1Genetic Structure and Diversity of Parental Cultivars Involved in China Mainland Sugarcane Breeding Programs as Inferred from DNA Microsatellites显示文摘To understand genetic structure and diversity of parental cultivars involved in China Mainland sugarcane breeding programs,92 elite parents and 4 wild relatives were genotyped with 18 microsatellite DNA markers.The genetic similarity(GS) values among the cultivars ranged from 0.346 to 0.960 with an average of 0.533.Among the introduced cultivars,India accessions had the closest genetic distance to China Mainland accessions(0.447),while Australia accessions have the furthest distance(0.503).A comparison of allelic diversity among geographical origins showed that there were 22 China Mainland specific alleles,of which 28% were derived from native S.spontaneaum germplasm in China.Model-based genetic structure,clustering,and principal components analyses consistently revealed there were five groups within the 96 accessions.Groups 1,2,4,and 5 consisted of all cultivars and group 3 only contained wild germplasm.Group 2 was characterized as the Introduction group with 46 cultivars predominantly introduced from Australia,Taiwan of China,India,and USA.Groups 1,4,and 5 consisted of cultivars mostly originated from China Mainland,defined as the Complex group,Yacheng lines group,and F134/CP72-1210 group,respectively,upon their pedigree.By understanding the genetic relationships among the parental cultivars,breeders can gain a rational basis for expanding the gene pool and select the best parental accessions for crossing.QI Yong-wen PAN Yong-bao LAO Fang-ye ZHANG Chui-ming FAN Li-na HE Hui-yi LIU Ruin WANG Qin-nan LIU Shao-mou LIU Fu-ye LI Qi-wei DENG Hai-hua 2012Journal of Integrative Agriculture2012,11,11:6
2Characterization of polyclonal antibodies against nonstructural protein 9 from the porcine reproductive and respiratory syndrome virus显示文摘Porcine reproductive and respiratory syndrome(PRRS) is considered to be one of the most important infectious diseases impacting the swine industry and is characterized by reproductive failure in late term gestation in sows and respiratory disease in pigs of all ages.The nonstructural protein 9 gene,Nsp9,encoding the RNA-dependent RNA polymerase,is generally regarded as fairly conserved when compared to other viral proteins.Antibodies against Nsp9 will be of great importance for the diagnosis and treatment of the causal agent,PRRS virus.A study was undertaken to generate polyclonal antibodies against the immunodominant Nsp9.For this purpose,the Nsp9 was expressed in Escherichia coli and subsequently used as an antigen to immunize New Zealand rabbits.Antiserum was identified via an indirect ELISA,and then verified based on the ability to react with both naturally and artificially expressed Nsp9.Results of virus neutralization test showed that this antiserum could not neutralize the PRRSV.Nevertheless,this antiserum as a diagnostic core reagent should prove invaluable for further investigations into the mechanism of PRRS pathogenesis.Mengmeng ZHAO Juanjuan QIAN Jiexiong XIE Tiantian CUI Songling FENG Guoqiang WANG Ruining WANG Guihong ZHANG 2016Frontiers of Agricultural Science and Engineering2016,3,2:2

4Efficient purification of cell culture-derived classical swine fever virus by ultrafiltration and size-exclusion chromatography显示文摘Large-scale production of cell culture-based classical swine fever virus(CSFV)vaccine is hampered by the adverse reactions caused by contaminants from host cell and culture medium.Hence,we have developed an efficient method for purifying CSFV from cell-culture medium.Pure viral particles were obtained with two steps of tangential-flow filtration(TFF)and size-exclusion chromatography(SEC),and were compared with particles from ultracentrifugation by transmission electron microscopy(TEM),infectivity and recovery test,and real time fluorescent quantitative PCR(FQ-PCR).TFF concentrated the virus particles effectively with a retention rate of 98.5%,and 86.2%of viral particles were obtained from the ultrafiltration retentate through a Sepharose 4 F F column on a biological liquid chromatography system.CSFV purified by TFF-SEC or ultracentrifugation were both biologically active from 1.0×10–4.25 TCID50·mL^(–1) to 3.0×10^(–6.25) TCID50·mL^(–1),but the combination of TFF and SEC produced more pure virus particles than by ultracentrifugation alone.In addition,pure CSFV particles with the expected diameter of 40–60 nm were roughly spherical without any visible contamination.Mice immunized with CSFV purified by TFF-SEC produced higher antibody levels compared with immunization with ultracentrifugation-purified CSFV(P<0.05).The purification procedures in this study are reliable technically and feasible for purification of large volumes of viruses.Ruining WANG Yubao ZHI Junqing GUO Qingmei LI Li WANG Jifei YANG Qianyue JIN Yinbiao WANG Yanyan YANG Guangxu XING Songlin QIAO Mengmeng ZHAO Ruiguang DENG Gaiping ZHANG 2015Frontiers of Agricultural Science and Engineering2015,2,3:1
5Intelligent sorting method for assembly line based on visual positioning and robotic arm model predictive control显示文摘The existing steering device in the fruit and vegetable packaging assembly line cannot adjust the attitude of lettuce to a unified attitude,affecting the input and packaging process of the packaging machine.This study proposes an intelligent assembly line sorting method based on the visual positioning and model predictive control of a robotic arm.First,lightweight improvement based on the YOLOv5 is realized,the lettuce stalk in the background of the conveyor belt is promptly identified,the image of the lettuce stalk in the anchor box area is processed,and the edge contour point set is determined to extract the pixel coordinates of the optimal grasp point and mirror inclination angle of the lettuce.For the intelligent assembly line system,a robot arm kinematics model is constructed and the robot kinematics inverse solutions are calculated.Additionally,the lettuce movement speeds are dynamically measured by the vision system.A combination of the model prediction control,dynamic tracking,and rapid sorting of the lettuce by the robot claw is realized.The results show that the average detection time of a single frame image in the visual positioning part is 0.014 s,which is reduced by 50%;the accuracy and recall are 98%and 95%,respectively.The detection time is significantly reduced by ensuring accuracy.Within the current speed range of the packaging assembly line conveyor belt,the manipulator can grasp lettuce at different speeds stably and fast;the average axial error,average radial error,and adjusted average inclination angle error are 0.71 cm,1.02 cm,and 3.79°,respectively,verifying the high efficiency and stability of the model.The proposed method of this study enables application in the intelligent sorting operation of industrial assembly lines.Ruining Zhang Wei Lu Xingliang Jian Hui Luo 2023International Journal of Agricultural and Biological Engineering2023,16,4:0
6AUDITORY BRAINSTEM RESPONSES IN SENILE PRESBYCUSIS PATIENTS OVER 90 YEARS显示文摘Objective To analyze the characteristics of auditory brainstem response(ABR) in presbycusis patients elder than 90 years. Methods Fourteen presbycusis patients elder than 90 years(presbycusis group, 91.1.4±1.3 years, 26 ears) and 9 normal-hearing young adults(control group, 22.7±1.2 years, 18 ears) participated in the study. Alternative click-evoked ABRs were recorded in both groups. The peak latency(PL) of peak I,Ⅲ, and V, and the inter-peak latency(IPI) of I-Ⅲ, Ⅲ-V, and I-V were compared between groups. Results In elder presbycusis patients, the occurrence rate of peak I and Ⅲ were both 76.9%, and that of peak V was84.6%. In presbycusis group, the peak latencies of I, Ⅲ, V were significantly longer than that of control group(P<0.001). There was no significant difference between groups in the IPI of peak I-I Ⅲ(P=0.298,peak Ⅲ-V(P=0.254) and peak I-V(P=0.364). 7FOLR1-induced folate deficiency reduces viral replication via modulating APOBEC3 family expression显示文摘Folate receptor alpha(FOLR1)is vital for cells ingesting folate(FA).FA plays an indispensable role in cell pro-liferation and survival.However,it is not clear whether the axis of FOLR1/FA has a similar function in viral replication.In this study,we used vesicular stomatitis virus(VSV)to investigate the relationship between FOLR1-mediated FA deficiency and viral replication,as well as the underlying mechanisms.We discovered that FOLR1 upregulation led to the deficiency of FA in HeLa cells and mice.Meanwhile,VSV replication was notably sup-pressed by FOLR1 overexpression,and this antiviral activity was related to FA deficiency.Mechanistically,FA deficiency mainly upregulated apolipoprotein B mRNA editing enzyme catalytic subunit 3B(APOBEC3B)expression,which suppressed VSV replication in vitro and in vivo.In addition,methotrexate(MTX),an FA metabolism inhibitor,effectively inhibited VSV replication by enhancing the expression of APOBEC3B in vitro and in vivo.Overall,our present study provided a new perspective for the role of FA metabolism in viral infections and highlights the potential of MTX as a broad-spectrum antiviral agent against RNA viruses.Jing Wu Yajing Han Ruining Lyu Fang Zhang Na Jiang Hongji Tao Qiao You Rui Zhang Meng Yuan Waqas Nawaz Deyan Chen Zhiwei Wu 2023Virologica Sinica2023,38,3:0
8Research progress on corrosion behaviors and biocompatibility of rare-earth magnesium alloys in vivo and in vitro显示文摘As yet,Mg alloys acting as the medical implants have drawn extensive attention,due to their spontaneous degrada bility,effective load-transmissibility and the excellent biocompatibility,particularly in bone tissue reconstruction and vascular radial-support.Regrettably,they were inevitably affected by the tension/compression-torsion,dynamic erosion and corrosion fatigue under complex service conditions,which lead to premature failure of implantation-materials.Micro-alloying addition is an effective way to delay the rapid degradation,especially in rare-earth micro-composite addition.It can not only reduce intensities of galvanic-corrosion by refining the grain sizes and adjusting the Volta-potentials distribution of the precipitates,but also modify the compositions and biocompatibility of the degradation products.Moreover,the higher compress tress on the surface can improve the stability and densification of the film layer,which enhanced the corrosion resistance.Thus,the latest research progress about in vivo/vitro degradation behavio rs and bioco mpatibility of rare-earth Mg alloys is reviewed;The internal relationships between rare-earth elements,phase features and degradation behaviors of Mg alloys are summarized.Moreover,the effects of rare-earth addition on the film-characteristics are deeply explained,and the induced mechanisms of rare earth elements on the biocompatibility are revealed.Yuan Zhang Yun Liu Ruining Zheng Yaqi Zheng Liansheng Chen 2023Journal of Rare Earths2023,41,12:0
