| 1 | Design and baseline data of a populationbased metabonomics study of eye diseases in eastern China: the Yueqing Ocular Diseases Investigation显示文摘Background:China is undergoing a massive transition toward an urban and industrial economy.These changes will restructure the demographics and economy which will eventually influence the future patterns of disease.The risk factors of vision-impairing eye diseases remain ambiguous and poorly understood.Metabolomics is an ideal tool to understand and shed light on the ocular disease mechanisms for earlier treatment.This article aims to describe the design,methodology and baseline data of the Yueqing Ocular Diseases Investigation(YODI),a developed county population-based study to determine the prevalence and primary causes of visual impairment;also with metabonomics analysis we aimed to identify,predict and suggest some preventive biomarkers that cause blindness.Methods:A population-based,cross-sectional study.Randomized clustering sampling was used to identify adults aged 50 years and older in Xiangyang Town,Yueqing county-level City.The interviews covered demographic,behavioral,ocular risk factors and mental health state.The ocular examination included visual acuity,autorefraction,intraocular pressure,anterior and posterior segment examinations,fundus photography,retinal tomography and angiography,and visual field testing.Anthropometric measurements included height and weight,waist and hip circumference,blood pressure,pulse rate,electrocardiogram,and abdominal ultrasound scan.A venous blood sample was collected for laboratory tests and metabonomics studies.Results:Of the 5319 individuals recruited for the YODI,4769(89.7%)subjects were enrolled for analyses.The median age was 62.0 years,and 45.6%were male.The educational level of illiteracy or semi-illiteracy,primary,middle and high school or above was 29.8%,45.5%,20.1%,and 3.3%,respectively.Majority of the participants were female,younger,and less educated when compared with nonparticipants.The average body mass index and waist-hip ratios were 24.4±3.4 kg/m^(2) and 0.9±0.1 respectively.Blood sample collection reached a sample size of 1909(479 from subjects with selfreported diabetes and 1430 from one-third of the 4290 subjects without self-reported diabetes).Conclusions:The YODI provides population-based data with a high response rate(89.7%)on the prevalence and primary causes of major vision-impairing eye diseases in developed county areas in eastern China.Metabonomics analysis from YODI will provide further association of metabolic characteristics with the visual impairment eye diseases.The risk prediction model could be created and has the potential to be generalized to developed eastern areas in China for prevention. | Yuxuan Deng Yuanbo Liang Sigeng Lin Liang Wen Jin Li Yue Zhou Meixiao Shen Jingwei Zheng Kemi Feng Yanting Sun Kwapong Willaim Robert Jia Qu Fan Lu | 2020 | Eye and Vision2020,7,1: | 4 |
| 2 | Reduced macular inner retinal thickness and microvascular density in the early stage of patients with dysthyroid optic neuropathy显示文摘Background:The goal was to investigate changes of the inner intra-retinal layer thicknesses and retinal capillary density(RCD)around the macula in thyroid-associated ophthalmopathy(TAO)patients with or without dysthyroid optic neuropathy(DON).Methods:Forty-four TAO patients including 23 non-DON and 21 DON patients,and 38 healthy participants were enrolled.Spectral domain optical coherence tomography equipped with Angiovue was used to obtain threedimensional retinal thickness maps and microvascular images of the superficial and deep retinal capillary layers(SRCL and DRCL,respectively)around the macula.Quantitative analyses were performed using a custom automated algorithm.Results:The thicknesses of the nerve fiber layer,ganglion cell layer+inner plexiform layer,and ganglion cell complex(GCC)as well as the RCDs in the SRCL and DRCL in both TAO groups were significantly decreased compared to the controls.In addition,the RCDs in DRCL of the DON group were further decreased compared to the non-DON group.GCC thickness in both TAO groups was positively correlated with the RCDs of the SRCL in the total annular zone and in the temporal,inferior,and nasal sectors.The areas under the receiver operating characteristic curves for the GCC thickness combined with the RCD were generally larger than those of each single indicator.Conclusions:Thinned inner intra-retinal layers and decreased RCDs in the TAO patients without DON revealed that morphological changes might precede visual dysfunction.The composite index of the retinal structure and the microvascular density might be valuable in the diagnosing,monitoring,and intervention for early DON. | Yufei Wu Yunhai Tu Chaoming Wu Lulu Bao Jianhua Wang Fan Lu Meixiao Shen Qi Chen | 2020 | Eye and Vision2020,7,1: | 3 |
| 3 | Machine learning helps improve diagnostic ability of subclinical keratoconus using Scheimpflug and OCT imaging modalities显示文摘Purpose:To develop an automated classification system using a machine learning classifier to distinguish clinically unaffected eyes in patients with keratoconus from a normal control population based on a combination of Scheimpflug camera images and ultra-high-resolution optical coherence tomography(UHR-OCT)imaging data.Methods:A total of 121 eyes from 121 participants were classified by 2 cornea experts into 3 groups:normal(50 eyes),with keratoconus(38 eyes)or with subclinical keratoconus(33 eyes).All eyes were imaged with a Scheimpflug camera and UHR-OCT.Corneal morphological features were extracted from the imaging data.A neural network was used to train a model based on these features to distinguish the eyes with subclinical keratoconus from normal eyes.Fisher’s score was used to rank the differentiable power of each feature.The receiver operating characteristic(ROC)curves were calculated to obtain the area under the ROC curves(AUCs).Results:The developed classification model used to combine all features from the Scheimpflug camera and UHR-OCT dramatically improved the differentiable power to discriminate between normal eyes and eyes with subclinical keratoconus(AUC=0.93).The variation in the thickness profile within each individual in the corneal epithelium extracted from UHR-OCT imaging ranked the highest in differentiating eyes with subclinical keratoconus from normal eyes.Conclusion:The automated classification system using machine learning based on the combination of Scheimpflug camera data and UHR-OCT imaging data showed excellent performance in discriminating eyes with subclinical keratoconus from normal eyes.The epithelial features extracted from the OCT images were the most valuable in the discrimination process.This classification system has the potential to improve the differentiable power of subclinical keratoconus and the efficiency of keratoconus screening. | Ce Shi Mengyi Wang Tiantian Zhu Ying Zhang Yufeng Ye Jun Jiang Sisi Chen Fan Lu Meixiao Shen | 2020 | Eye and Vision2020,7,1: | 3 |
| 4 | Choroidal thickness and choriocapillaris vascular density in myopic anisometropia显示文摘Background:This study aims to examine interocular differences in the choroidal thickness and vascular density of the choriocapillaris in anisometropic myopes and to further explore the relationship between choroidal blood flow and myopia.Methods:The sample comprised 44 participants with anisometropic myopia,aged 9 to 18 years,with normal bestcorrected visual acuity.All participants underwent a series of examinations,including spherical equivalent refraction(SER)and axial length(AL),measured by a Lenstar optical biometer and optical coherence tomography angiography(OCTA)scanner.OCT measured the choroidal thickness,vascular density,and flow voids of the choriocapillaris,and a customized algorithm was implemented in MATLAB R2017a with the post-correction of AL.The choroidal thickness was measured at the fovea and 0.5,1.0,1.5,2.0,2.5,and 3.0 mm nasally,temporally,inferiorly,and superiorly to the fovea.The vascular density and the flow voids of the choriocapillaris were measured at a 0.6-mm-diameter central circle,and the 0.6-2.5 mm diameter circle in the nasal,temporal,inferior,and superior regions.Repeated-measured ANOVAs were used to analyze the interocular differences.Partial correlations with the K value and age adjustments were used to study the relationships between the choroidal thickness,the choriocapillaris vascular density and flow voids,the SER and AL.Results:The choroidal thickness of the more myopic eyes was significantly thinner than less myopic eyes(P≤0.001),and the flow voids in the more myopic eyes were more than less myopic eyes(P=0.002).There was no significant difference in the vascular density of the choriocapillaris between the more and less myopic eyes(P=0.525).However,when anisometropia was more than 1.50 D,the vascular density of choriocapillaris in the more myopic eyes was significantly less than the less myopic eyes(P=0.026).The interocular difference of the choroidal thickness was significantly correlated with the interocular difference in SER and AL in the center,superior,and inferior regions but not in the nasal or temporal regions.The interocular differences of the vascular density and the flow voids of the choriocapillaris were not correlated with the interocular difference of SER and AL.Conclusions:The choroidal thickness is thinner in the more myopic eyes.The flow void is increased,and the vascular density of the choriocapillaris is reduced in the more myopic eyes of children with anisometropia exceeding 1.50 D. | Xinting Liu Zhiyi Lin Feifu Wang Xiaoyi Peng Wenwen He Dan Chen Meixiao Shen Fan Lu Jun Jiang | 2021 | Eye and Vision2021,8,1: | 3 |
| 5 | Associated risk factors in the early stage of diabetic retinopathy显示文摘Background:To investigate the retinal capillary density(RCD)of the macula using optical coherence tomography angiography(OCT-A)in type 2 diabetic patients and to further determine the association with risk factors.Methods:A total of 212 eyes from 212 subjects were recruited;subjects included diabetics with no retinopathy(NDR,n=90 eyes),diabetics with mild retinopathy DR(MDR,n=36 eyes),and healthy participants(Control,n=86 eyes).All participants underwent OCT-A scanning.RCD was quantified by superficial and deep retinal capillary layers(SRCL and DRCL)from OCT-A images.Results:RCD in SRCL and DRCL was lower in NDR(P<0.001)as well as in MDR(P<0.001)when compared with control eyes.Diabetic patients were subdivided according to individual risk factors,complications related to diabetes,and hyperglycemia.Diabetic patients showed lower RCD in both the SRCL and DRCL when compared with healthy controls.Diabetics with age>55y,HbA1c>7%had significantly reduced DRCL(P<0.05)when compared with the other group of diabetics(age<55y,HbA1c<7%).Diabetics with a blood urea nitrogen(BUN)>8.2 mmol/L had significantly reduced SRCL and DRCL when compared to the other group of diabetics.Conclusions:Risk factors including older age,higher level of HbA1c,LDL-C and BUN,were associated with lower RCDs found in type 2 diabetic patients with and without mild DR by OCT-A.The impairment of retinal capillary by OCT-A may play a key role in the early monitoring of management in diabetes. | Fan Tan Qi Chen Xiran Zhuang Chaoming Wu Yanying Qian Yuanyuan Wang Jianhua Wang Fan Lu Meixiao Shen Yingzi Li | 2019 | Eye and Vision2019,6,1: | 2 |

| 13 | Modeling of gonioscopic anterior chamber angle grades based on anterior segment optical coherence tomography显示文摘Background:To quantitatively assess anterior chamber angle(ACA)structure by anterior segment optical coherence tomography(AS-OCT)and develop a model to evaluate angle width as defined by gonioscopy.Methods:The ACAs of each quadrant were evaluated by gonioscopy,classified by the Scheie grading system,and assigned into one of the three grades:small angle(SA),moderate angle(MA),and large angle(LA).The eyes were imaged by AS-OCT,and ACA structural parameters including angle opening distance at the scleral spur(AODSS)and at 750μm anterior to the scleral spur(AOD750),trabecular-iris space area at 750μm anterior to the scleral spur(TISA750),and a newly defined parameter“light intersection distance”(LID),were measured.The ACA structural data were used to construct an ordered logistic regression model for assignment of ACAs to one of the three angle grades.The validity of the model was then tested.Results:A total of 169 quadrants from 53 subjects were included in the analysis,of which 111 quadrants were included in the modeling data and 58 in the testing data.In pairwise comparisons of SA,MA,and LA by ANOVA,the measured parameters were as follows:AOD750(0.174±0.060 vs.0.249±0.068 vs.0.376±0.114 mm;P<0.001),TISA750(0.075±0.035 vs.0.117±0.036 vs.0.181±0.062 mm^(2);P<0.001),and LID(−0.300±0.187 vs.-0.085±0.170 vs.0.122±0.156 mm;P<0.001).The ACA grading model based on LID showed a relatively high correction rate of 72.4%,and the model efficiency,calculated using the receiver operating characteristic,showed an area under the curve of 0.740.Weighted kappa statistics showed a good agreement for multiple ACA grades(0.772).Conclusions:The AS-OCT-based multiple ACA grades model was demonstrated as a non-contact approach for ACA assessment with high speed and high spatial resolution,providing guidance for diagnosis of angle closure. | Yingying Dai Shaodan Zhang Meixiao Shen Yuheng Zhou Mengyi Wang Jie Ye Dexi Zhu | 2020 | Eye and Vision2020,7,1: | 0 |
| 14 | Long scan depth optical coherence tomography on imaging accommodation: impact of enhanced axial resolution, signal-to-noise ratio and speed显示文摘Background:Spectral domain optical coherence tomography(SD-OCT)was a useful tool to study accommodation in human eye,but the maximum image depth is limited due to the decreased signal-to-noise ratio(SNR).In this study,improving optical resolutions,speeds and the SNR were achieved by custom built SD-OCT,and the evaluation of the impact of the improvement during accommodation was investigated.Methods:Three systems with different spectrometer designs,including two Charge Coupled Device(CCD)cameras and one Complementary Metal-Oxide-Semiconductor Transistor(CMOS)camera,were tested.We measured the point spread functions of a mirror at different positions to obtain the axial resolution and the SNR of three OCT systems powered with a light source with a 50 nm bandwidth,centered at a wavelength of 840 nm.Two normal subjects,aged 26 and 47,respectively,and one 75-year-old patient with an intraocular lens implanted were imaged.Results:The results indicated that spectrometers using cameras with 4096 camera pixels optimized the axial resolutions,due to the use of the full spectrum provided by the light source.The CCD camera system with 4096 pixels had the highest SNR and the best image quality.The system with the CMOS camera with 4096 pixels had the highest speed but had a compromised SNR compared to the CCD camera with 4096 pixels.Conclusions:Using these three OCT systems,we imaged the anterior segment of the human eye before and after accommodation,which showed similar results among the different systems.The system using the CMOS camera with an ultra-long scan depth,high resolution and high scan speed exhibited the best overall performance and therefore was recommended for imaging real-time accommodation. | Yilei Shao Aizhu Tao Hong Jiang Meixiao Shen Dexi Zhu Fan Lu Carol LKarp Yufeng Ye Jianhua Wang | 2018 | Eye and Vision2018,5,1: | 0 |

| 19 | Visual acuity is correlated with ischemia and neurodegeneration in patients with early stages of diabetic retinopathy显示文摘Purpose:We investigated the effects of retinal ischemia,neurodegeneration,and subclinical edema on best-corrected visual acuity(BCVA)in the early stages of diabetic retinopathy(DR).Methods:Ischemia was evaluated by the microvascular parameters measured by optical coherence tomography angiography.Neurodegeneration and subclinical edema were identified by the intraretinal layer thickness obtained by optical coherence tomography.Eyes with nonproliferative diabetic retinopathy(n=132)from 89 patients were analyzed.Eyes were classified as having normal BCVA(n=88[66.7%],Snellen equivalent≥20/20)or decreased BCVA(n=44[33.3%],Snellen equivalent<20/20).The prevalence of ischemia,neurodegeneration,and subclinical edema was explored in patients with and without decreased BCVA,and correlations between BCVA and these pathological pathways were determined.Results:Vessel density in the deep retinal capillary plexus(DRCP)and thickness of ganglion cell layer plus inner plexiform layer(GCL-IPL)were significantly lower in eyes with decreased BCVA compared with eyes with normal BCVA(both P<0.05).In the final multiple regression predictive model,age,DRCP vessel density,and GCL-IPL thickness(all P≤0.044)were predictors of BCVA.DRCP vessel density and GCL-IPL thickness have an interactive effect on visual acuity.The proportions of ischemia and neurodegeneration were significantly higher in eyes with decreased BCVA than in eyes with normal BCVA(P=0.001 and P=0.004,respectively).Conclusion:During the natural course of the early stages of DR,ischemia and neurodegeneration were the main disease pathways associated with visual acuity,and the mechanisms varied among patients. | Jin Li Yue Zhou Feng Chen Yingzi Li Rong Zhou Chaoming Wu Huankai Yu Zhiyang Lin Ce Shi Gu Zheng Yilei Shao Qi Chen Fan Lu Meixiao Shen | 2021 | Eye and Vision2021,8,1: | 0 |
