1QTG-Seq Accelerates QTL Fine Mapping through QTL Partitioning and Whole-Genome Sequencing of Bulked Segregant Samples显示文摘Deciphering the genetic mechanisms underlying agronomic traits is of great importance for crop improvement. Most of these traits are controlled by multiple quantitative trait loci (QTLs), and identifying the underlying genes by conventional QTL fine-mapping is time-consuming and labor-intensive. Here, we devised a new method, named quantitative trait gene sequencing (QTG-seq), to accelerate QTL fine-mapping. QTGseq combines QTL partitioning to convert a quantitative trait into a near-qualitative trait, sequencing of bulked segregant pools from a large segregating population, and the use of a robust new algorithm for identifying candidate genes. Using QTG-seq, we fine-mapped a plant-height QTL in maize (Zea mays L.), qPH7, to a 300-kb genomic interval and verified that a gene encoding an NF-YC transcription factor was the functional gene. Functional analysis suggested that qPH7-encoding protein might influence plant height by interacting with a CO-like protein and an AP2 domain-containing protein. Selection footprint ana卜 ysis indicated that qPH7 was subject to strong selection during maize improvement. 2Natural products as LSD1 inhibitors for cancer therapy显示文摘Natural products generally fall into the biologically relevant chemical space and always possess novel biological activities, thus making them a rich source of lead compounds for new drug discovery. With the recent technological advances, natural product-based drug discovery is now reaching a new era. Natural products have also shown promise in epigenetic drug discovery, some of them have advanced into clinical trials or are presently being used in clinic. The histone lysine specific demethylase1(LSD1), an important class of histone demethylases, has fundamental roles in the development of various pathological conditions. Targeting LSD1 has been recognized as a promising therapeutic option for cancer treatment. Notably, some natural products with different chemotypes including protoberberine alkaloids, flavones, polyphenols, and cyclic peptides have shown effectiveness against LSD1. These natural products provide novel scaffolds for developing new LSD1 inhibitors. In this review, we mainly discuss the identification of natural LSD1 inhibitors, analysis of the co-crystal structures of LSD1/natural product complex, antitumor activity and their modes of action. We also briefly discuss the challenges faced in this field. 4β-catenin activation in hair follicle dermal stem cells in duces ectopic hair outgrowth and skin fibrosis显示文摘Hair follicle dermal sheath(DS)harbors hair follicle dermal stem cells(hfDSCs),which can be recruited to replenish DS and dermal papilla(DP).Cultured DS cells can differentiate into various cell lineages in vitro.However,it is unclear how its plasticity is modulated in vivo.Wnt/β-catenin signaling plays an important role in maintaining stem cells of various lineages and is required for HF development and regeneration.Here we report that activation ofβ-catenin in DS generates ectopic HF outgrowth(EF)by reprogramming HF epidermal cells and DS cells themselves,and endows DS cells with hair inducing ability.Epidermal homeostasis of pre-existing HFs is disrupted.Additionally,cell-autonomous progressive skin fibrosis is prominent in dermis,where the excessive fibroblasts largely originate from DS.Gene expression analysis of purified DS cells with activatedβ-catenin revealed significantly increased expression of Bmp,Fgf,and Notch ligands and administration of Bmp,Fgf,or Notch signaling inhibitor attenuates EF formation.In summary,our findings advance the current knowledge of high plasticity of DS cells and provide an insight into understanding how Wnt/β-catenin signaling controls DS cell behaviors.Yixin Tao Qingchun Yang Lei Wang Jie Zhang Xuming Zhu Qianqian Sun Yunbin Han Qian Luo Yushu Wang Xizhi Guo Ji Wu Baojie Li Xiao Yang Lin He Gang Ma 2019Journal of Molecular Cell Biology2019,11,1:4

6Hydrological Impacts of Climate Change on Streamflow of Dongliao River Watershed in Jilin Province,China显示文摘The impacts of future climate change on streamflow of the Dongliao River Watershed located in Jilin Province,China have been evaluated quantitatively by using a general circulation model(HadCM3)coupled with the Soil and Water Assessment Tool(SWAT)hydrological model.The model was calibrated and validated against the historical monitored data from 2005 to 2009.The streamflow was estimated by downscaling HadCM3 outputs to the daily mean temperature and precipitation series,derived for three 30-year time slices,2020s,2050s and 2080s.Results suggest that daily mean temperature increases with a changing rate of 0.435℃per decade,and precipitation decreases with a changing rate of 0.761 mm per decade.Compared with other seasons,the precipitation in summer shows significant downward trend,while a significant upward trend in autumn.The annual streamflow demonstrates a general downward trend with a decreasing rate of 0.405 m3/s per decade.The streamflow shows significant downward and upward trends in summer and in autumn,respectively.The decreasing rate of streamflow in summer reaches 1.97 m 3 /s per decade,which contributes primarily to the decrease of streamflow.The results of this work would be of great benifit to the design of economic and social development planning in the study area.ZHANG Lei LU Wenxi YANG Qingchun AN Yongkai LI Di GONG Lei 2012Chinese Geographical Science2012,22,5:3

8Primary environmental factors controlling gully distribution at the local and regional scale:An example from Northeastern China显示文摘Gully erosion is the main cause of global land degradation.The factors controlling gully erosion at watershed scale have been extensively studied,but the spatial pattern of gullies and their primary environmental factors are unclear.In order to explore the primary factors at the regional scale of over thousands of square kilometers,34 sample areas were selected in northeastern China and 10 environ-mental factors were proposed.Based on gully interpretations from high-resolution Google Earth images and spatial analysis,kernel density estimation(with polyline features),and correlation analysis,the results show that the maximum kernel density(KD)of the 34 sample areas ranged from 0.11 to 87.16,and there were significant positive correlations between the average KD and the maximum value and standard deviation.At the regional scale,topography(relief amplitude,slope,surface roughness,surface incision,and elevation with correlation coefficients(p)of 0.60,0.59,0.57,0.48,and 0.41 respectively)was the primary environmental factor,with a significant positive correlation with KD.The correlation co-efficients of the normalized difference vegetation index(NDVI)(p=032),annual precipitation(p=0.19),and soil(silt,sand,and clay content with p of 0.139,-0.117,and 0.085,respectively)decreased succes-sively with the KD.At sample area scale,there were six areas with weak correlations between envi-ronmental factors and KD,20 areas with topographic factors,six areas with annual precipitation,and two areas with NDVI as the primary factor.The correlation between soil texture and KD was low.There were different positive or negative correlations between the environmental factors and KD.The spatial dis-tribution of gullies in northeastern China is heterogeneous,and the primary environmental factors were diverse.This study will help to understand the spatial pattern and formation mechanism of gullies at large scale.Yushu Zhou Bin Zhang Wei Qin Qingchun Deng Jun Luo Hui Liu Dan Yang Houling Wang Ying Zhao 2021International Soil and Water Conservation Research2021,9,1:2
9A warmer but drier Marine Isotope Stage 11 during the past 650 ka as revealed by the thickest loess on the western Chinese Loess Plateau显示文摘Marine Isotope Stage 11(MIS 11; ca. 423–362 ka) is generally considered to be the best analogue for the present interglacial(Holocene), and investigation of it will improve our understanding of current climate variability and assist in predictions of future climate change. However, many recent studies primarily focus on the structure and duration of MIS 11. Little research has focused on climate warmth and stability recorded in the Chinese loess-paleosol sequences(LPS) during the S4 paleosol formation(equivalent to MIS 11). On the basis of previous work, this study presents a high-resolution record(ca. 75 a/cm) that spans from MIS 1 to MIS 15, as preserved in the thickest known Jingyuan loess section on the western Chinese Loess Plateau(CLP). This LPS is almost 165 m thick and was sampled from the upper part of L6 to the modern soil at 2-cm depth intervals. Measurements of magnetic susceptibility, mean grain size and >63 μm particle content, carbonate content, total organic carbon, and soil color of samples were made to reconstruct the paleoclimate variation, and a grain-size age model was used to constrain the chronological framework. The primary results show that a generally warm-humid climate dominated the S4 paleosol development, but the climate condition was extremely unstable during the whole of MIS 11. Two obviously different climate regimes controlled the MIS 11 climate variation: the early part of MIS 11 was extremely warm and stable, but the latter part was relatively cool(non-glacial) and unstable. This climate pattern was consistent with records on the central CLP and wavelet analysis suggested that it was forced by the 65°N insolation variability modulated by a quasi-100-ka cycle. In addition, a multi-proxy comparative study on the climate conditions during S0 to S5 paleosol development indicates that the period of S4 development might be the warmest interglacial of the past 650 ka. However, the climate condition during S4 development was not the most humid episode as recorded in Xifeng and Luochuan loess sections on the central CLP. 10Thermal behavior of an isolator with mode transition inducing back-pressure of a dual-mode scramjet显示文摘Combustion mode transition is a valuable and challenging research area in dual-mode scramjet engines.The thermal behavior of an isolator with mode transition inducing backpressure is investigated by direct-connect dual-mode scramjet experiments and theoretical analysis.Combustion experiments are conducted under the incoming airflow conditions of total temperature1270 K and Mach 2.A small increment of the fuel equivalence ratio is scheduled to trigger mode transition.Correspondingly,the variation of the coolant flow rate is very small.Based on the measured wall pressures,the heat-transfer model can quantify the thermal state variation of the engine with active cooling.Compared with the combustor,mode transition has a greater effect on the isolator thermal behavior,and it significantly changes the isolator heat-flux and wall temperature.To further study the isolator thermal behavior from flight Mach 4 to Mach 7,a theoretical analysis is carried out.Around the critical point of combustion mode transition,sudden changes of the isolator flowfield and thermal state are discussed.Yang Qingchun Bao Wen Chetehouna Khaled Zhang Silong Gascoin Nicolas 2017Chinese Journal of Aeronautics2017,30,2:2

13Technical and economic analysis of an oil shale comprehensive utilization process with solid heat carrier technology显示文摘To relieve the shortage of oil,exploration of oil shale attracts increasingly attention from different countries,especially in China.Facing with the disadvantages of conventional oil shale retorting technology,more and more attention has been paid to the comprehensive utilization of oil shale for utilizing oil shale efficiently and in an environment-friendly manner.However,there are few systematic studies on comprehensive utilization of oil shale.Thus,this paper is focused on establishing the complete mathematical model of an oil shale comprehensive utilization process with solid heat carrier technology(OSCU-SHC).Moreover,its techno-economic performance is analyzed and compared to a conventional oil shale retorting process with solid heat carrier(OSRSHC).Several key parameters are investigated and optimized during the simulation process.For example,the optimal retorting temperature is set to 500℃.The comparison results show that the OSCU-SHC process has better techno-economic performance since it increases the exergy efficiency by 9.26%,total income by 29.02%,and return on investment by 4.85%.Qingchun Yang Xufang Li Yu Qian Dawei Zhang 2018Carbon Resources Conversion2018,1,3:1
14Fabrication of magnetically responsive anti-fouling and easy-cleaning nanofiber membrane and its application for efficient oil-water emulsion separation显示文摘The magnetically responsive anti-fouling nanofiber membrane(MRANM)was fabricated for efficient oilwater emulsion separation,which could be cleaned using oscillating magnetic field.MRANM was prepared by grafting superparamagnetic Fe_(3)O_(4) nanoparticles onto the surface of electrospun polyacrylonitrile nanofiber membrane(PANM).Compared with PANM,the water contact angle of MRANM decreased from 104°to 0°,indicating that the hydrophilicity of the membrane was significantly improved.For the emulsions of hexadecane,octane and rapeseed oil,the separation efficiency was 98.04%,96.59%and 92.67%,respectively.After the treatments in oscillating magnetic field,the separation efficiency kept above 95%after 8 times recycling,which indicated that the MRANM had good regenerability and reusability.The as-fabricated membrane with magnetic responsiveness facilitated an effective method for solving the membrane fouling problem during practical applications of separation high viscosity oil-water emulsion.Yajie Wang Zhiwei Guo Yujie Yang Yanxiang Li Qingchun Guo Peilin Cui Wangliang Li 2022Chinese Journal of Chemical Engineering2022,35,1:1

20Influence of semi-solid isothermal treatment on eutectic carbide in Cr12MoV steel显示文摘In-situ observation of eutectic solidification of Cr12MoV steel was conducted using high temperature confocal laser-scanning microscopy. The semi-solid isothermal treatment temperature of the steel was determined by thermodynamic calculation using Thermo-cal software. At the same time, the infl uences of isothermal treatment temperature and time on eutectic carbides in the steel were also studied. The results show that when the liquid metal cools at the rate of 47 °C·min-1, the eutectic reaction occurs rapidly at 1,214.7 °C in one second with the reticular liquid around austenite dendrites, transforming into a network of eutectic structure. After being held at 1,300 °C for 30 min, the carbide network is broken due to the impingement of refi ned primary austenite dendrites and secondary dendrites arms, and the thickness of eutectic structure is reduced.Li Qingchun Yang Bo Li Chong Wang Chao Chang Guowei 2014China Foundry2014,11,6:1
