13D bioprinting of integral ADSCs-NO hydrogel scaffolds to promote severe burn wound healing显示文摘Severe burns are challenging to heal and result in significant death throughout the world.Adiposederived mesenchymal stem cells(ADSCs)have emerged as a promising treatment for fullthickness burn healing but are impeded by their low viability and efficiency after grafting in vivo.Nitric oxide(NO)is beneficial in promoting stem cell bioactivity,but whether it can function effectively in vivo is still largely unknown.In this study,we bioprinted an efficient biological scaffold loaded with ADSCs and NO(3D-ADSCs/NO)to evaluate its biological efficacy in promoting severe burn wound healing.The integral 3D-ADSCs/NO hydrogel scaffolds were constructed via 3D bioprinting.Our results shown that 3D-ADSCs/NO can enhance the migration and angiogenesis of Human Umbilical Vein Endothelial Cells(HUVECs).Burn wound healing experiments in mice revealed that 3D-ADSCs/NO accelerated the wound healing by promoting faster epithelialization and collagen deposition.Notably,immunohistochemistry of CD31 suggested an increase in neovascularization,supported by the upregulation of vascular endothelial growth factor(VEGF)mRNA in ADSCs in the 3D biosystem.These findings indicated that 3D-ADSC/NO hydrogel scaffold can promote severe burn wound healing through increased neovascularization via the VEGF signalling pathway.This scaffold may be considered a promising strategy for healing severe burns.Yu Wu Tangzhao Liang Ying Hu Shihai Jiang Yuansen Luo Chang Liu Guo Wang Jing Zhang Tao Xu Lei Zhu 2021Regenerative Biomaterials2021,8,3:2

3Effect of low dose fractionated radiation on reversing cisplatin resistance in ovarian carcinoma via VEGF and mTOR显示文摘Objective To investigate the mechanism of low-dose fractionated radiation on reversing cisplatin resistance in ovarian carcinoma via vascular endothelial growth factor(VEGF) and mammalian target of rapamycin(mTOR) in vivo.Methods Human cisplatin-resistant ovarian carcinoma cells(SKOV3/DDP) were injected into nude mice to establish ovarian cancer xenografts. The mice were randomly divided into three groups: a control group, a low-dose fractionated radiation(LDRFT) group, and a conventional-dose radiation group. Each group was exposed to 0 cGy, 50 cGy, and 200 cGy radiation, respectively, for 4 weeks, up to a total of 8.0 Gy. Mice in the LDFRT group were irradiated twice daily with 6 hour intermissions on day 1 and 2 of every week for a total of 4 weeks. Conventional-dose group mice were given a single 200 cGy radiation dose on the first day each week for a total of 4 weeks. Maximum horizontal and vertical diameters of the tumors were measured every other day and used to create a tumor growth curve. After 4 weeks of irradiation, we dissected the tumor tissue and calculated the tumor inhibition rate. RT-PCR detected the expression of VEGF and m TOR, and Western blots detected the expression of corresponding proteins.Results Both LDRFT and conventional-dose radiation inhibited the growth of tumor cells, and growth of tumors in the two radiation groups compared with growth in the control group were significantly different(P < 0.05). The rate of tumor inhibition in the LDFRT group(37.5603%) was lower than in the conventionaldose group(47.4446%), but there was no significant difference(P 0.05). Compared with the other two groups, the m RNA expression of VEGF was significantly lower in the LDFRT group(P < 0.05), but there was no obvious difference between the conventional-dose and control groups. There was no obvious difference in the m RNA expression of m TOR among the three groups, but the expression of the protein p-m TOR was lower in the LDFRT group(P < 0.05), as confirmed by Western blotting.Conclusion LDFRT is as effective at inhibiting the growth of tumor cells as conventional-dose radiation. 4Identification of potential immune-related prognostic biomarkers of lung cancer using gene co-expression network analysis显示文摘Objective The objective of this study was to identify new carcinogenetic hub genes and develop the integration of differentially expressed genes to predict the prognosis of lung cancer.Methods GSE139032 microarray data packages were downloaded from the Gene Expression Omnibus for planning,testing,and review of data.We identified KRT6C,LAMC2,LAMB3,KRT6A,and MYEOV from a key module for validation.Results We found that the five genes were related to a poor prognosis,and the expression levels of these genes were associated with tumor stage.Furthermore,Kaplan-Meier plotter showed that the five hub genes had better prognostic values.The mean levels of methylation in lung adenocarcinoma(LUAD)were significantly lower than those in healthy lung tissues for the hub genes.However,gene set enrichment analysis(GSEA)for single hub genes showed that all of them were immune-related.Conclusion Our findings demonstrated that KRT6C,LAMC2,LAMB3,KRT6A,and MYEOV are all candidate diagnostic and prognostic biomarkers for LUAD.They may have clinical implications in LUAD patients not only for the improvement of risk stratification but also for therapeutic decisions and prognosis prediction.Aixia Chen Shengnan Zhao Fei Zhou Hongying Lv Donghai Liang Tao Jiang Rui Liu Lijin Zhu Jingyu Cao Shihai Liu Hongsheng Yu 2020Oncology and Translational Medicine2020,6,6:1
5Study on UV Shielding TiO_2 Thin Film Prepared by Sol-Gel Method显示文摘Titanium dioxide (TiO_2) thin films were prepared on microscopes slides by sol-gel and dip-coating processes from specially formulated sols.The results show that there exists anatase and rutile structure of TiO_2 when heat treatment temperature is 450℃,and at 800℃,TiO_2 particle size is of below 100 nm and rutile structure is presented.In the range of 360 nm~400 nm the transmittance of TiO_2 sol increases with the increasing of the concentration of Ti(OC_4H_9)_4 in ethanol solution. 11Study on the Milk Yield Pattern of Maiwa Yak显示文摘The change in lactation of Maiwa yak was studied based on the data of locating Maiwa yak in warm seasons during 2003- 2009. The results showed that the milk yield was increasing gradually during days 30- 70 after calving,reached the peak at days 70- 110,and decreased gradually to no milking whenafter. 12Physiological and Biochemical Indexes of Tibetan Sheep at Different Months of Age显示文摘To study the changes in physiological and biochemical indexes of Tibetan sheep from the plateau area at different months of age,we measured the physiological and biochemical indexes of Tibetan sheep reared in Hongyuan County of Sichuan Province according to their growth stages. The results showed that the respiration,body temperature and heart rate of Tibetan sheep at 3- 24 months of age were( 24. 60 ± 6. 33)-( 50. 30 ± 21. 18) times / min,( 39. 14 ± 0. 28) ℃-( 39. 64 ± 0. 17) ℃ and( 91. 00 ± 13. 27)-( 112. 00 ± 8. 69) times/min,respectively. The blood physiological indexes of Tibetan sheep including white blood cell count( WBC),hematocrit( HCT) and red blood cell distribution width coefficient of variation( RDW- CV) at 3,6,15 and 18 months of age were relatively lower than those at 9,12,21 and 24 months of age. The blood physiological indexes including red blood cell count( RBC),mean corpuscular capacity( MCV),mean corpuscular hemoglobin concentration( MCHC) and mean corpuscular hemoglobin( MCH) at 3,6,15 and 18 months of age were relatively higher than those at 9,12,21 and 24 months of age,while hemoglobin( HGB) and platelet count( PLT) did not vary with months of age. 13Hsa_circ_0002137 stabled by LIN28B promotes osteosarcoma cell growth through the hsa-miR-1246/BCL2 axis显示文摘Circular RNAs(circRNAs)are a novel class of non-coding RNA that have recently shown to have huge capabilities in the regulation of gene expression at the posttranscriptional level.Growing evidence has indicated that circRNAs could serve as competing endogenous RNAs(ceRNAs)to sponge microRNAs(miRNAs)and suppress functions of targeted miRNAs.Osteosarcoma(OS)is the most common malignant primary bone cancer.Hsa_circ_0002137 is upregulated in OS.However,the role of hsa_circ_0002137 in OS remains unclear.Using miRNA pull-down assay,we showed that cir_0002137 sponged hsa-miR-1246,and BCL2 apoptosis regulator(BCL2)mRNA was a potential target of hsa-miR-1246 in human osteosarcoma(HOS)cells.Further,we found that hsa_cir_0002137 could enhance the expression of BCL2 hsa-miR-1246 and promote HOS cell growth through sponging hsa-miR-1246.Moreover,RNA binding protein immunoprecip itation(RIP)assay revealed that lin-28 homolog B(LIN28B)protein associated with hsa_circ_0002137,and LIN28B could increase hsa_circ_0002137 stability and thus accelerate OS cell growth.Our work was the first to study the functions of hsa_circ_0002137,has-miR-1246 and LIN28B in OS,and these results may provide novel therapeutic targets for OS treatment.FEI ZHANG JIANZHONG LAI RONGHAN HE YI SHI KUN XU SHIHAI JIANG TANGZHAO LIANG WANYU ZHAO WEIDA REN LEI ZHU SONG JIN KUN WANG 2022BIOCELL2022,46,3:0
14Seasonal Changes of Physiological and Biochemical Indexes of Tibetan Sheep显示文摘To investigate the seasonal changes in physiological and biochemical indexes of Tibetan sheep in Hongyuan area,we measured the physiological and biochemical indexes of Tibetan sheep at four different seasons. The results showed that the body temperature,respiration and heart rate of Tibetan sheep at four seasons were 39. 18- 39. 60 ℃,27. 20- 49. 04 times / min,91. 87- 106. 25 times / min,respectively. The indexes of WBC,MCV,MCH and MCHC in autumn and winter were significantly higher than those in spring and summer( P < 0. 01),while the indexes of RBC,HCT and RDW-CV in spring and summer were significantly higher than those in autumn and winter( P < 0. 01). The PLT did not vary greatly in spring,summer or autumn,but were all extremely higher than that in winter( P< 0. 01). The HGB maintained stable in the four seasons. The indexes of TP,ALB and GLO in summer and autumn were extremely higher than those in winter and spring( P < 0. 01),while the indexes of ALP and PCHE in summer and autumn were extremely lower than those in winter and spring( P < 0. 01). The blood physiological indexes of WBC,HCT and RDW-CV at 3,6,15 and 18 months of age were lower than those at 9,12,21 and 24 months of age. On the contrary,the indexes of RBC,MCV,MCH and MCHC at 3,6,15 and 18 months of age were higher than those at 9,12,21 and 24 months of age,while HGB and PLT did not vary with months of age. For blood biochemical indexes,ALT,AST,TP,ALB,LDH and CHOL reached the maximums at 21 months of age,while ALP,PCHE and GLU reached the maximums at 3 months of age. Besides,the GLO and GA reached the maximums at 18 and 24 months of age,respectively. 16Low-dose fractionated radiation reverses cisplatin resistance in ovarian cancer cells via PI3K/AKT/GSK-3β signaling显示文摘Objective To investigate whether low-dose fractionated radiation(LDFRT) could enhance cisplatin sensitivity in drug-resistant human ovarian cancer cells SKOV3/DDP, and to further explore the underlying mechanism.Methods SKOV3/DDP ovarian cancer cells were divided into three groups as follows: control, LDFRT, and conventional-dose radiation groups. Cells from all three groups were treated with different concentrations of cisplatin(0, 1.25, 2.5, 5, 10, and 20 μg/m L) for 48 h. The proliferation inhibition rate was investigated using the cell counting kit 8(CCK8). The rate of apoptosis was determined by flow cytometry(FCM). Protein levels of AKT, P-AKT, GSK-3β, P-GSK-3β, P21, cyclin D1, and P27 were examined by Western blotting. Results As expected, LDFRT significantly reduced the half-maximal inhibitory concentration(IC50) of cisplatin and promoted apoptosis in SKOV3/DDP cells. 17Analysis of Physiological and Biochemical Indexes of White Suffolk Sheep,White-tibetan Sheep and Tibetan Sheep显示文摘In order to compare the physiological and biochemical indexes of different sheep populations in plateau area,we measured the physiological and biochemical indexes of White Suffolk sheep,White-tibetan sheep and Tibetan sheep in four seasons.The body temperature,respiration and heart rate of White Suffolk sheep,White-tibetan sheep and Tibetan sheep throughout the year were(38.96±0.23)-(40.80±0.74)℃,(27.20±7.73)-(74.56±10.91)times/min,and(83.10±2.42)-(106.25±7.07)times/min,respectively.The main blood physiological indexes of White-tibetan sheep were between those of Suffolk sheep and Tibetan sheep or close to that of Tibetan sheep,the indexes of WBC in summer,HGB and RDW-CV in autumn,RDW-CV in winter were close to that of White Suffolk sheep.The main blood biochemical indexes of White-tibetan sheep were between that of White Suffolk sheep and Tibetan sheep or close to that of Tibetan sheep,but the indexes of ALB in spring,AST in summer,ALP and CHOL in autumn,and ALP in winter were close to that of White Suffolk sheep.It revealed that the adaptability of White-tibetan sheep was prior to White Suffolk sheep,and tended to Tibetan sheep in the plateau environment.Zhou Mingliang Chen Minghua Wu Weisheng Xie Rongqing Jiang Shihai Yang Pinggui 2016Animal Husbandry and Feed Science2016,8,3:0
