outgoing viscous shock profile. When the initial data is suitably close to the corresponding outgoing viscous shock profile which is suitably away from the boundary, it is proved that the unique global solution exists in time and tends toward the properly shifted shock profile as the time goes to infinity.
