| 6 | Enhanced CD19 activity in B cells contributes to immunodeficiency in mice deficient in the ICF syndrome gene Zbtb24显示文摘Immunodeficiency,centromeric instability,and facial anomalies(ICF)syndrome is a rare autosomal recessive disorder characterized by DNA hypomethylation and antibody deficiency.It is caused by mutations in DNMT3B,ZBTB24,CDCA7,or HELLS.While progress has been made in elucidating the roles of these genes in regulating DNA methylation,little is known about the pathogenesis of the life-threatening hypogammaglobulinemia phenotype.Here,we show that mice deficient in Zbtb24 in the hematopoietic lineage recapitulate the major clinical features of patients with ICF syndrome.Specifically,Vav-Cre-mediated ablation of Zbtb24 does not affect lymphocyte development but results in reduced plasma cells and low levels of IgM,IgG1,and IgA.Zbtb24-deficient mice are hyper and hypo-responsive to T-dependent and T-independent type 2 antigens,respectively,and marginal zone B-cell activation is impaired.Mechanistically,Zbtb24-deficient B cells show severe loss of DNA methylation in the promoter region of Il5ra(interleukin-5 receptor subunit alpha),and Il5ra derepression leads to elevated CD19 phosphorylation.Heterozygous disruption of Cd19 can revert the hypogammaglobulinemia phenotype of Zbtb24-deficient mice.Our results suggest the potential role of enhanced CD19 activity in immunodeficiency in ICF syndrome. | Zhengzhou Ying Swanand Hardikar Joshua B.Plummer Tewfik Hamidi Bin Liu Yueping Chen Jianjun Shen Yunxiang Mu Kevin M.McBride Taiping Chen | 2023 | Cellular & Molecular Immunology2023,20,12: | 0 |
| 7 | Meta-Analysis of Smoking and the Risk of Gastric Cancer among the Chinese Population显示文摘OBJECTIVE To reevaluate the effect of tobacco smoking on therisk of developing gastric cancer among the Chinese population.METHODS Thirty articles from the literature both in Chineseand English from January,1988 to present were identified andfrom which adjusted odd ratios(ORs)or relative risks(RRs)werecombined by meta-analysis.Generalized least squares(GLS)fortrend estimation of summarized dose-response data was carriedout.All the analyses were performed using software of STATAversion 10.0.RESULTS Comparing current smokers with subjects who havenever smoked,the summary effect values on gastric cancer witha 95% confidence interval(CI)were 1.67(1.43-1.96)for case-control studies and 1.52(1.17-1.96)for cohort studies,respectively.The combined effect values with a 95%CI for the comparison ofsmoking quantity of current smokers with the referent groupwere 1.41(1.15-1.72)for case-control studies and 1.24(1.02-1.52)for cohort studies,respectively.The combined effect values witha 95%CI for the comparison of accumulative years smoked ofcurrent smokers with the referent group were 1.07(0.89-1.28)for case-control studies and 1.28(0.95-1.74)for cohort studies,respectively.Dose-response meta-analysis showed that the gastriccancer risk will increase 50%(OR=1.50,95%CI:1.34-1.67)witheach 20 cigarette per day increment and increase 14%(OR=1.14,95%CI:1.10-1.18)with each 10 year increment of smoking.CONCLUSION The gastric cancer risk among the Chinesepopulation is significantly associated with tobacco smoking andthe smoking quantity per day.Smoking cessation should be morestrongly advocated. | Na Liu Yueping Shen Liqiang Qin Tianfeng He Yinmei Liu | 2009 | Clinical oncology and cancer resexreh2009,6,4: | 0 |
| 8 | Influence of the Kunlun Mountain M_S8.1 Earthquake on Horizontal Crustal Deformation in the Sichuan and Yunnan Area显示文摘In order to track the space-time variation of regional strain field holistically(in a large scale) and to describe the regional movement field more objectively,the paper uses a nonlinear continuous strain model focused on extracting medium-low frequency strain information on the basis of a region with no rotation.According to the repeated measurements(1999~2001~2004) from GPS monitoring stations in the Sichuan and Yunnan area obtained by the Project of 'China Crust Movement Measuring Network',and with the movement of 1999~2001(stage deformation background) as the basic reference,we separated the main influencing factors of the Kunlun Mountain M-S8.1 earthquake in 2001 from the data of 2001 and 2004,and the results indicate:(1) the Kunlun Mountain M-S8.1 earthquake has a discriminating effect on the Sichuan and Yunnan area,moreover,the deformation mode and background had not only certain similitude but also some diversity;(2) The movement field before the earthquake was very ordinal,while after the earthquake,order and disorder existed simultaneously in the displacement field;The displacement quantities of GPS monitoring stations were generally several millimeters;(3) The principal strain field before earthquake was basically tensile in an approximate EW direction and compressive in the SN direction,and tension was predominant.After the earthquake,the principal strain field in the Sichuan area was compressive in the EW direction and tensile in the SN direction,and the compression was predominant.In the Yunnan area,it was tensional in the NE direction and compressive in the NW direction,and tension was predominant;(4) The surficial strain before the earthquake was dominated by superficial expansion,the contractive area being located basically in the east boundary of Sichuan and Yunnan block and its neighborhood.After the earthquake,the Sichuan area was surface contractive(the further north,the greater it was),and south of it was an area of superficial expansion.Generally speaking,the Kunlun Mountain M-S8.1 earthquake played an active role in the accumulation of energy in the Sichuan and Yunnan area.Special attention shall be focused on the segment of Xichang-Dongchuan and its neighborhood. | Yang Guohua Jiang Zaiseng Zhang Fengshuang Liu Xia Han Yueping Shen Wuchun Wang Li | 2007 | Earthquake Research in China2007,21,3: | 0 |
| 9 | A highly selective fluorescent probe for visualizing dry eye disease-associated viscosity variations显示文摘Dry eye disease(DED) is a multifactorial chronic inflammatory disease of the ocular surface with complex and unclear etiology. The development of reliable detection tools for the pathology of DED will benefit its treatment, but it is still lacking. In parallel, it has been discovered recently that viscosity changes are involved in inflammation processes. In this regard, we constructed a fluorescent probe V5with an asymmetric donor-acceptor-donor(D-A-D) feature after rational structural modulation for viscosity detection during DED progression. The probe manifested a remarkable fluorescence enhancement(110 folds) in highly viscous conditions without interferences from polarity and reactive species. Specifically, no aggregation effect of the probe was found in glycerol. Moreover, viscosity increment in human corneal epithelial cells(HCECs) induced by hyperosmosis and inflammation was monitored, and ferroptosis in HCECs also led to the viscosity elevation. A reactive oxygen species(ROS)-dependent viscosity changes during DED progression is demonstrated. Finally, viscosity change in corneal epithelial cell layer from mice treated by scopolamine was also visualized for the first time.
