2IL-25 stimulates M2 macrophage polarization and thereby promotes mitochondrial respiratory capacity and lipolysis in adipose tissues against obesity显示文摘Obesity and associated metabolic diseases are characterized by a chronic low-grade inflammatory state with the infiltration of many inflammatory cells,especially macrophages.Immune molecules,including some cytokines,have a close relationship with metabolism.Interleukin(IL)-25 is a member of the IL-17 cytokine family that can regulate macrophages and alleviate some metabolic dysfunction;however,its role and mechanisms in lipid metabolism remain to be extensively clarified.Human serum and liver biopsy specimens,high-fat diet-induced obesity mice and DB/DB(Lepr−/−)animal models were used to examine IL-25 expression in obesity and nonalcoholic fatty liver diseases(NAFLD).To observe the role of IL-25 in lipid metabolism,model mice were administered with IL-25 or adoptively transferred with IL-25-educated macrophages in vivo,whereas bone marrow-derived macrophages,the macrophage cell line RAW264.7 and adipocytes differentiated from 3T3-L1 were used in vitro.IL-25 was decreased in NAFLD patients and obese mice.In addition,IL-25 reduced body weight gain and lipid accumulation,enhanced lipid uptake by macrophages and increased the expression of lipolysis andβ-oxidation enzymes via alternatively activating macrophages.IL-25 also promoted lipolysis and suppressed lipogenesis in adipocytes co-cultured with the IL-25-educated macrophages.Furthermore,IL-25 improved the mitochondrial respiratory capacity and oxygen consumption rate of macrophages and produced more NAD+/NADH and ATP.In conclusion,IL-25 can stimulate M2 macrophage polarization and thereby promote lipolysis and mitochondrial respiratory capacity,highlighting the potential for IL-25 to be used as a therapeutic agent against obesity and associated metabolic syndromes.Juan Feng Lingyi Li Zhiying Ou Qiao Li Baoyong Gong Zhenxian Zhao Weiwei Qi Ti Zhou Jun Zhong Weibin Cai Xia Yang Aiping Zhao Guoquan Gao Zhonghan Yang 2018Cellular & Molecular Immunology2018,15,5:10
3Clinical and genetic findings in a Chinese family with VDR-associated hereditary vitamin D-resistant rickets显示文摘Hereditary vitamin D-resistant rickets(HVDRR) is a rare autosomal recessive disorder characterized by severe rickets,hypocalcemia,hypophosphatemia,secondary hyperparathyroidism,and elevated alkaline phosphatase.This disorder is caused by homogeneous or heterogeneous mutations affecting the function of the vitamin D receptor(VDR),which lead to complete or partial target organ resistance to the action of 1,25-dihydroxy vitamin D.A non-consanguineous family of Chinese Han origin with one affected individual demonstrating HVDRR was recruited,with the proband evaluated clinically,biochemically and radiographically.To identify the presence of mutations in the VDR gene,all the exons and exon–intron junctions of the VDR gene from all family members were amplified using PCR and sequenced.The proband showed rickets,progressive alopecia,hypocalcemia,hypophosphatemia,secondary hyperparathyroidism,and elevated alkaline phosphatase.She also suffered from epilepsy,which is rarely seen in patients with HVDRR.Direct sequencing analysis revealed a homozygous missense mutation c.122G4A(p.C41Y) in the VDR gene of the proband,which is located in the first zinc finger of the DNA-binding domain.Both parents had a normal phenotype and were found to be heterozygous for this mutation.We report a Chinese Han family with one individual affected with HVDRR.A homozygous missense mutation c.122G4A(p.C41Y) in the VDR gene was found to be responsible for the patient's syndrome.In contrast to the results of treatment of HVDRR in other patients,our patient responded well to a supplement of oral calcium and a low dose of calcitriol.Qianqian Pang Xuan Qi Yan Jiang Ou Wang Mei Li Xiaoping Xing Jin Dong Weibo Xia 2016Bone Research2016,4,1:9

5Vertical Distribution and Seasonal Fluctuation of Nematode Trophic Groups as Affected by Land Use显示文摘A field investigation was conducted at the Shenyang Experimental Station of Ecology, Chinese Academy of Sciences, in an aquic brown soil of Northeast China under three land use types (cropland, abandoned cropland, and woodland) in order to evaluate whether the vertical distribution and seasonal fluctuation for the number of total nematodes and trophic groups could reflect soil ecosystem differences and to determine the relationships between soil chemical properties and soil nematodes. The majority of soil nematodes were present in the 0-20 cm soil layers, and for these land use types plant parasites were the most abundant trophic group. In the abandoned cropland the numbers of plant parasites reached a peak on the August sampling date, whereas the cropland and woodland peaked on the October sampling date. Meanwhile, in all land use types the number of total nematodes, bacterivores, plant parasites, and omnivores-predators was negatively (P < 0.05, except for bacterivores in cropland, which was not significant) correlated with bulk density, and positively (P < 0.05, except for fungivores in abandoned cropland, which was not significant) correlated with total organic carbon and total nitrogen.MENG Fan-Xiang OU Wei LI Qi JIANG Yong WEN Da-Zhong 2006Pedosphere2006,16,2:7

7Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 8Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. 9Insights into triterpene synthesis and unsaturated fatty-acid accumulation provided by chromosomallevel genome analysis of Akebia trifoliata subsp . australis显示文摘Akebia trifoliata subsp.australis is a well-known medicinal and potential woody oil plant in China.The limited genetic information available for A.trifoliata subsp.australis has hindered its exploitation.Here,a high-quality chromosomelevel genome sequence of A.trifoliata subsp.australis is reported.The de novo genome assembly of 682.14 Mb was generated with a scaffold N50 of 43.11 Mb.The genome includes 25,598 protein-coding genes,and 71.18%(485.55 Mb)of the assembled sequences were identi fi ed as repetitive sequences.An ongoing massive burst of long terminal repeat(LTR)insertions,which occurred~1.0 million years ago,has contributed a large proportion of LTRs in the genome of A.trifoliata subsp.australis.Phylogenetic analysis shows that A.trifoliata subsp.australis is closely related to Aquilegia coerulea and forms a clade with Papaver somniferum and Nelumbo nucifera,which supports the well-established hypothesis of a close relationship between basal eudicot species.The expansion of UDP-glucoronosyl and UDP-glucosyl transferase gene families and fi-amyrin synthase-like genes and the exclusive contraction of terpene synthase gene families may be responsible for the abundant oleanane-type triterpenoids in A.trifoliata subsp.australis.Furthermore,the acyl-ACP desaturase gene family,including 12 stearoyl-acyl-carrier protein desaturase(SAD)genes,has expanded exclusively.A combined transcriptome and fatty-acid analysis of seeds at fi ve developmental stages revealed that homologs of SADs,acyl-lipid desaturase omega fatty acid desaturases(FADs),and oleosins were highly expressed,consistent with the rapid increase in the content of fatty acids,especially unsaturated fatty acids.The genomic sequences of A.trifoliata subsp.australis will be a valuable resource for comparative genomic analyses and molecular breeding.Hui Huang Juan Liang Qi Tan Linfeng Ou Xiaolin Li Caihong Zhong Huilin Huang Ian Max Moller Xianjin Wu Songquan Song 2021Horticulture Research2021,8,1:5

18Abdominal hemorrhage after peritoneal dialysis catheter insertion:A rare cause of luteal rupture:A case report显示文摘BACKGROUND Abdominal hemorrhage is a complication of peritoneal dialysis catheter(PDC)insertion that cannot be neglected,and its causes are mainly related to surgical injury.This article reports a case of massive abdominal hemorrhage that was caused by a rare rupture of corpus luteum shortly after PDC during the initiation of peritoneal dialysis(PD)insertion.CASE SUMMARY A 37-year-old woman was surgically placed a Tenckhoff catheter because of endstage renal disease.On the third postoperative day,the color of the abdominal drainage fluid was pink,and deepened gradually.It turned pale after initiating conservative treatment.On the tenth postoperative day,the color of the abdominal drainage fluid suddenly turned dark red,and the color progressively deepened.The patient’s hemoglobin dropped from 88 g/L to 57 g/L.Abdominal computed tomography(CT)indicated abdominal effusion and a high-density shadow in the abdominal cavity.The surgeon performed a laparotomy and found that the corpus luteum had ruptured on the right side and a left ovarian blood body had formed.The gynecologist repaired the ovary and performed a bilateral oophoroplasty.After the operation,the patient stopped bleeding and hemodialysis was temporarily stopped.PD was resumed after half a month.The patient’s condition improved,and she was discharged 14 d after the laparotomy.CONCLUSION If abdominal hemorrhage occurs in women of childbearing age after PDC insertion,luteal rupture should be considered as the cause.Lin-Wang Gan Qian-Cheng Li Zhao-Lan Yu Li-Ling Zhang Qi Liu Ying Li San-Tao Ou 2021World Journal of Clinical Cases2021,9,22:1
