1The CRISPR/Cas9 induces large genomic fragment deletions of MSTN and phenotypic changes in sheep显示文摘The CRISPR/Cas9 system has been extensively used to engineer genetic loci for the generation of knockouts, insertions, and point mutations in animal models. However, many mutations that have been reported in animals are small insertions or deletions. This study used the CRISPR/Cas9 system to induce large DNA fragment deletions in MSTN via three guide RNAs in sheep. This successfully achieved the precise gene editing of the ovine MSTN gene by injecting both Cas9 m RNA and sg RNAs into embryos at the one-cell stage. Of 10 edited animals, 3 animals(30%) exhibited large genomic fragment deletions(~5 kb). Furthermore, the body weights of these 3 animals were significantly different(P0<0.0001, P15=0.001, P30=0.005, P60=0.027) between lambs with large deletions and wildtype lambs. In addition, the edited lambs were also significantly different(P0<0.0001, P15<0.0001, P30=0.002, P60=0.011) compared with wildtype. These results suggest that the generated MSTN knockout sheep is a reliable and effective animal model for further study. 2Constraint-induced movement therapy promotes motor function recovery and downregulates phosphorylated extracellular regulated protein kinase expression in ischemic brain tissue of rats显示文摘Motor function impairment is a common outcome of stroke.Constraint-induced movement therapy(CIMT)involving intensive use of the impaired limb while restraining the unaffected limb is widely used to overcome the effects of‘learned non-use’and improve limb function after stroke.However,the underlying mechanism of CIMT remains unclear.In the present study,rats were randomly divided into a middle cerebral artery occlusion(model)group,a CIMT+model(CIMT)group,or a sham group.Restriction of the affected limb by plaster cast was performed in the CIMT and sham groups.Compared with the model group,CIMT significantly improved the forelimb functional performance in rats.By western blot assay,the expression of phosphorylated extracellular regulated protein kinase in the bilateral cortex and hippocampi of cerebral ischemic rats in the CIMT group was significantly lower than that in the model group,and was similar to sham group levels.These data suggest that functional recovery after CIMT may be related to decreased expression of phosphorylated extracellular regulated protein kinase in the bilateral cortex and hippocampi.Bei Zhang Qiang He Ying-ying Li Ce Li Yu-long Bai Yong-shan Hu Feng Zhang 2015Neural Regeneration Research2015,10,12:5

4HMGB1 induces secretion of matrix vesicles which participate in microcalcification of atherosclerotic plaques显示文摘AIM: Early calcification of atherosclerotic plaques are colocalized with macrophage and high mobility group box 1( HMGB1),a cytokine associated with biomineralizing process under physiological and pathological conditions. Our study aims to evaluate whether HMGB1 induces ectopic mineralization via promoting the secretion of matrix vesicles( MVs) from macrophages. METHODS: HMGB1 was added to the medium of macrophages,the secretion of MVs in the supernatant was tested by flow cytometry analysis. The mineral deposition in calcifying medium was detected by Alizarin Red staining and von Kossa staining. Transmission electron microscopy showed the formation of hydroxyapatite crystals in MVs. Then we subcutaneous injection into mice with MVs to induce regional mineralization. RESULTS: HMGB1 significantly promoted secretion of MVs from macrophages as raveled by flow cytometry analysis. TNAP activity,considered as a marker of MVs maturation,was higher in HMGB1-induced MVs compared to the control-MVs. HMGB1-MVs also led to mineral deposition in an in vitro MVs-collagen mineralization model. Subcutaneous injection into mice with MVs derived from HMGB1-treated cells showed a greater potential to initiate regional mineralization. Mechanistic experiments revealed that HMGB1 activated neutral sphingomyelinase 2( n SMase2) that involved the receptor for advanced glycation end products( RAGE) and p38MAPK( upstream of n SMase2). Inhibition of n SMase2 with GW4869 or p38 MAPK with SB-239063 prevented MVs secretion and mineral deposition. CONCLUSIONS: HMGB1 induces MVs secretion from macrophages at least in part,via the RAGE / p38 MAPK /n SMase2 signaling pathway. 5Encapsulation of lyophilized platelet-rich fibrin in alginate-hyaluronic acid hydrogel as a novel vascularized substitution for myocardial infarction显示文摘Cardiovascular diseases such as myocardial infarction(MI)are among the major causes of death worldwide.Although intramyocardial injection of hydrogels can effectively enhance the ventricular wall,this approach is limited because of its restriction to the poor vascularization in the infarcted myocardium.Here,we reported a new type of hydrogel composed of alginate(ALG)and hyaluronic acid(HA)with lyophilized platelet-rich fibrin(Ly-PRF)for releasing abundant growth factors to realize their respective functions.The results of in vitro studies demonstrated favorable mechanical property and release ability of ALG-HA with Ly-PRF.When injected into the infarcted myocardium,this composite hydrogel preserved heart function and the Ly-PRF within the hydrogel promoted angiogenesis and increased vascular density in both infarcted and border zone,which rescued the ischemic myocardium.These beneficial effects were also accompanied by macrophage polarization and regulation of myocardial fibrosis.Moreover,the autologous origin of Ly-PRF with ALG-HA hydrogel offers myriad advantages including safety profile,easiness to obtain and cost-effectiveness.Overall,this study demonstrated the versatile therapeutic effects of a novel composite hydrogel ALG-HA with Ly-PRF,which optimizes a promising vascularized substitution strategy for improving cardiac function after MI.Bei Qian Qi Yang Mingliang Wang Shixing Huang Chenyu Jiang Hongpeng Shi Qiang Long Mi Zhou Qiang Zhao Xiaofeng Ye 2022Bioactive Materials2022,7,1:2
6Clinical application of free/total PSA ratio in the diagnosis of prostate cancer in men over 50 years of age with total PSA levels of 2.0-25.0 ng ml^(−1) in Western China显示文摘The goal of this study was to investigate the clinical application of free/total prostate-specific antigen(F/T PSA)ratio,considering the new broad serum total PSA(T-PSA)“gray zone”of 2.0–25.0 ng ml^(−1)in differential diagnosis of prostate cancer(PCa)and benign prostate diseases(BPD)in men over 50 years in Western China.A total of 1655 patients were included,528 with PCa and 1127 with BPD.Serum T-PSA,free PSA(F-PSA),and F/T PSA ratio were analyzed.Receiver operating characteristic curves were used to assess the efficiency of PSA and F/T PSA ratio.There were 47.4%of cancer patients with T-PSA of 2.0–25.0 ng ml^(−1).When T-PSA was 2.0–4.0 ng ml^(−1),4.0–10.0 ng ml^(−1),and 10.0–25.0 ng ml^(−1),the area under the curve(AUC)of F/T PSA ratio was 0.749,0.769,and 0.761,respectively.The best AUC of F/T PSA ratio was 0.811 when T-PSA was 2.0–25.0 ng ml^(−1),with a specificity of 0.732,a sensitivity of 0.788,and an optimal cutoff value of 15.5%.The AUC of F/T PSA ratio in different age groups(50–59 years,60–69 years,70–79 years,and≥80 years)was 0.767,0.806,0.815,and 0.833,respectively,and the best sensitivity(0.857)and specificity(0.802)were observed in patients over 80 years.The T-PSA trend was in accordance with the Gleason score,tumor node metastasis(TNM)stage,and American Joint Committee on Cancer prognosis group.Therefore,the F/T PSA ratio can facilitate the differential diagnosis of PCa and BPD in the broad T-PSA“gray zone”.Serum T-PSA can be a Gleason score and prognostic indicator.Xue-Dan Gao Qiang Miao Jun-Long Zhang Jian-Zhao Zhai Xue-Mei Gui Yi-Han Cai Qian Niu Bei Cai 2022Asian Journal of Andrology2022,24,2:2

10A study of laparoscopic extraperitoneal sigmoid colostomy after abdomino-perineal resection for rectal cancer显示文摘Objective:To established a procedure for laparoscopic extraperitoneal ostomy after abdomino-perineal resection(APR)and study safety aspects and complications.Method:From July 2011 to July 2012,36 patients with low rectal cancer undergoing APR were included in the study and divided into extraperitoneal ostomy group(n=18)and intraperitoneal ostomy group(n=18).Short-and long-term complications were compared between the two groups.All patients were followed up and the median duration was 17 months(range:12-24).Results:The rates of short-term complication related to colostomies were comparable between the two groups,except the rate for stoma edema was higher in the extraperitoneal group(33.3%vs 0%;P=0.008).In the intraperitoneal ostomy group,two patients developed stoma prolapse,one had stoma stenosis,and two had parastomal hernia.In contrast,no long-term complications related to colostomies occurred in the extraperitoneal ostomy group.The rate of long-term complication was lower in the extraperitoneal ostomy group(0%vs 22.2%;P=0.036).Conclusion:The laparoscopic extraperitoneal ostomy is a relatively simple and safe procedure,with fewer long-term complications related to colostomy.However the follow-up period was not too long and needs to be extended.Jin Heiying Du Yonghong Wang Xiaofeng Yao Hang Wu Kunlan Zhang Bei Zhang Jinhao Leng Qiang 2014Gastroenterology Report2014,2,1:1
11Identification and validation of Temozolomide resistance hub gene LncRNA MOCS2-DT in glioblastoma显示文摘Background:Temozolomide(TMZ)is an important adjuvant chemotherapy drug after glioma surgery.However,with the progress of treatment,most patients will gradually develop drug resistance.Long non-coding RNA(lncRNA)in the prediction and reversal of chemotherapy drug resistance possesses great potential.Methods:1.The clinical data and RNA expression matrix of glioma patients were downloaded from the Chinese Glioma Genome Atlas(CGGA),and the clinical data and RNA expression profiles of 47 patients who only received TMZ chemotherapy after surgery were selected;2.WGCNA was performed on RNA expression profiles to find modules highly correlated with survival time and lncRNAs with high connectivity in modules.Core lncRNA were further screened from the GEO database(P<0.05);survival analysis,correlation analysis,enrichment analysis and co-expression analysis were performed;and 3.A TMZ concentration gradient induced glioma-resistant cells(U251TR).Cell proliferation toxicity assays and real-time quantitative PCR were used to verify the core lncRNA and identify the biological characteristics of U251TR.Results:1.According to the WGCNA analysis,it was found that the pink module is highly correlated with survival time and that there are 17 core lncRNAs with high connectivity in the pink module.2.The expression of the core lncRNA MOCS2-DT in the U251 resistant strain was significantly lower than that in the sensitive strain(P<0.05).3.U251TR cells had significantly larger cell bodies with irregular shapes.U251TR cells had a stronger tolerance to TMZ than U251 cells,and the resistance index was about four times higher(P<0.05).4.The expression levels of MGMT and ABCG2 in U251TR cells were significantly higher than those in parental U251 cells(P<0.05),and the expression levels of MOCS2-DT in U251TR cells were significantly lower than those in parental U251 cells(P<0.05).5.The survival time and survival rate of patients with low expression of MOCS2-DT were significantly lower than those of patients with high expression of MOCS2-DT in patients who only received TMZ chemotherapy after surgery.6.The enrichment analysis results show that the mRNA co-expressed with MOCS2-DT is enriched in synapse organization,regulation of membrane potential and signal release,indicating that MOCS2-DT is closely related to these functions.Conclusion:Targeted regulation of MOCS2-DT may reverse the drug resistance of glioma,improve the prognosis of patients and prolong the survival time of patients.Lang Li Qiang Gu Jia-Hao Yao Bei Zhou Fa-Yan Dong Xue-Feng Wang 2023Medical Theory and Hypothesis2023,6,2:0

13One-pot and One-step Cu(0)-mediated Reversible-Deactivation Radical Polymerization of N-Isopropylacrylamide(NIPAM) in Water显示文摘Copper(0)-mediated reversible-deactivation radical polymerization(Cu(0)-mediated RDRP) of the water-soluble monomer Nisopropylacrylamide(NIPAM) has been challenging with the problems of high dispersity, poor control over the molecular weights(MWs) or complex or multi reaction steps, etc. In this work, we report the well-controlled polymerization of NIPAM in water via a facile one-pot and one-step Cu(0)-mediated RDRP. The results of this approach show that the key for kicking off the Cu(0)-mediated NIPAM RDRPs is to ensure sufficient Cu~I at the very beginning, and the key to achieve a well-controlled chain growth is to provide adequate deactivation strength during the polymerization process. For NIPAM, which has a high propagation rate constant, the deactivation control can be effectively enhanced by extra adding deactivator(i.e., Cu~II) to the system. Moreover, a low reaction temperature(4 ℃) is necessary in the controlled synthesis of higher MW poly(Nisopropylacrylamide)(PNIPAM) to avoid the compromise in control caused by the phase transition from its lower critical solution temperature(LCST). 14A Spatial Supply-Demand Imbalance Between Rural Primary and Secondary Schools in Shaanxi Province Based on Road Network Analysis显示文摘It is of great significance to optimize the spatial matching of basic education facilities in rural areas and promote the equalization of urban and rural basic public services and the overall revitalization of rural areas.Based on the theory of spatial accessibility and spatial balance of supply and demand,this paper constructs the evaluation system of rural basic education facilities and layout rationality.In this study,we took 42 primary and secondary schools and 2089 rural areas with concentrated population in six township areas of Hancheng City as the research subjects.We used ArcGIS network analysis,population coupling model,and other methods,from the administrative scale and rural population concentration area scale for evaluation.The accessibility of basic education facilities and its coupling relationship with the spatial distribution of student population describe the spatial and geographical pattern of basic education facilities in rural areas.The results show that the accessibility of rural administrative scale and the spatial balance of supply and demand of population concentration area scale are associated with great problems,the differences between towns are obvious,and the overall level of the best performing towns is still low.On the scale of population concentration area,the overall coupling degree is too low,the'dislocation'distribution of educational facilities and student population is obvious,and the matching degree of the student population and school in different towns is significantly different,showing a more obvious'core-periphery'spatial structure characteristics centered on the eastern and southeast regions.The terrain characteristics of rural areas in Shaanxi province are prominent,and the accessibility of the road network is particularly significant.Therefore,optimizing the construction of rural roads and optimizing school layout according to local conditions will become an important solution to promote the balanced allocation of basic education resources in Shaanxi province.Ziwei Qiang Bei Jin Shiyuan Xu 2022Journal of Contemporary Educational Research2022,6,2:0

17Modeling iron release from cast iron pipes in an urban water distribution system caused by source water switch显示文摘Significant iron release from cast iron pipes in water distribution systems(WDSs),which usually occurs during the source water switch period,is a great concern of water utilities because of the potential occurrence of'red water'and customer complaints.This study developed a new method which combined in-situ water stagnation experiments with mathematical models and numerical simulations to predict the iron release caused by source water switch.In-situ water stagnation experiments were conducted to determine the total iron accumulation in nine cast iron pipes in-service in Beijing when switching the local water to tre ated Danjiangkou Reservior water.Results showe d that the difference in the concentration increment of total iron in 24 hr(ΔCITI,24),i.e.short-term iron release,caused by source water switch was mainly dependent on the difference in the key quality parameters(pH,hardness,nitrate,Larson Ratio and dissolved oxygen(DO))between the two source waters.The iron release rate(RFe)after switch,i.e.long-term iron release,was closely related to the pipe properties as well as the DO and total residual chlorine(TRC)concentrations.Mathematical models ofΔCITI,24 and RFe were developed to quantitatively reveal the relationship between iron release and the key quality parameters.The RFe model could successfully combine with EPANET-MSX,a numerical simulator of water quality for WDSs to extend the iron release modeling from pipe level to network level.The new method is applicable to predicting iron release during source water switch,thus facilitating water utilities to take preventive actions to avoid'red water'.Xiaodan Lin Qiang Xu Yuxian Li Bei Zhao Li Li Zhimin Qiang 2021Journal of Environmental Sciences2021,33,12:0
