1Abundance of antibiotic resistance genes and their association with bacterial communities in activated sludge of wastewater treatment plants: Geographical distribution and network analysis显示文摘Wastewater treatment plants(WWTPs) are deemed reservoirs of antibiotic resistance genes(ARGs). Bacterial phylogeny can shape the resistome in activated sludge. However, the co-occurrence and interaction of ARGs abundance and bacterial communities in different WWTPs located at continental scales are still not comprehensively understood. Here, we applied quantitative PCR and Miseq sequence approaches to unveil the changing profiles of ARGs(sul1, sul2, tet W, tet Q, tet X), int I1 gene, and bacterial communities in 18 geographically distributed WWTPs. The results showed that the average relative abundance of sul1 and sul2 genes were 2.08 × 10^(-1) and 1.32 × 10^(-1) copies/16 S rRNA copies, respectively. The abundance of tet W gene was positively correlated with the Shannon diversity index(H′), while both studied sul genes had significant positive relationship with the int I1 gene. The highest average relative abundances of sul1, sul2, tet X, and int I1 genes were found in south region and oxidation ditch system. Network analysis found that 16 bacterial genera co-occurred with tet W gene. Co-occurrence patterns were revealed distinct community interactions between aerobic/anoxic/aerobic and oxidation ditch systems. The redundancy analysis model plot of the bacterial community composition clearly demonstrated that the sludge samples were significant differences among those from the different geographical areas,and the shifts in bacterial community composition were correlated with ARGs. 2A fundamental theorem for eco-environmental surface modelling and its applications显示文摘We propose a fundamental theorem for eco-environmental surface modelling(FTEEM) in order to apply it into the fields of ecology and environmental science more easily after the fundamental theorem for Earth’s surface system modeling(FTESM). The Beijing-Tianjin-Hebei(BTH) region is taken as a case area to conduct empirical studies of algorithms for spatial upscaling, spatial downscaling, spatial interpolation, data fusion and model-data assimilation, which are based on high accuracy surface modelling(HASM), corresponding with corollaries of FTEEM. The case studies demonstrate how eco-environmental surface modelling is substantially improved when both extrinsic and intrinsic information are used along with an appropriate method of HASM. Compared with classic algorithms, the HASM-based algorithm for spatial upscaling reduced the root-meansquare error of the BTH elevation surface by 9 m. The HASM-based algorithm for spatial downscaling reduced the relative error of future scenarios of annual mean temperature by 16%. The HASM-based algorithm for spatial interpolation reduced the relative error of change trend of annual mean precipitation by 0.2%. The HASM-based algorithm for data fusion reduced the relative error of change trend of annual mean temperature by 70%. The HASM-based algorithm for model-data assimilation reduced the relative error of carbon stocks by 40%. 3Immunity-and-matrix-regulatory cells derived from human embryonic stem cells safely and effectively treat mouse lung injury and fibrosis显示文摘Lung injury and fibrosis represent the most significant outcomes of severe and acute lung disorders,including COVID-19.However,there are still no effective drugs to treat lung injury and fibrosis.In this study,we report the generation of clinical-grade human embryonic stem cells(hESCs)-derived immunity-and matrix-regulatory cells(IMRCs)produced under good manufacturing practice requirements,that can treat lung injury and fibrosis in vivo.We generate IMRCs by sequentially differentiating hESCs with serum-free reagents.IMRCs possess a unique gene expression profile distinct from that of umbilical cord mesenchymal stem cells(UCMSCs),such as higher expression levels of proliferative,immunomodulatory and anti-fibrotic genes.Moreover,intravenous delivery of IMRCs inhibits both pulmonary inflammation and fibrosis in mouse models of lung injury,and significantly improves the survival rate of the recipient mice in a dose-dependent manner,likely through paracrine regulatory mechanisms.IMRCs are superior to both primary UCMSCs and the FDA-approved drug pirfenidone,with an excellent efficacy and safety profile in mice and monkeys.In light of public health crises involving pneumonia,acute lung injury and acute respiratory distress syndrome,our findings suggest that IMRCs are ready for clinical trials on lung disorders.Jun Wu Dingyun Song Zhongwen Li Baojie Guo Yani Xiao Wenjing Liu Lingmin Liang Chunjing Feng Tingting Gao Yanxia Chen Ying Li Zai Wang Jianyan Wen Shengnan Yang Peipei Liu Lei Wang Yukai Wang Liang Peng Glyn Nigel Stacey Zheng Hu Guihai Feng Wei Li Yan Huo Ronghua Jin Ng Shyh-Chang Qi Zhou Liu Wang Baoyang Hu Huaping Dai Jie Hao 2020Cell Research2020,30,9:7
4Realization of population inversion between 7S_(1/2) and 6P_(3/2) levels of cesium for four-level active optical clock显示文摘We demonstrate experimentally the population inversion between 7S1/2 and 6P3/2 levels of cesium in thermal cesium cell with a 455.5 nm pumping laser.We calculate the relative population probabilities at each level theoretically with the density matrix method.In a steady state,5.8% atoms are at 7S1/2 level and 2.9% at 6P3/2 level,which builds up the population inversion between the two levels.We obtain the fluorescence spectra produced in thermal cesium cell in our experiment.The measured relative intensity of each available fluorescence spectral line in the experiment agrees very well with the theoretical result.The demonstrated population inversion between 7S1/2 and 6P3/2 levels can be used to construct an active optical clock of four-level system with a wavelength of 1469.9 nm.WANG YanFei WANG DongYing ZHANG TongGang HONG YeLong ZHANG ShengNan TAO ZhiMing XIE XiaoPeng CHEN JingBiao 2013Science China(Physics,Mechanics & Astronomy)2013,56,6:6
5Recent developments in multimodality fluorescence imaging probes显示文摘Multimodality optical imaging probes have emerged as powerful tools that improve detection sensitivity and accuracy, important in disease diagnosis and treatment. In this review, we focus on recent developments of optical fluorescence imaging(OFI) probe integration with other imaging modalities such as X-ray computed tomography(CT), magnetic resonance imaging(MRI), positron emission tomography(PET), single-photon emission computed tomography(SPECT), and photoacoustic imaging(PAI). The imaging technologies are briefly described in order to introduce the strengths and limitations of each techniques and the need for further multimodality optical imaging probe development. The emphasis of this account is placed on how design strategies are currently implemented to afford physicochemically and biologically compatible multimodality optical fluorescence imaging probes. 6Superior adsorption performance of graphitic carbon nitride nanosheets for both cationic and anionic heavy metals from wastewater显示文摘Water pollution caused by highly toxic Cd(II), Pb(II), and Cr(VI) is a serious problem. In the present work,a green and low-cost adsorbent of g-C_3N_4 nanosheets was developed with superior capacity for both cationic and anionic heavy metals. The adsorbent was easily fabricated through one-step calcination of guanidine hydrochloride with thickness less than 1.6 nm and specific surface area of 111.2 m^2·g^(-1). Kinetic and isotherm studies suggest that the adsorption is an endothermic chemisorption process, occurring on the energetically heterogeneous surface based on a hybrid mechanism of multilayer and monolayer adsorption. The tri-s-triazine units and surface N-containing groups of g-C_3N_4 nanosheets are proposed to be responsible for the adsorption process.Further study on pH demonstrates that electrostatic interaction plays an important role. The maximum adsorption capacity of Cd(II), Pb(II), and Cr(VI) on g-C_3N_4 nanosheets is 123.205 mg·g^(-1), 136.571 mg·g^(-1),and 684.451 mg·g^(-1), respectively. The better adsorption performance of the adsorbent than that of the recently reported nanomaterials and low-cost adsorbents proves its great application potential in the removal of heavy metal contaminants from wastewater. 7A comparative study of distant hybridization in plants and animals显示文摘Distant hybridization refers to crosses between two different species, genera, or higher-ranking taxa, which can break species limits, increase genetic variation, and combine the biological characteristics of existing species. It is an important way of creating genetic variation, fertile strains, and excellent characteristics in new strains and populations. Combining analyses and summaries from many inter-related documents in plants and animals, both domestic and international, including examples and long-standing research on distant hybridization in fish from our laboratory, we summarize and compare the similarities and differences in plant and animal distant hybridization. In addition, we analyze and review the biological characteristics of their different ploidy progenies and the possible causes of disparity in survival rates. Mechanisms of sterility in animal and plant distant hybrids are also discussed, and research methods for the study of biological characteristics of hybrids, including morphology, cytology,and molecular cytogenetics are presented. This paper aims to provide comprehensive research materials and to systematically compare the general and specific characteristics of plant and animal hybrids with regards to reproduction, genetics, growth traits,and other biological characteristics. 8Facile Approach to Synthesize Gold Nanorod@Polyacrylic Acid/Calcium Phosphate Yolk–Shell Nanoparticles for Dual-Mode Imaging and pH/NIR-Responsive Drug Delivery显示文摘A facile strategy to fabricate gold nanorod@polyacrylic acid/calcium phosphate(Au NR@-PAA/Ca P) yolk–shell nanoparticles(NPs) composed with a PAA/Ca P shell and an Au NR yolk is reported. The asobtained Au NR@PAA/Ca P yolk–shell NPs possess ultrahigh doxorubicin(DOX) loading capability(1 mg DOX/mg NPs), superior photothermal conversion property(26%)and p H/near-infrared(NIR) dual-responsive drug delivery performance. The released DOX continuously increased due to the damage of the Ca P shell at low p H values. When the DOX-loaded Au NR@PAA/Ca P yolk–shell NPs wereexposed to NIR irradiation, a burst-like drug release occurs owing to the heat produced by the Au NRs. Furthermore,Au NR@PAA/Ca P yolk–shell NPs are successfully employed for synergic dual-mode X-ray computed tomography/photoacoustic imaging and chemo-photothermal cancer therapy. 10Enhancing recovery and sensitivity studies in an unconventional tight gas condensate reservoir显示文摘The recovery factor from tight gas reservoirs is typically less than 15%, even with multistage hydraulic fracturing stimulation. Such low recovery is exacerbated in tight gas condensate reservoirs, where the depletion of gas leaves the valuable condensate behind. In this paper, three enhanced gas recovery(EGR) methods including produced gas injection,CO_2 injection and water injection are investigated to increase the well productivity for a tight gas condensate reservoir in the Montney Formation, Canada. The production performance of the three EGR methods is compared and their economic feasibility is evaluated. Sensitivity analysis of the key factors such as primary production duration, bottom-hole pressures,and fracture conductivity is conducted and their effects on the well production performance are analyzed. Results show that, compared with the simple depletion method, both the cumulative gas and condensate production increase with fluids injected. Produced gas injection leads to both a higher gas and condensate production compared with those of the CO_2 injection, while waterflooding suffers from injection difficulty and the corresponding low sweep efficiency. 11Three-dimensional domain swapping as a mechanism to lock the active conformation in a super-active octamer of SARS-CoV main protease显示文摘Proteolytic processing of viral polyproteins is indispensible for the lifecycle of coronaviruses.The main protease(M^(pro))of SARS-CoV is an attractive target for anti-SARS drug development as it is essential for the polyprotein processing.M^(pro) is initially produced as part of viral polyproteins and it is matured by autocleavage.Here,we report that,with the addition of an N-terminal extension peptide,M^(pro) can form a domain-swapped dimer.After complete removal of the extension peptide from the dimer,the mature M^(pro) self-assembles into a novel super-active octamer(AO-M^(pro)).The crystal structure of AO-M^(pro) adopts a novel fold with four domainswapped dimers packing into four active units with nearly identical conformation to that of the previously reported M^(pro) active dimer,and 3D domain swapping serves as a mechanism to lock the active conformation due to entanglement of polypeptide chains.Compared with the previously well characterized form of M^(pro),in equilibrium between inactive monomer and active dimer,the stable AO-M^(pro) exhibits much higher proteolytic activity at low concentration.As all eight active sites are bound with inhibitors,the polyvalent nature of the interaction between AO-M^(pro) and its polyprotein substrates with multiple cleavage sites,would make AO-M^(pro) functionally much more superior than the M^(pro) active dimer for polyprotein processing.Thus,during the initial period of SARS-CoV infection,this novel active form AOM^(pro) should play a major role in cleaving polyproteins as the protein level is extremely low.The discovery of AOM^(pro) provides new insights about the functional mechanism of M^(pro) and its maturation process.Shengnan Zhang Nan Zhong Fei Xue Xue Kang Xiaobai Ren Jiaxuan Chen Changwen Jin Zhiyong Lou Bin Xia 2010Protein & Cell2010,1,4:2
12Replanting of broadleaved trees alters internal nutrient cycles of native and exotic pines in subtropical plantations of China显示文摘Background:The replanting of broadleaved trees in pure coniferous plantations is widely implemented,as mixed plantations are generally more stable and functional.However,the effect of interspecific interactions between broadleaved and coniferous trees on internal nutrient cycles of conifers remains unclear.Methods:We selected pure coniferous plantations of a native(Pinus massoniana)and an exotic(P.elliottii)pine species and their corresponding mixed plantations with broadleaved trees(Schima superba)in subtropical China,and measured the nitrogen(N)and phosphorus(P)contents in the rhizosphere soils,fine roots,twigs,needles and needle litter of pines.We calculated the root capture,needle resorption and translocation of N and P by pines to determine the mobility of nutrients in trees.Results:Although the N and P in the rhizosphere soils increased due to the replanting of broadleaved trees,the N and P contents in the aboveground tissues of the two pine species did not increase in mixed plantations.Mixed planting had a negative effect on the N and P capture of native pine and a positive effect on that of exotic pine.The N and P resorption efficiencies increased in native pine but were unchanged in exotic pine after the replanting of S.superba.Native pine preferentially employed an aboveground nutrient resorption strategy,whereas exotic pine tended to adopt a belowground nutrient capture strategy after replanting.Translocation of N and P in trees was detected,which reflected the trade-offs between root nutrient capture and needle nutrient resorption.Conclusions:The effect of mixed planting varied between the species of native and exotic pines,and the internal nutrient cycles of both pine species might be dominated by interspecific interaction effects on nutrients rather than soil nutrients.Our study highlights the importance of selecting suitable broadleaved species for replanting in coniferous plantations.Yong Lin Chengkang Xia Gaoyang Wu Fangchao Wang Shengnan Wang Yuanqiu Liu Fusheng Chen 2022Forest Ecosystems2022,9,5:2

16Molecular cloning and expression analysis of the Rf-m candidate genes encoding pentatricopeptide repeat proteins in soybean显示文摘Cytoplasmic male sterility(CMS)-restorer system is a useful tool to exploit heterosis in soybean.The major restorer gene for the M-type CMS is known as Rf-m,located in the 162.4-kb region on chromosome 16.Sequence analysis has revealed that the Rf-m locus in Glycine max consists of seven penta tricopeptide repeat(GmPPR)genes.The deduced amino acid sequences contain 8 to 14 PPR motifs,and a phylogenetic analysis grouped these GmPPR proteins into two PPR subfamilies:Glyma.16G161800 belongs to the PLS subfamily,and the P subfamily consists.of Glyma.16G161900,Glyma 16G162000,Glyma.16G162100,Glyma.16G162700,Glyma.16G162800,and Gly-ma 16G163100.The phylogenetic analysis of seven GmPPR proteins and 27 other plant PPR proteins also showed that proteins in the same subfamilies cluster together.Comparative sequence analysis was conducted using the seven Rf-m candidate GmPPR genes from the sterile line W931A,the maintainer line W931B,and the restorer line WR016,the result showed that Glyma 16G161900 had higher polymorphism than the other candidate genes.Based on real-time quantitative RT-PCR data,all seven GmPPR genes were differentially expressed but showed constitutive expression in roots,stems,leaves,and pollen grains.Additionally,the expression level of Gly-ma 16G161900 in the sterile line W931 A was significantly higher in all tissues than in the restorer line WR016.Taken together,these results suggest that Glyma 16G161900 is the most likely candidate for the restorer gene Rf-m.This study is the first report and analysis of candidate fertility restorer(Rf)genes encoding PPR proteins in soybean.Dagang Wang Shengnan Chen Jiekun Li Qian Wu Guoyu Hu Zhiping Huang 2021Oil Crop Science2021,6,3:1

19An integrated spectroscopic strategy to trace the geographical origins of emblic medicines: Application for the quality assessment of natural medicines显示文摘Emblic medicine is a popular natural source in the world due to its outstanding healthcare and therapeutic functions.Our preliminary results indicated that the quality of emblic medicines might have an apparent regional variation.A rapid and effective geographical traceability system has not been designed yet.To trace the geographical origins so that their quality can be controlled,an integrated spectroscopic strategy including spectral pretreatment,outlier diagnosis,feature selection,data fusion,and machine learning algorithm was proposed.A featured data matrix(245220)was successfully generated,and a carefully adjusted RF machine learning algorithm was utilized to develop the geographical traceability model.The results demonstrate that the proposed strategy is effective and can be generalized.Sensitivity(SEN),specificity(SPE)and accuracy(ACC)of 97.65%,99.85%and 97.63%for the calibrated set,as well as 100.00%predictive efficiency,were obtained using this spectroscopic analysis strategy.Our study has created an integrated analysis process for multiple spectral data,which can achieve a rapid,nondestructive and green quality detection for emblic medicines originating from seventeen geographical origins.Luming Qi Furong Zhong Yang Chen Shengnan Mao Zhuyun Yan Yuntong Ma 2020Journal of Pharmaceutical Analysis2020,10,4:1
20BET bromodomain inhibitor JQ1 regulates spermatid development by changing chromatin conformation in mouse spermatogenesis显示文摘As a BET bromodomain inhibitor, JQ1 has been proven have efficacy against a number of different cancers. In terms of male reproduction, JQ1 may be used as a new type of contraceptive, since JQ1 treatment in male mice could lead to germ cell defects and a decrease of sperm motility, moreover, this effect is reversible. However, the mechanism of JQ1 acting on gene regulation in spermatogenesis remains unclear. Here, we performed single-cell RNA sequencing (scRNA-seq) on mouse testes treated with JQ1 or vehicle control to determine the transcriptional regulatory function of JQ1 in spermatogenesis at the single cell resolution. We confirmed that JQ1 treatment could increase the numbers of somatic cells and spermatocytes and decrease the numbers of spermatid cells. Gene Ontology (GO) analysis demonstrated that differentially expressed genes which were down-regulated after JQ1 injection were mainly enriched in “DNA conformation change” biological process in early developmental germ cells and “spermatid development” biological process in spermatid cells. ATAC-seq data further confirmed that JQ1 injection could change the open state of chromatin. In addition, JQ1 could change the numbers of accessible meiotic DNA double-stranded break sites and the types of transcription factor motif that functioned in pachytene spermatocytes and round spermatids.
