3Clinical trial perspective for adult and juvenile Huntington's disease using genetically-engineered mesenchymal stem cells显示文摘Progress to date from our group and others indicate that using genetically-engineered mesenchymal stem cells(MSC) to secrete brain-derived neurotrophic factor(BDNF) supports our plan to submit an Investigational New Drug application to the Food and Drug Administration for the future planned Phase 1 safety and tolerability trial of MSC/BDNF in patients with Huntington's disease(HD). There are also potential applications of this approach beyond HD. Our biological delivery system for BDNF sets the precedent for adult stem cell therapy in the brain and could potentially be modified for other neurodegenerative disorders such as amyotrophic lateral sclerosis(ALS), spinocerebellar ataxia(SCA), Alzheimer's disease, and some forms of Parkinson's disease. The MSC/BDNF product could also be considered for studies of regeneration in traumatic brain injury, spinal cord and peripheral nerve injury. 4Effect of Silicon on Growth, Physiology, and Cadmium Translocation of Tobacco (Nicotiana tabacum L.) in Cadmium-Contaminated Soil显示文摘Silicon(Si) offers beneficial effect on plants under cadmium(Cd) stress such as promoting plant growth and increasing resistance to heavy metal toxicity. In this study, a pot experiment was performed to study the role of Si in alleviating Cd toxicity in tobacco(Nicotiana tabacum L.) plants on a yellow soil taken from Guiyang, China. Nine treatments consisting of three concentrations of Cd(0, 1, and 5 mg kg^(-1)) together with three Si levels(0, 1, and 4 g kg^(-1)) were established. Plant growth parameters, Cd concentration,and the malondialdehyde(MDA), chlorophyll, and carotenoid contents were determined 100 d after transplanting of tobacco seedlings.Application of exogenous Si enhanced the growth of tobacco plants under Cd stress. When 5 mg kg^(-1) Cd was added, Si addition at 1 and 4 g kg^(-1) increased root, stem, and leaf biomass by 26.1%–43.3%, 33.7%–43.8%, and 50.8%–69.9%, respectively, compared to Si addition at 0 g kg^(-1). With Si application, the transfer factor of Cd in tobacco from root to shoot under both 1 and 5 mg kg^(-1) Cd treatments decreased by 21%. The MDA contents in the Si-treated tobacco plants declined by 5.5%–17.1% compared to those in the non-Si-treated plants, indicating a higher Cd tolerance. Silicon application also increased the chlorophyll and carotenoid contents by 33.9%–41% and 25.8%–47.3% compared to the Cd only treatments. 6Rational construction of self-assembly azobenzene derivative monolayers with photoswitchable surface properties显示文摘Photo-responsive azobenzene(ABZ) derivatives with different end groups(R) as photoswitchable molecules were employed to construct self-assembled monolayers(SAMs) on silicon substrate by using3-glycidoxypropyltrimethoxysilane(GPTS) as the bridging molecules. The assembly process was optimized by changing various parameters, including the type and concentration of ABZ derivatives,reaction time, etc. The obtained SAMs were fully characterized and evaluated using UV spectroscopy,atomic force microscope(AFM), elllipsometer, static contact angle and X-ray photoelectron spectroscopy(XPS). It is found that the end group property of azobenzene derivatives is critical to the obtained SAMs' photoresponsive properties. Compared with hydrophobic compounds(4-(4'-aminophenylazo) benzoic acid, ABZ-CF_3),the hydrophilic compounds(4-(4'-aminophenylazo) benzoic acid, ABZ-COOH) show excellent reversible photoswitching performance with a large contact angle change of 35° under optimized process, and the SAMs are removable by thermal treatment at 240℃ in air for only 5 min.Yu Zhang Peng Gao Qin Yue Peter Trefonas Yonghui Deng 2018Chinese Chemical Letters2018,29,11:1

17Phylogenetic patterns suggest frequent multiple origins of secondary metabolites across the seed-plant‘tree of life’显示文摘To evaluate the phylogenetic patterns of the distribution and evolution of plant secondary metabolites(PSMs),we selected 8 classes of PSMs and mapped them onto an updated phylogenetic tree including 437 families of seed plants.A significant phylogenetic signal was detected in 17 of the 18 tested seed-plant clades for at least 1 of the 8 PSM classes using the D statistic.The phylogenetic signal,nevertheless,indicated weak clustering of PSMs compared to a random distribution across all seed plants.The observed signal suggests strong diversifying selection during seed-plant evolution and/or relatively weak evolutionary constraints on the evolution of PSMs.In the survey of the current phylogenetic distributions of PSMs,we found that multiple origins of PSM biosynthesis due to external selective forces for diverse genetic pathways may have played important roles.In contrast,a single origin of PSMs seems rather uncommon.The distribution patterns for PSMs observed in this study may also be useful in the search for natural compounds for medicinal purposes.Yongzeng Zhang Tao Deng Lu Sun Jacob B.Landis Michael J.Moore Hengchang Wang Yuehua Wang Xiaojiang Hao Jijun Chen Shenghong Li Maonian Xu Pema-Tenzin Puno Peter H.Raven Hang Sun 2021National Science Review2021,8,4:1
