1Proanthocyanidins Inhibit Seed Germination by Maintaining a High Level of Abscisic Acid in Arabidopsis thaliana显示文摘Proanthocyanidins (PAs) are the main products of the flavonoid biosynthetic pathway in seeds, but their biological function during seed germination is still unclear. We observed that seed germination is delayed with the increase of exogenous PA concentration in Arabidopsis. A similar inhibitory effect occurred in peeled Brassica napus seeds, which was observed by measuring radicle elongation. Using abscisic acid (ABA), a biosynthetic and metabolic inhibitor, and gene expression analysis by real-time polymerase chain reaction, we found that the inhibitory effect of PAs on seed germination is due to their promotion of ABA via de novo biogenesis, rather than by any inhibition of its degradation. Consistent with the relationship between PA content and ABA accumulation in seeds, PA-deficient mutants maintain a lower level of ABA compared with wild-types during germination. Our data suggest that PA distribution in the seed coat can act as a doorkeeper to seed germination. 2ZBP1 mediates interferon-induced necroptosis显示文摘Interferons(IFNs)play an important role in immunomodulatory and antiviral functions.IFN-induced necroptosis has been reported in cells deficient in receptor-interacting protein kinase 1(RIPK1),Fas-associated protein with death domain(FADD),or caspase-8,but the mechanism is largely unknown.Here,we report that the DNA-dependent activator of IFN regulatory factors(ZBP1,also known as DAI)is required for both type Ⅰ(β)and type Ⅱ(γ)IFN-induced necroptosis.We show that L929 fibroblast cells became susceptible to IFN-induced necroptosis when RIPK1,FADD,or Caspase-8 was genetically deleted,confirming the antinecroptotic role of these proteins in IFN signaling.We found that the pronecroptotic signal from IFN stimulation depends on new protein synthesis and identified ZBP1,an IFN-stimulated gene(ISG)product,as the de novo synthesized protein that triggers necroptosis in IFN-stimulated cells.The N-terminal domain(ND)of ZBP1 is important for ZBP1–ZBP1 homointeraction,and its RHIM domain in the C-terminal region interacts with RIPK3 to initiate RIPK3-dependent necroptosis.The antinecroptotic function of RIPK1,FADD,and caspase-8 in IFN-treated cells is most likely executed by caspase-8-mediated cleavage of RIPK3,since the inhibitory effect on necroptosis was eliminated when the caspase-8 cleavage site in RIPK3 was mutated.ZBP1-mediated necroptosis in IFN-treated cells is likely physiologically relevant,as ZBP1 KO mice were significantly protected against acute systemic inflammatory response syndrome(SIRS)induced by TNF+IFN-γ.Daowei Yang Yaoji Liang Shubo Zhao Yan Ding Qiuyu Zhuang Qilin Shi Tingting Ai Su-Qin Wu Jiahuai Han 2020Cellular & Molecular Immunology2020,17,4:5
3Efficacy of Qingre Huashi decoction(清热化湿方) on infection of Helicobacter pylori:inhibiting adhesion,antioxidant,and anti-inflammation显示文摘OBJECTIVE: To investigate the phytochemical profile of Qingre Huashi decoction( 清 热 化 湿 方, QHD) and evaluate the mechanisms rationalizing the use of QHD against Helicobacter pylori(H. pylori)-associated gastritis. METHODS: QHD is composed of 11 herbs, which was prepared by a fixed Pharmacy and concentrated into clear paste. High-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry(HPLC-QTOF/MS) was used to detect the phytochemical profile of QHD. The toxicity of QHD against H. pylori and human gastric epithelial cells was evaluated by the toxicology test and cell counting kit-8 assay. The adhesion model was constructed by incubating H. pylori and gastric mucosal epithelial cells for 2 h. The urease assay was used to examine the antiadhesion effects of QHD, and gene expression of adhesins was evaluated by quantitative polymerase chain reaction. The antioxidant activity was assessed by DCFHDA labeling. To evaluate the anti-inflammatory effect, the levels of pro-inflammatory cytokines in the culture supernatant were detected by enzyme-linked immunosorbent assay. RESULTS: HPLC-QTOF/MS profiling indicated the presence of primary compounds 1-20 in QHD. Drug concentration was determined as 1, 2, and 5 mg/m L by the toxic concentration of QHD against H. pylori and human gastric epithelial cells. QHD prevented H. pylori adhesion to the human gastric epithelial cells and reduced levels of reactive oxygen species. QHD also reduced the level of interleukin-8 and other proinflammatory cytokines that were upregulated by H. pylori infection. 5An on-Si directional second harmonic generation amplifier for MoS2/WS2 heterostructure显示文摘Transition metal dichalcogenides(TMD)heterostructure is widely applied for second harmonic generation(SHG)and holds great promises for laser source,nonlinear switch,and optical logic gate.However,for atomically thin TMD heterostructures,low SHG conversion efficiency would occur due to reduction of light-matter interaction length and lack of phase matching.Herein,we demonstrated a facile directional SHG amplifier formed by MoS2/WS2 monolayer heterostructures suspended on a holey SiO_(2)/Si substrate.The SHG enhancement factor reaches more than two orders of magnitude in a wide spectral range from 355 to 470 nm,and the radiation angle is reduced from 38°to 19°indicating higher coherence and better emission directionality.The giant SHG enhancement and directional emission are attributed to the great excitation and emission field concentration induced by a self-formed vertical Fabry-Pérot microcavity.Our discovery gives helpful insights for the development of two-dimensional(2D)nonlinear optical devices.Jiaxing Du Jianwei Shi Chun Li Qiuyu Shang Xinfeng Liu Yuan Huang Qing Zhang 2023Nano Research2023,16,3:0
6Targeted screening of an anti-inflammatory polypeptide from Rhopilema esculentum Kishinouye cnidoblasts and elucidation of its mechanism in alleviating ulcerative colitis based on an analysis of the gut microbiota and metabolites显示文摘Ulcerative colitis(UC)is a recurrent inflammatory bowel disease that imposes a severe burden on families and society.In recent years,exploiting the potential of marine bioactive peptides for the treatment of diseases has become a topic of intense research interest.This study revealed the mechanism underlying the protective effect of the dominant polypeptide PKKVV(Pro-Lys-Lys-Val-Val)of Rhopilema esculentum cnidoblasts against DSS-induced UC through a combined analysis of the metagenome and serum metabolome.Specifically,the polypeptide composition of R.esculentum cnidoblasts was determined by matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF/TOF-MS).Molecular docking showed that the dominant peptide PKKVV could bind better with tumor necrosis factor-α(TNF-α)than the original ligand.Subsequent animal experiments suggested that PKKVV could modulate disorganized gut microorganisms in mice with UC;affect serum metabolites through the arachidonic acid,glycerophospholipid and linoleic acid metabolism pathways;and further alleviate UC symptoms.This study provides a reference for the comprehensive development of marine bioactive substances and nonpharmaceutical treatments for UC.Ziyan Wang Qiuyue Shi Ying Feng Jiaojiao Han Chenyang Lu Jun Zhou Zhonghua Wang Xiurong Su 2024Food Science and Human Wellness2024,13,3:0

9Effect of Counterparts and Applied Load on the Tribological Behavior of the Graphene–Nickel Matrix Self-Lubricating Composite显示文摘Graphene-oxide (GO) has been recognized as an excellent lubrication material owing to its two-dimensional structure and weak interlayer interactions. However, the functional groups of GO that can contribute to anti-friction, anti-wear, and superlubricity are yet to be elucidated. Hence, further improvement in GO-family materials in tribology and superlubricity fields is impeded. In this study, macroscale superlubricity with a coefficient of friction of less than 0.01 is achieved by exploiting the high adhesive force between amino groups within aminated GO (GO–NH_(2)) nanosheets and SiO_(2). It was observed that GO–NH_(2) nanosheets form a robust adsorption layer on the worn surfaces owing to the high adsorption of amino groups. This robust GO–NH_(2) adsorption layer not only protects the contact surfaces and contributes to low wear, but also causes the shearing plane to transform constantly from solid asperities (high friction) into GO–NH_(2) interlayers (weak interlayer interactions), resulting in superlubricity. A SiO_(2)-containing boundary layer formed by tribochemical reactions and a liquid film are conducive to low friction.
