1Single-cell brain atlas of Parkinson's disease mouse model显示文摘Parkinson's disease(PD)is a neurodegenerative disease,leading to the impairment of movement execution.PD pathogenesis has been largely investigated,either limited to bulk transcriptomic levels or at certain cell types,which failed to capture the cellular heterogeneity and intrinsic interplays among distinct cell types.Here,we report the application of single-nucleus RNA-seq on midbrain,striatum,and cerebellum of theα-syn-A53 T mouse,a well-established PD mouse model,and matched controls,generating the first single cell transcriptomic atlas for the PD model mouse brain composed of 46,174 individual cells.Additionally,we comprehensively depicte the dysfunctions in PD pathology,covering the elevation of NF-k B activity,the alteration of ion channel components,the perturbation of protein homeostasis network,and the dysregulation of glutamatergic signaling.Notably,we identify a variety of cell types closely associated with PD risk genes.Taken together,our study provides valuable resources to systematically dissect the molecular mechanism of PD pathogenesis at the single-cell resolution,which facilitates the development of novel approaches for diagnosis and therapies against PD.Jixing Zhong Gen Tang Jiacheng Zhu Weiying Wu Ge Li Xiumei Lin Langchao Liang Chaochao Chai Yuying Zeng Feiyue Wang Lihua Luo Jiankang Li Fang Chen Zhen Huang Xiuqing Zhang Yu Zhang Hongde Liu Xin Qiu Shengping Tang Dongsheng Chen 2021Journal of Genetics and Genomics2021,48,4:1
2Screening for musculoskeletal system malformations and birth injuries in newborns:Results of a screening program in two hospitals in Shenzen,China显示文摘Importance:There are a variety of musculoskeletal malformations and injuries that can occur in newborns.These can be a significant cause of perinatal death or a reason for miscarriage and can lead to long-term functional issues if not managed appropriately.There is no systematic and well-established screening program for neonatal musculoskeletal malformations and injuries in China now.Objective:To report the incidence and types of congenital musculoskeletal malformations in two hospitals in Shenzhen City,to explore and discuss the details of the screening procedure and improve future prevention and treatment.Methods:From October 2013 to May 2014,2564 one-day-old newborns were screened by a pediatric orthopedic physical examination,in combination with ultrasonography when required,and the incidence and variety of diseases were recorded statistically.Results:Among 2564 screened newborns,the following musculoskeletal conditions were identified:congenital muscular torticollis(CMT)(seven cases,0.27%),hip subluxation(four cases,0.16%),hip dysplasia(47 cases,1.83%),congenital talipes equinovarus(CTEV)(two cases,0.08%),congenital talipes calcaneovalgus(15 cases,0.58%),polydactyly(nine cases,0.35%),syndactyly(one case,0.04%),and spinal hemivertebra(one case,0.04%).Additionally,there were five(0.19%)neonates with birth injuries.Interpretation:It is feasible to carry out neonatal screening and identification of musculoskeletal malformations and birth injuries in China.This is helpful as timely detection and early intervention for many of these conditions can avoid permanent functional impairment in these children.Zhu Xiong Zhenhui Zhao Hansheng Deng Xin Qiu Weiqing Li Xiaodi Chen Yu Tang Shuai Han Jiaxin Zhao Ting Cai Xian Liu Shuaidan Zeng Shengping Tang 2022Pediatric Investigation2022,6,3:1
3Activated carbon prepared from waste tire pyrolysis carbon black via CO_(2)/KOH activation used as supercapacitor electrode显示文摘As the quantity of waste tires increases,more pyrolysis carbon black(CBp),a type of low value-added carbon black,is being produced.However,the application of CBp has been limited.Therefore,it is necessary to identify and expand applications of CBp.This work focuses on the preparation of activated carbon(AC)from CBp using the physicochemical activation of carbon dioxide(CO_(2))and potassium hydroxide(KOH).Thereafter,AC is applied to the electrode of the electrical double-layer capacitor(EDLC).The AC prepared by CO_(2)/KOH activation exhibited a hierarchical pore structure.The specific surface area increased from 415 to 733 m^(2)g^(−1),and in combination with low ash content of 1.51%,ensured abundant ion diffusion channels and active sites to store charge.The EDLC comprising the AC(AC-2)electrode prepared by excitation of CO_(2)(300 sccm)and KOH had a reasonable gravimetric specific capacitance of 192 F g^(−1)at 0.5 A g^(−1),and exhibited a good rate capability of 73%at 50 A g^(−1)in a three-electrode system.Moreover,the EDLC device comprising the AC-2 electrode delivered excellent cycling stability(capacitance retention of 106%after 10000 cycles at 2 A g^(−1)in a two-electrode system).Furthermore,a symmetric supercapacitor based on an AC electrode that exhibits a supreme energy density of 4.7 Wh kg^(−1)and a maximum power density of 6362.6 W kg^(−1)is demonstrated.HOU ShengPing ZHANG Da XIE ZhiPeng KANG Yao TANG ZhengGang DAI YongNian LEI Yong CHEN Jian LIANG Feng 2022Science China(Technological Sciences)2022,65,10:1

5New strategy for reversing biofilm-associated antibiotic resistance through ferrocene-substituted carborane ruthenium(Ⅱ)-arene complex显示文摘Bacterial biofilms are inherently resistant to antimicrobial agents and are difficult to eradicate with conventional antimicrobial agents, resulting in many persistent and chronic bacterial infections. In this contribution, a new strategy for reversing the biofilm-associated antibiotic resistance has been explored by induction of a carborane ruthenium(II)-arene complex (FcRuSB). Our results demonstrate that the FcRuSB could be utilized as an inducer to efficiently reverse the biofilm-associated antibiotic resistance of multidrug-resistant (MDR) clinical isolates of Staphylococcus aureus and Pseudomonas aeruginosa. The induced effect of FcRuSB is correlated with a considerable decrease in the expression of extracellular matrix proteins (EMP) of the two strains. 6The OsBSK1-2-MAPK module regulates blast resistance in rice显示文摘Receptor-like cytoplasmic kinase OsBSK1-2 was reported to play an important role in regulation of response to rice blast,but the signaling pathway remained unknown.In this study,we identified OsMAPKKK18 and previously uncharacterized MAPKKKs OsMAPKKK16 and OsMAPKKK19 that interact with OsBSK1-2.Expression of all three MAPKKKs was induced by Magnaporthe oryzae infection,and all three induced cell death when transiently expressed in Nicotiana benthamiana leaves.Knockout of OsMAPKKK16 and OsMAPKKK18 compromised blast resistance and overexpression of OsMAPKKK19 increased blast resistance,indicating that all three MAPKKKs are involved in regulation of rice blast response.Furthermore,both OsMAPKKK16 and OsMAPKKK19 interacted with and phosphorylated OsMKK4 and OsMKK5,and chitin-induced MAPK activation was suppressed in osmapkkk16 and osbsk1-2 mutants.OsMAPKKK18 was earlier reported to interact with and phosphorylate OsMKK4 and affect chitin-induced MAPK activation,suggesting that OsBSK1-2 is involved in regulation of immunity through multiple MAPK signaling pathways.Unlike BSK1 in Arabidopsis,OsBSK1-2 was not involved in response to avirulent M.oryzae strains.Taken together,our results revealed important roles of OsMAPKKK16/18/19 and a OsBSK1-2-OsMAPKKK16/18/19-OsMKK4/5 module in regulating response to rice blast.Shengping Li Xinquan Xiang Zhijuan Diao Na Xia Ling Lu Jing Zhang Zhiwei Chen Dingzhong Tang 2024The Crop Journal2024,12,1:0
