1Graft-accelerated virus-induced gene silencing facilitates functional genomics in rose flowers显示文摘Rose has emerged as a model ornamental plant for studies of flower development, senescence, and morphology, as well as the metabolism of floral fragrances and colors.Virus-induced gene silencing(VIGS) has long been used in functional genomics studies of rose by vacuum infiltration of cuttings or seedlings with an Agrobacterium suspension carrying TRV-derived vectors. However, VIGS in rose flowers remains a challenge because of its low efficiency and long time to establish silencing. Here we present a novel and rapid VIGS method that can be used to analyze gene function in rose,called ‘graft-accelerated VIGS', where axil ary sprouts are cut from the rose plant and vacuum infiltrated with Agrobacterium. The inoculated scions are then grafted back onto the plants to flower and silencing phenotypes can be observed within 5 weeks, post-infiltration. Using this new method, we successfully silenced expression of the RhDFR_1, RhA G, and RhNUDX_1 in rose flowers, and affected their color, petal number, as well as fragrance, respectively. This grafting method will facilitate high-throughput functional analysis of genes in rose flowers. 2Inheritance of Perpetual Blooming in Rosa chinensis ‘Old Blush'显示文摘Perpetual blooming is one of the most important biological and economical traits in modern rose, while the genetic basis underlining the control of this trait is poorly investigated. With an aim in dissecting the genetic determinism of perpetual blooming, we developed six rose populations(OB, W, F1, F2, BC1 OB and BC1W) derived from a WOB population [interspecific diploid hybridization between Rosa chinensis ‘Old Blush'(OB) and R. wichuriana ‘Basye's Thornless'(W)]. Perpetual blooming is absent both in a F1 population with 296 individuals and a BC1 W population(W as the backcross parent) with 150 individuals. However, the perpetual blooming trait showed a typical 3︰1 segregation in a backcross population BC1 OB with OB as the backcross parent. In this population with 300 individuals, 83 plants had the perpetual blooming phenotype while the other 217 featured non-perpetual blooming, indicating that the perpetual blooming trait is very likely controlled by two recessive genes in R. chinensis(rpb1 and rpb2). In the present paper, numerical analysis of the melt flow in the Czochralski (CZ) crystal growth configuration, the three dimensional (3D) modeling, the simulation of melt flow under the magnetic field, the inverse modeling and the time-dependent simulation are reviewed. 4Monitoring the Cultivated Slope Land in the Three Gorges Reservoir Area Based on Remote Sensing and GIS显示文摘By means of combining auto-extraction with manual interpretation, the current distribution information about cultivated land is obtained. The distribution information of 1992 is extracted from the dynamic polygons of 2002. The monitoring mini-system of the cultivated slope land is established. In the system, detailed surveys, focused on the resources of cultivated slope land, are carried out. The results indicate that the area of the cultivated slope land is very large. Meanwhile, there are lots of cultivated steep slopes with gradient above 35°.The areas of steep land cultivated had been slowly reduced from 1992 to 2002. At the same time, the pressures of returning farm land to forestry are great in all counties. The conflicts between population growth, insufficient grain supply and stagnant economic development sharpen increasingly. 5Comparison of measurements and simulation results in 300mm CZ silicon crystal growth显示文摘A special thermal modeling tool, CrysVUn, which was developed by Crystal Growth Laboratory (CGL) of Fraunhofer Institute of Integrated Systems and Devices Technology in Erlangen of Germany, was used for numerical analysis of growth interface situation. The heat transportation, argon flow and melt convection have been considered. Cauchy's first and second laws of motion have been the governing partial equations for stress calculation. 6Effect of thermal shield and gas flow on thermal elastic stresses in 300mm silicon crystal显示文摘The thermal elastic stresses induced in 300 mm Si crystal may be great troubles because it can incur the generation of dislocations and undesirable excessive residual stresses. A special thermal modeling tool, CrysVUn, was used for numerical analysis of thermal elastic stresses and stress distribution of 300 mm Si crystal under the consideration of different thermal shields and gas flow conditions. The adopted governing partial equations for stress calculation are Cauchy′s first and second laws of motion. It is demonstrated that the presence and shape of thermal shield, the gas pressure and velocity can strongly affect von Mises stress distribution in Si crystal. With steep-wall shield, however, the maximal stress and ratio of high stress area are relatively low. With slope-wall shield or without shield, both maximal stress and ratio of high stress area are increased in evidence. Whether thermal shields are used or not, the increase of gas flow velocity could raise the stress level. In contrast, the increase of gas pressure cannot result in so significant effect. 7Remote Sense and GIS-Based Division of Landslide Hazard Degree in Wanzhou District of the Three Gorges Reservoir Area显示文摘An evaluation model divided landslide hazard degrees in Wanzhou District of Three Gorges Reservoir Area. The model was established by GIS techniques and took land use/cover, stratum characters, slope aspect, slope gradient, elevation difference and slope shape as evaluation factors. The data of land use/cover were obtained by remote sensing, and the weights of the factors mentioned above were established by the analytic hierarchy process (AHP). The results indicate, low danger areas in the studied area account for 66.51%, and high danger areas and very high danger areas occupy 1/3 of the total area. The regions of high and very high danger are mainly located around the urban area of Wanzhou District and on the banks of the Yangtze River with a relatively large area , where collapse and landslide directly threats densely populated areas and Three Gorges Reservoir. Slope destabilization, if occurs, will bring huge loss to social economy. 8Overexpression of the mTERT gene by adenoviral vectors promotes the proliferation of neuronal stem cells in vitro and stimulates neurogenesis in the hippocampus of mice显示文摘We sought to construct the adenoviral vector carrying the gene encoding mouse telomerase reverse transcriptase(mTERT),as well as detect its expression and effect on the proliferation of neuronal stem cells.mTERT was am-plified by RT-PCR and then the eukaryotic expression vector of pDC-EGFP-TERT was constructed.After DNA sequence analysis,we detected that there were 293 cells transfected with pDC-EGFP-TERT and helper adenovirus plasmid pBHG lox ΔE1,and three Cre using Lipofectamine 2000 mediation,named Ad-mTERT-GFP,to pack-age adenoviral particles.The Ad-mTERT-GFP was used to infect neuronal stem cells and then the expression and activity of mTERT were detected.In addition,Bromodeoxyuridine labeling test identified the impact of mTERT overexpression on proliferation of neuronal stem cells.The recombinant adenoviral vector confirmed that mTERT was successfully constructed.Overexpression of mTERT stimulated the proliferation of neuronal stem cells both in vitro and in vivo.mTERT overexpression via adenoviral vector carrying mTERT cDNA upregulated the ability of proliferation in neuronal stem cells.Mengying Liu Yao Hu Lijuan Zhu Chen Chen Yu Zhang Weixiang Sun Qigang Zhou 2012The Journal of Biomedical Research2012,26,5:1
9Microstructure of flow pattern defects in boron-doped Czochralski-grown silicon显示文摘The morphology and microstructure of flow pattern defects (FPDs) in lightly boron-doped Czochralski-grown silicon (Cz-Si) crystals were investigated using optical microscopy and atomic force microscopy. The experimental results showed that the morphology of FPDs was parabola-like with several steps. Single-type and dual-type voids were found on the tip of FPDs and two heaves exist on the left and right sides of the void. All the results have proved that FPDs were void-type defects. 10Optimization of heat shield for single silicon crystal growth by using numerical simulation显示文摘In integrated circuit-grade single silicon Czochralski growth, the position and material of heat shield are main parameters affecting the heat exchange and crystal growth condition. By optimizing the above parameters, we attempted to increase the growth rate and crystal quality. Numerical simulation proved to verify the results before and after optimization. 11Numerical simulation of effects of cusp magnetic field on oxygen concentration of 300 mm CZ-Si显示文摘In magnetic Czochralski (MCZ) silicon growth, the distance and diameter of the electrified coils may affect the magnetic field intensity and melt flow. By changing the above parameters, the optimum geometric configuration of the coils was attempted. Through analyses of the oxygen concentration distribution of the crystal/melt interface, axial and radial velocity distribution of melt and the magnetic field intensity in the melt, it is found that smaller diameter of coils contributes to reducing the needed current intensity and production costs. 15CATase-immobilized hydrogel platform molded by photo-enzyme coupling polymerization for effectively preventing postoperative abdominal adhesion显示文摘Redox homeostasis,which is regulated by enzymes acting as regulatory valves,is crucial for maintaining the proper functioning of biomolecules and a stable microenvironment for physiological processes by modulating the homeostasis of reactive oxygen species(ROS).Antioxidant enzymes in biocatalysis are used in the prevention or treatment of oxidative stress-related disease by counteracting the harmful effects of ROS.However,designing a system that can efficiently immobilize antioxidant enzymes with high catalytic activity and stability is still challenging.Bioinspired by photo-biocatalysis,a novel and effective catalase(CATase)-immobilized hydrogel platform has been developed by the proposed photo-enzymatic coupled radical polymerization strategy of the visible light coupling with the porphyrin-centered CATase.The higher catalytic stability and activity can therefore be achieved due to the preferential polymerization of CATase-immobilized hydrogel platform with a favorable three-dimensional network of enhanced coupling efficacy between light and enzymes.The mechanisms of free radical-initiated polymerization as well as the antioxidant cycle in the photo-CATase coupling system have been explored.Intriguingly,the CATase-immobilized hydrogel platform affords an unprecedented antioxidant ability to scavenge ROS and provide an effective cellular protection mechanism against external oxidative stress.Additionally,the CATase-immobilized hydrogel platform can effectively prevent peritoneal adhesion by reducing the expression of inflammatory cytokines.Therefore,the novel CATase-immobilized hydrogel platform is a potential candidate for physical barriers that effectively prevent postoperative adhesion formation,offering a new anti-adhesion strategy for clinical applications.Min Hu Mingyue Shi Xia Wang Yujing Tang Dingze Zhou Kai Pan Qigang Wang 2023Science China Chemistry2023,66,9:0
16Comparative transcriptomic and metabonomic analysis revealed the relationships between biosynthesis of volatiles and flavonoid metabolites in Rosa rugosa显示文摘Rosa rugosa is not only cultivated as a landscaping plant,but also used in cosmetics,the medical and food industries.However,little information is currently available on the gene regulatory networks involved in its scent and color biosynthesis and metabolism.In this study,R.rugosa Thunb.f.rosea Rehd with red petals(RR)and its white petal variant(WR),were used to study the molecular mechanisms in flower color and scent.Sixty-five differential flavonoid metabolites and 15 volatiles were found to have significant differences between RR and WR.Correspondingly,the key regulators(MYB-bHLH-WD40)of anthocyanin synthesis pathway and their structural genes involved in flavonoid biosynthesis,benzenoid/phenylpropanoid biosynthesis,terpenoid biosynthesis pathways were also found to be differentially expressed by comparative transcriptome.Further,qPCR permitted the identification of some transcripts encoding proteins that were putatively associated with scent and color biosynthesis in roses.Particularly,the results showed that the ACT gene(encoding CoA geraniol/citronellol acetyltransferase,GeneID:112190420),which expressed lower in WR,was involved in three pathways:flavonoid biosynthesis,phenylpropanoid biosynthesis and terpenoid biosynthesis,however,GT5(anthocyanin glycosylation gene,GeneID:112186660),expressed higher in WR,was involved in both flavonoid and phenylpropanoid biosynthesis pathways.These results suggested that ACT and GT5 might play important roles in regulating the relationship of color pigmentation and volatile emission.Dedang Feng Hao Zhang Xianqin Qiu Hongying Jian Qigang Wang Ningning Zhou Yu Ye Jun Lu Huijun Yan Kaixue Tang 2021Ornamental Plant Research2021,1,1:0
17Current Situation of Land Use in Three-gorges Reservoir Region in 2010显示文摘Land is rare natural resource.Production and construction of all sectors in a region must be based on land.Thus,overall research and analysis on current situation of land use can reveal scope,depth and reasonableness of land use,is helpful for analyzing internal mechanism land use change,and can reflect production scale,level and characteristics,and also can provide basis for optimization and adjustment of land use structure.Based on RS and GIS technologies,with the aid of TM image data of the Three-gorges Reservoir Region in 2010,the data of current situation of land use in Three-gorges Reservoir Region was obtained,and current land use situation was analyzed using geographimetrics and landscape ecology methods.Results show that since natural,social and economic conditions are different,land type diversity,combination type and location index of counties in the Three-gorges Reservoir Region are varied.In the land use diversity index,Xingshan County has the most single land use type(mainly forest land); in land use degree,Yuzhong District has the highest land use degree; in the integrated index of land use,Changshou District has the lowest integrated index of land use.This study is expected to provide reference and basis for formulating policies of protecting ecological environment of the Three-gorges Reservoir Region.Fuhai WANG Qigang ZHOU Fei YANG Pengwu YANG Yao WANG 2014Asian Agricultural Research2014,6,7:0
18Genome-wide identification and evolutionary analysis of MLO gene family in Rosaceae plants显示文摘Mutants lacking wild-type MLO(Mildew resistance Locus O)proteins show broad-spectrum resistance to the powdery mildew fungus,and dysregulated cell death control,with spontaneous cell death in response to developmental or abiotic stimuli.In order to understand the evolution and divergence patterns of the MLO gene family in Rosaceae plants,we analysed systematically genome-wide data from Fragaria vesca,Prunus persica,Prunus mume,Malus domestica,Pyrus bretschneideri and Rubus occidentalis based on bioinformatics methods.Using three phylogenetic methods(the neighbour-joining,maximum likelihood,and Bayesian methods),we identified 117 MLO genes from 6 Rosaceae species.The results of all three phylogenetic analysis methods supported that these genes were divided into six clades.Conserved motif analysis found that only motif 2 was present in all MLO proteins and had 3 nearly invariant amino acid residues.The findings indicated that motif 2 might be shared by the MLO gene family.The structural features of these genes showed large variations in sequence length among different species,although the lengths and the numbers of exons exhibited high degrees of similarity.Selective pressure analysis showed extremely significant differences in all 6 clades,with 2,1,and 1 site(s)under significant positive selection detected in clades III,IV,and VI,respectively.These positive selection sites were important driving forces for the promotion of the functional differentiation of the MLO genes.Functional divergence analysis showed that the significantly divergent sites were located within the domains of the MLO genes.Functional distance analysis showed that the clade V had more conservative functions and might have retained more original functions during the evolutionary process.However,clade I may have undergone extensive altered functional constraints as a specialised functional role.Moreover,the most original function of the MLO genes in Rosaceae could be related to the evolution of their resistance to powdery mildew,which then gradually evolved into functions such as the regulation of flower development,the control of root morphology,and seed evolution due to the different evolutionary rates after gene duplication.These results provide a theoretical basis for further studies of the molecular evolutionary patterns of the plant MLO gene family.Yongxian Tian Qigang Wang Hao Zhang Ningning Zhou Huijun Yan Hongying Jian Shubin Li Guisheng Xiang Kaixue Tang Xianqin Qiu 2022Horticultural Plant Journal2022,8,1:0
19GIS-Based Spatial Analysis and Modeling for Landslide Hazard Assessment:A Case Study in Upper Minjiang River Basin显示文摘By analyzing the topographic features of past landslides since 1980s and the main land-cover types (including change information) in landslide-prone area, modeled spatial distribution of landslide hazard in upper Minjiang River Basin was studied based on spatial analysis of GIS in this paper. Results of GIS analysis showed that landslide occurrence in this region closely related to topographic feature. Most areas with high hazard probability were deep-sheared gorge. Most of them in investigation occurred assembly in areas with elevation lower than 3 000 m, due to fragile topographic conditions and intensive human disturbances. Land-cover type, including its change information, was likely an important environmental factor to trigger landslide.
