| 2 | Changing patterns and survival improvements of young breast cancer in China and SEER database, 1999-2017显示文摘Objective: Breast cancer in young females was usually considered more aggressive and requires aggressive therapy. We investigated whether early detection and improved treatments changed the patterns of characteristics,management and outcomes of young breast cancer patients over time.Methods: Females under 40 years of age diagnosed with breast cancer during the periods 1999-2017 and1999-2015 were identified in the Fudan University Shanghai Cancer Center(FUSCC) and the population-based Surveillance, Epidemiology, and End Results(SEER) registry, respectively. Clinicopathologic characteristics and treatment information were collected. Patients diagnosed before 2013 were followed up.Results: The proportions of young breast cancer patients were 15.0% and 5.3% in the FUSCC and SEER cohorts, respectively. In the FUSCC cohort, there was a significant increase in the proportion of ductal carcinoma in situ(DCIS)(from 8.8% to 16.9%;P<0.0001) and it remained stable in SEER cohort. The proportion of T1-stage tumors increased dramatically in the FUSCC cohort(from 35.3% to 41.9%;P=0.008), whereas it decreased in SEER cohort(from 42.4% to 33.0%;P<0.0001). The percentage of estrogen receptor(ER)-positive cancers was consistently increased in both the invasive ductal carcinoma(IDC) and DCIS patients in the two cohorts. Breastconserving surgery and immediate implant reconstruction after mastectomy both exhibited increased use over time in the FUSCC cohort. Both the FUSCC and SEER cohorts showed a significantly better prognosis in the recent time period.Conclusions: With the increased early-stage and ER-positive diseases in young patients as well as better systemic treatment strategies, improved survival has been observed in recent years. | 4 | Molecular mechanisms of apoptosis in hepatocellular carcinoma cells induced by ethanol extracts of Solanum lyratum Thumb through the mitochondrial pathway显示文摘AIM To explore the induction effects and mechanism of Solanum lyratum Thumb(ST) on human hepatocellularcarcinoma SMMC-7721 cells through the mitochondrial pathway.METHODS The experiments were conducted on three groups: an experimental group (with ST ethanol extracts' concentration being 2.5, 5 and 10 mg/L), a negative control group (with only nutrient solution, 0 mg/L ST ethanol extracts), and a positive control group (2.5 mg/L DDP). The inhibition rate of cell proliferation was checked by using the methyl thiazolyl tetrazolium method, and cell apoptosis was tested by TUNEL method. Furthermore, RT-PCR was used to examine m RNA expression of Fas, Fas L, caspase-8, caspase-3, p53 and Bcl-2 genes.RESULTS Compared with the negative control group, the inhibition and apoptosis rates of the experimental group with different concentrations of ST extracts on human hepatocellular carcinoma SMMC-7721 cells significantly increased(P<0.05). Besides, the m RNA expression of Fas L and Bcl-2 significantly decreased(P<0.05) while the m RNA expression of Fas, caspase-8, caspase-3 and p53 increased significantly. | 5 | A smart safe rechargeable zinc ion battery based on sol-gel transition electrolytes显示文摘Thermal runaway has been a long-standing safety issue impeding the development of high-energydensity batteries. Physical safety designs such as employing circuit-breakers and fuses to batteries are limited by small operating voltage windows and no resumption of original working condition when it is cooled down. Here we report a smart thermoresponsive polymer electrolyte that can be incorporated inside batteries to prevent thermal runaway via a fast and reversible sol-gel transition, and successfully combine this smart electrolyte with a rechargeable Zn/a-MnO_2 battery system. At high temperature, battery operation is inhibited as a result of the increased internal resistance caused by the gelation of liquid electrolyte. After cooling down, the electrolyte is spontaneously reversed to sol state and the electrochemical performance of the battery is restored. | 6 | Mineral Chemistry and Crystallization Conditions of the Late Cretaceous Mamba Pluton from the Eastern Gangdese, Southern Tibetan Plateau显示文摘The Late Cretaceous Mamba granodiorite belongs to a part of the Mesozoic Gangdese continental magmatic belt. No quantitative mineralogical study has been made hitherto, and hence the depth at which it formed is poorly constrained. Here we present mineralogical data for the Mamba pluton, including host rocks and their mafic microgranular enclaves(MMEs), to provide insights into their overall crystallization conditions and information about magma mixing. All amphiboles in the Mamba pluton are calcic, with ~B(Ca+Na)>1.5, and Si=6.81–7.42 apfu for the host rocks and Si=6.77–7.35 apfu for the MMEs. The paramount cation substitutions in amphibole include edenite type and tschermakite type. Biotites both in the host rocks and the MMEs collectively have high Mg O(13.19 wt.%–13.03 wt.%) contents, but define a narrow range of Al apfu(atoms per formula unit) variations(2.44–2.57). The oxygen fugacity estimates are based on the biotite compositions cluster around the NNO buffer. The calculated pressure ranges from 1.2 to 2.1 kbar according to the aluminum-in-hornblende barometer. The computed pressure varies from 0.9 to 1.3 kbar based on the aluminum-in-biotite barometer which corresponds to an average depth of ca. 3.9 km. Besides, the estimates of crystallization pressures vary from 0.8 to 1.4 kbar based on the amphibole barometer proposed by Ridolfi et al.(2010), which can be equivalent to the depths ranging from 3.1 to 5.2 km. The MMEs have plagioclase oscillatory zonings and quartz aggregates, probably indicating the presence of magma mixing. Besides, core-to-rim element variations(Rb, Sr, Ba, and P) for the K-feldspar megacrysts serve as robust evidence to support magma mixing and crystal fractionation. | 7 | Cloning and characterization of squalene synthase and cycloartenol synthase from Siraitia grosvenorii显示文摘Mogrosides and steroid saponins are tetracyclic triterpenoids found in Siraitia grosvenorii.Squalene synthase(SQS) and cycloartenol synthase(CAS) are key enzymes in triterpenoid and steroid biosynthesis.In this study,full-length cDNAs of SgSQS and SgCAS were cloned by a rapid amplification of cDNA-ends with polymerase chain reaction(RACE-PCR) approach.The SgSQS cDNA has a 1254 bp open reading frame(ORF) encoding 417 amino acids,and the SgCAS cDNA contains a 2298 bp ORF encoding 765 amino acids.Bioinformatic analysis showed that the deduced SgSQS protein has two transmembrane regions in the C-terminal.Both SgSQS and SgCAS have significantly higher levels in fruits than in other tissues,suggesting that steroids and mogrosides are competitors for the same precursors in fruits.Combined in silico prediction and subcellular localization,experiments in tobacco indicated that SgSQS was probably in the cytoplasm or on the cytoskeleton,and SgCAS was likely located in the nucleus or cytosol.These results will provide a foundation for further study of SgSQS and SgCAS gene functions in S.grosvenorii,and may facilitate improvements in mogroside content in fruit by regulating gene expression. | Huan Zhao Qi Tang Changming Mo Longhua Bai Dongping Tu Xiaojun Ma | 2017 | Acta Pharmaceutica Sinica B2017,7,2: | 10 |
| 8 | Clinicopathological and molecular genetic analysis of 4 typical Chinese HNPCC families显示文摘AIM: To study the clinicopathological and molecular genetic characteristics of typical Chinese hereditary nonpolyposis cotorectal cancer (HNPCC) families.METHODS: Four typical Chinese HNPCC families were analyzed using microdissection, microsatellite instability analysis, immunostaining of hMSH2 and hMLH1 proteins and direct DNA sequencing of hMSH2 and hMLH1 genes.RESULTS: All five tumor tissues of 4 probands from the 4typical Chinese HNPCC families showed microsatellite instability at more than two loci (MSI-H or RER +phenotype). Three out of the 4 cases lost hMSH2 protein expression and the other case showed no hMLH1 protein expression. Three pathological germline mutations (2 in hMSH2 and 1 in hMLH1 ), which had not been reported previously, were identified. The same mutations were also found in otler affected members of two HNPCC families,respectively.CONCLUSION: Typical Chinese HNPCC families showed relatively frequent germline mutation of mismstch repair genes. High-level microsateliita instability and loss of expression of mismatch repair genes correlated closely with germline mutation of mismatch repair genes. Microsatellite instability analysis and immunostaining of miamstch repair gens might serve as effective screening methods before direct DNA sequencing. | 12 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 14 | Arsenic trioxide alleviates acute graft-versus-host disease by modulating macrophage polarization显示文摘This study aimed to explore macrophage polarization in acute graft-versus-host disease after hematopoietic stem cell transplantation, and investigated if arsenic trioxide(ATO) could correct this imbalance. In the colon of GVHD mice, we found that the number of F4/80+iNOS+ cells as well as the expression intensity of TNF-α and IL-1β was greater in the GVHD group than in the BM group, whereas the number of F4/80+CD206+ cells and the expression intensity of IL-10 and TGF-β was greater in the BM group than in the GVHD group. We investigated the effect of ATO on GVHD mice, and found that ATO treatment clearly improved the survival of the mice and reduced the severity of GVHD. In addition, ATO reduced the number of F4/80+iNOS+ cells, and increased the number of F4/80+CD206+ cells in the colon of GVHD mice. Furthermore, ATO sharply decreased CD86 and CD80 expression, and increased CD163 and CD206 expression in macrophages induced from aGVHD patients. Therefore,ATO can modulate the M1 and M2 phenotype in GVHD mice or in macrophages from aGVHD patients. | 15 | Computer-aided ionic liquid design for alkane/cycloalkane extractive distillation process显示文摘A computer-aided ionic liquid design(CAILD) study is presented for the frequently encountered alkane/cycloalkane separations in petrochemical industry. Exhaustive experimental data are first collected to extend the UNIFAC-IL model for this system, where the proximity effect in alkanes and cycloalkanes is considered specifically by defining distinct groups. The thermodynamic performances of a large number of ILs for 4 different alkane/cycloalkane systems are then compared to select a representative example of such separations. By applying n-heptane/methylcyclohexane extractive distillation as a case study, the CAILD task is cast as a mixed-integer nonlinear programming(MINLP) problem based on the obtained task-specific UNIFAC-IL model and two semi-empirical models for IL physical properties. | 16 | Overexpression of GmBIN2,a soybean glycogen synthase kinase 3 gene,enhances tolerance to salt and drought in transgenic Arabidopsis and soybean hairy roots显示文摘Glycogen synthase kinase 3(GSK3)is a kind of serine/threonine kinase widely found in eukaryotes.Many plant GSK3 kinases play important roles in regulating stress responses.This study investigated BRASSINOSTEROID-INSENSITIVE 2(GmBIN2)gene,a member of the GSK3 protein kinase family in soybean and an orthologue of Arabidopsis BIN2/At SK21.GmBIN2 expression was increased by salt and drought stresses,but was not significantly affected by the ABA treatment.To examine the function of GmB IN2,transgenic Arabidopsis and transgenic soybean hairy roots were generated.Overexpression of GmBIN2 in Arabidopsis resulted in increased germination rate and root length compared with wild-type plants under salt and mannitol treatments.Overexpression of GmBIN2 increased cellular Ca^(2+) content and reduced Na+content,enhancing salt tolerance in transgenic Arabidopsis plants.In the soybean hairy root assay,overexpression of GmBIN2 in transgenic roots also showed significantly higher relative root growth rate than the control when subjected to salt and mannitol treatments.Measurement of physiological indicators,including proline content,superoxide dismutase(SOD)activity,and relative electrical conductivity,supported this conclusion.Furthermore,we also found that GmBIN2 could up-regulate the expression of some stress-related genes in transgenic Arabidopsis and soybean hairy roots.Overall,these results indicated that GmBIN2 improved tolerance to salt and drought in transgenic Arabidopsis and soybean hairy roots. | WANG Ling-shuang CHEN Qing-shan XIN Da-wei QI Zhao-ming ZHANG Chao LI Si-nan JIN Yang-mei LI Mo MEI Hong-yao SU Anoyu WU Xiao-xia | 2018 | Journal of Integrative Agriculture2018,17,9: | 4 |
| 17 | The Effect of Body Pitching on Leg-Spring Behavior in Quadruped Running显示文摘In this paper we investigated how the running speed would affect the dynamics of body pitching, and whether body inertiais important for animals. Passive trotting of spring-mass model and passive bounding of spring-beam model were studied atdifferent speeds for different sets of body parameters respectively. Furthermore, different body inertias were used in bounding.We found that running speed exerts effect on leg performance by means of centrifugal force. The centrifugal force can be understoodas an enhancement to the natural frequency of the spring-mass system. The disadvantage of body pitching may beoffset by the great increase in centrifugal force at high speed. | 20 | Cloning and expression of SgCYP450-4from Siraitia grosvenorii显示文摘CYP450 plays an essential role in the development and growth of the fruits of Siraitia grosvenorii. However, little is known about the SgCYP450-4 gene in S. grosvenorii. Here, based on transcriptome data, a full-length c DNA sequence of SgCYP450-4 was cloned by reverse transcriptasepolymerase chain reaction(RT-PCR) and rapid-amplification of c DNA ends(RACE) strategies. SgCYP450-4 is 1677 bp in length(Gen Bank accession No. AEM42985.1) and contains a complete open reading frame(ORF) of 1422 bp. The deduced protein was composed of 473 amino acids, the molecular weight is 54.01 kDa, the theoretical isoelectric point(PI) is 8.8, and the protein was predicted to possess cytochrome P450 domains. SgCYP450-4 gene was highly expressed in root, diploid fruit and fruit treated with hormone and pollination. At 10 days after treatment with pollination and hormones, the expression of SgCYP450-4had the highest level and then decreased over time, which was consistent with the development of fruits of S.Grosvenorii. Hormonal treatment could significantly induce the expression of SgCYP450-4.
