| 4 | Effect of low dose fractionated radiation on reversing cisplatin resistance in ovarian carcinoma via VEGF and mTOR显示文摘Objective To investigate the mechanism of low-dose fractionated radiation on reversing cisplatin resistance in ovarian carcinoma via vascular endothelial growth factor(VEGF) and mammalian target of rapamycin(mTOR) in vivo.Methods Human cisplatin-resistant ovarian carcinoma cells(SKOV3/DDP) were injected into nude mice to establish ovarian cancer xenografts. The mice were randomly divided into three groups: a control group, a low-dose fractionated radiation(LDRFT) group, and a conventional-dose radiation group. Each group was exposed to 0 cGy, 50 cGy, and 200 cGy radiation, respectively, for 4 weeks, up to a total of 8.0 Gy. Mice in the LDFRT group were irradiated twice daily with 6 hour intermissions on day 1 and 2 of every week for a total of 4 weeks. Conventional-dose group mice were given a single 200 cGy radiation dose on the first day each week for a total of 4 weeks. Maximum horizontal and vertical diameters of the tumors were measured every other day and used to create a tumor growth curve. After 4 weeks of irradiation, we dissected the tumor tissue and calculated the tumor inhibition rate. RT-PCR detected the expression of VEGF and m TOR, and Western blots detected the expression of corresponding proteins.Results Both LDRFT and conventional-dose radiation inhibited the growth of tumor cells, and growth of tumors in the two radiation groups compared with growth in the control group were significantly different(P < 0.05). The rate of tumor inhibition in the LDFRT group(37.5603%) was lower than in the conventionaldose group(47.4446%), but there was no significant difference(P 0.05). Compared with the other two groups, the m RNA expression of VEGF was significantly lower in the LDFRT group(P < 0.05), but there was no obvious difference between the conventional-dose and control groups. There was no obvious difference in the m RNA expression of m TOR among the three groups, but the expression of the protein p-m TOR was lower in the LDFRT group(P < 0.05), as confirmed by Western blotting.Conclusion LDFRT is as effective at inhibiting the growth of tumor cells as conventional-dose radiation. | 5 | Identification of potential immune-related prognostic biomarkers of lung cancer using gene co-expression network analysis显示文摘Objective The objective of this study was to identify new carcinogenetic hub genes and develop the integration of differentially expressed genes to predict the prognosis of lung cancer.Methods GSE139032 microarray data packages were downloaded from the Gene Expression Omnibus for planning,testing,and review of data.We identified KRT6C,LAMC2,LAMB3,KRT6A,and MYEOV from a key module for validation.Results We found that the five genes were related to a poor prognosis,and the expression levels of these genes were associated with tumor stage.Furthermore,Kaplan-Meier plotter showed that the five hub genes had better prognostic values.The mean levels of methylation in lung adenocarcinoma(LUAD)were significantly lower than those in healthy lung tissues for the hub genes.However,gene set enrichment analysis(GSEA)for single hub genes showed that all of them were immune-related.Conclusion Our findings demonstrated that KRT6C,LAMC2,LAMB3,KRT6A,and MYEOV are all candidate diagnostic and prognostic biomarkers for LUAD.They may have clinical implications in LUAD patients not only for the improvement of risk stratification but also for therapeutic decisions and prognosis prediction. | Aixia Chen Shengnan Zhao Fei Zhou Hongying Lv Donghai Liang Tao Jiang Rui Liu Lijin Zhu Jingyu Cao Shihai Liu Hongsheng Yu | 2020 | Oncology and Translational Medicine2020,6,6: | 1 |
| 6 | Effect of processing parameters on the microstructure and mechanical properties of TiAl/Ti_(2)AlNb laminated composites显示文摘In order to improve the intrinsic brittleness of TiAl alloys,Ti_(2)AlNb alloys with outstanding ductility and toughness at room temperature,and good high-temperature performance are competitive candidates in constructing the TiAl-based laminated composites.In this work,TiAl/Ti_(2)AlNb laminated composites are successfully synthesized by vacuum hot pressing combined with the foil-foil(sheet)metallurgy.Under the pressure of 65 MPa,different holding time and temperature of hot pressing are tried and the optimized fabrication parameter is acquired as 1050℃/120 min/65 MPa.Along with the changes of processing parameters,the defect,microstructure,interface,phase transformation and the corresponding mechanical properties are detailly discussed.The results show that the TiAl/Ti_(2)AlNb laminated composite fabricated at 1050℃ for 2 h achieves a good metallurgical interface bonding.The corresponding interface microstructure is composed of region I and region II.The region I consists of O,α_(2)and B2/βphase,and region II is made up ofα2.Subsequently,the tensile tests indicate that the composite synthesized at 1050℃ for 2 h possesses a maximum strength of 812 MPa and a total elongation of 1.31%at room temperature,and a strength of 539.71 MPa and the highest total elongation of 10.34%at 750℃.The well synergistic deformation ability between the interface and the two base alloys endows the composite an excellent tensile performance.Moreover,the composite processed at 1050℃ for 2 h behaves the best fracture toughness in both arrester orientation and divider orientation with the value of 32.6 MPa.m^(1/2)and 30.1 MPa.m^(1/2),respectively.The Ti_(2)AlNb alloy in the laminated structure effectively release the stress around the crack tip and plays a role in toughening.Further,crack deflection,crack bridging,crack blunting and fragmentation also make contributions to enhance the fracture toughness of the laminated composites. | Donghai Li Binbin Wang Liangshun Luo Xuewen Li Yanjin Xu BinQiang Li Diween Hawezy Liang Wang Yanqing Su Jingjie Guo Hengzhi Fu | 2022 | Journal of Materials Science & Technology2022,,14: | 1 |

| 9 | Urban Air Pollution May Enhance COVID-19 Case-Fatality and Mortality Rates in the United States显示文摘BACKGROUND The novel human coronavirus disease 2019(COVID-19)pandemic has claimed more than 600,000 lives worldwide,causing tremendous public health,social,and economic damages.Although the risk factors of COVID-19 are still under investigation,environmental factors,such as urban air pollution,may play an important role in increasing population susceptibility to COVID-19 pathogenesis.METHODS We conducted a cross-sectional nationwide study using zero-inflated negative binomial models to estimate the association between longterm(2010–2016)county-level exposures to NO2,PM2.5,and O3 and county-level COVID-19 case-fatality and mortality rates in the United States.We used both single-and multi-pollutant models and controlled for spatial trends and a comprehensive set of potential confounders,including state-level test positive rate,county-level health care capacity,phase of epidemic,population mobility,population density,sociodemographics,socioeconomic status,race and ethnicity,behavioral risk factors,and meteorology.RESULTS From January 22,2020,to July 17,2020,3,659,828 COVID-19 cases and 138,552 deaths were reported in 3,076 US counties,with an overall observed case-fatality rate of 3.8%.County-level average NO2 concentrations were positively associated with both COVID-19 case-fatality rate and mortality rate in single-,bi-,and tri-pollutant models.When adjusted for co-pollutants,per interquartile-range(IQR)increase in NO2(4.6 ppb),COVID-19 case-fatality rate and mortality rate were associated with an increase of 11.3%(95%CI 4.9%–18.2%)and 16.2%(95%CI 8.7%–24.0%),respectively.We did not observe significant associations between COVID-19 case-fatality rate and long-term exposure to PM2.5 or O3,although per IQR increase in PM2.5(2.6 mg/m3)was marginally associated,with a 14.9%(95%CI 0.0%–31.9%)increase in COVID-19 mortality rate when adjusted for co-pollutants.DISCUSSION Long-term exposure to NO2,which largely arises from urban combustion sources such as traffic,may enhance susceptibility to severe COVID-19 outcomes,independent of long-termPM2.5 and O3 exposure.The results support targeted public health actions to protect residents from COVID-19 in heavily polluted regions with historically high NO2 levels.Continuation of current efforts to lower traffic emissions and ambient air pollution may be an important component of reducing population-level risk of COVID-19 case fatality and mortality. | Donghai Liang Liuhua Shi Jingxuan Zhao Pengfei Liu Jeremy A.Sarnat Song Gao Joel Schwartz Yang Liu Stefanie T.Ebelt Noah Scovronick Howard H.Chang | 2020 | The Innovation2020,1,3: | 1 |

| 13 | Benefit of adjuvant chemoradiotherapy in patients with pathologically lymph node-positive and locally advanced gastric cancer显示文摘Objective This study aimed to compare the effectiveness of adjuvant chemoradiotherapy(CRT)and adjuvant chemotherapy(ChT)for T3–4/N+gastric cancer(GC)following D2/R0 dissection,and identify the specific subgroups that could benefit from adjuvant CRT.Methods All eligible patients were divided into the CRT group and ChT group.We assessed the survival outcomes and patterns of recurrence for each group,and determined the prognostic factors for survival by performing Cox proportional risk regression analyses.Results A total of 192 gastric cancer patients were included in the study.The estimated 3-year and 5-year disease-free survival(DFS)probabilities in the CRT and ChT groups were 52.9%vs.36.7%(P=0.024)and 41.2%vs.31.1%(P=0.148),respectively,and the estimated 3-year and 5-year overall survival(OS)probabilities were 82.4%vs.70.0%(P=0.044)and 52.0%vs.35.6%(P=0.022).Patients in the CRT group had a lower risk of locoregional recurrence than those in the ChT group(20.6%vs.34.4%;P=0.031).The subset analyses revealed that patients with stage N1–2 disease were more likely to benefit from adjuvant CRT than from adjuvant ChT(DFS:53.1%vs.36.4%;P=0.039;OS:53.1%vs.38.6%;P=0.036).Conclusion For locally advanced gastric cancer patients with LN+,adjuvant CRT showed superior survival benefits compared with adjuvant ChT alone.Patients with N1–2 achieved better survival from adjuvant CRT. | Shanhui Zhang Fei Zhou Donghai Liang Hongying Lv Hongsheng Yu | 2020 | Oncology and Translational Medicine2020,6,2: | 0 |
| 14 | Low-dose fractionated radiation reverses cisplatin resistance in ovarian cancer cells via PI3K/AKT/GSK-3β signaling显示文摘Objective To investigate whether low-dose fractionated radiation(LDFRT) could enhance cisplatin sensitivity in drug-resistant human ovarian cancer cells SKOV3/DDP, and to further explore the underlying mechanism.Methods SKOV3/DDP ovarian cancer cells were divided into three groups as follows: control, LDFRT, and conventional-dose radiation groups. Cells from all three groups were treated with different concentrations of cisplatin(0, 1.25, 2.5, 5, 10, and 20 μg/m L) for 48 h. The proliferation inhibition rate was investigated using the cell counting kit 8(CCK8). The rate of apoptosis was determined by flow cytometry(FCM). Protein levels of AKT, P-AKT, GSK-3β, P-GSK-3β, P21, cyclin D1, and P27 were examined by Western blotting. Results As expected, LDFRT significantly reduced the half-maximal inhibitory concentration(IC50) of cisplatin and promoted apoptosis in SKOV3/DDP cells.
