1Establishment of cell clones with different metastatic potential from the metastatic hepatocellular carcinoma cell line MHCC97显示文摘ALM To establish clone cells with different metastatic potential for the study of metastasis-related mechanisms. METHODS Cloning procedure was performed on parental hepatocellular carcinoma (HCC) cell line MHCC97. andbiological characteristics of the target clones selected by in vivo screening were studied.``RESULTS Two clones with high MHCC97-H and IowMHCC9--L1 metastatic potential were isolated from theparent cell line. Compared with MHCC97-L. MHCC97-H hadsmaller cell size average cell diameter 43 um vs 50 μmand faster in vitro and in vivo growth rate tumor celldoubling time was 34.2 h vs 60.0 h. The main ranges ofchromosomes were 5.5 58 in MHCC97-H and 57 62 inMHCC97-L. Boyden chamber in vitro invasion assay demonstrated that the number of penetrating cells through the artificial basement membrane was 137.5 - 11 .0) cellsfield for MHC_C99--H vs 17.7 - 6.3) field for MHCC97-L.The proportions of cells in GO Gl phase. 2Metastatic human hepatocellular carcinoma models in nude mice and cell line with metastatic potential显示文摘Metastatic human HCC model is needed for the studies on mechanism and intervention of metastatic recurrence. By using orthotopic implantation of histologically intact tissues of 30 surgical specimens, a patient like metastatic model of human HCC in nude mice (LCI-D20)and a Iow metastatic model of human HCC in nude mice LCI-D35 ) have been established. All mice with transplanted LCI-D20 tumors exhibited extremely high metastatic ability including spontaneous metastasis to liver, lungs, lymph nodes and peritoneal seeding.Remarkable difference was also found in expression of some of the invasiveness related genes and growth factors between the LCI-D20 and LCI-D35 tumors. PAI-Iincreased gradually following tumor progression in LCID20 model, and correlated with tumor size and AFP level,Phasic expression of tissue intercellular adhesion molecule-I in this model was also observed. Using corneal micropocket model, it was demonstrated that the vascular response induced by LCI-D20 tumor was stronger than that induced by LCI-D35 tumor. Similar report on metastatic human HCC model in nude mice and human HCC cell line with metastatic potential was rarely found in the literature. 4High Cr white cast iron/carbon steel bimetal liner by lost foam casting with liquid-liquid composite process显示文摘Liners in wet ball mill for mineral processing industry must bear abrasive wear and corrosive wear, and consequently,the service life of the liner made from traditional materials,such as Hadfield steel and alloyed steels,is typically less than ten months.Bimetal liner,made from high Cr white cast iron and carbon steel,has been successfully developed by using liquid-liquid composite lost foam casting process.The microstructure and interface of the composite were analyzed using optical microscope,SEM,EDX and XRD.Micrographs indicate that the boundary of bimetal combination regions is staggered like dogtooth,two liquid metals are not mixed,and the interface presents excellent metallurgical bonding state.After heat treatment,the composite liner specimens have shown excellent properties,including hardness>61 HRC,fracture toughnessα k >16.5 J·cm-2 and bending strength >1,600 MPa.Wear comparison was made between the bimetal composite liner and alloyed steel liner in an industrial hematite ball mill of WISCO,and the results of eight-month test in wet grinding environment have proved that the service life of the bimetal composite liner is three times as long as that of the alloyed steel liner.Xiao Xiaofeng Ye Shengping YinWeixin Zhou Xiaoguang Xue Qiong 2012China Foundry2012,9,2:10
5Dopamine D4 Receptor Gene Associated with the Frontal-Striatal-Cerebellar Loop in Children with ADHD: A Resting-State fMRI Study显示文摘Attention deficit hyperactivity disorder(ADHD)is a common childhood neuropsychiatric disorder that has been linked to the dopaminergic system. This study aimed to investigate the effects of regulation of the dopamine D4 receptor(DRD4) on functional brain activity during the resting state in ADHD children using the methods of regional homogeneity(Re Ho) and functional connectivity(FC). Resting-state functional magnetic resonance imaging data were analyzed in 49 children with ADHD. All participants were classified as either carriers of the DRD44-repeat/4-repeat(4 R/4 R) allele(n = 30) or the DRD42-repeat(2 R) allele(n = 19). The results showed that participants with the DRD4 2 R allele had decreased Re Ho bilaterally in the posterior lobes of the cerebellum, while Re Ho was increased in the left angular gyrus. Compared with participants carrying the DRD4 4 R/4 R allele, those with the DRD4 2 R allele showed decreased FC to the left angular gyrus in the left striatum, right inferior frontal gyrus, and bilateral lobes of the cerebellum. The increased FC regions included the left superior frontal gyrus, medial frontal gyrus, and rectus gyrus. These data suggest that the DRD4 polymorphisms are associated with localized brain activity and specific functional connections, including abnormality in the frontal-striatal-cerebellar loop. 9Latest Wuchiapingian to Earliest Triassic Conodont Zones and δ13 Ccarb Isotope Excursions from Deep-Water Sections in Western Hubei Province, South China显示文摘Deep-water facies sections have advantages of recording complete information across the Permian-Triassic boundary(PTB). Here we present a detailed study on the conodont biostratigraphy and carbon isotope profile ranges from the Wuchiapingian-Changhsingian boundary(WCB) to the PTB of two deep-water facies sections at Zhuqiao and Shiligou in the Middle Yangtze region, western Hubei, South China. Fifteen species and three genera are identified. Eight conodont zones are recognized which in ascending order are the Clarkina orientalis, C. wangi, C. subcarinata, C. changxingensis, C. yini, C. meishanensis, Hindeodus parvus and Isarcicella isarcica zones. The onset of deposition of the deep-water siliceous strata of Dalong Formation in western Hubei began in the Late Wuchiapingian and persisted to the Late Changhsingian. Carbon isotope negative excursions occur near both the WCB and PTB in both sections. The WCB δ13 Ccarb negative excursion is in the C. orientalis and C. wangi zones. The PTB δ13 Ccarb negative excursion began in the C. yini Zone and extended to the I. isarcica Zone. The absence of several Changhsingian zones may indicate the difficulty of extracting conodonts from siliceous strata or the presence of an intra-Changhsingian hiatus.Bo Yang Hanxiao Li Paul B. 10Characterization of astrocytes and microglial cells in the hippocampal CA1 region after transient focal cerebral ischemia in rats treated with Ilexonin A显示文摘Ilexonin A is a compound isolated from the root of Ilex pubescens,a traditional Chinese medicine.Ilexonin A has been shown to play a neuroprotective role by regulating the activation of astrocytes and microglia in the peri-infarct area after ischemia.However,the effects of ilexonin A on astrocytes and microglia in the infarct-free region of the hippocampal CA1 region remain unclear.Focal cerebral ischemia models were established by 2-hour occlusion of the middle cerebral artery in rats.Ilexonin A(20,40 or 80 mg/kg)was administered immediately after ischemia/reperfusion.The astrocyte marker glial fibrillary acidic protein,microglia marker Iba-1,neural stem cell marker nestin and inflammation markers were detected by immunohistochemistry and western blot assay.Expression levels of tumor necrosis factor-αand interleukin 1βwere determined by enzyme linked immunosorbent assay in the hippocampal CA1 tissue.Astrocytes were activated immediately in progressively increasing numbers from 1,3,to 7 days post-ischemia/reperfusion.The number of activated astrocytes further increased in the hippocampal CA1 region after treatment with ilexonin A.Microglial cells remained quiescent after ischemia/reperfusion,but became activated after treatment with ilexonin A.Ilexonin A enhanced nestin expression and reduced the expression of tumor necrosis factor-αand interleukin 1βin the hippocampus post-ischemia/reperfusion.The results of the present study suggest that ilexonin A has a neuroprotective effect in the hippocampus after ischemia/reperfusion,probably through regulating astrocytes and microglia activation,promoting neuronal stem cell proliferation and reducing the levels of pro-inflammatory factors.This study was approved by the Animal Ethics Committee of the Fujian Medical University Union Hospital,China.Ai-Ling Xu Guan-Yi Zheng Hui-Ying Ye Xiao-Dong Chen Qiong Jiang 2020Neural Regeneration Research2020,15,1:5

12Identification of coastal water quality by multivariate statistical techniques in two typical bays of northern Zhejiang Province,East China Sea显示文摘The Hangzhou Bay(HZB) and Xiangshan Bay(XSB), in northern Zhejiang Province and connect to the East China Sea(ECS) were considerably affected by the consequence of water quality degradation. In this study, we analyzed physical and biogeochemical properties of water quality via multivariate statistical techniques. Hierarchical cluster analysis(HCA) grouped HZB and XSB into two subareas of different pollution sources based on similar physical and biogeochemical properties. Principal component analysis(PCA) identified three latent pollution sources in HZB and XSB respectively and emphasized the importance of terrestrial inputs, coastal industries as well as natural processes in determining the water quality of the two bays. 13The Genomes of the Allohexaploid Echinochloa crus-galli and Its Progenitors Provide Insights into Polyploidization-Driven Adaptation显示文摘The hexaploid species Echinochloa crus-galli is one of the most detrimental weeds in crop fields,especially in rice paddies.Its evolutionary history is similar to that of bread wheat,arising through polyploidization after hybridization between a tetraploid and a diploid species.In this study,we generated and analyzed high-quality genome sequences of diploid(E.haploclada),tetraploid(E.oryzicola),and hexaploid(E.crus-galli)Echinochloa species.Gene family analysis showed a significant loss of disease-resistance genes such as those encoding NB-ARC domain-containing proteins during Echinochloa polyploidization,contrary to their significant expansionduring wheat polyploidization,suggesting that natural selection might favor reduced investment in resistance in this weed to maximize its growth and reproduction.In contrast to the asymmetric patterns of genome evolution observed in wheat and other crops,no significant differences in selection pressure were detected between the subgenomes in E.oryzicola and E.crus-galli.In addition,distinctive differences in subgenome transcriptome dynamics during hexaploidization were observed between E.crus-galli and bread wheat.Collectively,our study documents genomic mechanisms underlying the adaptation of a major agricultural weed during polyploidization.The genomic and transcriptomic resources of three Echinochloa species and new insights into the polyploidization-driven adaptive evolution would be useful for future breeding cereal crops.Chu-Yu Ye Dongya Wu Lingfeng Mao Lei Jia Jie Qiu Sangting Lao Meihong Chen Bowen Jiang Wei Tang Qiong Peng Lang Pan Lifeng Wang Xiaoxiao Feng Longbiao Guo Chulong Zhang Elizabeth A.Kellogg Kenneth M.Olsen Lianyang Bai Longjiang Fan 2020Molecular Plant2020,13,9:4

15New preparation of Mg-Li-Al alloys by electrolysis in molten salt显示文摘This work presents a study on electrochemical formation of Mg-Li-Al alloys on an inert electrode(Mo electrode) in a molten KCl-LiCl-AlCl3-MgCl2-KF system.It aims at preparation Mg-Li-Al metal alloy directly under an optimal electrolytic parameters.Main factors which affect current efficiency are investigated.We have discussed the co-electrodeposition conditions and the effect of technical parameters on these experiments,and gained a good electrolytic process.The result of calculation shows that co-electrodeposition Mg,Li and Al occurs at the cathode current density higher than 9 A/cm2 and electrolytic voltage between 6 to 9 V.And at the optimal parameters,12-13 A/cm2,620-640 ℃,the highest current effciency reaches as high as 86%.Lithium combined with magnesium forms Li0.92Mg4.08 and Li3Mg7,and aluminum mainly exists in a form of Mg0.58Al0.42,distributing in the alpha phases and beta phases.Wei HAN Qiong CHEN Ke YE Yongde YAN Milin ZHANG 2010Acta Metallurgica Sinica(English Letters)2010,23,2:3
16Study on the Dynamics of Blending of Glucomannan with Carrageenan and Guaran Gum显示文摘In order to prove up the interaction between konjac glucomannan (KGM) and other amyloses, by using KGM, guaran gum and carrageenan as materials, their blending in different external conditions was modeled by means of molecular dynamics (MD) method. The result showed that 323 K was a significant turning point for the formation of hydrogen bond, and KGM and guaran gum were just juxtaposed together via intermolecular hydrogen bond; but with the addition of carrageenan, KGM, guaran gum and carrageenan were spirally twisted together, and when their blending proportion is 3:2:9 the combination was the most compact. 17Co-infection of SARS-COV-2 and Influenza A Virus:A Case Series and Fast Review显示文摘Coronavirus disease 2019(COVID-19)occurs in the influenza season and has become a global pandemic.The present study aimed to examine severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)co-infection with influenza A virus(IAV)in an attempt to provide clues for the antiviral interventions of co-infected patients.We described two patients who were co-infected with SARS-CoV-2 and IAV treated at Wuhan Union Hospital,China.In addition,we performed a review in PubMed,Web of Science and CNKI(from January 1 up to November 1,2020)with combinations of the following key words:“COVID-19,SARS-COV-2,influenza A and co-infection”.A total of 28 co-infected patients were enrolled in the analysis.Of the 28 patients,the median age was 54.5 years(IQR,34.25–67.5)and 14 cases(50.0%)were classified as severe types.The most common symptoms were fever(85.71%),cough(82.14%)and dyspnea(60.71%).Sixteen patients had lymphocytopenia on admission and 23 patients exhibited abnormal radiological changes.The median time from symptom onset to hospital admission was 4 days(IQR,3–6),and the median time of hospital stay was 14 days(IQR,8.5–16.75).In conclusion,patients with SARSCOV-2 and IAV co-infection were similar to those infected with SARS-COV-2 alone in symptoms and radiological images.SARS-COV-2 co-infection with IAV could lead to more severe clinical condition but did not experience longer hospital stay compared with patients infected with SARSCOV-2 alone.Xuan XIANG Zi-hao WANG Lin-lin YE Xin-liang HE Xiao-shan WEI Yan-ling MA Hui LI Long CHEN Xiao-rong WANG Qiong ZHOU 2021Current Medical Science2021,41,1:2

19Reduction in interhemispheric functional connectivity in the dorsal visual pathway in unilateral acute open globe injury patients: a resting-state fMRI study显示文摘This study investigated the changes in interhemispheric functional connectivity(FC) of the whole brain in open globe injury(OGI) patients, using voxel-mirrored homotopic connectivity(VMHC), and their relationships with clinical features. Totally, 16 male and 2 female acute OGI patients and 18 sex, age, and education-matched healthy volunteers were enrolled in the study. All subjects were scanned through functional magnetic resonance imaging(f MRI). Receiver operating characteristic(ROC) curves analyses had been used to identify the VMHC in these brain areas could be used as biomarkers to distinguish OGI and from healthy control(HC). The mean VMHC values in multiple brain areas and clinical OGI manifestations were evaluated with a Pearson correlation analysis. OGI patients had significantly decreased VMHC in the bilateral calcarine/lingual/cuneus(BA18, 19, 30) and middle occipital gyrus(BA18, 19). 20Real-time fluorescent quantitative immuno-PCR method for determination of fluoranthene in water samples with a molecular beacon显示文摘A reliable and sensitive competitive real-time fluorescent quantitative immuno-PCR (RTFQ-IPCR) assay using a molecular beacon was developed for the determination of trace fluoranthene (FL) in the environment.Under optimized assay conditions,FL can be determined in the concentration range from 1 fg/mL to 100 ng/mL,with y=0.194x + 7.859,and a correlation coefficient of 0.967 was identified,with a detection limit of 0.6 fg/mL.Environmental water samples were successfully analyzed,recovery was between 90% and 116%,with intra-day relative standard deviation (RSD) of 6.7%-12.8% and inter-day RSD of 8.4%-15.2%.The results obtained from RTFQ-IPCR were confirmed by ELISA,showing good accuracy and suitability to analyze FL in field samples.As a highly sensitive method,the molecular beacon-based RTFQ-IPCR is acceptable and promising for providing reliable test results to make environmental decisions.Qiyan Ye Huisheng Zhuang Chun Zhou Qiong'e Wang 2010Journal of Environmental Sciences2010,22,5:2
