| 1 | Interleukin-33 drives hepatic fibrosis through activation of hepatic stellate cells显示文摘Liver fibrosis is a consequence of chronic liver disease,causing morbidity and mortality.Interleukin-33(IL-33)is a critical mediator of inflammation,which may be involved in the development of liver fibrosis.Here,we investigated the role of IL-33 in human patients and experimental bile-duct ligation(BDL)-induced fibrosis in mice.We report increased hepatic IL-33 expression in the murine BDL model of fibrosis and in surgical samples obtained from patients with liver fibrosis.Liver injury,inflammatory cell infiltration and fibrosis were reduced in the absence of the IL-33/ST2 receptor,and the activation of hepatic stellate cells(HSCs)was decreased in ST2-deficient mice.Recombinant IL-33 activated HSCs isolated from C57BL/6 mice,leading to the expression of IL-6,TGF-β,α-SMA and collagen,which was abrogated in the absence of ST2 or by pharmacological inhibition of MAPK signaling.Finally,administration of recombinant IL-33 significantly increased hepatic inflammation in sham-operated BL6 mice but did not enhance BDL-induced hepatic inflammation and fibrosis.In conclusion,BDL-induced liver inflammation and fibrosis are dependent on ST2 signaling in HSCs,and therefore,the IL-33/ST2 pathway may be a potential therapeutic target in human patients with chronic hepatitis and liver fibrosis. | Zhongming Tan Qianghui Liu Runqiu Jiang Long Lv Siamak S Shoto Isabelle Maillet Valerie Quesniaux Junwei Tang Wenjie Zhang Beicheng Sun Bernhard Ryffel | 2018 | Cellular & Molecular Immunology2018,15,4: | 24 |

| 3 | Targets and molecular mechanisms of triptolide in cancer therapy显示文摘Triptolide(TPL/TL) is a natural drug with novel anticancer effects. Preclinical studies indicated that TPL inhibits cell proliferation, induces cell apoptosis, inhibits tumor metastasis and enhances the effect of other therapeutic methods in various cancer cell lines. Multiple molecules and signaling pathways, such as caspases, heat-shock proteins, NF-κB, and deoxyribonucleic acid(DNA) repair-associated factors, are associated with the anti-cancer effect. TPL also improves chemoradiosensitivity in cancer therapy. Phase I trials indicate the potential clinical value of TPL use. | 4 | Therapeutic potential of intravenous Xuebijing on transforming growth factor β_1 and procollagen type III peptide in patients with acute paraquat poisoning显示文摘OBJECTIVE:Paraquat(PQ) poisoning-induced pulmonary fibrosis causes asphyxiation and death.The therapeutic potential of intravenous Xuebijing therapy in PQ poisoning patients and its underlying immunomodulatory effects on transforming growth factor(TGF)-β1 and procollagen type III peptide(PIIIP) were investigated.METHODS:Thirty-six acute PQ poisoning patients were randomly assigned to conventional therapy(Group A) and intravenous Xuebijing administration plus conventional therapy(Group B).Twenty volunteers served as controls(Group C).Blood samples were collected upon admission(day 0) and at post-treatment days 5,10,and 14.TGF-β1 and PIIIP concentrations were determined by ELISA and analyzed for intra-and inter-group differences over time.One-month follow-up was conducted for determining the mortality rate.RESULTS:TGF-β1 and PIIIP levels were significantly higher in PQ poisoning patients and increased over time(Groups A and B vs C,P<0.01).However,the TGF-β1 and PIIIP levels were consistently significantly lower in Group B compared with those of Group A(P<0.01).The 1-month mortality rate was also lower in Group B compared with that of Group A(P<0.05).PQ poisoning patients showed remarkably high levels of TGF-β1 and PIIIP,which increased as PQ-induced pulmonary fibrosis progressed.CONCLUSION:Treatment with intravenous Xuebijing plus conventional therapy significantly lowered TGF-β1 and PIIIP levels,which indicates therapeutic efficacy in the treatment of PQ poisoning patients. | Xuebing Zheng Xiaoli Sun Piyong Ma Zhongming Liu Nan Jiang | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 11 |

| 6 | Chemical characteristics of precipitation at Nanping Mangdang Mountain in eastern China during spring显示文摘To study the characteristics of precipitation in eastern China,an automatic sampler was used to collect rainwater samples from 19 precipitation events at Mangdang Mountain,Nanping City,Fujiang Province,in the spring of 2009.We used ion chromatography to analyze the ionic components and concentrations,and inductively coupled plasma mass spectrometry (ICP-MS) to analyze element compositions and contents.The results demonstrated remarkable acidic characteristics:in more than 80% of precipitation events the pH was less than 5.6,with an average of 4.81.Mass concentration results showed SO 4 2 was the main anionic component (36.2% of the total anion mass),while NH 4+ was the main cationic component (47.7% of the total cation mass) and main ion for acidity neutralization in the rainwater.Organic acid content accounted for 30.9% of total anion mass.The main trace metals were Ca,K,and Na.The SO 4 2/NO 3 ratio was 1.4,indicating that precipitation in this region was influenced by complex air pollution-the product of individual coal-burning combined with vehicle exhaust pollution.Correlation analysis of the chemical composition of the precipitation indicated that acidity in this region was determined by a combination of all acidic and neutralization ions rather than any single ion component.The results also showed that Na+ and Cl contributions were mainly by seawater;Mg 2+ by seawater and crustal materials;the NH 4+,K+,Ca 2+,NO 3 and SO 4 2 by anthropogenic sources;the trace metals were from the Earth's crust;and organic acids were potentially from combustion of biomass. | Yanli Cheng Ying Liu Mingqun Huo Qian Sun Huixiang Wang Zhongming Chen Yuhua Bai | 2011 | Journal of Environmental Sciences2011,23,8: | 8 |

| 8 | RP11-40C6.2 Inactivates Hippo Signaling by Attenuating YAP1 Ubiquitylation in Hepatitis B Virus-associated Hepatocellular Carcinoma显示文摘Background and Aims:Chronic hepatitis caused by hepatitis B virus(HBV)infection is a leading cause of hepatocellular carcinoma(HCC).We investigated the roles of oncogenic HBV infection-associated long noncoding RNAs in HCC.Methods:Bioinformatics analysis of data from the Cancer Genome Atlas(TCGA)was performed to screen potential oncogenic HBV-related lncRNAs.Next,we assessed their expression in clinical samples and investigated their correlation with clinical characteristics.The detailed oncogenic effects were analyzed by performing in vitro and in vivo studies.Results:RP11-40C6.2,an HBV infection-related lncRNA,was identified by analysis of the TCGA–Liver Hepatocellular Carcinoma database.Gene Set Enrichment Analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis of differentially expressed genes revealed a strong association of RP11-40C6.2 with the Hippo signaling pathway.RP11-40C6.2 was overexpressed in HCC patients with HBV infection compared to those without HBV infection.RP11-40C6.2 transcription showed a positive association with HBV-X protein(HBx),but not HBV core protein(HBc)expression,both of which are carcinogenic proteins.Luciferase gene reporter and ChIP assays revealed that YAP1/TAZ/TEADs complex enhanced RP11-40C6.2 transcription by binding to its promoter area.RP11-40C6.2 showed oncogenic characteristics in HCC cell lines and in animal models that were mediated via activation of YAP1.In vitro ubiquitylation assay revealed that RP11-40C6.2 can promote the stabilization of YAP1 by stopping phosphorylation at its s127 residue and further stopping its degradation through binding to 14-3-3.Conclusions:RP11-40C6.2 is an HBV infection-related lncRNA that exerts its oncogenic effects by targeting the Hippo signaling pathway. | Han Zhuo Chen Wu Junwei Tang Feihong Zhang Zhenggang Xu Dongwei Sun Yue Teng Zhongming Tan | 2023 | Journal of Clinical and Translational Hepatology2023,11,2: | 3 |

| 11 | Construction of miRNA-mRNA network reveals crucial miRNAs and genes in acute myocardial infarction显示文摘Acute myocardial infarction(AMI)is a severe cardiovascular disease.This study aimed to identify crucial microRNAs(miRNAs)and mRNAs in AMI by establishing a miRNA-mRNA network.The microarray datasets GSE31568,GSE148153,and GSE66360 were downloaded from the Gene Expression Omnibus(GEO)database.We identified differentially expressed miRNAs(DE-miRNAs)and mRNAs(DE-mRNAs)in AMI samples compared with normal control samples.The consistently changing miRNAs in both GSE31568 and GSE148153 datasets were selected as candidate DE-miRNAs.The interactions between the candidate DE-miRNAs and DE-mRNAs were analyzed,and a miRNA-mRNA network and a protein-protein interaction network were constructed,along with functional enrichment and pathway analyses.A total of 209 DE-miRNAs in the GSE31568 dataset,857 DE-miRNAs in the GSE148153 dataset,and 351 DE-mRNAs in the GSE66360 dataset were identified.Eighteen candidate DE-miRNAs were selected from both the GSE31568 and GSE148153 datasets.Furthermore,miR-646,miR-127-5p,miR-509-5p,miR-509-3-5p,and miR-767-5p were shown to have a higher degree in the miRNA-mRNA network.THBS-1 as well as FOS was a hub gene in the miRNA-mRNA network and the protein-protein interaction(PPI)network,respectively.CDKN1A was important in both miRNA-mRNA network and PPI network.We established a miRNA-mRNA network in AMI and identified five miRNAs and three genes,which might be used as biomarkers and potential therapeutic targets for patients with AMI. | Kai Wang Zhongming Li Wenjie Ma Yan Sun Xianling Liu Lijun Qian Jian Hong Dasheng Lu Jing Zhang Di Xu | 2021 | The Journal of Biomedical Research2021,35,6: | 2 |

| 18 | High precision nuclear mass predictions towards a hundred kilo-electron-volt accuracy显示文摘Mass is a fundamental property and an important fingerprint of atomic nucleus.It provides an extremely useful test ground for nuclear models and is crucial to understand energy generation in stars as well as the heavy elements synthesized in stellar explosions.Nuclear physicists have been attempting at developing a precise,reliable,and predictive nuclear model that is suitable for the whole nuclear chart,while this still remains a great challenge even in recent days.Here we employ the Fourier spectral analysis to examine the deviations of nuclear mass predictions to the experimental data and to present a novel way for accurate nuclear mass predictions.In this analysis,we map the mass deviations from the space of nucleon number to its conjugate space of frequency,and are able to pin down the main contributions to the model deficiencies.By using the radial basis function approach we can further isolate and quantify the sources.Taking a pedagogical mass model as an example,we examine explicitly the correlation between nuclear effective interactions and the distributions of mass deviations in the frequency domain.The method presented in this work,therefore,opens up a new way for improving the nuclear mass predictions towards a hundred kilo-electron-volt accuracy,which is argued to be the chaos-related limit for the nuclear mass predictions. | Zhongming Niu Haozhao Liang Baohua Sun Yifei Niu Jianyou Guo Jie Meng | 2018 | Science Bulletin2018,63,12: | 1 |
