| 3 | Combined deficiency of SLAMF8 and SLAMF9 prevents endotoxin-induced liver inflammation by downregulating TLR4 expression on macrophages显示文摘Classical signaling lymphocyte activating molecule(SLAM)family receptors are abundant within many types of immune cells,whereas the nonclassical SLAM family receptors SLAMF8 and SLAMF9,which uniquely lack cytoplasmic signaling motifs,are highly expressed by myeloid cells.Due to the potential redundancy,whether these two receptors regulate macrophage function remains largely unknown.Here,we show that SLAMF8 and SLAMF9 co-regulate macrophage-mediated liver inflammation.To overcome the redundancy,we generated mice that simultaneously lacked SLAMF8 and SLAMF9 using CRISPR-Cas9 technology.Although macrophage differentiation was not altered by the combined deficiency of SLAMF8 and SLAMF9,the loss of these two receptors significantly protected against lipopolysaccharide(LPS)-induced liver injury.SLAMF8 and SLAMF9 double-deficient mice had a prolonged survival rate and less infiltration of inflammatory cells.The depletion of macrophages using clodronate liposomes abolished the effects of SLAMF8 and SLAMF9 deficiencies on LPS-induced liver injury,which demonstrates that these receptors are required for macrophage activation following LPS challenge.Moreover,the deficiency of SLAMF8 and SLAMF9 suppressed the secretion of inflammatory cytokines by downregulating the expression of Toll-like receptor-4(TLR4),a receptor that specifically binds LPS,which led to decreased mitogen-activated protein kinases(MAPK)signaling activation.Notably,combined injections of truncated extracellular SLAMF8 and SLAMF9 proteins significantly alleviated LPS-induced liver injury.Thus,our findings provide insights into the role of SLAMF8 and SLAMF9 in endotoxin-induced liver injury and suggest that SLAMF8 and SLAMF9 are potential therapeutic targets for acute hepatic injury. | Xiaokang Zeng Guangao Liu Wanwen Peng Junming He Chenxu Cai Wei Xiong Shasha Chen Meixiang Yang Zhongjun Dong | 2020 | Cellular & Molecular Immunology2020,17,2: | 5 |

| 5 | High-throughput sequencing exclusively identified a novel Torque teno virus genotype in serum of a patient with fatal fever显示文摘Torque teno virus(TTV) has been found to be prevalent world-wide in healthy populations and in patients with various diseases, but its etiological role has not yet been determined. Using high-throughput unbiased sequencing to screen for viruses in the serum of a patient with persistent high fever who died of suspected viral infection and prolonged weakness, we identified the complete genome sequence of a TTV(isolate Hebei-1). The genome of TTV-Hebei-1 is 3649 bp in length, encoding four putative open reading frames, and it has a G+C content of 49%. Genomic comparison and a BLASTN search revealed that the assembled genome of TTV-Hebei-1 represented a novel isolate, with a genome sequence that was highly heterologous to the sequences of other reported TTV strains. A phylogenetic tree constructed using the complete genome sequence showed that TTV-Hebei-1 and an uncharacterized Taiwan Residents strain, TW53A37, constitute a new TTV genotype. The patient was strongly suspected of carrying a viral infection and died eventually without any other possible causes being apparent. No virus other than the novel TTV was identified in his serum sample. | 7 | Community structure and carbon and nitrogen storage of sagebrush desert under grazing exclusion in Northwest China显示文摘Overgrazing is regarded as one of the key factors of vegetation and soil degradation in the arid and semi-arid regions of Northwest China.Grazing exclusion(GE)is one of the most common pathways used to restore degraded grasslands and to improve their ecosystem services.Nevertheless,there are still significant controversies concerning GE’s effects on grassland diversity as well as carbon(C)and nitrogen(N)storage.It remains poorly understood in the arid desert regions,whilst being essential for the sustainable use of grassland resources.To assess the effects of GE on community characteristics and C and N storage of desert plant community in the arid desert regions,we investigated the community structure and plant biomass,as well as C and N storage of plants and soil(0-100 cm depth)in short-term GE(three years)plots and adjacent long-term freely grazing(FG)plots in the areas of sagebrush desert in Northwest China,which are important both for spring-autumn seasonal pasture and for ecological conservation.Our findings indicated that GE was beneficial to the average height,coverage and aboveground biomass(including stems,leaves and inflorescences,and litter)of desert plant community,to the species richness and importance values of subshrubs and perennial herbs,and to the biomass C and N storage of aboveground parts(P<0.05).However,GE was not beneficial to the importance values of annual herbs,root/shoot ratio and total N concentration in the 0-5 and 5-10 cm soil layers(P<0.05).Additionally,the plant density,belowground biomass,and soil organic C concentration and C storage in the 0-100 cm soil layer could not be significantly changed by short-term GE(three years).The results suggest that,although GE was not beneficial for C sequestration in the sagebrush desert ecosystem,it is an effective strategy for improving productivity,diversity,and C and N storage of plants.As a result,GE can be used to rehabilitate degraded grasslands in the arid desert regions of Northwest China. | DONG Yiqiang SUN Zongjiu AN Shazhou JIANG Shasha WEI Peng | 2020 | Journal of Arid Land2020,12,2: | 3 |
| 8 | Intensification of the East Asian Monsoon in Southern China at about300-400 kaBP Inferred from Eolian Deposits in the Middle-lower Reaches of the Yangtze River显示文摘In the East Asian monsoon region,eolian deposits widely distributed in the middle-lower reaches of the Yantgze River are among the best materials available for studies on Quaternary climate change in the subtropical zone of Southern China.Typical eolian deposits in this region include upper Xiashu Loess(XL) and underlying Vermiculated Red Soil(VRS) layers.In this paper,chronological and paleoclimatic studies are conducted on an eolian deposit sequence near Jiujiang(JJ) city in northern Jiangxi province.A magnetostratigraphic study,combined with optically stimulated luminescence(OSL) dating,is conducted on the JJ section and provides further evidence that eolian deposits in the middle-lower reaches of the Yangtze River have been formed since the late Early Pleistocene,and that the boundary age between the XL and VRS layers is about 300-400 kaBP.In grain-size records of the JJ section,the median grain-size and content of the >30 μm size fraction increase sharply after 300-400 kaBP,representing an East Asian winter monsoon intensification event.Further pollen analysis reveals differing pollen assemblages before and after 300-400 kaBP:there is an evident increase in plants adapted to grow in a warm humid environment after 300-400 kaBP,implying an increase in precipitation caused by intensification of the East Asian summer monsoon.Global ice volume and uplift of the Tibet Plateau(TP) are regarded as crucial factors influencing variations of the East Asian monsoon on a long-term scale.The deep-sea δ^(18)O record,which reflects variations in global ice volume,shows no obvious change after 300-400 kaBP.Moreover,the influence of global ice volume changes on the East Asian summer and winter monsoons is inverse;the global ice volume increase(decrease) implies a strengthened(weakened) winter monsoon and weakened(strengthened) summer monsoon.We therefore interpret the coupled intensifications of the East Asian summer and winter monsoons at about 300-400 kaBP to the uplift of the TP in the Middle Pleistocene.This climate event is also documented in eolian deposits from the southern margin of the Chinese Loess Plateau(CLP) and from the desert-loess transitional belt.However,it is not recorded in the loess-paleosol sequences from the central part of the CLP,thereby indicating differing climate responses to TP uplift in different regions,which requires further study. | QI Lin QIAO Yansong LI Yuehui WANG Yan PENG Shasha HE Zexin YANG Shuaibin HAN Chao ZHANG Xujiao | 2017 | Acta Geologica Sinica(English Edition)2017,91,3: | 3 |
| 9 | Efficacy of Rebixiao Chinese herbal tablets and Chinese formula granules in acute gout arthritis patients:a randomized,multicenter,double-blind,controlled trial显示文摘OBJECTIVE:To evaluate the clinical efficacy and safety of Rebixiao(RBX)Chinese herbal tablets(CHT)and Chinese formula granules(CFG)in the treatment of acute gout arthritis(AGA).METHODS:This randomized,multicenter,double-blind,controlled trial included 165 AGA patients with the damp-heat symptom pattern who were randomly divided into an RBX CHT group and an RBX CFG group and treated for 7 d at three centers.The total effective rates of the joint symptom score,Traditional Chinese Medicine(TCM)symptoms score,erythrocyte sedimentation rate(ESR),and C-reactive protein(CRP)were used to evaluate the clinical efficacy.Safety assessments were also performed.RESULTS:Of the 165 enrolled patients,147 completed the clinical observation.There was no difference in baseline between the two groups.The total effective rates of the joint symptom score were 94.36%and 97.36%,and the total effective rates of the TCM symptoms score were 95.77%and 97.36%in the CFG group and CHT group,respectively.No statistical difference was found between the two groups(P>0.05).Additionally,ESR and CRP were similar in both groups(P>0.05).Furthermore,treatment efficacy regarding TCM and joint symptoms,the ESR,and CRP were consistent within each center and among the different centers(P>0.05).In addition,the incidence of adverse events was 4.22%and 2.63%in the CFG group and CHT group,respectively,and no difference was observed between the two groups(P>0.05).CONCLUSION:RBX CFG and CHT have significant and similar efficacy in the treatment of AGA,and CFG did not increase adverse side effects. | Wang Shasha Zhang Ying Dai Min Yang Min Leng Wenfei Peng Xiufeng Li Guojun Yang Jinbing Gan Jianping Zhu Fenglin Li Yanping Wu Bin | 2020 | Journal of Traditional Chinese Medicine2020,40,4: | 3 |
| 10 | The Uniform Distribution of SiC Particles in an A356-SiC_p Composite Produced by the Tilt-blade Mechanical Stirring显示文摘The tilt-blade mechanical stirring of A356-2.5vol.%SiCp liquid was conducted in a cylindrical crucible to solve the problem of nonhomogeneous radial distribution of SiC particles in conventional straight-blade mechanical stirring. The radial distribution of SiC particles in A356 liquid was studied under the conditions of 25° for horizontal tilt angle α of the blade, 200 r/min for rotating speed of stirrer and 10 mm/s for speed of moving up and down of stirrer. The results show that there exists a nonlinear relationship between circumferential tilt angle β of the blade and radial relative deviation d of SiCp content in A356 liquid. When β is smaller than 26°, the d of SiCp content in A356 liquid between the center and the periphery of crucible decreases with increasing β. Conversely, when β is bigger than 26 , d increases with increasing β. Only when β is about 26°, d can be equal to nought i.e. uniform radial distribution of SiC particles can be realized. | 11 | Effects of thyme(Thymus vulgaris L.)addition on the volatile compounds of mutton broth during boiling显示文摘To investigate the effects of thyme(Thymus vulgaris L.)addition on the flavor modification of mutton broth during boiling,three mutton-broth samples with various thyme contents 0.06%(S1),0.30%(S2),and1.50%(S3)were analyzed and compared,without thyme as control(0.00%,S0).The meaty,fatty,pastoral flavor and spicy were chosen as sensory attributes to evaluate the flavor of the mutton-broth samples.Sensory results were significantly different(P<0.001),with S2 having the optimum overall acceptability.A total of 99 volatile compounds were identified by gas chromatography-mass spectrometry,among which19 compounds were considered as the odor-active compounds according to their odor activity values.Significant changes(P<0.05)appeared in most volatiles in S0 with thyme addition,especially aldehydes.Free fatty acids(FFAs)were also identified,and all of them significantly increased with increased thyme(P<0.05).Correlation analysis of odor-active compounds,FFAs,and sensory attributes through partial least squares regression indicated the important volatiles and FFAs remarkably contributed to the mutton broth samples,and further confirmed that the 0.30%of thyme may be a desirable addition amount for the sensory characteristics of mutton broth. | Shasha Qi Ping Zhan Honglei Tian Peng Wang Xueping Ma Kaixuan Li | 2022 | Food Science and Human Wellness2022,11,2: | 2 |

| 15 | The histone methyltransferase SUVR2 promotes DSB repair via chromatin remodeling and liquidliquid phase separation显示文摘Maintaining genomic integrity and stability is particularly important for stem cells,which are at the top of the cell lineage origin.Here,we discovered that the plant-specific histone methyltransferase SUVR2 maintains the genome integrity of the root tip stem cells through chromatin remodeling and liquid-liquid phase separation(LLPS)when facing DNA double-strand breaks(DSBs).The histone methyltransferase SUVR2(MtSUVR2)has histone methyltransferase activity and catalyzes the conversion of histone H3 lysine 9 monomethylation(H3K9me1)to H3K9me2/3 in vitro and in Medicago truncatula.Under DNA damage,the proportion of heterochromatin decreased and the level of DSB damage marker y-H2AX increased in suvr2 mutants,indicating that MtSUVR2 promotes the compaction of the chromatin structure through H3K9 methylation modification to protect DNA from damage.Interestingly,MtSUVR2 was induced by DSBs to phase separate and form droplets to localize at the damage sites,and this was confirmed by immunofluorescence and fluorescence recovery after photobleaching experiments.The IDR1 and lowcomplexity domain regions of MtSUVR2 determined its phase separation in the nucleus,whereas the IDR2 region determined the interaction with the homologous recombinase MtRAD51.Furthermore,we found that MtSUVR2 drove the phase separation of MtRAD51 to form'DNA repair bodies,'which could enhance the stability of MtRAD51 proteins to facilitate error-free homologous recombination repair of stem cells.Taken together,our study reveals that chromatin remodeling-associated proteins participate in DNA repair through LLPS. | Qianwen Liu Peng Liu Tuo Ji Lihua Zheng Chen Shen Shasha Ran Jinling Liu Yafei Zhao Yiding Niu Tao Wang Jiangli Dong | 2022 | Molecular Plant2022,15,7: | 1 |
