Maternal genetic history of ancient Tibetans over the past 4000 years显示文摘The settlement of the Tibetan Plateau epitomizes human adaptation to a high-altitude environment that poses great challenges to human activity. Here,we reconstruct a 4000-year maternal genetic history of Tibetans using 128 ancient mitochondrial genome data from 37 sites in Tibet. The phylogeny of haplotypes M9a1a,M9a1b,D4g2,G2a’c,and D4i show that ancient Tibetans share the most recent common ancestor with ancient Middle and Upper Yellow River populations around the Early and Middle Holocene. In addition, the connections between Tibetans and Northeastern Asians vary over the past 4000 years, with a stronger matrilineal connection between the two during 4000 BPe3000 BP,and a weakened connection after 3000 BP,that are coincident with climate change, followed by a reinforced connection after the Tubo period(1400 BPe1100 BP).Besides,an over 4000-year matrilineal continuity is observed in some of the maternal lineages. We also find the maternal genetic structure of ancient Tibetans is correlated to the geography and interactions between ancient Tibetans and ancient Nepal and Pakistan populations.Overall, the maternal genetic history of Tibetans can be characterized as a long-term matrilineal continuity with frequent internal and external population interactions that are dynamically shaped by geography, climate changes,as well as historical events. | Ganyu Zhang Can Cui Shargan Wangdue Hongliang Lu Honghai Chen Lin Xi Wei He Haibing Yuan Tinley Tsring Zujun Chen Feng Yang Tashi Tsering Shuai Li Norbu Tashi Tsho Yang Yan Tong Xiaohong Wu Linhui Li Yuanhong He Peng Cao Qingyan Dai Feng Liu Xiaotian Feng Tianyi Wang Ruowei Yang Wanjing Ping Ming Zhang Xing Gao Yichen Liu Wenjun Wang Qiaomei Fu | 2023 | Journal of Genetics and Genomics2023,50,10: | 1

Maternal genetic history of southern East Asians over the past 12,000 years显示文摘Southern East Asia, including Guangxi and Fujian provinces in China,is home to diverse ethnic groups, languages, and cultures.Previous studies suggest a high complexity regarding population dynamics and the history of southern East Asians.However,large-scale genetic studies on ancient populations in this region are hindered by limited sample preservation. Here, using highly efficient DNA capture techniques,we obtain 48 complete mitochondrial genomes of individuals from Guangxi and Fujian in China and reconstruct their maternal genetic history over the past 12,000 years. We find a strong connection between southern East Asians dating to~12,000-6000 years ago and present-day Southeast Asians. In addition, stronger genetic affinities to northern East Asians are observed in historical southern East Asians than Neolithic southern East Asians, suggesting increased interactions between northern and southern East Asians over time.Overall,we reveal dynamic connections between ancient southern East Asians and populations located in surrounding regions,as well as a shift in maternal genetic structure within the populations over time. | Yalin Liu Tianyi Wang Xichao Wu Xuechun Fan Wei Wang Guangmao Xie Zhen Li Qingping Yang Peng Cao Ruowei Yang Feng Liu Qingyan Dai Xiaotian Feng Wanjing Ping Bo Miao Yun Wu Yichen Liu Qiaomei Fu | 2021 | Journal of Genetics and Genomics2021,48,10: | 1
VHH212 nanobody targeting the hypoxia-inducible factor 1α suppresses angiogenesis and potentiates gemcitabine therapy in pancreatic cancer in vivo显示文摘Objective:We aimed to develop a novel anti-HIF-1αintrabody to decrease gemcitabine resistance in pancreatic cancer patients. Methods:Surface plasmon resonance and glutathione S-transferase pull-down assays were conducted to identify the binding affinity and specificity of anti-HIF-1αVHH212[a single-domain antibody(nanobody)].Molecular dynamics simulation was used to determine the protein-protein interactions between hypoxia-inducible factor-1α(HIF-1α)and VHH212.The real-time polymerase chain reaction(PCR)and Western blot analyses were performed to identify the expressions of HIF-1αand VEGF-A in pancreatic ductal adenocarcinoma cell lines. The efficiency of the VHH212 nanobody in inhibiting the HIF-1 signaling pathway was measured using a dual-luciferase reporter assay. Finally,a PANC-1 xenograft model was developed to evaluate the anti-tumor efficiency of combined treatment.Immunohistochemistry analysis was conducted to detect the expressions of HIF-1αand VEGF-A in tumor tissues.Results:VHH212 was stably expressed in tumor cells with low cytotoxicity, high affinity, specific subcellular localization, and neutralization of HIF-1αin the cytoplasm or nucleus. The binding affinity between VHH212 and the HIF-1αPAS-B domain was 42.7 n M.Intrabody competitive inhibition of the HIF-1αheterodimer with an aryl hydrocarbon receptor nuclear translocator was used to inhibit the HIF-1/VEGF pathway in vitro.Compared with single agent gemcitabine,co-treatment with gemcitabine and a VHH212-encoding adenovirus significantly suppressed tumor growth in the xenograft model with 80.44%tumor inhibition. Conclusions:We developed an anti-HIF-1αnanobody and showed the function of VHH212 in a preclinical murine model of PANC-1 pancreatic cancer. The combination of VHH212 and gemcitabine significantly inhibited tumor development. These results suggested that combined use of anti-HIF-1αnanobodies with first-line treatment may in the future be an effective treatment for pancreatic cancer. | Guangbo Kang Min Hu He Ren Jiewen Wang Xin Cheng Ruowei Li Bo Yuan Yasmine Balan Zixuan Bai He Huang | 2021 | Cancer Biology & Medicine2021,18,3: | 1

Large electrocaloric effect over a wide temperature span in lead-free bismuth sodium titanate-based relaxor ferroelectrics显示文摘For efficient solid-state refrigeration technologies based on electrocaloric effect(ECE),it is a great challenge of simultaneously obtaining a large adiabatic temperature change(DT)within a wide temperature span(Tspan)in lead-free ferroelectric ceramics. Here,we studied the electrocaloric effect(ECE)in(1-x)(Na_(0.5)Bi_(0.5))TiO_(3)-xCaTiO_(3)((1-x)NBT-xCT)and explored the combining effect of morphotropic phase boundary(MPB)and relaxor feature. The addition of CT not only constructs a MPB region with the coexistence of rhombohedral and orthorhombic phases, but also enhances the relaxor feature. The ECE peak appears around the freezing temperature(Tf),and shifts toward to lower temperature with the increasing CT amount. The directly measured ECE result shows that the ceramic of x=0.10,which is in the MPB region, has an optimal ECE property of DTmax=1.28 K@60℃under 60 kV/cm with a wide Tspan of 65C.The enhanced ECE originates from the electric-field-induced transition between more types of polar nanoregions and long-range ferroelectric macrodomains. For the composition with more relaxor feature in the MPB region, such as x?0.12,the ECE is relatively weak under low electric fields but it exhibits a sharp increment under a sufficiently high electric field. This work provides a guideline to develop the solidestate cooling devices for electronic components. | Xiaopo Su Junjie Li Yuxuan Hou Ruowei Yin Jianting Li Shiqiang Qin Yanjing Su Lijie Qiao Chuanbao Liu Yang Bai | 2023 | Journal of Materiomics2023,9,2: | 0
Correlation between multi-factor phase diagrams and complex electrocaloric behaviors in PNZST antiferroelectric ceramic system显示文摘Ferroelectric(FE)phase transition with a large polarization change benefits to generate large electrocaloric(EC)effect for solid-sate and zero-carbon cooling application. However, most EC studies only focus on the single-physical factor associated phase transition.Herein,we initiated a comprehensive discussion on phase transition in Pb_(0.99)Nb_(0.02)[(Zr_(0.6)Sn_(0.4))1−xTix]_(0.98)O_(3)(PNZST100x)antiferroelectric(AFE)ceramic system under the joint action of multi-physical factors, including composition, temperature, and electric field.Due to low energy barrier and enhanced zero-field entropy, the multi-phase coexistence point(x=0.12)in the composition–temperature phase diagram yields a large positive EC peak of maximum temperature change(ΔT_(max))=2.44 K(at 40 kV/cm).Moreover,the electric field–temperature phase diagrams for four representative ceramics provide a more explicit guidance for EC evolution behavior.Besides the positive EC peaks near various phase transition temperatures, giant positive EC effects are also brought out by the electric field-induced phase transition from tetragonal AFE(AFET)to low-temperature rhombohedral FE(FER),which is reflected by a positive-slope boundary in the electric field–temperature phase diagram, while significant negative EC responses are generated by the phase transition from AFET to high-temperature multi-cell cubic paraelectric(PEMCC)with a negative-slope phase boundary. This work emphasizes the importance of phase diagram covering multi-physical factors for high-performance EC material design. | Junjie Li Ruowei Yin Jianting Li Xiaopo Su Yanjing Su Lijie Qiao Yang Baia | 2023 | Journal of Advanced Ceramics2023,12,3: | 0
Ancient DNA reveals the maternal genetic history of East Asian domestic pigs显示文摘Zoo-archaeological and genetic evidence suggest that pigs were domesticated independently in Central China and Eastern Anatolia along with the development of agricultural communities and civilizations. However, the genetic history of domestic pigs, especially in China, has not been fully explored. In this study,we generate 42 complete mitochondrial DNA sequences from~7500-to 2750-year-old individuals from the Yellow River basin. Our results show that the maternal genetic continuity of East Asian domestic pigs dates back to at least the Early to Middle Neolithic. In contrast, the Near Eastern ancestry in European domestic pigs saw a near-complete genomic replacement by the European wild boar. The majority of East Asian domestic pigs share close haplotypes, and the most recent common ancestor of most branches dates back to less than 20,000 years before present, inferred using new substitution rates of whole mitogenomes or combined protein-coding regions. Two major population expansion events of East Asian domestic pigs coincided with changes in climate, widespread adoption of introduced crops, and the development of agrarian societies. These findings add to our understanding of the maternal genetic composition and help to complete the picture of domestic pig evolutionary history in East Asia. | Ming Zhang Yichen Liu Zhipeng Li Peng Lü Jacob D.Gardner Maolin Ye Jihuai Wang Miaomiao Yang Jing Shao Weilin Wang Qingyan Dai Peng Cao Ruowei Yang Feng Liu Xiaotian Feng Lizhao Zhang Enwei Li Yunzheng Shi Zehui Chen Shilun Zhu Weiwei Zhai Tao Deng Ziyuan Duan E.Andrew Bennett Songmei Hu Qiaomei Fu | 2022 | Journal of Genetics and Genomics2022,49,6: | 0
Dynamics and controls of ecosystem multiserviceability across the Qingzang Plateau显示文摘Ecosystem multiserviceability(EMS),a comprehensive and significant ecological indicator, reflects the capacity of ecosystems to offer multiple services concurrently.Intensified climate change and human activity are continu-ously altering ecosystem functions, services, and EMSs.However, numerous studies have only focused on one or a few ecosystem services, rarely taking into account spatial-temporal distribution and drivers of EMS on behalf of different agencies. We calculated EMS including pastoralist(PA),environmental protection agency(EPA),bio-diversity conservation agency(BCA),and climate change mitigation agency(CCMA)using grassland production, habitat quality, water conservation, and carbon sequestration. Then, the effects of geographical features, climate factors, and human activities on spatial-temporal patterns of EMS were explored. The result indicated that EMS showed a decreasing tendency from the southeast to northwest on the Qingzang Plateau(QZP).Meanwhile, there were no obvious fluctuations in four simulated scenarios(PA,EPA,BCA and CCMA)among different vegetation types during 2000 to 2015.Notably,EMS of all simulated scenarios decreased in the alpine steppe ecosystem, but negligible changes were found in other ecosystems from 2015 to 2020.Moreover,the relative importance of precipitation in annual mean value(from 2000 to 2020)of PA,EPA,BCA and CCMA were 0.13,0.11,0.30 and 0.19,respectively.Overall,precipitation played the dominant role on the dynamics of EMS,followed by elevation and human footprint. Our findings highlighted that understanding the patterns and drivers of EMS could provide a reference for the regional management and maintenance of ecosystem stability on QZP. | Ruowei Li Guodong Han Jian Sun Tiancai Zhou Junhe Chen Wen He Yi Wang | 2023 | Geography and Sustainability2023,4,4: | 0
