1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Genomic structure of metabotropic glutamate receptor 7 and comparison of genomic structures of extracellular domains of mGluR family显示文摘Metabotropic glutamate receptor 7, coupled with a chemical neurotransmitter L-glutamate, plays an important role in the development of many psychiatric and neurological disorders. To study the biological and genetic mechanism of the mGluR7-related diseases, a physical map covering the full-length mGluR7 genomic sequence has been constructed through seed clone screening and fingerprinting database searching. These BAC clones in the physical map have been sequenced with shotgun strategy and assembled by Phred-Phrap-Consed software; the error rate of the final genoniic sequence is less than 0.01%. mGluR7 spans 880 kb genoniic region, the GC content and repeat content of mGluR7 genoniic sequence are 38% and 37.5% respectively. mGluR7 has a typical 'house-keeping' promoter and consists of 11 exons, with introns ranging from 6 kb to 285 kb. mGluR7a and mGluR7b are two known alternatively splicing variants. 4Role of humidity on the structures and properties of regenerated silk fibers显示文摘Silk fiber was processed from highly concentrated spinning dope to solid fibers along with water removal. To understand the mechanism of water removal during silk fiber spinning process, a microfluidic chip was designed and applied to investigate the structures and mechanical properties of two kinds of regenerated silk fibroin fibers dry-spun at different relative humidity. The experimental results showed that the diameters of the fibers spun at 40% RH are always larger than the fibers spun at 50% RH due to different removal rates of water. The fibers spun at low humidity contain more β-sheet structure and lower degree of chain orientation and crystalline orientation. 19A host-guest approach to fabricate metallic cobalt nanoparticles embedded in silk-derived N-doped carbon fibers for efficient hydrogen evolution显示文摘Hydrogen evolution reaction(HER) plays a key role in generating clean and renewable energy. As the most effective HER electrocatalysts, Pt group catalysts suffer from severe problems such as high price and scarcity. It is highly desirable to design and synthesize sustainable HER electrocatalysts to replace the Pt group catalysts. Due to their low cost, high abundance and high activities, cobalt-incorporated N-doped nanocarbon hybrids are promising candidate electrocatalysts for HER. In this report, we demonstrated a robust and eco-friendly host-guest approach to fabricate metallic cobalt nanoparticles embedded in N-doped carbon fibers derived from natural silk fibers. Benefiting from the onedimensional nanostructure, the well-dispersed metallic cobalt nanoparticles and the N-doped thin graphitized carbon layer coating, the best Cobased electrocatalyst manifests low overpotential(61 mV@10 mA/cm^2) HER activity that is comparable with commercial 20% Pt/C, and good stability in acid. 20Molecular basis and mechanism of action of Albizia julibrissin in depression treatment and clinical application of its formulae显示文摘Albizzia julibrissin is empirically used as an antidepressant in clinical practice.Preclinical studies have indicated that its total extracts or bioactive constituents exerted antidepressant-like responses in animal models,providing the molecular basis to reveal its underlying mechanism of action.While attempts have been made to understand the antidepressant effect of A.julibrissin,many fundamental questions regarding its mechanism of action remain to be addressed at the molecular and systems levels.In this review,we conclusively discussed the mechanism of action of A.julibrissin and A.julibrissin formulae by reviewing recent preclinical and clinical studies conducted by using depressive animal models and depressive patients.Several representative bioactive constituents and formulae were highlighted as examples,and their mechanisms of action were discussed.In addition,some representative A.julibrissin formulae that have been shown to be compatible with conventional antidepressants in clinical practice were also reviewed.Furthermore,we discussed the future research directions to reveal the underlying mechanism of A.julibrissin at the molecular and systems levels in depression treatment.The integrated study using both the molecular and systematic approaches is required not only for improving our understanding of its molecular basis and mechanisms of action,but also for providing a way to discover novel agents or approaches for the effective and systematic treatment of depression.Bishan Huang Yingyao Wu Chan Li Qingfa Tang Yuanwei Zhang 2023Chinese Herbal Medicines2023,15,2:0
