1An artificial chromosome for data storage显示文摘DNA digital storage provides an alternative for information storage with high density and long-term stability.Here,we report the de novo design and synthesis of an artificial chromosome that encodes two pictures and a video clip.The encoding paradigm utilizing the superposition of sparsified error correction codewords and pseudo-random sequences tolerates base insertions/deletions and is well suited to error-prone nanopore sequencing for data retrieval.The entire 254 kb sequence was 95.27%occupied by encoded data.The Transformation-Associated Recombination method was used in the construction of this chromosome from DNA fragments and necessary autonomous replication sequences.The stability was demonstrated by transmitting the data-carrying chromosome to the 100 th generation.This study demonstrates a data storage method using encoded artificial chromosomes via in vivo assembly for write-once and stable replication for multiple retrievals,similar to a compact disc,with potential in economically massive data distribution.Weigang Chen Mingzhe Han Jianting Zhou Qi Ge Panpan Wang Xinchen Zhang Siyu Zhu Lifu Song Yingjin Yuan 2021National Science Review2021,8,5:3
2Loss of Lkb1 impairs Treg function and stability to aggravate graft-versus-host disease after bone marrow transplantation显示文摘Accumulating evidence suggests that a reduction in the number of Foxp3^(+) regulatory T cells(Tregs)contributes to the pathogenesis of acute graft-versus-host disease(aGVHD),which is a major adverse complication that can occur after allogeneic hematopoietic stem cell transplantation(allo-HSCT).However,the precise features and mechanism underlying the defects in Tregs remain largely unknown.In this study,we demonstrated that Tregs were more dramatically decreased in bone marrow compared with those in peripheral blood from aGVHD patients and that bone marrow Treg defects were negatively associated with hematopoietic reconstitution.Tregs from aGVHD patients exhibited multiple defects,including the instability of Foxp3 expression,especially in response to IL-12,impaired suppressor function,decreased migratory capacity,and increased apoptosis.Transcriptional profiling revealed the downregulation of Lkb1,a previously identified critical regulator of murine Treg identity and metabolism,and murine Lkb1-regulated genes in Tregs from aGVHD patients.Foxp3 expression in human Tregs could be decreased and increased by the knockdown and overexpression of the Lkb1 gene,respectively.Furthermore,a loss-of-function assay in an aGVHD murine model confirmed that Lkb1 deficiency could impair Tregs and aggravate disease severity.These findings reveal that Lkb1 downregulation contributes to multiple defects in Tregs in human aGVHD and highlight the Lkb1-related pathways that could serve as therapeutic targets that may potentially be manipulated to mitigate aGVHD.Xiuhua Su Qianqian Wang Wei Guo Xiaolei Pei Qing Niu Maolan Liu Yuanyuan Liu Song Chen Sizhou Feng Yi He Donglin Yang Rongli Zhang Qiaoling Ma Weihua Zhai Aiming Pang Jialin Wei Yong Huang Yuechen Luo Mingzhe Han Xiaoming Feng Erlie Jiang 2020Cellular & Molecular Immunology2020,17,5:3
3Pathological matrix stiffness promotes cardiac fibroblast differentiation through the POU2F1 signaling pathway显示文摘Cardiac fibroblast(CF)differentiation into myofibroblasts is a crucial cause of cardiac fibrosis,which increases in the extracellular matrix(ECM)stiffness.The increased stiffness further promotes CF differentiation and fibrosis.However,the molecular mechanism is still unclear.We used bioinformatics analysis to find new candidates that regulate the genes involved in stiffnessinduced CF differentiation,and found that there were binding sites for the POU-domain transcription factor,POU2F1(also known as Oct-1),in the promoters of 50 differentially expressed genes(DEGs)in CFs on the stiffer substrate.Immunofluorescent staining and Western blotting revealed that pathological stiffness upregulated POU2F1 expression and increased CF differentiation on polyacrylamide hydrogel substrates and in mouse myocardial infarction tissue.A chromatin immunoprecipitation assay showed that POU2F1 bound to the promoters of fibrosis repressors IL1R2,CD69,and TGIF2.The expression of these fibrosis repressors was inhibited on pathological substrate stiffness.Knockdown of POU2F1 upregulated these repressors and attenuated CF differentiation on pathological substrate stiffness(35 kPa).Whereas,overexpression of POU2F1 downregulated these repressors and enhanced CF differentiation.In conclusion,pathological stiffness upregulates the transcription factor POU2F1 to promote CF differentiation by inhibiting fibrosis repressors.Our work elucidated the crosstalk between CF differentiation and the ECM and provided a potential target for cardiac fibrosis treatment.Mingzhe Li Jimin Wu Guomin Hu Yao Song Jing Shen Junzhou Xin Zijian Li Wei Liu Erdan Dong Ming Xu Youyi Zhang Han Xiao 2021Science China(Life Sciences)2021,64,2:3

5Structure-preserving properties of Strmer-Verlet scheme for mathematical pendulum显示文摘The structure-preserving property, in both the time domain and the frequency domain, is an important index for evaluating validity of a numerical method. Even in the known structure-preserving methods such as the symplectic method, the inherent conservation law in the frequency domain is hardly conserved. By considering a mathematical pendulum model, a Strmer-Verlet scheme is first constructed in a Hamiltonian framework. The conservation law of the Strmer-Verlet scheme is derived, including the total energy expressed in the time domain and periodicity in the frequency domain. To track the structure-preserving properties of the Strmer-Verlet scheme associated with the conservation law, the motion of the mathematical pendulum is simulated with different time step lengths. The numerical results illustrate that the Strmer-Verlet scheme can preserve the total energy of the model but cannot preserve periodicity at all. A phase correction is performed for the Strmer-Verlet scheme. 6Mechanical performance analysis and stiffness test of a new type of suspension bridge显示文摘A new type of suspension bridge is proposed based on the gravity stiffness principle.Compared with a conventional suspension bridge,the proposed bridge adds rigid webs and cross braces.The rigid webs connect the main cable and main girder to form a truss that can improve the bending stiffness of the bridge.The cross braces connect the main cables to form a closed space truss structure that can improve the torsional stiffness of the bridge.The rigid webs and cross braces are installed after the construction of a conventional suspension bridge is completed to resist different loads with different structural forms.A new type of railway suspension bridge with a span of 340 m and a highway suspension bridge with a span of 1020 m were designed and analysed using the finite element method.The stress,deflection of the girders,unbalanced forces of the main towers,and natural frequencies were compared with those of conventional suspension bridges.A stiffness test was carried out on the new type of suspension bridge with a small span,and the results were compared with those for a conventional bridge.The results showed that the new suspension bridge had a better performance than the conventional suspension bridge.Xia QIN Mingzhe LIANG Xiaoli XIE Huilan SONG 2021Frontiers of Structural and Civil Engineering2021,15,5:1

11Structure-preserving Analysis on Folding and Unfolding Process of Undercarriage显示文摘The main idea of the structure-preserving method is to preserve the intrinsic geometric properties of the continuous system as much as possible in numerical algorithm design. The geometric constraint in the multi-body systems, one of the difficulties in the numerical methods that are proposed for the multi-body systems, can also be regarded as a geometric property of the multi-body systems. Based on this idea, the symplectic precise integration method is applied in this paper to analyze the kinematics problem of folding and unfolding process of nose undercarriage. The Lagrange governing equation is established for the folding and unfolding process of nose undercarriage with the generalized defined displacements firstly. And then, the constrained Hamiltonian canonical form is derived from the Lagrange governing equation based on the Hamiltonian variational principle. Finally, the symplectic precise integration scheme is used to simulate the kinematics process of nose undercarriage during folding and unfolding described by the constrained Hamiltonian canonical formulation.
