1AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge.The viral pathogen responsible,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein,which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear,studies have shown that ACE2 expression is extremely low in various human tissues,especially in the respiratory tract.Thus,other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist.In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together,our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies.Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li 2021Cell Research2021,31,2:39
2Neoadjuvant chemotherapy plus intensity-modulated radiotherapy versus concurrent chemoradiotherapy plus adjuvant chemotherapy for the treatment of locoregionally advanced nasopharyngeal carcinoma:a retrospective controlled study显示文摘Background:In the era of intensity-modulated radiotherapy(IMRT),the role of neoadjuvant chemotherapy(NAC)for locoregionally advanced nasopharyngeal carcinoma(NPC)is under-evaluated.The aim of this study was to compare the efficacy of NAC plus IMRT and concurrent chemoradiotherapy(CCRT)plus adjuvant chemotherapy(AC)on locoregionally advanced NPC.Methods:Between January 2004 and December 2008,240 cases of locoregionally advanced NPC confirmed by pathologic assessment in Sun Yat-sen University Cancer Center were reviewed.Of the 240 patients,117 received NAC followed by IMRT,and 123 were treated with CCRT plus AC.The NAC+IMRT group received a regimen that included cisplatin and 5-fluorouracil(5-FU).The CCRT+AC group received cisplatin concurrently with radiotherapy,and subsequently received adjuvant cisplatin and 5-FU.The survival rates were assessed by Kaplan-Meier analysis,and the survival curves were compared using a log-rank test.Multivariate analysis was conducted using the Cox proportional hazard regression model.Results:The 5-year overall survival(OS),locoregional relapse-free survival(LRRFS),distant metastasis-free survival(DMFS),and disease-free survival(DFS)were 78.0,87.9,79.0,and 69.8%,respectively,for the NAC+IMRT group and78.7,84.8,76.2,and 65.6%,respectively,for the CCRT+AC group.There were no significant differences in survival between the two groups.In multivariate analysis,age(<50 years vs.>50 years)and overall stage(Ⅲvs.Ⅳ)were found to be independent predictors for OS and DFS;furthermore,the overall stage was a significant prognostic factor for DMFS.Compared with the CCRT+AC protocol,the NAC+IMRT protocol significantly reduced the occurrence rates of grade 3-4 nausea-vomiting(6.5 vs.1.5%,P=0.023)and leukopenia(9.7 vs.0.8%,P=0.006).Conclusions:The treatment outcomes of the NAC+IMRT and CCRT+AC groups were similar.Distant metastasis remained the predominant mode of treatment failure.Wen-Ze Qiu Pei-Yu Huang Jun-Li Shi Hai-Qun Xia Chong Zhao Ka-Jia Cao 2016Chinese Journal of Cancer2016,35,1:22

5Effects of tourmaline on nitrogen removal performance and biofilm structures in the sequencing batch biofilm reactor显示文摘The effects of tourmaline on nitrogen removal performance and biofilm structures were comparatively investigated in two identical laboratory-scale sequencing batch biofilm reactors(SBBRs)(denoted SBBR1 and SBBR2) at different nitrogen loading rates(NLRs) varying from(0.24 ± 0.01) to(1.26 ± 0.02) g N/(L·day). SBBR1 was operated in parallel with SBBR2, but SBBR1 was filled with polyurethane foam loaded tourmaline(TPU) carriers and another(SBBR2) filled with polyurethane foam(PU) carriers. Results obtained from this study showed that the excellent and stable performance of SBBR1 was obtained. Ammonia nitrogen removal and total nitrogen removal were higher in SBBR1 than that in SBBR2 with increase of NLR. At an NLR of(0.24 ± 0.01) g N/(L·day), the majority of the spherical and elliptical bacteria were surrounded by the extracellular polymeric substance(EPS) and bacillus or filamentous bacteria in two SBBRs biofilms. When NLR increased to(1.26 ± 0.02) g N/(L·day), the clusters were more obvious in the SBBR1 biofilm than that in the SBBR2 biofilm. Bacteria in SBBR1 were inclined to synthesis more EPS, and the formed EPS could protect the bacteria from free ammonia(FA) under extreme condition NLR(1.26 ± 0.02) g N/(L·day). 6Development and validation of an endoscopic images-based deep learning model for detection with nasopharyngeal malignancies显示文摘Background:Due to the occult anatomic location of the nasopharynx and frequent presence of adenoid hyperpla-sia,the positive rate for malignancy identification during biopsy is low,thus leading to delayed or missed diagnosis for nasopharyngeal malignancies upon initial attempt.Here,we aimed to develop an artificial intelligence tool to detect nasopharyngeal malignancies under endoscopic examination based on deep learning.Methods:An endoscopic images-based nasopharyngeal malignancy detection model(eNPM-DM)consisting of a fully convolutional network based on the inception architecture was developed and fine-tuned using separate training and validation sets for both classification and segmentation.Briefly,a total of 28,966 qualified images were collected.Among these images,27,536 biopsy-proven images from 7951 individuals obtained from January 1st,2008,to December 31st,2016,were split into the training,validation and test sets at a ratio of 7:1:2 using simple randomiza-tion.Additionally,1430 images obtained from January 1st,2017,to March 31st,2017,were used as a prospective test set to compare the performance of the established model against oncologist evaluation.The dice similarity coef-ficient(DSC)was used to evaluate the efficiency of eNPM-DM in automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images,by comparing automatic segmentation with manual segmenta-tion performed by the experts.Results:All images were histopathologically confirmed,and included 5713(19.7%)normal control,19,107(66.0%)nasopharyngeal carcinoma(NPC),335(1.2%)NPC and 3811(13.2%)benign diseases.The eNPM-DM attained an overall accuracy of 88.7%(95%confidence interval(CI)87.8%-89.5%)in detecting malignancies in the test set.In the prospective comparison phase,eNPM-DM outperformed the experts:the overall accuracy was 88.0%(95%CI 86.1%-89.6%)vs.80.5%(95%CI 77.0%-84.0%).The eNPM-DM required less time(40 s vs.110.0±5.8 min)and exhibited encouraging performance in automatic segmentation of nasopharyngeal malignant area from the background,with an average DSC of 0.78±0.24 and 0.75±0.26 in the test and prospective test sets,respectively.Conclusions:The eNPM-DM outperformed oncologist evaluation in diagnostic classification of nasopharyngeal mass into benign versus malignant,and realized automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images.Chaofeng Li Bingzhong Jing Liangru Ke Bin Li Weixiong Xia Caisheng He Chaonan Qian Chong Zhao Haiqiang Mai Mingyuan Chen Kajia Cao Haoyuan Mo Ling Guo Qiuyan Chen Linquan Tang Wenze Qiu Yahui Yu Hu Liang Xinjun Huang Guoying Liu Wangzhong Li Lin Wang Rui Sun Xiong Zou Shanshan Guo Peiyu Huang Donghua Luo Fang Qiu Yishan Wu Yijun Hua Kuiyuan Liu Shuhui Lv Jingjing Miao Yanqun Xiang Ying Sun Xiang Guo Xing Lv 2018Cancer Communications2018,38,1:9
7Is laparoscopy equal to laparotomy in detecting and treating small bowel injuries in a porcine model?显示文摘AIM: To evaluate the safety and effectiveness of laparoscopy compared with laparotomy for diagnosing and treating small bowel injuries (SBIs) in a porcine model. METHODS: Twenty-eight female pigs were anesthetized and laid in the left recumbent position. The SBI model was established by shooting at the right lower quadrant of the abdomen. The pigs were then randomized into either the laparotomy group or the laparoscopy group. All pigs underwent routine exploratory laparotomy or laparoscopy to evaluate the abdominal injuries, particularly the types, sites, and numbers of SBIs. Traditional open surgery or therapeutic laparoscopy was then performed. All pigs were kept alive within the observational period (postoperative 72 h). The postoperative recovery of each pig was carefully observed. RESULTS: The vital signs of all pigs were stable within 1-2 h after shooting and none of the pigs died from gunshot wounds or SBIs immediately. The SBI model was successfully established in all pigs and definitively diagnosed with single or multiple SBIs either by exploratory laparotomy or laparoscopy. Compared with exploratory laparotomy, laparoscopy took a significantly longer time for diagnosis (41.27 ± 12.04 min vs 27.64 ± 13.32 min, P = 0.02), but the time for therapeutic laparoscopy was similar to that of open surgery. The length of incision was significantly reduced in the laparoscopy group compared with the laparotomy group (5.27 ± 1.86 cm vs 15.73 ± 1.06 cm, P < 0.01). In the final post-mortem examination 72 h after surgery, both laparotomy and laparoscopy offered a definitive diagnosis with no missed injuries. Postoperative complications occurred in four cases (three following laparotomy and one following laparoscopy, P = 0.326). The average recovery period for bowel function, vital appearance, and food re-intake after laparoscopy was 10.36 ± 4.72 h, 14.91 ± 3.14 h, and 15.00 ± 7.11 h, respectively. All of these were significantly shorter than after laparotomy (21.27 ± 10.17 h, P = 0.004; 27.82 ± 9.61 h, P < 0.001; and 24.55 ± 9.72 h, respectively, P = 0.016). 10Clinical observations of osteoporotic vertebral compression fractures by using mineralized collagen modified polymethylmethacrylate bone cement显示文摘To investigate the clinical outcomes of the treatment of osteoporotic vertebral compression fractures(OVCFs)by using mineralized collagen(MC)modified polymethylmethacrylate(PMMA)bone cement.52 cases(52 vertebras)who sustained OVCF treated with MC modified PMMA bone cement from July 2014 to December 2015 were reviewed retrospectively.All the cases(52 patients,52 vertebras)included 8 males and 44 females with an average age of 74.83(ranging from 57-to 90-years old).The visual analogue scale(VAS),vertebral body height,Cobb angle,CT values preand post-operation as well as incidence of complications were used to be observed.All the patients underwent the surgery were successfully followed-up with an average period of 13.54 months(ranging from 6 to 23 months).The patients can ambulate at the second day after the operation.The VAS scores 2 days after the operation and during the last follow-up were significantly decreased compared with that before the operation(P<0.05);the average vertebral height and local Cobb angle had significant recovery(P<0.05);the CT value of the treated vertebra significantly increased compared with that before the operation(P<0.05).MC with good osteogenic activity and degradation properties can effectively improve the mechanical properties and biocompatibility of the PMMA bone cement,thus obtain better clinical results.XiWang Jian-Ming Kou Yang Yue An-Ze Shao Xue-Jun Jia Jian-Wen Hou Chong Gao Zhi-Ye Qiu Xiu-Mei Wang Xi-Sheng Weng 2017Regenerative Biomaterials2017,4,2:5
11Interval finite difference method for steady-state temperature field prediction with interval parameters显示文摘A new numerical technique named interval finite difference method is proposed for the steady-state temperature field prediction with uncertainties in both physical parameters and boundary conditions. Interval variables are used to quantitatively describe the uncertain parameters with limited information. Based on different Taylor and Neumann series, two kinds of parameter perturbation methods are presented to approximately yield the ranges of the uncertain temperature field. 12Key stress extraction and equivalent test method for hybrid DC circuit breaker显示文摘Firstly, relevant stress properties of millisecond level breaking process and microsecond level commutation process of hybrid HVDC circuit breaker are studied in detail on the basis of the analysis for the application environment and topological structure and operating principles of hybrid circuit breakers, and key stress parameters in transient state process of two time dimensions are extracted. The established digital simulation circuit for PSCAD/EMTDC device-level operation of the circuit breaker has verified the stress properties of millisecond level breaking process and microsecond level commutation process. Then, equivalent test method, circuits and parameters based on LC power supply are proposed on the basis of stress extraction. Finally, the results of implemented breaking tests for complete 200 kV circuit breaker, 100 kV and 50 kV circuit breaker units, as well as single power electronic module have verified the accuracy of the simulation circuit and mathematical analysis. 13Clinical observation of mineralized collagen bone grafting after curettage of benign bone tumors显示文摘Curettage of benign bone tumor is a common cause for bone defect.For such bone defect repair,autogenous bone,allogeneic bone and traditional artificial bone graft substitutes have many disadvantages.In recent years,a biomimetic mineralized collagen(MC)with similar composition and microstructures to the natural bone matrix was developed and used for treating various bone defects.In this work,a retrospective study analyzed clinical outcomes of patients treated with curettage of benign bone tumors and bone grafting with MC,in comparison to another group treated with the same surgical method and autogenous bone.Lane-Sandhu X-ray score of the autogenous bone group was superior to the MC group at 1 month after the operation,but the two groups had no statistical difference at 6 and 12 months.The MC group was better in Musculoskeletal Tumor Society scoring at 1 and 6 months after the operation,and the two groups had no statistical difference at 12 month.Therefore,the MC performed not as good as autogenous bone in early stage of bone healing but achieved comparable outcomes in long-term follow-ups.Moreover,the MC has advantages in function recovery and avoided potential complications induced by harvesting autogenous bone.Chong Gao Zhi-Ye Qiu Jian-Wen Hou Wei Tian Jian-Ming Kou Xi Wang 2020Regenerative Biomaterials2020,7,6:3
14Fuzzy finite difference method for heat conduction analysis with uncertain parameters显示文摘A new numerical technique named as fuzzy finite difference method is proposed to solve the heat conduction problems with fuzzy uncertainties in both the physical parameters and initial/boundary conditions.In virtue of the level-cut method,the difference discrete equations with fuzzy parameters are equivalently transformed into groups of interval equations.New stability analysis theory suited to fuzzy difference schemes is developed.Based on the parameter perturbation method,the interval ranges of the uncertain temperature field can be approximately predicted.Subsequently,fuzzy solutions to the original difference equations are obtained by the fuzzy resolution theorem.Two numerical examples are given to demonstrate the feasibility and efficiency of the presented method for solving both steady-state and transient heat conduction problems.Chong Wang Zhi-Ping Qiu 2014Acta Mechanica Sinica2014,30,3:3

17Modified perturbation method for eigenvalues of structure with interval parameters显示文摘In overcoming the drawbacks of traditional interval perturbation method due to the unpredictable effect of ignoring higher order terms,a modified parameter perturbation method is presented to predict the eigenvalue intervals of the uncertain structures with interval parameters.In the proposed method,interval variables are used to quantitatively describe all the uncertain parameters.Different order perturbations in both eigenvalues and eigenvectors are fully considered.By retaining higher order terms,the original dynamic eigenvalue equations are transformed into interval linear equations based on the orthogonality and regularization conditions of eigenvectors.The eigenvalue ranges and corresponding eigenvectors can be approximately predicted by the parameter combinatorial approach.Compared with the Monte Carlo method,two numerical examples are given to demonstrate the accuracy and efficiency of the proposed algorithm to solve both the real eigenvalue problem and complex eigenvalue problem.WANG Chong QIU ZhiPing 2015Science China(Physics,Mechanics & Astronomy)2015,58,1:2
