High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population. We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable. Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping, candidate gene validation, and developing functional markers for genomics-based breeding in wheat. | Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu | 2020 | Molecular Plant2020,13,9: | 8

Myofiber necroptosis promotes muscle stem cell proliferation via releasing Tenascin-C during regeneration显示文摘Necroptosis,a form of programmed cell death,is characterized by the loss of membrane integrity and release of intracellular contents, the execution of which depends on the membrane-disrupting activity of the Mixed Lineage Kinase Domain-Like protein(MLKL)upon its phosphorylation. Here we found myofibers committed MLKL-dependent necroptosis after muscle injury.Either pharmacological inhibition of the necroptosis upstream kinase Receptor Interacting Protein Kinases 1(RIPK1)or genetic ablation of MLKL expression in myofibers led to significant muscle regeneration defects.By releasing factors into the muscle stem cell(MuSC)microenvironment, necroptotic myofibers facilitated muscle regeneration.Tenascin-C(TNC),released by necroptotic myofibers, was found to be critical for MuSC proliferation. The temporary expression of TNC in myofibers is tightly controlled by necroptosis;the extracellular release of TNC depends on necroptotic membrane rupture.TNC directly activated EGF receptor(EGFR)signaling pathway in MuSCs through its N-terminus assembly domain together with the EGF-like domain. These findings indicate that necroptosis plays a key role in promoting MuSC proliferation to facilitate muscle regeneration. | Shen'ao Zhou Wei Zhang Gaihong Cai Yingzhe Ding Caixia Wei Sheng Li Yu Yang Jie Qin Dan Liu Hao Zhang Xiexiang Shao Jianhua Wang Hongye Wang Wenjun Yang Huating Wang She Chen Ping Hu Liming Sun | 2020 | Cell Research2020,30,12: | 4
Nanoenzyme engineered neutrophil-derived exosomes attenuate joint injury in advanced rheumatoid arthritis via regulating inflammatory environment显示文摘Rheumatoid arthritis(RA)is a chronic inflammatory disease characterized by synovitis and destruction of cartilage, promoted by sustained inflammation. However, current treatments remain unsatisfactory due to lacking of selective and effective strategies for alleviating inflammatory environments in RA joint.Inspired by neutrophil chemotaxis for inflammatory region,we therefore developed neutrophil-derived exosomes functionalized with sub-5 nm ultrasmall Prussian blue nanoparticles(uPB-Exo)via click chemistry, inheriting neutrophil-targeted biological molecules and owning excellent anti-inflammatory properties.uPB-Exo can selectively accumulate in activated fibroblast-like synoviocytes, subsequently neutralizing pro-inflammatory factors, scavenging reactive oxygen species, and alleviating inflammatory stress. In addition,uPB-Exo effectively targeted to inflammatory synovitis, penetrated deeply into the cartilage and real-time visualized inflamed joint through MRI system, leading to precise diagnosis of RA in vivo with high sensitivity and specificity.Particularly,uPB-Exo induced a cascade of anti-inflammatory events via Th17/Treg cell balance regulation, thereby significantly ameliorating joint damage. Therefore, nanoenzyme functionalized exosomes hold the great potential for enhanced treatment of RA in clinic. | Lei Zhang Ziguo Qin Han Sun Xiang Chen Jian Dong Siyu Shen Liming Zheng Ning Gu Qing Jiang | 2022 | Bioactive Materials2022,7,12: | 4

Neuroprotective role of edaravone and the effects of endoplasmic reticulum stress in an adult rat model of focal cerebral ischemia/reperfusion显示文摘BACKGROUND: Endoplasmic reticulum (ER) stress impairs ER functions and leads to the accumulation of unfolded or misfolded proteins in the ER lumen. ER stress-induced cell death plays an important role in cerebral ischemia. Edaravon (3-methyl-1-phenyl-2-pyrazolin-5-one) is a potent and novel scavenger of free radicals that inhibit delayed neuronal death, as demonstrated by in vitro and animal studies. However, its effect on ER stress and induced neuronal apoptosis in a rat model of brief middle cerebral artery occlusion remains unclear. OBJECTIVE: To explore the effects of edaravone on the expression of ER stress-related factors and neuronal apoptosis, based on the hypothesis that edaravone influences ER stress in a rat model of cerebral ischemia/reperfusion. DESIGN, TIME AND SETTING: A randomized, controlled, animal study was performed at the Laboratory of Department of Neurology, Xiangya Hospital and the Department of Laboratory Animals, Xiangya Medical College, Central South University in China from June 2005 to May 2006. MATERIALS: Edaravone was purchased from Simcere Pharmaceutical Group, China. METHODS: A total of 216 adult, male, Sprague Dawley rats were randomly assigned to sham-surgery, model and edaravone groups, with 72 rats in each group. Brief middle cerebral artery occlusion was established in the model and edaravone groups. In addition, the edaravone group rats were injected with 3 mg/kg edaravone through the tail vein. MAIN OUTCOME MEASURES: RNA-dependent protein kinase-like endoplasmic reticulum eukaryotic translation initiation factor 2α kinase (PERK) and C/EBP homology protein (CHOP) mRNA expression in the ischemic parietal cortex was determined by reverse transcription- polymerase chain reaction; phosphorylated PERK and CHOP protein expression was detected by immunohistochemistry; neuronal apoptosis was detected by TdT-mediated-dUTP nick end labeling. RESULTS: Neurological deficit scores were significantly reduced in the edaravone group compared to the model group at 12, 24, and 72 hours following reperfusion (P < 0.05). In addition, PERK and CHOP mRNA as well as phosphorylated PERK and CHOP protein expression were significantly reduced in the edaravone group compared to the model group at 1, 3, and 6 hours following reperfusion (P < 0.05, P < 0.01). CHOP mRNA expression was decreased in the edaravone group compared to the model group at 3, 6, 12, and 24 hours following reperfusion (P < 0.01), while CHOP protein expression was less than the model group at 6, 12, and 24 hours following reperfusion (P < 0.05). CONCLUSION: Edaravone treatment resulted in decreased PERK and CHOP expression following ischemia/reperfusion, as well as reduced neuronal apoptosis. Edaravone exhibited a neuroprotective role by inhibiting endoplasmic reticulum stress. | Xiangmin Shen Liming Tan Yunhai Liu Hainan Zhang Chunyu Wang Qidong Yang QingHuang Lin Zhou Zhenyu Tang | 2010 | Neural Regeneration Research2010,5,3: | 2

Constructing an unbalanced structure toward high working voltage for improving energy density of non-aqueous carbon-based electrochemical capacitors显示文摘The energy density of non-aqueous carbon-based electrochemical capacitors(cEC)is mainly determined by the specific capacitance and operational voltage range. In this study,we propose to construct an unbalanced structure to make full use of stable voltage range for improving energy density. The stable voltage range is firstly carefully explored using cyclic voltammetry. Then an unbalanced carbon-based electrochemical capacitor(ucEC)is constructed with an optimized positive electrode to negative electrode weight ratio and voltage range. Its electrochemical performance is comprehensively investigated, including energy density, power density as well as cycle life. The ucEC is capable to deliver an improved energy density up to 64.9 Wh/kg(1.4 times as high as a general cEC)without sacrificing the power density and cycle life. The electrode properties after cycling are also analyzed, illustrating the change of electrode potential caused by unbalanced structure. The proposed structure demonstrates a great potential for improving the energy density at little cost of electrode design and cell configuration. | Junsheng Zheng Nan Qin Liming Jin Xin Guo Chao Shen Qiang Wu Jim PZheng | 2020 | Chinese Chemical Letters2020,31,3: | 2
Nogo-A and Nogo-A receptor expression in periventricular white matter of experimental autoimmune encephalomyelitis rats显示文摘BACKGROUND:Previous studies have focused on the correlation between Nogo-A expression and multiple sclerosis or between Nogo-A receptor(NgR) expression and multiple sclerosis in the central nervous system.Expression patterns of Nogo-A and NgR remain poorly understood in rat models of experimental autoimmune encephalomyelitis(EAE).OBJECTIVE:To observe dynamic changes in Nogo-A and NgR protein expression, and to verify the correlation between Nogo-A and NgR protein,as well as expression patterns at various time points,in periventricular tissue of EAE rats.DESIGN,TIME AND SETTING:A neuroimmunological, randomized, controlled experiment was performed at the Clinical Institute of Hunan People's Hospital of China from September to November 2008.MATERIALS:Immunohistochemistry(streptavidin-biotin-peroxidase complex method) kit was purchased from Boster,China.METHODS:A total of 60 female,Wistar rats, aged 6-8 weeks, were randomly assigned to EAE and control groups(n = 30,respectively).Guinea pig spinal cord homogenate,self-made complete Freund's adjuvant(0.2 mL/100 g),and pertussis vaccine(0.2 mL) were subcutaneously injected into the hindlimb foot pad of rats from the EAE group to create rat models of EAE.Complete Freund's adjuvant(0.2 mL) was infused into rats from the control group.MAIN OUTCOME MEASURES:Nogo-A and NgR protein expression was determined in periventricular white matter using immunohistochemical methods.Neurological scores were determined in all rats.RESULTS:Rats from the EAE group developed acute-onset EAE following immunization. The pathogenetic symptoms reached a peak on day 15,and neurological scores were also greatest at this time point.Neurological scores decreased with recovery of the illness.Nogo-A was shown to be expressed in neuronal cells and oligodendrocytes, and expression increased 11 days after immunization(P < 0.01),decreased by day 13(P < 0.01),and then increased again by day 15.Nogo-A expression remained greater in the EAE group compared with the control group at day 30(P < 0.01).In the EAE group,NgR protein was primarily expressed on the surface of neuronal bodies and axons.NgR expression increased 13-18 days after immunization(P < 0.01 or P < 0.05).CONCLUSION:Nogo-A and NgR protein expression altered with disease course in periventricular white matter of EAE rats. Results suggested that Nogo-A and NgR were involved in EAE occurrence. | Liming Tan Ying Chi Chunyu Wang1, Jun Wu1, Hainan Zhang1, Xiangmin Shen1, ShujuanDuan1, Bo JiangI Jun Wu Hainan Zhang Xiangmin Shen Shujuan Duan Bo Jiang | 2010 | Neural Regeneration Research2010,5,8: | 2
Solvothermal-assisted assembly of MoS2 nanocages on graphene sheets to enhance the electrochemical performance of lithium-ion battery显示文摘An ordered hollow M0S2 nanocages/RGO nanocomposite is constructed by a simple solvothermal-assisted assembly method combined with freeze-drying and annealing. In this novel nanostructure, hollow M0S2 nanocages are homogeneously distributed on graphene sheets with a tight bond of C-O-Mo.The nanosized and hollow MoS2 nanocages can effectively accommodate the huge volume change during charge/discharge process and increase the number of electrochemical reaction active sites, accelerating the kinetics of lithiation/delithiation.The tight C-O-Mo bond between graphene and MoS2 further reinforces the structural stability, thus improve the electrical conductivity and substantially enhance the lithium storage performance of M0S2 anode material. As a result, this novel nanocomposite shows a long-cycle stability of 717.4 mAh·g^-1 after 800 cycles at a high current density of 3 A·g^-1,exhibiting great potential as an anode nanocomposite for advanced lithium-ion batteries. | Dafang He Yi Yang Zhenmin Liu Jin Shao Jian Wu Shun Wang Liming Shen Ningzhong Bao | 2020 | Nano Research2020,13,4: | 2
Curcumin alleviates TGF-β1-induced fibrosis in NRK-49F cells via suppression of miR-21 expression, and regulation of the TGF-β1/smad3 signaling pathway显示文摘Objective:Overexpression of microRNA-21(miR-21)has been well-recognized during fibrosis in a wide range of tissue types through regulation of fibrogenesis via diverse pathways.Curcumin exhibits antifibrotic effects in various organs. We and others have previously demonstrated that curcumin has positive effects on the protection against fibrosis. However, the effect of curcumin on miR-21 expression has not been reported. Methods:In this study, transforming growth factor-b1(TGF-b1)-stimulated rat renal fibroblast cells(NRK-49F)were transfected with miR-21 inhibitors, then the expression of Smad3,a-smooth muscle actin(a-SMA),type I collagen(COL1A1),and type III collagen(COL3A1)was determined using quantitative real-time-PCR(qRT-PCR)and western blot analysis. The effect of curcumin on production of the extracellular matrix was evaluated using immunofluorescence,qRT-PCR,and western blot analysis.Results:TGF-b1 stimulation upregulated the expression of miR-21 and induced fibrogenesis in NRK-49F cells.Transfection with miR-21 inhibitors selectively decreased smad3 activity, and noticeably reduced the expression of a-SMA,COL1A1,and COL3A1.Curcumin treatment significantly inhibited the expressions of smad3,a-SMA,COL1A1,COL3A1,as well as miR-21 expression in NRK-49F cells in a dosedependent manner. Conclusion:Curcumin exerted its anti-fibrotic effects by targeting the TGF-b1/smad3 signaling pathway and suppressing miR-21 expression, thereby provides novel insight in the protective effects of curcumin against fibrosis in various organs. | Lan Zhang Yi Liu Xiting Wang Cheng Li Mengmeng Shen Liming Di Shuangshuang He Yu Li | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,1: | 2
Synergistic effects of autophagy/mitophagy inhibitors and magnolol promote apoptosis and antitumor efficacy显示文摘Mitochondria as a signaling platform play crucial roles in deciding cell fate. Many classic anticancer agents are known to trigger cell death through induction of mitochondrial damage.Mitophagy, one selective autophagy,is the key mitochondrial quality control that effectively removes damaged mitochondria. However, the precise roles of mitophagy in tumorigenesis and anticancer agent treatment remain largely unclear. Here,we examined the functional implication of mitophagy in the anticancer properties of magnolol,a natural product isolated from herbal Magnolia officinalis.First,we found that magnolol induces mitochondrial depolarization, causes excessive mitochondrial fragmentation, and increases mitochondrial reactive oxygen species(mtROS).Second, magnolol induces PTEN-induced putative kinase protein 1(PINK1)-Parkin-mediated mitophagy through regulating two positive feedforward amplification loops.Third, magnolol triggers cancer cell death and inhibits neuroblastoma tumor growth via the intrinsic apoptosis pathway. Moreover, magnolol prolongs the survival time of tumor-bearing mice. Finally, inhibition of mitophagy by PINK1/Parkin knockdown or using inhibitors targeting different autophagy/mitophagy stages significantly promotes magnolol-induced cell death and enhances magnolol's anticancer efficacy, both in vitro and in vivo.Altogether, our study demonstrates that magnolol can induce autophagy/mitophagy and apoptosis, whereas blockage of autophagy/mitophagy remarkably enhances the anticancer efficacy of magnolol, suggesting that targeting mitophagy may be a promising strategy to overcome chemoresistance and improve anticancer therapy. | Yancheng Tang Liming Wang Tao Yi Jun Xu Jigang Wang Jiang-Jiang Qin Qilei Chen Ka-Man Yip Yihang Pan Peng Hong Yingying Lu Han-Ming Shen Hu-Biao Chen | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 2
Molecular design of organic dyes based on vinylene hexylthiophene bridge for dye-sensitized solar cells显示文摘Three donor-(π-spacer)-acceptor(D-π-A) organic dyes, containing different groups(triphenylamine,di(p-tolyl)phenylamine,and 9-octylcarbazole moieties) as electron donors, were designed and synthesized.Nanocrystalline TiO2 dye-sensitized solar cells were fabricated by using these dyes.It was found that the variation of electron donors in the D-π-A dyes played an important role in modifying and tuning photophysical properties of organic dyes.Under standard global AM 1.5 solar condition, the DSSC based on the dye D2 showed the best photovoltaic performance:a short-circuit photocurrent density(Jsc) of 13.93 mA/cm2,an open-circuit photovoltage(Voc) of 0.71 V,and a fill factor(FF) of 0.679,corresponding to solar-to-electric power conversion efficiency(η) of 6.72%. | LIU DaXi ZHAO Bin SHEN Ping HUANG Hui LIU LiMing TAN SongTing | 2009 | Science China Chemistry2009,52,8: | 2
