| 2 | Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual. | Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang | 2020 | Protein & Cell2020,11,4: | 44 |
| 3 | Enhanced debromination of 4-bromophenol by the UV/sulfite process:Efficiency and mechanism显示文摘Halogenated aromatic compounds have attracted increasing concerns due to their toxicity and persistency in the environment, and dehalogenation is one of the promising treatment and detoxification methods. Herein, we systematically studied the debromination efficiency and mechanism of para-bromophenol(4-BP) by a recently developed UV/sulfite process. 4-BP underwent rapid degradation with the kinetics accelerated with the increasing sulfite concentration, pH(6.1–10) and temperature, whereas inhibited by dissolved oxygen and organic solvents. The apparent activation energy was estimated to be 27.8 kJ/mol. The degradation mechanism and pathways of 4-BP were explored by employing N_2O and nitrate as the electron scavengers and liquid chromatography/mass spectrometry to identify the intermediates. 4-BP degradation proceeded via at least two pathways including direct photolysis and hydrated electron-induced debromination. The contributions of both pathways were distinguished by quantifying the quantum yields of 4-BP via direct photolysis and hydrated electron production in the system. 4-BP could be readily completely debrominated with all the substituted Br released as Br-, and the degradation pathways were also proposed. | 7 | Antidepressant-like effects and memory enhancement of a herbal formula in mice exposed to chronic mild stress显示文摘Shen Yuan Gan(SYG)is a Chinese herbal prescription composed of total saponins of Panax ginseng and total oligosaccharide esters of Polygala tenuifolia(2:1).Our previous studies have demonstrated that SYG has antidepressant-like effects in various mouse models of behavioral depression.The present study aimed to test whether SYG affected chronic mild stress(CMS)-induced depression and cognitive impairment in mice.We found that a 5-week CMS schedule induced significant degradation of the coat state,decreased sucrose intake in the sucrose-preference test,and increased the latency to feed in the noveltysuppressed feeding test.All of these CMS-induced changes were ameliorated by SYG(100 and 200mg/kg)and fluoxetine(10 mg/kg).In addition,SYG restored the decreased monoamine neurotransmitter concentrations(serotonin,dopamine,norepinephrine and acetylcholine)induced by CMS in the prefrontal cortex.Interestingly,SYG ameliorated CMS-induced cognitive impairment in the step-through test,and increased the acetylcholine level in the prefrontal cortex.These results suggest that SYG has an antidepressant-like action and enhances cognition by modulating the serotonin,dopamine,norepinephrine,and acetylcholine levels in the prefrontal cortex. | Xiu-Ping Sun Si-Di Li Zhe Shi Teng-Fei Li Rui-Le Pan Qi Chang Chuan Qin Xin-Min Liu | 2013 | Neuroscience Bulletin2013,29,6: | 6 |

| 11 | High thermal stability Cu_(2)O@OZrP micro-nano hybrids for melt-spun excellent antibacterial activity polyester fibers显示文摘Public safety incidents caused by bacterial infections have attracted widespread attention towards antibacterial textiles(fibers,fabrics,etc.).Nevertheless,it is still challenging to efficiently load inorganic nano-antibacterial materials in polymer fibers.In this work,zirconium phosphate(ZrP,layered micro-nano materials)was utilized as a micro-nano carrier.The octadecyl triphenyl phosphonium bromide(OTP)was intercalated between the ZrP sheets by the ion exchange method to improve the carrier-polymer compatibility and the antibacterial performance.Through in-situ chemical reduction,the ultra-small nano-sized cuprous oxide(Cu_(2)O<5 nm)was loaded on the outer surface of ZrP to realize the uniform and stable dispersion of the Cu_(2)O on the carrier and improve the antibacterial performance.The ZrP nanosheets loaded with Cu_(2)O and OTP(Cu_(2)O@OZrP)had excellent antibacterial properties,and the antibacterial rate against E.coli,S.aureus and C.albicans was more than 99%.The intercalation amount of OTP in Cu_(2)O@OZrP can reach 16%,and the thermal stability was excellent and a significant increase in the Zeta potential.Indeed,the decomposition temperature was greater than 350℃,which was suitable for high-temperature melt processing of polymers.Consequently,we prepared PET/Cu_(2)O@OZrP fibers using polyethylene glycol terephthalate(PET),which accounts for 70%of the total chemical fibers,as the fiber matrix.PET/Cu_(2)O@OZrP fibers exhibited excellent mechanical property and antibacterial performance when the content of Cu_(2)O@OZrP was only 0.2%.The antibacterial rate against five types of bacteria including super bacteria(MRSA,VRE)was more than 99%. | Jialiang Zhou Yaping Wang Weinan Pan Hengxue Xiang Peng Li Zhe Zhou Meifang Zhu | 2021 | Journal of Materials Science & Technology2021,,22: | 4 |
| 12 | Delayed improvements in visual memory task performance among chronic schizophrenia patients after high-frequency repetitive transcranial magnetic stimulation显示文摘BACKGROUND Cognitive impairments are core characteristics of schizophrenia,but are largely resistant to current treatments.Several recent studies have shown that highfrequency repetitive transcranial magnetic stimulation(rTMS)of the left dorsolateral prefrontal cortex(DLPFC)can reduce negative symptoms and improve certain cognitive deficits in schizophrenia patients.However,results are inconsistent across studies.AIM To examine if high-frequency rTMS of the DLPFC can improve visual memory deficits in patients with schizophrenia.METHODS Forty-seven chronic schizophrenia patients with severe negative symptoms on stable treatment regimens were randomly assigned to receive active rTMS to the DLPFC(n=25)or sham stimulation(n=22)on weekdays for four consecutive weeks.Patients performed the pattern recognition memory(PRM)task from the Cambridge Neuropsychological Test Automated Battery at baseline,at the end of rTMS treatment(week 4),and 4 wk after rTMS treatment(week 8).Clinical symptoms were also measured at these same time points using the Scale for the Assessment of Negative Symptoms(SANS)and the Positive and Negative Syndrome Scale(PANSS).RESULTS There were no significant differences in PRM performance metrics,SANS total score,SANS subscores,PANSS total score,and PANSS subscores between active and sham rTMS groups at the end of the 4-wk treatment period,but PRM performance metrics(percent correct and number correct)and changes in these metrics from baseline were significantly greater in the active rTMS group at week 8 compared to the sham group(all P<0.05).Active rTMS treatment also significantly reduced SANS score at week 8 compared to sham treatment.Moreover,the improvement in visual memory was correlated with the reduction in negative symptoms at week 8.In contrast,there were no between-group differences in PANSS total score and subscale scores at either week 4 or week 8(all P>0.05).CONCLUSION High-frequency transcranial magnetic stimulation improves visual memory and reduces negative symptoms in schizophrenia,but these effects are delayed,potentially due to the requirement for extensive neuroplastic changes within DLPFC networks. | Xiang-Dong Du Zhe Li Nian Yuan Ming Yin Xue-Li Zhao Xiao-Li Lv Si-Yun Zou Jun Zhang Guang-Ya Zhang Chuan-Wei Li Hui Pan Li Yang Si-Qi Wu Yan Yue Yu-Xuan Wu Xiang-Yang Zhang | 2022 | World Journal of Psychiatry2022,12,9: | 3 |
| 13 | Oncogenic role of microRNA-423-5p in hepatocellular carcinoma显示文摘BACKGROUND: It has been found that micro RNA-423-5p(mi R423-5p) is an oncogenic factor and frequently upregulated in gastric carcinoma. However,the involvement of mi R423-5p in hepatocellular carcinoma(HCC) has been rarely reported. The aim of this study was to assess whether mi R423-5p is aberrantly expressed in HCC tissues,and to characterize its roles in the cancerous biology of HCC.METHODS: HCC and corresponding nonmalignant tissues were obtained from 115 patients during liver transplantation to detect the expression level of mi R423-5p. The mi R423-5p mimic and inhibitor were transfected into LM3 cell line. Cell viability assay,cell cycle analysis,transwell invasion and migration experiments were used to evaluate the oncogenic role of mi R423-5p.RESULTS: mi R423-5p was significantly upregulated in HCC compared with nonmalignant tissues,and this upregulation was negatively associated with recurrence-free survival. For patients beyond the Milan criteria,low expression of mi R423-5p was correlated with better prognosis. Functional analysis showed that mi R423-5p enhanced the proliferative,invasive and migratory capacity of HCC cells.CONCLUSIONS: mi R423-5p contributed to the tumorigenesis and progression of HCC. | 16 | Genetic parameters and trends for production and reproduction traits of a Landrace herd in China显示文摘The objectives of this study were to estimate the genetic parameters and the breeding progress in a Landrace herd in China, and to predict the potential benefits by applying new breeding technology. Hereby, the performance records from a Landrace swine herd in China, composing over 33 000 pigs born between 2001 and 2013, were collected on six economically important traits, i.e., average daily gain between 30–100 kg(ADG), adjusted backfat thickness at 100 kg(BF), adjusted days to 30 kg(D30), adjusted days to 100 kg(D100), number born alive(NBA), and total number born(TNB). The genetic parameters were estimated by restricted maximum likelihood via DMU, and realized genetic trends were analyzed. Based on the real population structure and genetic parameters obtained from this herd, the potential genetic trends by applying genomic selection(GS) were predicted via a computer simulation study. Results showed that the heritability estimates in this Landrace herd were 0.55(0.02), 0.42(0.01), and 0.12(0.01), for BF, D100, and TNB, respectively. Favorable genetic trends were obtained for D100, BF, and TNB due to direct selection, for ADG and NBA due to indirect selection. Long-term selection against D100 did not improve D30, though they are highly genetically correlated(0.64). Appling GS in such a swine herd, the genetic gain can be increased by 25%, or even larger for traits with low heritability or individuals without phenotypes before selection. | 20 | Development and validation of a questionnaire-based risk scoring system to identify individuals at high risk for gastric cancer in Chinese populations显示文摘Objective:This study aimed to develop and validate a risk scoring system to identify high-risk individuals carrying malignant lesions in stomach for tailored gastric cancer screening.Methods:A gastric cancer risk scoring system(GC-RSS)was developed based on questionnaire-based predictors for gastric cancer derived from systematic literature review.To assess the capability of this system for discrimination,risk scores for 8,214 and 7,235 outpatient subjects accepting endoscopic examination in two endoscopy centers,and 32,630 participants in a community-based cohort in China were calculated to plot receiver operating characteristic curves and generate area under the curve(AUC).To evaluate the performance of GC-RSS,the screening proportion,sensitivity and detection rate ratio compared to universal screening were used under different risk score cutoff values.Results:GC-RSS comprised nine predictors including advanced age,male gender,low body mass index(<18.5 kg/m^(2)),family history of gastric cancer,cigarette smoking,consumption of alcohol,preference for salty food,irregularity of meals and consumption of preserved food.This tool performed well in determining the risk of malignant gastric lesions with AUCs of 0.763,0.706 and 0.696 in three validation sets.When subjects with risk scores≥5 were evaluated with endoscopy,nearly 50%of these endoscopies could be saved with a detection rate of over 1.5 times achieved.When the cutoff was set at 8,only about 10%of subjects with the highest risk would be offered endoscopy,and detection rates for gastric cancer could be increased 2-4 fold compared to universal screening.Conclusions:An effective questionnaire-based GC-RSS was developed and validated.This tool may play an important role in establishing a tailored screening strategy for gastric cancer in China. | Ren Zhou Hongchen Zheng Mengfei Liu Zhen Liu Chuanhai Guo Hongrui Tian Fangfang Liu Ying Liu Yaqi Pan Huanyu Chen Zhe Hu Hong Cai Zhonghu He Yang Ke | 2021 | Chinese Journal of Cancer Research2021,33,6: | 3 |
