1Association of KIR Genotypes and Haplotypes with Susceptibility to Chronic Hepatitis B Virus Infection in Chinese Han Population显示文摘Killer immunoglobulin-like receptor (KIR) genes can regulate the activation of NK and T cells upon interaction with HLA class I molecules. Hepatitis B virus (HBV) infection has been regarded as a multi-factorial disorder disease. Previous studies revealed that KIRs were involved in HCV and HIV infection or clearance. The aim of this study was to explore the possibility of the inheritance of KIR genotypes and haplotypes as a candidate for susceptibility to persistent HBV infection or HBV clearance. The sequence specific primer polymerase chain reaction (SSP-PCR) was employed to identify the KIR genes and pseudogenes in 150 chronic hepatitis B (CHB) patients, 251 spontaneously recovered (SR) controls, and 412 healthy controls. The frequencies of genotype G, M, FZ1 increased in CHB patients compared with healthy control subjects. The frequency of genotype AH was higher in SR controls than that in both CHB patients and healthy controls. The carriage frequencies of genotype G and AH were higher; while, the frequencies of AF and AJ were lower in SR controls than those in healthy control subjects. The frequency of A haplotype was lower, whereas, the frequency of B haplotype was higher in CHB patients and SR controls than those in healthy controls. In healthy controls, haplotype 4 was found lower compared with that in CHB patients and SR controls and the frequency of haplotype 5 was higher in SR controls than that in other two groups. Based on these findings, it seems that the genotypes M and FZ1 are HBV susceptive genotypes; AH, on the other hand, may be protective genotypes that facilitate the clearance of HBV. 2Single-Atom Catalysts for Electrochemical Hydrogen Evolution Reaction: Recent Advances and Future Perspectives显示文摘Hydrogen,a renewable and outstanding energy carrier with zero carbon dioxide emission,is regarded as the best alternative to fossil fuels.The most preferred route to large-scale production of hydrogen is by water electrolysis from the intermittent sources(e.g.,wind,solar,hydro,and tidal energy).However,the efficiency of water electrolysis is very much dependent on the activity of electrocatalysts.Thus,designing high-effective,stable,and cheap materials for hydrogen evolution reaction(HER)could have a substantial impact on renewable energy technologies.Recently,single-atom catalysts(SACs)have emerged as a new frontier in catalysis science,because SACs have maximum atom-utilization efficiency and excellent catalytic reaction activity.Various synthesis methods and analytical techniques have been adopted to prepare and characterize these SACs.In this review,we discuss recent progress on SACs synthesis,characterization methods,and their catalytic applications.Particularly,we highlight their unique electrochemical characteristics toward HER.Finally,the current key challenges in SACs for HER are pointed out and some potential directions are proposed as well.Zonghua Pu Ibrahim Saana Amiinu Ruilin Cheng Pengyan Wang Chengtian Zhang Shichun Mu Weiyue Zhao Fengmei Su Gaixia Zhang Shijun Liao Shuhui Sun 2020Nano-Micro Letters2020,12,2:10
3Modeling cavitation flow of cryogenic fluids with thermodynamic phase-change theory显示文摘Cavitation is the formation of vapor bubbles within a liquid where the flow dynamics causes the local static pressure to drop below the vapor pressure. The so-called full cavitation model (FCM) developed by Singhal has been widely used in numerical modeling of the cavitation flow for thermosensible and non-thermosensible fluids. Within the FCM, the bubble size is taken to be equivalent to the maximum possible value to forego the calculation of bubble number density. We developed a new cavitation model by recalculating the bubble radius in FCM to account for the effects of local pressure. The new model was obtained by combining the thermodynamic phase-change theory and the Young-Laplace equation with the assumption of thermodynamic equilibrium during the cavitation process. The cavitation calculations were performed based on the mathematical framework of the homogeneous equilibrium flow model and the transport-equation-based model for vapor phase mass fraction. The model was validated by modeling the cavitating flow of liquid nitrogen and liquid hydrogen through NASA hydrofoil and Ogive with consideration of the phase-change thermal effects. 4Mortality outcomes of low-dose computed tomography screening for lung cancer in urban China:a decision analysis and implications for practice显示文摘Background: Mortality outcomes in trials of low-dose computed tomography(CT) screening for lung cancer are inconsistent. This study aimed to evaluate whether CT screening in urban areas of China could reduce lung cancer mortality and to investigate the factors that associate with the screening effect.Methods: A decision tree model with three scenarios(low-dose CT screening, chest X-ray screening, and no screening) was developed to compare screening results in a simulated Chinese urban cohort(100,000 smokers aged45-80 years). Data of participant characteristics were obtained from national registries and epidemiological surveys for estimating lung cancer prevalence. The selection of other tree variables such as sensitivities and specificities of low-dose CT and chest X-ray screening were based on literature research. Differences in lung cancer mortality(primary outcome), false diagnoses, and deaths due to false diagnosis were calculated. Sensitivity analyses were performed to identify the factors that associate with the screening results and to ascertain worst and optimal screening effects considering possible ranges of the variables.Results: Among the 100,000 subjects, there were 448,541, and 591 lung cancer deaths in the low-dose CT, chest X-ray, and no screening scenarios, respectively(17.2% reduction in low-dose CT screening over chest X-ray screening and 24.2% over no screening). The costs of the two screening scenarios were 9387 and 2497 false diagnoses and 7and 2 deaths due to false diagnosis among the 100,000 persons, respectively. The factors that most influenced death reduction with low-dose CT screening over no screening were lung cancer prevalence in the screened cohort, lowdose CT sensitivity, and proportion of early-stage cancers among low-dose CT detected lung cancers. 5Gene expression profiling of rat livers with Yin-deficiency-heat syndrome显示文摘OBJECTIVE: To explore the nature of 'Yin internal heat caused byYin-deficiency,' in terms of the theory of Traditional Chinese Medicine, by studying energy metabolism in rats with Yin-deficiency-heat syndrome and analyzing the gene expression profile of their livers. METHODS: A Yin-deficiency-heat syndrome model was induced in rats using three Chinese medicinal herbs. Glycogen and triglycerides in blood plasma, and the enzyme activity of ATP in livers were measured colorimetrically. Triiodothyronine (T3 ), thyroxine (T4 ), and thyroid stimulating hormone levels in blood plasma were also measured with enzyme linked immunosorbent assay. The gene expression profile of livers was detected with gene chip analysis. Differentially expressed genes were screened out and classified according to Gene Ontology. The accuracy of results were examined with reversetranscription-polymerase chain reaction. RESULTS: Compared with the control group, body weight (P<0.05) and hepatic glycogen (P<0.05) were significantly lower in the Yin-deficiency-heat syndrome group. Moreover, toe temperature (P< 0.01) and triglyceride (P<0.05), Na+ -K+ -ATPase (P< 0.01), Mg2+ -ATPase (P<0.01), T3 (P<0.05), and T4 (P< 0.01) levels were significantly higher. There were 99 differentially expressed genes in livers from the Yin-deficiency-heat syndrome group. Genes were mainly related to sterol synthesis (Pc=0.0392), defense response (Pc=0.0448), and sterol metabolism (Pc=0.0533). CONCLUSION: Abnormal expression genes in rats with Yin-deficiency-heat syndrome prompted the synthesis and metabolism of cholesterol, increased energy consumption, and reduced defense response. 6Defect properties in a VTaCrW equiatomic high entropy alloy(HEA)with the body centered cubic(bcc)structure显示文摘We report first-principles results of the point defect properties in a V-Ta-Cr-W high-entropy alloy(HEA)with the body-centered cubic(bcc)structure.Different from the widely-investigated face-centered cubic(fcc)HEAs,the local lattice distortion is more pronounced in bcc ones,which has a strong influence on the defect properties and defect evolution under irradiation.Due to the large size of Ta,the exchange between vacancies and Ta exhibits lower energy barriers.On the other hand,interstitial dumbbells containing V and Cr possess lower formation energies.These defect energetics predicts an enrichment of V and Cr and a depletion of Ta andWin the vicinity of defect sinks.Besides,we find that interstitial dumbbells favor the[110]orientation in the HEA,instead of[111]direction in most nonmagnetic bcc metals,which helps to slow down interstitial diffusion significantly.Consequently,the distribution of migration energies for vacancies and interstitials exhibit much larger overlap regions in the bcc HEA compared to fcc HEAs,leading to the good irradiation resistance by enhancing defect recombination.Our results suggest that HEAs with the bcc structure may bear excellent irradiation tolerance due to the particular defect properties.Shijun Zhao 2020Journal of Materials Science & Technology2020,44,9:6
7Formal analysis of TPM2.0 key management APIs显示文摘The trusted platform module(TPM),a system component implemented on physical resources,is designed to enable computers to achieve a higher level of security than the security level that it is possible to achieve by software alone.For this reason,the TPM provides a way to store cryptographic keys and other sensitive data in its memory,which is shielded from access by any entity other than the TPM.Users who want to use those keys and data to achieve some security goals are restricted to interact with the TPM through its APIs defined in the TPM specification.Therefore,whether the TPM can provide Protected Capabilities it claimed depends to a large extent on the security of its APIs.In this paper,we devise a formal model,which is accessible to a fully mechanized analysis,for the key management APIs in the TPM2.0 specification.We identify and formalize security properties of these APIs in our model and then successfully use the automated prover Tamarin to obtain the first mechanized analysis of them.The analysis shows that the key management subset of TPM APIs preserves the secrecy of non-duplicable keys for unbounded numbers of fresh keys and handles.The analysis also reports that the key duplication mechanism,used to duplicate a key between two hierarchies,is vulnerable to impersonation attacks,which enable an adversary to recover the duplicated key of the originating hierarchy or import his own key into the destination hierarchy.Aiming at avoiding these vulnerabilities,we proposean approach,which restricts the originating and destination TPMs to authenticate each other’s identity during duplication.Then we formally demonstrate that our approach maintains the secrecy of duplicable keys when they are duplicated.Qianying Zhang Shijun Zhao Yu Qin Dengguo Feng 2014Chinese Science Bulletin2014,59,32:5
8Contribution of Baroreflex Afferent Pathway to NPY-Mediated Regulation of Blood Pressure in Rats显示文摘Neuropeptide Y (NPY), a metabolism-related cardiovascular factor, plays a crucial role in blood pressure(BP) regulation via peripheral and central pathways. The expression of NPY receptors (Y1R/Y2R) specific to baroreflex afferents impacts on the sexually dimorphic neural control of circulation. This study was designed to investigate the expression profiles of NPY receptors in the nodose ganglion (NG) and nucleus tractus solitary (NTS) under hypertensive conditions. To this end, rats with hypertension induced by NG-nitro-L-arginine methylester (L-NAME) or high fructose drinking (HFD), and spontaneously hypertensive rats (SHRs) were used to explore the effects/mechanisms of NPY on BP using functional, molecular, and electrophysiological approaches. The data showed that BP was elevated along with baroreceptor sensitivity dysfunction in model rats;Y1R was up-or down-regulated in the NG or NTS of male and female HFD/L-NAME groups,while Y2R was only down-regulated in the HFD groups as well as in the NG of the male L-NAME group. In SHRs,Y1R and Y2R were both down-regulated in the NTS, and not in the NG. In addition to NPY-mediated energy homeostasis, leptin-melanocortin activation may be essential for metabolic disturbance-related hypertension. We found that leptin and a-melanocyte stimulating hormone (aMSH) receptors were aberrantly down-regulated in HFD rats. In addition, a-MSH concentrations were reduced and NPY concentrations were elevated in the serum and NTS at 60 and 90 min after acute leptin infusion. Electrophysiological recordings showed that the decay time-constant and area under the curve of excitatory post-synaptic currents were decreased by Y1R activation in A-types, whereas, both were increased by Y2R activation in Ah-or C-types. 9Strengthening in Al-,Mo-or Ti-doped CoCrFeNi high entropy alloys:A parallel comparison显示文摘In the current work,a parallel comparison of the influence of Al,Mo and Ti,on the microstructure and strengthening of the CoCrFeNi alloy was conducted.To achieve this,inconsistencies on variables including the extent of alloying,thermomechanical processing and property-evaluation method were avoided.Microstructurally,following cold-rolling,annealing of the 4 at.%Al-doped alloys at 800-1000℃ did not result in phase separation;nevertheless,that of the 4 at.%Mo-and Ti-doped alloys led to the respective formation ofσandηphase and,consequently,caused extra strengthening through the Orowan dislocation bypassing mechanism.Our systematic qualitative analysis and DFT calculations showed that Al and Ti are more effective than Mo in reducing the stacking fault energy(SFE)of the CoCrFeNi alloy,because they can induce more considerable deformation of electronic density,making the gliding of atomic layers easier.Following identical thermomechnical processing,Al-,Mo-,and Ti-doping causes different extent of solid solution strengthening and grain boundary strengthening.Mo causes the most pronounced solid solution strengthening but does not benefit the grain boundary strengthening;in contrast,the effectiveness of grain boundary strengthening is boosted by the doping Al and Ti.Current analyses support that Labusch instead of Fleischer mechanism is applicable to explain the differences in solid solution strengthening,and the observed differences in grain boundary strengthening arise from the different tendency of Al,Mo and Ti to reduce the SFE of CoCrFeNi.In addition,we determined the value of the dimensionless parameter f in the Labusch model for CoCrFeNi-based alloys and observed a close relation between Hall-Petch slope and SFE.Although more in-depth studies are needed to provide full and mechanistic understandings,both these findings in fact presents significant values toward designing novel singlephase high-strength CoCrFeNi-based alloys through manipulating the solid solution and grain boundary strengthening by compositional tuning.Xi Li Zhongtao Li Zhenggang Wu Shijun Zhao Weidong Zhang Hongbin Bei Yanfei Gao 2021Journal of Materials Science & Technology2021,,35:4
10Neural stem cell transplantation in the hippocampus of rats with cerebral ischemia/reperfusion injury Activation of the phosphatidylinositol-3 kinase/Akt pathway and increased brain-derived neurotrophic factor expression显示文摘The phosphatidylinositol-3 kinase (PI3K)/Akt pathway and brain-derived neurotrophic factor (BDNF) are involved in neurological functional recovery following cerebral ischemia. Therefore, we hypothesized that mechanisms of neuroprotection by transplantation of neural stem cells (NSCs) on cerebral ischemia contributed to activation of the PI3K/Akt pathway and enhanced BDNF expression. In the present study, Wortmannin (a specific, covalent inhibitor of PI3K) was administered adjacent to ischemic hippocampus by stereotactic transplantation to further confirm the neuroprotective mechanisms of NSC transplantation following cerebral ischemia. Results showed that focal infarct volume was significantly smaller in the NSCs group, but the neurological behavior score in the NSC group was significantly greater than the middle cerebral artery occlusion model group, Wortmannin treatment group, and NSCs + Wortmannin treatment group. Protein expression of BDNF was significantly greater in the NSC group compared with the Wortmannin treatment group and NSCs + Wortmannin treatment group. 12Formal Analysis of Trusted Platform Module Commands for Compromising User Key显示文摘The Trusted Platform Module (TPM) is a dedicated hardware chip designed to provide a higher level of security for computing platform. All TPM functionalities are implemented in TPM commands to achieve specific security goals. We attempt to analyze the security properties of these commands, especially the key management API. Our study utilizes applied pi calculus to formalize the commands and determine how their security properties affect TPM key management. The attacker is assumed to call TPM commands without bounds and without knowing the TPM root key, expecting to obtain or replace the user key. The analysis goal in our study is to guarantee the corresponding property of API execution and the integrity of API data. We analyze the security properties of TPM commands with a process reduction method, identify the key-handle hijack attack on a TPM newly created key, and propose reasonable solutions to solve the problem. Then, we conduct an experiment involving a key-handle attack, which successfully replaces a user key with an attacker's key using malicious TPM software. This paper discloses the weakness of the relationship between the key handle and the key object. 13Local-environment dependence of stacking fault energies in concentrated solid-solution alloys显示文摘Concentrated solid-solution alloys(CSAs)based on 3d transition metals have demonstrated extraordinary mechanical properties and radiation resistance associated with their low stacking fault energies(SFEs).Owing to the intrinsic disorder,SFEs in CSAs exhibit distributions depending on local atomic configurations.In this work,the distribution of SFEs in equiatomic CSAs of NiCo,NiFe,and NiCoCr are investigated based on empirical potential and first-principles calculations.We show that the calculated distribution of SFEs in chemically disordered CSAs depends on the stacking fault area using empirical potential calculations.Based on electronic structure calculations,we find that local variations of SFEs in CSAs correlate with the charge density redistribution in the stacking fault region.We further propose a bond breaking and forming model to understand and predict the SFEs in CSAs based on the local structure alone.It is shown that the perturbation induced by a stacking fault is localized in the first-nearest planes for NiCo,but extends up to the third nearest planes for NiFe and NiCoCr because of partially filled d electrons in Fe and Cr.Shijun Zhao Yuri Osetsky G.Malcolm Stocks Yanwen Zhang 2019npj Computational Materials2019,,1:2
14Ascorbate peroxidase 1 confers resistance to southern corn leaf blight in maize显示文摘Southern corn leaf blight (SCLB), caused by Bipolarismaydis, is one of the most devastatingdiseases affecting maize production. However,only one SLCB resistance gene, conferring partialresistance, is currently known, underscoring theimportance of isolating new SCLB resistancerelatedgenes. Here, we performed a comparativeproteomic analysis and identified 258 proteinsshowing differential abundance during the maizeresponse to B. maydis. These proteins included anascorbate peroxidase (Zea mays ascorbate peroxidase1 (ZmAPX1)) encoded by a gene locatedwithin the mapping interval of a previously identifiedquantitative trait locus associated with SCLBresistance. ZmAPX1 overexpression resulted inlower H_(2)O_(2) accumulation and enhanced resistanceagainst B. maydis. Jasmonic acid (JA)contents and transcript levels for JA biosynthesisand responsive genes increased in ZmAPX1-overexpressing plants infected with B. maydis,whereas Zmapx1 mutants showed the oppositeeffects. We further determined that low levels of H_(2)O_(2) are accompanied by an accumulation of JAthat enhances SCLB resistance. These resultsdemonstrate that ZmAPX1 positively regulatesSCLB resistance by decreasing H_(2)O_(2) accumulationand activating the JA-mediated defensesignaling pathway. This study identified ZmAPX1as a potentially useful gene for increasing SCLBresistance. 17Genomic Variations in the Tea Leafhopper Reveal the Basis of Its Adaptive Evolution显示文摘Tea green leafhopper(TGL),Empoasca onukii,is of biological and economic interest.Despite numerous studies,the mechanisms underlying its adaptation and evolution remain enigmatic.Here,we use previously untapped genome and population genetics approaches to examine how the pest adapted to different environmental variables and thus has expanded geographically.We complete a chromosome-level assembly and annotation of the E.onukii genome,showing notable expansions of gene families associated with adaptation to chemoreception and detoxification.Genomic signals indicating balancing selection highlight metabolic pathways involved in adaptation to a wide range of tea varieties grown across ecologically diverse regions.Patterns of genetic variations among 54 E.onukii samples unveil the population structure and evolutionary history across different tea-growing regions in China.Our results demonstrate that the genomic changes in key pathways,including those linked to metabolism,circadian rhythms,and immune system functions,may underlie the successful spread and adaptation of E.onukii.This work highlights the genetic and molecular basis underlying the evolutionary success of a species with broad economic impacts,and provides insights into insect adaptation to host plants,which will ultimately facilitate more sustainable pest management.Qian Zhao Longqing Shi Weiyi He Jinyu Li Shijun You Shuai Chen Jing Lin Yibin Wang Liwen Zhang Guang Yang Liette Vasseur Minsheng You 2022Genomics, Proteomics & Bioinformatics2022,20,6:1
18Modulating effects of Astragalus polysaccharide on immune disorders via gut microbiota and the TLR4/NF-κB pathway in rats with syndrome of dampness stagnancy due to spleen deficiency显示文摘The syndrome of dampness stagnancy due to spleen deficiency(DSSD)is relatively common globally.Although the pathogenesis of DSSD remains unclear,evidence has suggested that the gut microbiota might play a significant role.Radix Astragali,used as both medicine and food,exerts the effects of tonifying spleen and qi.Astragalus polysaccharide(APS)comprises a macromolecule substance extracted from the dried root of Radix Astragali,which has many pharmacological functions.However,whether APS mitigates the immune disorders underlying the DSSD syndrome via regulating gut microbiota and the relevant mechanism remains unknown.Here,we used DSSD rats induced by high-fat and low-protein(HFLP)diet plus exhaustive swimming,and found that APS of moderate molecular weight increased the body weight gain and immune organ indexes,decreased the levels of interleukin-1β(IL-1β),IL-6,and endotoxin,and suppressed the Toll-like receptor 4/nuclear factor-κB(TLR4/NF-κB)pathway.Moreover,a total of 27 critical genera were significantly enriched according to the linear discriminant analysis effect size(LEfSe).APS increased the diversity of the gut microbiota and changed its composition,such as reducing the relative abundance of Pseudoflavonifractor and Paraprevotella,and increasing that of Parasutterella,Parabacteroides,Clostridium XIVb,Oscillibacter,Butyricicoccus,and Dorea.APS also elevated the contents of short-chain fatty acids(SCFAs).Furthermore,the correlation analysis indicated that 12 critical bacteria were related to the body weight gain and immune organ indexes.In general,our study demonstrated that APS ameliorated the immune disorders in DSSD rats via modulating their gut microbiota,especially for some bacteria involving immune and inflammatory response and SCFA production,as well as the TLR4/NF-κB pathway.This study provides an insight into the function of APS as a unique potential prebiotic through exerting systemic activities in treating DSSD.Wenxiao ZHAO Chenchen DUAN Yanli LIU Guangying LU Qin LYU Xiumei LIU Jun ZHENG Xuelian ZHAO Shijun WANG Haijun ZHAO 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,7:1
