| 1 | Relationship between plasma D(-)-lactate and intestinal damage after severe injuries in rats显示文摘AIM To explore the kinetic changes in plasma D (-)lactate and lipopolysaccharide ( LPS ) levels, and investigate whether D (-)-lactate could be used as a marker of intestinal injury in rats following gut ischemia/reperfusion, burn, and acute necrotizing pancreatitis (ANP).METHODS Three models were developed in rats:gut ischemia/reperfusion obtained by one hour of superior severe burn injury created by 30% of total body surface induced by continuous inverse infusion of sodium taurocholate and trypsin into main pancreatic duct.Plasma levels of D( - )-lactate in systemic circulation and LPS in portal circulation were measured by enzymaticspectrophotometric method and limulus amebocyte lysate (LAL) test kit, respectively. Tissue samples of intestine were taken for histological analysis.RESULTS One hour gut ischemia followed by reperfusion injuries resulted in a significant elevation in plasma D( - )lactate and LPS levels, and there was a significant correlation between the plasma D( - )-lactate and LPS (r =0.719, P<0.05). The plasma concentrations of D(-)lactate and LPS increased significantly at 6 h postburn,and there was also a remarkable correlation between them (r = 0.877, P < 0.01). D ( - )-lactate and LPS levels elevated significantly at 2 h after ANP, with a similar significant correlation between the two levels (r-0.798,P < 0.01 ). The desquamation of intestine villi and infiltration of inflammatory cells in the lamina propria were observed in all groups.CONCLUSION The changes of plasma D( - )-lactate levels in systemic blood paralleled with LPS levels in the portal vein blood. | 9 | Riboflavin-induced Priming for Pathogen Defense in Arabidopsis thaliana显示文摘Riboflavin (vitamin B2) participates in a variety of redox processes that affect plant defense responses. Previously we have shown that riboflavin induces pathogen resistance in the absence of hypersensitive cell death (HCD) in plants. Herein, we report that ribo-avin induces priming of defense responses in Arabidopsis thaliana toward infection by virulent Pseudomonas syringae pv. tomato DC3000 (Pst). Induced resistance was mechanistically connected with the expression of defense response genes and cellular defense events, including H2O2 burst, HCD, and callose deposition in the plant. Riboflavin treatment and inoculation of plants with Pst were neither active but both synergized to induce defense responses. The priming process needed NPR1 (essential regulator of systemic acquired resistance) and maintenance of H2O2 burst but was independent of salicylic acid, jasmonic acid, ethylene, and abscisic acid. | 10 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 12 | Selective tropism of liver stem cells to hepatocellular carcinoma in vivo显示文摘AIM: To investigate the selective tropism of liver stem cells to hepatocellular carcinoma (HCC) in an animal model and its feasibility as a vector to deliver therapeutic genes for targeted therapy of HCC. METHODS: WB-F344, a kind of rat liver stem cell, was infected with recombinant virus to establish a cell line with stable, high-level expressing enhanced green fluorescent protein (EGFP). An animal model of HCC in Wistar rats was established by implanting HCC cells (CBRH7919) combined with an immunosuppressive drug. EGFP labeled liver stem cells were injected into caudal veins of the animals and distribution was observed at different time points after injection. SDF-1 and c-kit expression in non-tumor liver and tumor tissue were analysed by immunohistochemistry for the relationshiop between the expression and migration of liver stem cells. Furthermore, hepatic stem cells were injected via the portal vein, hepatic artery, caudal vein, or directly into the pericancerous liver tissue, respectively, and effects on migration, localization, and proliferation of the hepatic stem cells within the tumor tissue were observed and analyzed. RESULTS: Recombinant adenovirus could deliver the EGFP gene to hepatic stem cells. A new stem cell line, named WB-EGFP, was established that stably expressed EGFP. WB-EGFP cells still showed selective tropism towards HCC and EGFP expression was stable in vivo. According to immunohistochemistry results, SDF-1 may not be related to the mechanisms of tropism of hepatic stem cells. Different application sites affected the distribution of liver stem cells. Injection via the portalvein was superior with regard to selective migration, localization, and proliferation of the hepatic stem cells within the tumor tissue. CONCLUSION: Liver stem cells have the biological behavior of selective migration to HCC in vivo and they could localize and proliferate within HCC tissue stably expressing the target gene. | 14 | HOXC10 up-regulation promotes gastric cancer cell proliferation and metastasis through MAPK pathway显示文摘Objective: As an important regulator of embryonic morphogenesis,homeodomain-containing gene 10(HOXC10) has been found to promote progression of human cancers and its expression indicates poor survival outcome.However,very few studies are available on the role of HOXC10 in gastric carcinoma.Therefore,the aim of this study was to determine the role of HOXC10 in gastric cancer and the potential mechanism underlying its function for cancer biology.Methods: A primary gastric cancer mouse model was obtained via intra-gastric wall injection of gastric cancer cells and was used to evaluate the function of HOXC10 during gastric cancer progression in vivo.Immunohistochemistry was performed to visualize and measure HOXC10 protein expression in gastric cancer tissue.Cells were transfected with plasmids to increase the expression of HOXC10,and si RNA transfection was performed to suppress HOXC10 expression.Reverse transcription polymerase chain reaction(RT-PCR) and western blotting were utilized to measure m RNA and protein expression,respectively.Proliferation,migration,and invasion were investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay,wound healing assay,and matrigel invasion assay in vitro,respectively.Results: HOXC10 expression was significantly increased in gastric cancer tissues compared to matched normal tissues.HOXC10 up-regulation significantly increased tumor volumes in nude mice.Plasmid transfection significantly increased HOXC10 protein and m RNA expressions and effectively promoted cell proliferation.Moreover,HOXC10 up-regulation significantly promoted migration and invasion of gastric cancer cells.Mechanistic investigation showed that HOXC10 up-regulation significantly increased m RNA and protein expression of mitogen-activated protein kinase(MAPK) signaling related genes,including c-myc,c-jun and p53,while also modulating the phosphorylation of c-Jun N-terminal kinase(JNK),extracellular signal-regulated kinase(ERK) and P38 but not their total protein levels.Conclusions: This study demonstrated the tight link between HOXC10 and gastric cancer cell proliferation and metastasis via involvement of the MAPK pathway. | Chen Guo Jianing Hou Sheng Ao Xingming Deng Guoqing Lyu | 2017 | Chinese Journal of Cancer Research2017,29,6: | 10 |

| 18 | AtMGT7:An Arabidopsis Gene Encoding a Low-Affinity Magnesium Transporter显示文摘Magnesium(Mg2+) is one of the essential cations in all cells. Although the Mg2+ transport mechanism has been welldocumented in bacteria,less is known about Mg2+ transporters in eukaryotes. The AtMGT gene family encoding putative magnesium transport proteins had been described previously. We report here that one of the Arabidopsis MGT family members,the AtMGT7 gene,encodes two mRNAs that have resulted from alternative splicing variants,designated AtMGT7a and AtMGT7b. Interestingly,the two mRNA variants were expressed with different patterns with AtMGT7a expressing in all organs,but AtMGT7b appearing only in root and flowers. The AtMGT7a variant functionally complemented a bacterial mutant lacking Mg2+ transport capacity,whereas AtMGT7b did not. The 63Ni2+ tracer uptake analysis in the bacterial model showed that AtMGT7a mediated low-affinity transport of Mg2+. Consistent with the complementation assay result,63Ni2+ tracer uptake analysis revealed that AtMGT7b did not transport Mg2+.
