| 2 | Exosomal piRNA profiling revealed unique circulating piRNA signatures of cholangiocarcinoma and gallbladder carcinoma显示文摘Cholangiocarcinoma(CCA)and gallbladder carcinoma(GBC)are biliary tract cancers with poor five-year survival and high recurrence rates.Both CCA and GBC patients suffer from lack of circulating diagnostic biomarkers at the early stage.Extracellular vesicles,especially exosomes,have been emerged as promising diagnostic sources for cancers due to easy and quick accessibility.Hence,identification of exosomal biomarkers provides a novel strategy for CCA and GBC diagnosis.Here,five CCA patients and four GBC patients were enrolled for exosomal small RNA sequencing.Our data showed that exosomal piwi-interacting RNA(piRNA)populations were altered in the plasma of CCA and GBC patients.In comparison to healthy individuals,694 and 323 piRNAs were upregulated in CCA and GBC,respectively,while 36 and 191 piRNAs were down regulated.Interestingly,sequencing results predicted that piR-2660989,piR-10506469,piR-20548188,piR-10822895,piR-hsa-23209,and piR-18044111 were upregulated in both CCA and GBC plasma.Importantly,we further included blood samples from 50 health individuals,40 CCA patients,and 25 GBC patients and found that piR-10506469 were significantly increased in the exosomes of plasma from both CCA and GBC patients.Moreover,we analyzed the expression levels of differentially expressed exosomal piRNAs in the plasma of CCA and GBC patient before and after surgeries and found that piR-10506469 and piR-20548188 were significantly decreased in patients underwent surgeries.Taken together,our data revealed that exosomal piRNAs those are differentially expressed in CCA and GBC plasma may serve as potential biomarkers for the diagnosis of CCA and GBC. | Xinjin Gu Chen Wang Hui Deng Chong Qing Rong Liu Sanhong Liu Xinying Xue | 2020 | Acta Biochimica et Biophysica Sinica2020,52,5: | 7 |
| 3 | The histone H3 lysine-27 demethylase Jmjd3 plays a critical role in specific regulation of Th17 cell differentiation显示文摘Interleukin(IL)17-producing T helper(Th17)cells play critical roles in the clearance of extracellular bacteria and fungi as well as the pathogenesis of various autoimmune diseases,such as multiple sclerosis,psoriasis,and ulcerative colitis.Although a global transcriptional regulatory network of Th17 cell differentiation has been mapped recently,the participation of epigenetic modifications in the differentiation process has yet to be elucidated.We demonstrated here that histone H3 lysine-27(H3K27)demethylation,predominantly mediated by the H3K27 demethylase Jmjd3,crucially regulated Th17 cell differentiation.Activation of naı¨ve CD41 T cells immediately induced high expression of Jmjd3.Genetic depletion of Jmjd3 in CD41 T cells specifically impaired Th17 cell differentiation both in vitro and in vivo.Ectopic expression of Jmjd3 largely rescued the impaired differentiation of Th17 cells in vitro in Jmjd3-deficientCD41 T cells.Importantly,Jmjd3-deficient mice were resistant to the induction of experimental autoimmune encephalomyelitis(EAE).Furthermore,inhibition of the H3K27 demethylase activity with the specific inhibitor GSK-J4 dramatically suppressed Th17 cell differentiation in vitro.At the molecular level,Jmjd3 directly bound to and reduced the level of H3K27 trimethylation(me3)at the genomic sites ofRorc,which encodes the masterTh17 transcription factorRorgt,and Th17 cytokine genes such as Il17,Il17f,and Il22.Therefore,our studies established acritical role of Jmjd3-mediatedH3K27demethylation inTh17 cell differentiation andsuggest that Jmjd3 can be a novel therapeutic target for suppressing autoimmune responses. | Zhi Liu Wei Cao Longxia Xu Xi Chen Yu Zhan Qian Yang Sanhong Liu Pengfei Chen Yuhang Jiang Xiaohua Sun Yu Tao Yiming Hu Cuifeng Li Qi Wang Ying Wang Charlie Degui Chen Yufang Shi Xiaoren Zhang | 2015 | Journal of Molecular Cell Biology2015,7,6: | 6 |
| 4 | Elevating H3K27me3 level sensitizes colorectal cancer to oxaliplatin显示文摘Histone methylation is a context-dependent modification that regulates gene expression,and the trimethylation of histone H3 lysine 27(H3K27me3)usually induces gene silencing.Overcoming colorectal cancer(CRC)chemoresistance is currently a huge challenge,but the relationship between H3K27me3 modification and chemoresistance remains largely unclear.Here,we found that H3K27me3 levels positively correlated with the metastasis-free survival of CRC patients and a low H3K27me3 level predicted a poor outcome upon chemotherapeutic drug treatment.Oxaliplatin stimulation significantly induced the expression of H3K27 lysine demethylase 6A/6B(KDM6A/6B),thus decreasing the level of H3K27me3 in CRC cells.Elevation of H3K27me3 level through KDM6A/6B depletion or GSK-J4(a KDM6A/6B inhibitor)treatment significantly enhanced oxaliplatin-induced apoptosis.Conversely,when inhibiting the expression of H3K27me3 by EPZ-6438,an inhibitor of the histone methyltransferase EZH2,the proportion of apoptotic cells remarkably decreased.In addition,the combination of GSK-J4 and oxaliplatin significantly inhibited tumor growth in an oxaliplatin-resistant patient-derived xenograft model.Importantly,we revealed that oxaliplatin treatment dramatically induced NOTCH2 expression,which was caused by downregulation of H3K27me3 level on the NOTCH2 transcription initiation site.Thus,the activated NOTCH signaling promoted the expression of stemness-related genes,which resulted in oxaliplatin resistance.Furthermore,oxaliplatin-induced NOTCH signaling could be interrupted by GSK-J4 treatment.Collectively,our findings suggest that elevating H3K27me3 level can improve drug sensitivity in CRC patients. | Qi Wang Xi Chen Yuhang Jiang Sanhong Liu Hanshao Liu Xiaohua Sun Haohao Zhang Zhi Liu Yu Tao Cuifeng Li Yiming Hu Dandan Liu Deji Ye Yongzhong Liu Mingliang Wang Xiaoren Zhang | 2020 | Journal of Molecular Cell Biology2020,12,2: | 6 |

| 13 | CitationAS: A Tool of Automatic Survey Generation Based on Citation Content显示文摘Purpose: This study aims to build an automatic survey generation tool, named CitationAS, based on citation content as represented by the set of citing sentences in the original articles.Design/methodology/approach: Firstly, we apply LDA to analyse topic distribution of citation content. Secondly, in CitationAS, we use bisecting K-means, Lingo and STC to cluster retrieved citation content. Then Word2Vec, Word Net and combination of them are applied to generate cluster labels. Next, we employ TF-IDF, MMR, as well as considering sentence location information, to extract important sentences, which are used to generate surveys. Finally, we adopt manual evaluation for the generated surveys.Findings: In experiments, we choose 20 high-frequency phrases as search terms. Results show that Lingo-Word2Vec, STC-Word Net and bisecting K-means-Word2Vec have better clustering effects. In 5 points evaluation system, survey quality scores obtained by designing methods are close to 3, indicating surveys are within acceptable limits. When considering sentence location information, survey quality will be improved. Combination of Lingo, Word2Vec, TF-IDF or MMR can acquire higher survey quality.Research limitations: The manual evaluation method may have a certain subjectivity. We use a simple linear function to combine Word2Vec and Word Net that may not bring out their strengths. The generated surveys may not contain some newly created knowledge of some articles which may concentrate on sentences with no citing.Practical implications: CitationAS tool can automatically generate a comprehensive, detailed and accurate survey according to user's search terms. It can also help researchers learn about research status in a certain field.Originality/value: Citaiton AS tool is of practicability. It merges cluster labels from semantic level to improve clustering results.
