| 1 | Bevacizumab as a second- or later-line of treatment for metastatic colorectal cancer显示文摘AIM: To determine the efficacy of bevacizumab in patients with metastatic colorectal cancer (MCRC) who have failed prior chemotherapy without bevacizumab. METHODS: Between March 2002 and June 2010, 40 patients in South Korea with MCRC who were treated with bevacizumab plus chemotherapy as a second or later-line treatment were analyzed retrospectively for their overall response rate (ORR), overall survival (OS), and progression-free survival (PFS). The tumor responses were assessed using the RECIST (Response Evaluation Criteria in Solid Tumors) guidelines. RESULTS: All of the patients had progressed under prior chemotherapy without bevacizumab. Three patients (7.5%) exhibited an ORR, twenty one patients (52.5%) exhibited stable disease (SD), and fifteen patients (37.5%) exhibited disease progression. The median duration of the OS and PFS were 14.0 and 6.13 mo respectively. The median OSs were 16.60, 14.07 and 13.00 mo for second-line, third-line and fourth- or later-line treatments, respectively. The median PFSs were 7.23, 7.30 and 3.87 mo for the second-line, third-line and fourth- or later-line treatments, respectively. | 2 | Phase Ⅱ study of R-CVP followed by rituximab maintenance therapy for patients with advanced marginal zone lymphoma:consortium for improving survival of lymphoma(CISL)study显示文摘Background:The response rate and survival improvement for rituximab,a CD20-targeting monoclonal antibody,have been demonstrated in marginal zone lymphoma(MZL)as monotherapy and in combination with chemotherapeutic regimens,yet relapses still occur despite treatment completion.Thus,extending the period of remission in MZL patients remains an essential goal.This multicenter,single-arm,open-label phase II study evaluated the survival efficacy of 2 years of rituximab-maintenance therapy in patients with stage III-IV CD20-positive MZL who had responded to first-line R-CVP(rituximab,cyclophosphamide,vincristine,and prednisolone).The objective of this study was to determine whether rituximab maintenance following R-CVP warrants further investigation.Methods:Prior to rituximab-maintenance therapy,patients received 6-8 cycles of first-line R-CVP therapy for stage III-IV MZL.Rituximab(375 mg/m^(2)),cyclophosphamide(750 mg/m^(2)),and vincristine(1.4 mg/m^(2);maximum 2 mg)were administered via an intravenous infusion on day 1 of each 3-week cycle,while oral prednisolone(100 mg)was given on days 1-5 of each 3-week cycle.The patients who achieved complete response(CR),partial response(PR),or stable disease(SD)to R-CVP treatment,were prescribed rituximab-maintenance therapy which was administered intravenously at a dose of 375 mg/m^(2) every 8 weeks for up to 12 cycles.The primary endpoint was progression-free survival(PFS).Secondary endpoints were overall survival(OS)and treatment safety.Results:47 patients were enrolled,of whom,45(96%)received rituximab-maintenance treatment.Fifteen(33%)patients had nodal MZL.Following R-CVP first-line therapy,20(44%),22(49%),and 3(7%)patients achieved CR,PR,and SD,respectively.After a median follow-up of 38.2 months,their observed 3-year PFS rate was 81%.During the rituximab-maintenance,6 PR and 1 SD patients achieved CR following the administration of R-CVP.Elevated LDH and the presence of B symptoms were found to be significant prognostic factors for PFS(P=0.003)and demonstrated a 3-year OS rate of 90%.Rituximab-maintenance therapy was well tolerated,and the common treatment-emergent adverse events were sensory neuropathy(18%),myalgia(13%),fatigue(9%),and neutropenia(9%).Conclusion: Rituximab-maintenance therapy following first-line R-CVP demonstrated good PFS in patients with stage III-IV MZL, in addition to a favorable toxicity profile. | Sung Yong Oh Won Seog Kim Jin Seok Kim Seok Jin Kim Dok Hyun Yoon Deok-Hwan Yang Won Sik Lee Hyo Jung Kim Ho-Young Yhim Seong Hyun Jeong Jong Ho Won Suee Lee Jee Hyun Kong Sung-Nam Lim Jun Ho Ji Kyung A.Kwon Gyeong-Won Lee Jae Hoon Lee Ho Sup Lee Ho-Jin Shin Cheolwon Suh | 2019 | Cancer Communications2019，39，1： | 2 |

| 10 | Suppressed estrogen supply via extra-ovarian progesterone receptor membrane component 1 in menopause显示文摘In post-menopausal women,intra-mammary estrogen,which is converted from extra-ovarian estrone(E1),promotes the growth of breast cancer.Since the aromatase inhibitor letrozole does not suppress 17β-estradiol(E2)production from E1,high intra-mammary E1 concentrations impair letrozole's therapeutic efficacy.Progesterone receptor membrane component 1(Pgrmc1)is a non-classical progesterone receptor associated with breast cancer progression.In the present study,we introduced a Pgrmc1 heterozygous knockout(hetero KO)murine model exhibiting low Pgrmc1 expression,and observed estrogen levels and steroidogenic gene expression.Naïve Pgrmc1 hetero KO mice exhibited low estrogen(E2 and E1)levels and low progesterone receptor(PR)expression,compared to wild-type mice.In contrast,Pgrmc1 hetero KO mice that have been ovariectomized(OVX),including letrozole-treated OVX mice(OVX-letrozole),exhibited high estrogen levels and PR expression.Increased extra-ovarian estrogen production in Pgrmc1 hetero KO mice was observed with the induction of steroid sulfatase(STS).In MCF-7 cell,letrozole suppressed PR expression,but PGRMC1 knockdown increased PR and STS expression.Our presented results highlight the important role of Pgrmc1 in modulating estrogen production when ovary-derived estrogen is limited,thereby suggesting a potential therapeutic approach for letrozole resistance. | Sang RLee Hyun Yang Seong Lae Jo Young Ho Lee Hye Won Lee Bae-keun Park Eui-Ju Hong | 2021 | The Journal of Biomedical Research2021，35，3： | 1 |

| 15 | Effects of acupuncture stimulation on brain activation induced by cue-elicited alcohol craving显示文摘Acupuncture has been shown to be effective on alcohol use disorder.However,the underlying mechanism remains poorly understood.To investigate the effects of Shenmen(HT7)acupoint on brain activation induced by cue-elicited alcohol craving,30 right-handed healthy light to moderate alcohol drinkers were recruited from the community.They were randomly assigned to undergo acupuncture either at HT7(experimental acupoint,n=15)or Jingqu(LU8,control acupoint,n=15)acupoints.This randomized controlled study was performed in Daegu Haany University and Daegu-Gyeongbuk Medical Innovation Foundation,Republic of Korea.Recruitment and data collection were conducted from December 2018 to May 2019.The results showed that after acupuncture at HT7 acupoint,the activation of orbitofrontal cortex and dorsolateral prefrontal cortex was greatly increased,while the activation of dorsolateral prefrontal cortex was obviously reduced,and subject's craving for alcohol was reduced when he/she seeing alcohol-related video clips involving various alcohols(beer,wine,or soju)or drinking scenarios.Acupuncture at HT7 more greatly reduced subject's alcohol cravings than acupuncture at LU8 acupoint.These findings suggest that acupuncture can improve the self-control of mild to moderate social drinkers through the activation of the orbitofrontal cortex and dorsolateral prefrontal cortex,thereby reducing the craving for alcohol.The study protocol was approved by the Institutional Review Board of Daegu Haany University Korean Medicine Hospital,Republic of Korea(approval No.DHUMC-D-18026-PRO-02)on November 30,2018. | Hyeok Gyu Kwon Seong Hun Choi Joon Ho Seo Chae Ha Yang Mi Young Lee | 2022 | Neural Regeneration Research2022，17，5： | 1 |
