| 1 | Entecavir vs lamivudine therapy for na?ve patients with spontaneous reactivation of hepatitis B presenting as acute-on-chronic liver failure显示文摘AIM:To investigate the short-term and long-term efficacy of entecavir versus lamivudine in patients with spontaneous reactivation of hepatitis B presenting as acute-on-chronic liver failure(ACLF).METHODS:This was a single center,prospective cohort study.Eligible,consecutive hospitalized patients received either entecavir 0.5 mg/d or lamivudine 100mg/d.All patients were given standard comprehensive internal medicine.The primary endpoint was survival rate at day 60,and secondary endpoints were reduction in hepatitis B virus(HBV)DNA and alanine aminotransferase(ALT)levels,and improvement in Child-Turcotte-Pugh(CTP)and model for end-stage liver disease(MELD)scores at day 60 and survival rate at week 52.RESULTS:One hundred and nineteen eligible subjects were recruited from 176 patients with severe acute exacerbation of chronic hepatitis B:65 were included in the entecavir group and 54 in the lamivudine group(full analysis set).No significant differences were found in patient baseline clinical parameters.At day 60,entecavir did not improve the probability of survival(P=0.066),despite resulting in faster virological suppression(P<0.001),higher rates of virological response(P<0.05)and greater reductions in the CTP and MELD scores(all P<0.05)than lamivudine.Intriguingly,at week 52,the probability of survival was higher in the entecavir group than in the lamivudine group[42/65(64.6%)vs 26/54(48.1%),respectively;P=0.038].The pretreatment MELD score(B,1.357;95%Cl:2.138-7.062;P=0.000)and virological response at day30(B,1.556;95%Cl:1.811-12.411;P=0.002),were found to be good predictors for 52-wk survival.CONCLUSION:Entecavir significantly reduced HBV DNA levels,decreased the CTP and MELD scores,and thereby improved the long-term survival rate in patients with spontaneous reactivation of hepatitis B presenting as ACLF. | Yang Zhang Xiao-Yu Hu Sen Zhong Fang Yang Tao-You Zhou Guo Chen Yan-Yan Wang Jian-Xing Luo | 2014 | World Journal of Gastroenterology2014,20,16: | 42 |
| 2 | The intracellular mechanism of alpha-fetoprotein promoting the proliferation of NIH 3T3 cells显示文摘AIM The existence and properties of alpha-fetoprotein (AFP) receptor on the surface of NIH 3T3 cells and the effects of AFP on cellular signal transduction pathway were investigated. METHODS The effect of AFP on the proliferation of NIH 3T3 cells was measured by incorporation of 3H-TdR. Receptor-binding assay of 125I-AFP was performed to detect the properties of AFP receptor in NIH 3T3 cells. The influences of AFP on the [cAMP]i and the activities of protein kinase A (PKA) were determined. Western blot was used to detect the change of K-ras P21 protein expression. RESULTS The proliferation of NIH 3T3 cells treated with 0-80 mg/L of AFP was significantly enhanced. The Scatchard analysis indicated that there were two classes of binding sites with KD of 2.722×10-9M (Bmax=12810 sites per cell) and 8.931× 10-SM (Bmax=l19700 sites per cell) respectively. In the presence of AFP (20 mg/L), the content of cAMP and activities of PKA were significantly elevated . The level of K-ras P21 protein was upregulated by AFP at the concentration of 20 mg/L. The monoclonal antibody against AFP could reverse the effects of AFP on the cAMP content, PKA activity and the expression of K-ras p21 gene. | 3 | Effect of Sancaijiangtang on plasma nitric oxide and endothelin-1 levels in patients with type 2 diabetes mellitus and vascular dementia:a single-blind randomized controlled trial显示文摘OBJECTIVE: To observe the effect of Sancaijiangtang powders on plasma nitric oxide and endothelin-1 levels. We sought to identify the common pathological link and mechanism of action for Traditional Chinese medicine in type 2 diabetes mellitus and vascular dementia,and to explicate the material basis for treating the different diseases with the same method in Traditional Chinese Medicine.METHODS: In total,168 patients with type 2 diabetes mellitus and vascular dementia were enrolled in the study,and randomly divided into two groups by simple randomization. Patients in the treatment group received oral Sancaijiangtang powders with pioglitazone hydrochloride three times daily,while patients in the control group received pioglitazone hydrochloride alone. The treatment course was for12 weeks. Mini-mental state examinations(Chinese version) and Montreal Cognitive Assessments(Beijing version) were performed,and fasting plasma glucose,fasting insulin,hemoglobin A1 c,homeostasis model assessment of insulin resistance,plasma nitric oxide and endothelin-1 levels were measured before and after the treatment.RESULTS: The post-treatment levels for all measurements in both groups were better than pre-treatment levels(P < 0.05). The post-treatment levels for all measurements in the treatment group were better than the levels measured in the control group(P < 0.05).CONCLUSION: Type 2 diabetes mellitus and vascular dementia have common pathological mechanisms for insulin resistance and endothelium dysfunction. Sancaijiangtang powders could improve the release of nitric oxide and inhibit the secretion of endothelin-1. | 5 | CRISPR/Cas9-mediated PINK1 deletion leads to neurodegeneration in rhesus monkeys显示文摘Dear Editor,PINK1 mutations cause autosomal recessive and early-onset Parkinson’s disease(PD)with selective neurodegeneration.Unfortunately,current PINK1 knockout(KO)mouse1,2 and pig models3,4 are unable to recapitulate the selective and overt neurodegeneration seen in PD patients.Furthermore,endogenous Pink1 in the mouse brain is expressed at very low levels and can only be detected via immunoprecipitation,5 meaning that PINK1’s function in the mammalian brain needs to be assessed using larger animals that are closer to humans.We previously used CRISPR/Cas9 to target the monkey gene in one-cell stage embryos.6 Using the same approach,we designed two gRNAs to target exon 2(T1)and exon 4(T2),which encode a kinase domain in the PINK1 gene of rhesus monkeys(Fig.1a).CRISPR/Cas9 and PINK1 gRNAs were injected into one-cell stage rhesus monkey embryos.A T7E1 assay and sequencing of PCR products from the injected embryos showed high efficiency(61.5%)in targeting PINK1(Fig.1b and Supplementary information,Fig.S1).Transfer of 87 embryos to 28 surrogate rhesus monkeys resulted in 11 pregnancies(39.2%)(Fig.1b).Eleven fetuses developed to term and were born naturally.Of these live monkeys,eight carried PINK1 mutations(M),and three were wild type(WT).However,three mutant monkeys(M1,M3 and M4)were newborn triplets that struggled to survive and died 3-4 days after birth.One WT newborn monkey also died after a difficult labor.Another mutant monkey(M2)died 7 days after birth without noticeable warning signs or symptoms.The other three mutant monkeys(M6,M7 and M8)have lived for three years;M5,however,reduced its food intake and showed weakness at the age of 1.5 years,and died 30 days after anesthesia for MRI examination. | Weili Yang Yunbo Liu Zhuchi Tu Chong Xiao Sen Yan Xishan Ma Xiangyu Guo Xiusheng Chen Peng Yin Zhengyi Yang Su Yang Tianzi Jiang Shihua Li Chuan Qin Xiao-Jiang Li | 2019 | Cell Research2019,29,4: | 12 |
| 6 | Synthesis and thermal properties of poly(styrene-co-acrylonitrile)-graft-polyethylene glycol copolymers as novel solid–solid phase change materials for thermal energy storage显示文摘A novel poly(styrene-co-acrylonitrile)-graft-polyethylene glycol(SAN-g-PEG) copolymer was synthesized as new solid–solid phase change materials(SSPCMs) by grafting PEG to the main chain of poly(styrene-co-acrylonitrile). The chemical structure of the SAN-g-PEG was confirmed by the Fourier transform infrared(FT-IR) and proton nuclear magnetic resonance(1H NMR) spectroscopy techniques. The thermal energy storage properties and the storage durability of the SAN-g-PEG were investigated by differential scanning calorimetry(DSC). The SAN-g-PEG was endowed with the solid–solid phase transition temperatures within the range of 23–36 8C and the latent heat enthalpy ranged from 66.8 k J/kg to 68.3 k J/kg. Thermal cycling tests revealed that the SAN-g-PEG kept great heat storage durability after 1000 thermal cycles. | 8 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 13 | Distribution of Traditional Chinese Medicine patterns in 324 cases with hepatitis B-related acute-on-chronic liver failure:a prospective,cross-sectional survey显示文摘OBJECTIVE:To determine the distribution of Traditional Chinese Medicine(TCM) patterns in hepatitis B-related acute-on-chronic liver failure(HB-ACLF) in different stages for guiding clinical prescriptions and treatments.METHODS:A prospective,cross-sectional survey method was used in this study.A total of 324 cases with HB-ACLF in China were involved.RESULTS:The general frequency of TCM patterns in HB-ACLF were as follows:Heat Toxin Stagnation Pattern(134/324,41.36%),Damp-heat Obstruction Pattern(66/324,20.37%),Yang Qi Deficiency Pattern(52/324,16.05%),and Liver and Kidney Yin Deficiency Pattern(26/324,8.02%).In the early stage of HB-ACLF,there was a remarkably higher percentage of excessive patterns than those in the middle and late stage.The incidence of Heat Toxin Stagnation reached 58.57%(82/140) in the early stage,while it was 33.96%(36/106) in the middle stage and 20.51%(16/78) in the late stage.In the early stage of HB-ACLF,excessive patterns,such as the Heat Toxin Stagnation Pattern,were more prevalent than those in the middle and late stages(P<α ' = 0.003).However,in the late stage of HB-ACLF,deficient patterns,such as the Yang Qi Deficiency Pattern,were more prevalent than those in the early and middle stages.The Yang Qi Deficiency Pattern had a higher rate of 41.03%(32/78) in the late stage compared with that of 20.75%(22/106) in the middle stage and 8.57%(12/140,P<α ' =0.003) in the early stage.The distribution of the other patterns was not significant between the three stages(P>0.003).CONCLUSIONS:There are four major patterns of HB-ACLF,including the Heat Toxin Stagnation Pattern,the Damp-heat Obstruction Pattern,the Yang Qi Deficiency Pattern,and the Liver and Kidney Yin Deficiency Pattern.The Heat Toxin Stagnation and Yang Qi Deficiency Patterns are the representative patterns in the early and late stages of HB-ACLF.In the middle stage of HB-ACLF,the TCM patterns vary in a complicated manner,with no significant difference among the patterns.Treatment for HB-ACLF should vary with the different representative patterns in the early and late stages. | Xiaoyu Hu Yang Zhang Guo Chen Yibei Li Sen Zhong | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 6 |

| 16 | Co3O4 modified Ag/g-C3N4 composite as a bifunctional cathode for lithium-oxygen battery显示文摘Rechargeable lithium-oxygen(Li-O2)batteries have appeal to enormous attention because they demonstrate higher energy density than the state-of-the-art Li-ion batteries.Whereas,their practical application is impeded by several challenging problems,such as the low energy round trip efficiencies and the insufficient cycle life,due to the cathode passivation caused by the accumulation of discharge products.Developing efficient catalyst for oxygen reduction and evolution reactions is effective to reduce the overpotentials in Li-O2cells.In our work,we report a Co3O4modified Ag/g-C3N4nanocomposite as a bifunctional cathode catalyst for Li-O2cells.The g-C3N4substrate prevents the accumulation of Ag and Co3O4nanoparticles and the presence of Ag NPs improves the surface area of g-C3N4and electronic conductivity,significantly improving the oxygen reduction/evolution capabilities of Co3O4.Due to a synergetic effect,the Ag/g-C3N4/Co3O4nanocomposite demonstrates a higher catalytic activity than each individual constituent of Co3O4or Ag/g-C3N4for the ORR/OER on as catalysts in Li-O2cells.As a result,the Ag/gC3N4/Co3O4composite shows impressive electrochemical performance in a Li-O2battery,including high discharge capacity,small gap between charge and discharge potential,and high cycling stability. | Qi Guo Chenwei Zhang Chaofeng Zhang Sen Xin Pengchao Zhang Qiufan Shi Dawei Zhang Ya You | 2020 | Journal of Energy Chemistry2020,29,2: | 4 |
| 17 | Clinical significance of nerve growth factor and tropomyosin-receptor-kinase signaling pathway in intrahepatic cholangiocarcinoma显示文摘AIM:To investigate the correlation between nerve growth factor-tropomyosin-receptor-kinase(NGF-TrkA)signaling pathway and prognosis in intrahepatic cholangiocarcinoma(IHCC).METHODS:NGF and TrkA expression in 83 samples of IHCC was assessed by immunohistochemistry.Correlations between NGF-TrkA expression and clinicopathological features were analyzed byχ2 test.Moreover,we evaluated the association between NGF-TrkA and overall survival by univariate and multivariate analysis.With experiments in vitro,we investigated the crucial role of NGF-TrkA on proliferation and invasion of IHCC cells with recombinant NGF-βstimulation.RESULTS:We found that NGF and TrkA expression was significantly related with differentiation(P=0.024)and intraneural invasion(P=0.003),respectively.Additionally,double higher expression of NGF and TrkA was identified as an independent prognostic factor in IHCC(P=0.003).Moreover,we demonstrated that NGF-TrkA signaling pathway can promote IHCC proliferation and invasion.CONCLUSION:NGF-TrkA double higher expression is an independent prognostic factor in IHCC.NGF-TrkA pathway can promote IHCC progression,indicating that NGF-TrkA may become a potential drug target. | Xiao-Qing Yang Yun-Fei Xu Sen Guo Yi Liu Shang-Lei Ning Xiao-Fei Lu Hui Yang Yu-Xin Chen | 2014 | World Journal of Gastroenterology2014,20,14: | 4 |

| 20 | Effects of minor Nd and Er additions on the precipitation evolution and dynamic recrystallization behavior of Mg-6.0Zn-0.5Mn alloy显示文摘A new wrought magnesium(Mg)alloy based on Mg-6.0Zn-0.5Mn(ZM60)is developed,which performs excellent combination of high tensile yield strength and good ductility.We investigate the effects of micro-alloying on dynamic precipitation,dynamic recrystallization(DRX)and mechanical properties of ZM60 alloy.The co-addition of minor(0.6 wt%)neodymium(Nd)and(0.3 wt%)erbium(Er)can accelerate the twinning and DRX process of ZM60 alloy at the initial compression deformation stage.The dynamic precipitation process is also accelerated due to Nd and Er co-alloying.Dislocation accumulation disappears and a higher density of rodβ_(1) precipitates and some thick β_(2) precipitates in matrix and fine twins,which inhibits the growth of DRX grains in compressed ZM60-0.6Nd-0.3Er alloy.The as-extruded ZM60-0.6Nd-0.3Er alloy has a yield strength(YS)of 245.8 MPa,ultimate tensile strength(UTS)of 347.2 MPa and elongation(EL)of 16.3%.The yield strength and tensile strength are improved via minor Nd and Er additions due to fine complete DRX grains,second phase particles and high density of precipitates.The grain refinement,weakened reserved working hardening and weakened basal fiber texture improve the elongation of ZM60-0.6Nd-0.3Er alloy. | Bin-Jiang Lv Sen Wang Tie-Wei Xu Feng Guo | 2021 | Journal of Magnesium and Alloys2021,9,3: | 4 |
