2Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs.Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu 2020Cell Research2020,30,4:81
3Effects of different resuscitation fluid on severe acute pancreatitis显示文摘AIM: To compare effects of different resuscitation fluid on microcirculation, inflammation, intestinal barrier and clinical results in severe acute pancreatitis (SAP). METHODS: One hundred and twenty patients with SAP were enrolled at the Pancreatic Disease Institute between January 2007 and March 2010. The patients were randomly treated with normal saline (NS group), combination of normal saline and hydroxyethyl starch (HES) (SH group), combination of normal saline, hydroxyethyl starch and glutamine (SHG group) in resuscitation. The ratio of normal saline to HES in the SH and SHG groups was 3:1. The glutamine (20% glutamine dipeptide, 100 mL/d) was supplemented into the resuscitation liquid in the SHG group. Complications and outcomes including respiratory and abdominal infection, sepsis, abdominal hemorrhage, intra-abdominal hypertension, abdominal compartment syndrome (ACS), renal failure, acute respiratory distress syndrome (ARDS), multiple organ dysfunction syndrome (MODS), operation intervention, length of intensive care unit stay, length of hospital stay, and mortality at 60 d were compared. Moreover, blood oxygen saturation (SpO 2 ), gastric intramucosal pH value (pHi), intra-abdominal pressure (IAP), inflammation cytokines, urine lactulose/mannitol (L/M) ratio, and serum endotoxin were investigated to evaluate the inflammatory reaction and gut barrier. RESULTS: Compared to the NS group, patients in the SH and SHG groups accessed the endpoint more quickly (3.9 ± 0.23 d and 4.1 ± 0.21 d vs 5.8 ± 0.25 d, P < 0.05) with less fluid volume (67.26 ± 28.53 mL/kg/d, 61.79 ± 27.61 mL/kg per day vs 85.23 ± 21.27 mL/kg per day, P < 0.05). Compared to the NS group, incidence of renal dysfunction, ARDS, MODS and ACS in the SH and SHG groups was obviously lower. Furthermore, incidence of respiratory and abdominal infection was significantly decreased in the SH and SHG groups, while no significant difference in sepsis was seen. Moreover, less operation time was needed in the SH and SHG group than the NS group, but the difference was not significant. The mortality did not differ significantly among these groups. Blood SpO 2 and gastric mucosal pHi in the SH and SHG groups increased more quickly than in the NS group, while IAP was significantly decreased in the SH and SHG group. Moreover, the serum tumor necrosis factor-α, interleukin-8 and C-reactive protein levels in the SH and SHG groups were obviously lower than in the NS group at each time point. 4Progress in additive manufacturing on new materials: A review显示文摘Recent efforts and advances in additive manufacturing(AM) on different types of new materials are presented and reviewed. Special attention is paid to the material design of cladding layers, the choice of feedstock materials, the metallurgical behavior and synthesis principle during the AM process, and the resulted microstructures and properties, as well as the relationship between these factors. Thereafter,the trend of development in the future is forecasted, including: Effects of the particles size and size distribution of powders; Approaches for producing fine microstructures; Opportunities for creating new materials by AM; Wide applications in reconditioning of damaged components; Challenges for deep understanding and applications of the AMed new materials. 5Solution structure of the RNA recognition domain of METTL3-METTL14 N^6-methyladenosine methyltransferase显示文摘N^6-methyladenosine(m6A),a ubiquitous RNA modification,is installed by METTL3-METTL14 complex.The structure of the heterodimeric complex between the methyltransferase domains(MTDs)of METTL3 and METTL14 has been previously determined.However,the MTDs alone possess no enzymatic activity.Here we present the solution structure for the zinc finger domain(ZFD)of METTL3,the inclusion of which fulfills the methyltransferase activity of METTL3-METTL14.We show that the ZFD specifically binds to an RNA containing 5'-GGACU-3'consensus sequence,but does not to one without.The ZFD thus serves as the target recognition domain,a structural feature previously shown for DNA methyltransferases,and cooperates with the MTDs of METTL3-METTL14 for catalysis.However,the interaction between the ZFD and the specific RNA is extremely weak,with the binding affinity at several hundred micromolar under physiological conditions.The ZFD contains two CCCH-type zinc fingers connected by an anti-parallel P-sheet.Mutational analysis and NMR titrations have mapped the functional interface to a contiguous surface.As a division of labor,the RNA-binding interface comprises basic residues from zinc finger 1 and hydrophobic residues fromβ-sheet and zinc finger 2.Further we show that the linker between the ZFD and MTD of METTL3 is flexible but partially folded,which may permit the cooperation between the two domains during catalysis.Together,the structural characterization of METTL3 ZFD paves the way to elucidate the atomic details of the entire process of RNA m6A modification.Jinbo Huang Xu Dong Zhou Gong Ling-Yun Qin Shuai Yang Yue-Ling Zhu Xiang Wang Delin Zhang Tingting Zou Ping Yin Chun Tang 2019Protein & Cell2019,10,4:11
6Domino instability effect of surrounding rock-coal pillars in a room-and-pillar gob显示文摘To discuss the domino instability effect and large area roof falling and roof accidents of surrounding rockcoal pillars in a room-and-pillar gob,the equilibrium equation for a roof-coal pillar-floor system with the influence of mining floor was developed based on the engineering conditions of the surrounding rock in a room-and-pillar gob in the 3^(-2)coal seam of Tanggonggou mine.The conditions of system instability and the relationship between system stability and system stiffness were analyzed from an energetic point of view.Numerical simulation using the discrete element software UDEC was also carried out to simulate conditions causing the domino effect on surrounding rock-coal pillars in a 3^(-2)room-and-pillar gob.The results show that:if we want the system to destabilize,the collective energy in roof-and-floor must be larger than that in the coal pillar.When the stiffness of the coal pillars and the roof-and-floor are both greater than zero,the system is stable.When the stiffness of the coal pillars is negative but the summed stiffness of the coal pillars and roof-and-floor is larger than or equal to zero,the system is statically destroyed.When the sum of the coal pillars and the roof-floor stiffness is negative,the system suffers from severe damages.For equal advance distances of the coal mining face,the wider coal pillars can reduce the probability of domino type instability.Conversely,the smaller width pillars can increase the instability probability.Domino type instability of surrounding rock-coal pillars is predicted to be unlikely when the width of coal pillars is not less than 8 m.Li Chong Xu Jinhai Wang Zhongliang Qin Shuai 2013International Journal of Mining Science and Technology2013,23,6:7
7Degradation and mineralization of aniline by O_3/Fenton process enhanced using high-gravity technology显示文摘The degradation and mineralization of aniline(AN) using ozone combined with Fenton reagent(O_3/Fenton) in a rotating packed bed(RPB) was proposed in this study, and the process(RPB-O_3/Fenton) was compared with conventional O_3/Fenton in a stirred tank reactor(STR-O_3/Fenton) or single ozonation in an RPB(RPB-O_3). Effects of high gravity factor, H_2O_2 dosage, H_2O_2 dosing method and initial p H on the AN mineralization efficiency were investigated in the RPB-O_3/Fenton process. In addition, the behavior of Fe(II) was monitored at different H2 O2 dosing methods and p H values. Finally, the optimal operation conditions were determined with high gravity factor of100, initial pH of 5, Fe(II) concentration of 0.8 mmol·L^(-1) and H_2O_2 dosage of 2.5 ml. Under these conditions, for aniline wastewater at the volume of 1 L and concentration of 200 mg·L^(-1), a fast and thorough decay of AN was conducted in 10 min, and the TOC removal efficiency reached 89% in 60 min. The main intermediates of p-benzoquinone, nitrobenzene, maleic acid and oxalic acid were identified by liquid chromatography/mass spectroscopy(LC/MS), and the degradation pathways of AN in RPB-O_3/Fenton system were proposed based on experimental evidence. 9Discovery and implications of catenary-bead subaqueous dunes显示文摘Measurements of topography at the segment of bifurcation between the South Channel and North Channel in the Yangtze Estuary were conducted, and a new type of subaqueous dune was discovered. This structure, newly defined as a catenary-bead dune, consists of a catenary dune and its associated elliptical pit bedform. Based on this finding, the nomenclature of 'morphology of dune associated with accompanying bedform' is first proposed. The measured data indicate a mean height and wavelength of 1.29 m and 31.89 m, respectively; wavelength/height ratio(L/H) of 14 to 56; and elliptical pits of mean and maximum depth 0.98 m and 1.98 m, respectively. Flow information was obtained using an Acoustic Doppler Current Profile(ADCP), and the bed material components were gathered with a bottom sampler. The results show mean flood and ebb velocities of 0.27 and 0.78 m s?1, respectively, with shorter duration of flood tide than ebb tide. The silt, very fine sand, and fine sand fractions were within the ranges 21.6–23.4%, 28.2–32.2%, and 39.7–41.6%, respectively, revealing complex bed material composition. Water depth at the study site varies from 13 to 17 m. This finding will enrich the study of dunes and provide important data for geomorphological research. 11Microstructure and Mechanical Properties of Mg–3Al–Zn Magnesium Alloy Sheet by Hot Shear Spinning显示文摘Hot shear spinning experiments with Mg–3.0 Al–1.0 Zn–0.5 Mn(AZ31 B, wt%) magnesium alloy sheets were conducted at various temperatures, spindle speeds and feed ratios to investigate the effects of these processing parameters on the microstructure, crystallographic texture and mechanical properties. The AZ31 B sheet displayed good shear formability at temperatures from 473 to 673 K, spindle speeds from 300 to 600 rev/min and feed ratios from 0.1 to 0.5 mm/rev. During the dynamic recrystallization process, the grain size and texture were affected by the deformation temperature of the hot shear spinning process. Each of the spun sheets presented a strong basal texture, and the c-axis of most of the grains was parallel to the normal direction. The optimal hot shear spinning parameters were determined to be a temperature of 473 K, a spindle speed of 300 rev/min and a feed ratio of 0.1 mm/rev. The yield strength, ultimate tensile strength and elongation in the rolled direction reached 221 MPa, 288 MPa and 14.1%, and those in the transverse direction reached 205 MPa, 280 MPa and 12.4%, respectively. 12Rapid Identification of Legionella Pathogenicity by Surface-Enhanced Raman Spectroscopy显示文摘Objective To establish Surface-enhanced Raman Spectroscopy(SERS) can be used as a rapid and reliable method to distinguish virulent strain and mild strain of L. pneumophila. Methods We isolated and characterized of bacterial strains from ATCC and water samples strains, while we analyzed data from SERS technology using gold nanoparticles as a base and cell infections were employed to rapidly detect L. pneumophila strains. Origin 8.0 was used to collect Raman spectra, smooth and homogenize data, and to contrast spectra. Principal component analysis(PCA) was conducted to discriminate differences between groups using the multivariate analysis package Py Chem 3.0.5. Results Our results indicated that the peaks of high virulence strains reached ≥4000. This criterion was verified by subsequent cell experiments. In addition, we also conducted SERS rapid identification on the virulence of several collected clinical strains and obtained accurate results. 13Effects of Scutellaria Baicalensis on Activity and Biofilm Formation of Klebsiella Pneumoniae显示文摘Objective To explore the effects of Scutellaria baicalensis on activity and biofilm formation of Klebsiella pneumonia(Kp).Methods The broth and agar dilution methods were carried out to determine minimum inhibitory concentration and minimum bactericidal concentration of Scutellaria baicalensis for TW518.VITEK-32 system was used to assay TW518 susceptibility to antibiotics.Kp biofilms were formed in vitro and stained with Bac Light Live/Dead stain.The class integron geneⅠ1 m RNA expression was analyzed with RT-PCR.Results The minimum inhibitory concentration of Scutellaria baicalensis on TW518 identified as a Kp colony was 32 mg/ml,and minimum bactericidal concentration was 64 mg/ml.Scutellaria baicalensis and broad-spectrum penicillin,cephalosporin,quinolones,or beta-lactamase had synergistic bactericidal effects.Biofilm formation activity of Kp treated with Scutellaria baicalensis was significantly lower than that of the control group.And class integron geneⅠ1 m RNA expression of TW518 was significantly inhibited by Scutellaria baicalensis.Conclusions Scutellaria baicalensis has sterilization effect on Kp,and Scutellaria baicalensis could effectively inhibit Kp biofilm formation with prolonged treatment.Scutellaria baicalensis might inhibit Kp biofilm formation through down-regulating integron geneⅠ1 expression.Wei Chen Bei Li Shuai Li Yi-wen Ou Qin Ou 2016Chinese Medical Sciences Journal2016,31,3:3
14Relationship Between Perisylvian Essential Language Sites and Arcuate Fasciculus in the Left Hemisphere of Healthy Adults显示文摘Essential language sites and the arcuate fasciculus(AF) have been extensively researched. However, the relationship between them remains insufficiently studied,especially in healthy people. Navigated transcranial magnetic stimulation(n TMS) is increasingly used in language mapping. While enjoying the advantage of non-invasiveness, it is also capable of inducing a virtual lesion in the brain. Thus, it offers the possibility of using the virtuallesion method to study the healthy brain. This study combined n TMS and diffusion tensor imaging(DTI)tractography to investigate the relationship between essential language sites and the AF in 30 healthy right-handedvolunteers. A total of 143 essential language sites were identified using n TMS, and a total of 175 AF terminations were identified using DTI tractography. Sixty-six sites had a direct correlation with the AF, accounting for 46% of the total essential language sites. Forty-seven AF terminations harbored essential language sites, accounting for 27% of the total AF terminations. Upon data rendering to the cortical parcellation system, a region-related heterogeneity of the correlation rate was found. 16Epidemiological Characteristics of Plague in the Meriones unguiculatus Plague Focus--Inner Mongolia Autonomous Region,China,1950−2019显示文摘From 1950 to 2019,a total of 267 plague cases in humans were reported in the Inner Mongolia Autonomous Region with 133 deaths and 10,710 Yersinia pestis isolates;21 of these cases were reported from the Meriones unguiculatus(M.unguiculatus)plague focus with 6 deaths and 6,771 isolates.According to the plague situation in the Inner Mongolia Autonomous Region and the implementation of local preventive measures,the prevalence of plague in humans in the M.unguiculatus plague focus of the Inner Mongolia Autonomous Region could be divided into four stages.The first stage was the initial stage of plague prevention and control(1950–1959),with 0.80 cases annually and a case fatality rate of 37.50%.The second stage was the plague eradication stage(1960–1979),with 0.15 cases annually and a case fatality rate of 33.33%.The third stage was the plague surveillance stage(1980-1999),with 0.25 cases annually and a case fatality rate of 20.00%.The fourth stage is the comprehensive prevention and control stage under the emergency system(2000-2019),with 0.25 cases annually and a case fatality rate of 20.00%.The surveillance of rodent density(1981-2019)and studies on plague-related factors among M.unguiculatus have shown that the higher the M.unguiculatus density,the higher the nocturnal rodent capture rate(r=0.670,p<0.05)and the higher the indirect hemagglutination assay(IHA)positive rate of M.unguiculatus(r=0.344,p<0.05);the higher the percentage of hosts infected,the lower the M.unguiculatus density(r=-0.361,p<0.05)and the lower the IHA positive rate of M.unguiculatus(r=-0.337,p<0.05);the higher the percentage of nests infected with fleas,the lower the IHA positive rate of M.unguiculatus(r=-0.348,p<0.05).Together,these results suggest that it is necessary to simultaneously monitor the pathogens,serology,and vector index of M.unguiculatus to accurately reflect the plague prevalence among local animals.Although bubonic plague is the main plague type of the M.unguiculatus plague focus,severe pneumonic plague or septic plague may be secondary when the bubonic plague is misdiagnosed or not treated in time.In addition,the plague in animals is relatively virulent in the M.unguiculatus plague focus,and the risk of spreading to humans is higher.For plague in the Inner Mongolia Autonomous Region,comprehensive control efforts should be aimed at the M.unguiculatus focus,covering host animals,vector insects,and humans.Boxi Liu Dayu Zhang Yuhuang Chen Zhaokai He Jun Liu Dongyue Lyu Weiwei Wu Ran Duan Shuai Qin Junrong Liang Huaiqi Jing Xin Wang 2020China CDC weekly2020,2,49:3

18Degradation of Nitrobenzene Wastewater via Iron/Carbon Micro-electrolysis Enhanced by Ultrasound Coupled with Hydrogen Peroxide显示文摘The zero valent iron/granular active carbon(ZVI/GAC) micro-electrolysis enhanced by ultrasound(US) coupled with hydrogen peroxide(H_2O_2) was investigated for the deep degradation of nitrobenzene-containing wastewater. The results of scanning electron microscopy-energy dispersive X-rays analysis(SEM-EDS) demonstrated that continuously accelerated regeneration of ZVI and GAC in situ by US could improve the process for converting nitrobenzene(NB) to aniline(AN). H_2O_2 was decomposed catalytically by the byproduct Fe^(2+) ions generated in the micro-electrolysis process to hydroxyl radicals and the organic pollutants in the wastewater were finally mineralized to CO2 and H2O. Effects of the ZVI dosage, the ZVI/GAC mass ratio, the initial pH value and the H_2O_2 dosage on the efficiency for degradation of NB were studied in these experiments. The optimal operating conditions covered a ZVI dosage of 15 g/L, a ZVI/GAC mass ratio of 1:2,an initial pH value of 3 and a H_2O_2 dosage of 4 mL. In this case, the NB removal efficiency reached 97.72% and the total organic carbon(TOC) removal efficiency reached 73.42% at a NB concentration of 300 mg/L. The reduction of NB by USZVI/GAC followed the pseudo-first-order kinetics model, and the pseudo-first-order rate constants were given at different initial pH values.
