1Adenovirus-mediated gene delivery:Potential applications for gene and cell-based therapies in the new era of personalized medicine显示文摘With rapid advances in understanding molecular pathogenesis of human diseases in the era of genome sciences and systems biology,it is anticipated that increasing numbers of therapeutic genes or targets will become available for targeted therapies.Despite numerous setbacks,efficacious gene and/or cell-based therapies still hold the great promise to revolutionize the clinical management of human diseases.It is wildly recognized that poor gene delivery is the limiting factor for most in vivo gene therapies.There has been a long-lasting interest in using viral vectors,especially adenoviral vectors,to deliver therapeutic genes for the past two decades.Among all currently available viral vectors,adenovirus is the most efficient gene delivery system in a broad range of cell and tissue types.The applications of adenoviral vectors in gene delivery have greatly increased in number and efficiency since their initial development.In fact,among over 2000 gene therapy clinical trials approved worldwide since 1989,a significant portion of the trials have utilized adenoviral vectors.This review aims to provide a comprehensive overview on the characteristics of adenoviral vectors,including adenoviral biology,approaches to engineering adenoviral vectors,and their applications in clinical and preclinical studies with an emphasis in the areas of cancer treatment,vaccination and regenerative medicine.Current challenges and future directions regarding the use of adenoviral vectors are also discussed.It is expected that the continued improvements in adenoviral vectors should provide great opportunities for cell and gene therapies to live up to its enormous potential in personalized medicine.Cody S.Lee Elliot S.Bishop Ruyi Zhang Xinyi Yu Evan M.Farina Shujuan Yan Chen Zhao Zongyue Zeng Yi Shu Xingye Wu Jiayan Lei Yasha Li Wenwen Zhang Chao Yang Ke Wu Ying Wu Sherwin Ho Aravind Athiviraham Michael J.Lee Jennifer Moriatis Wolf Russell R.Reid Tong-Chuan He 2017Genes & Diseases2017,4,2:19
2Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases,which requires a thorough understanding of podocyte cell biology.As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago.However,the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells.In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites.We show the imPOD cells exhibit long-term high proliferative activity,which can be effectively reversed by FLP recombinase.The imPOD cells express most podocyte-related markers,including WT-1,Nephrin,Tubulin and Vinculin,but not differentiation marker Synaptopodin.The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions.Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan 2018Genes & Diseases2018,5,2:5
3The development of a sensitive fluorescent protein-based transcript reporter for high throughput screening of negative modulators of lncRNAs显示文摘While the human genome is pervasively transcribed,<2%of the human genome is transcribed into protein-coding mRNAs,leaving most of the transcripts as noncoding RNAs,such as microRNAs and long-noncoding RNAs(lncRNAs),which are critical components of epigenetic regulation.lncRNAs are emerging as critical regulators of gene expression and genomic stability.However,it remains largely unknown about how lncRNAs are regulated.Here,we develop a highly sensitive and dynamic reporter that allows us to identify and/or monitor negative modulators of lncRNA transcript levels in a high throughput fashion.Specifically,we engineer a fluorescent fusion protein by fusing three copies of the PEST destruction domain of mouse ornithine decarboxylase(MODC)to the C-terminal end of the codon-optimized bilirubin-inducible fluorescent protein,designated as dBiFP,and show that the dBiFP protein is highly destabilized,compared with the commonly-used eGFP protein.We further demonstrate that the dBiFP signal is effectively down-regulated when the dBiFP and mouse lncRNA H19 chimeric transcript is silenced by mouse H19-specific siRNAs.Therefore,our results strongly suggest that the dBiFP fusion protein may serve as a sensitive and dynamic transcript reporter to monitor the inhibition of lncRNAs by microRNAs,synthetic regulatory RNA molecules,RNA binding proteins,and/or small molecule inhibitors so that novel and efficacious inhibitors targeting the epigenetic circuit can be discovered to treat human diseases such as cancer and other chronic disorders.Zongyue Zeng Bo Huang Shifeng Huang Ruyi Zhang Shujuan Yan Xinyi Yu Yi Shu Chen Zhao Jiayan Lei Wenwen Zhang Chao Yang Ke Wu Ying Wu Liping An Xiaojuan Ji Cheng Gong Chengfu Yuan Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Rex C.Haydon Hue H.Luu Lan Zhou Russell R.Reid Tong-Chuan He Xingye Wu 2018Genes & Diseases2018,5,1:4
4Performance of plasma-sprayed CuNiIn coatings and Mo coatings subjected to fretting fatigue显示文摘During atmospheric plasma spraying,a cooling process usually plays an important role in the coating quality,especially for the oxidation containment of metallic coatings.CuNiIn and Mo coatings were prepared using atmospheric plasma spraying with different cooling processes.The obtained coatings were characterized in terms of microstructure,microhardness and tensile strengths.The relation between the coating microstructure and their fretting fatigue behavior was emphasized.The results show that the sensitivity of Mo coatings to the cooling process is lower than CuNiIn coatings.The resistance to fretting fatigue is determined by the coating microhardness,correlated with the contents of oxides and pores.The fretting wear mechanisms of both the coatings are galling,third body abrasive wear and material transfer.Shujuan Dong Yihui Wang Jinyan Zeng Xiong Yang Panpan Liang Dezheng Wang Huiqi Liao Hanlin Liao 2020Nano Materials Science2020,2,2:2

8An Old Story Retold: Loss of G1 Control Defines A Distinct Genomic Subtype of Esophageal Squamous Cell Carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC) has a high mortality rate. To determine the molecular basis of ESCC development, this study sought to identify characteristic genome-wide alterations in ESCC, including exonic mutations and structural alterations. The clinical implications of these genetic alterations were also analyzed. Exome sequencing and verification were performed for nine pairs of ESCC and the matched blood samples, followed by validation with additional samples using Sanger sequencing. Whole-genome SNP arrays were employed to detect copy number alteration(CNA) and loss of heterozygosity(LOH) in 55 cases, including the nine ESCC samples subjected to exome sequencing. A total of 108 non-synonymous somatic mutations(NSSMs) in102 genes were verified in nine patients. The chromatin modification process was found to be enriched in our gene ontology(GO) analysis. Tumor genomes with TP53 mutations were significantly more unstable than those without TP53 mutations. In terms of the landscape of genomic alterations, deletion of 9p21.3 covering CDKN2A/2B(30.9%), amplification of 11q13.3 covering CCND1(30.9%), and TP53 point mutation(50.9%) occurred in two-thirds of the cases. These results suggest that the deregulation of the G1 phase during the cell cycle is a key event in ESCC.Furthermore, six minimal common regions were found to be significantly altered in ESCC samples and three of them, 9p21.3, 7p11.2, and 3p12.1, were associated with lymph node metastasis. 9Downregulation of MANCR inhibits cancer cell proliferation in mantle cell lymphoma possibly by interacting with RUNX2显示文摘The mitotically associated lncRNA(MANCR)participates in breast cancer cell proliferation,while its involvement in other cancers is still unknown.In this study,we therefore studied the role of MANCR in mantle cell lymphoma(MCL).We found that serum MANCR and Runt-related transcription factor 2(RUNX2)were upregulated in MCL patients when compared with those in healthy controls.A positive correlation between serum MANCR and RUNX2 was found in MCL patients but not in controls.Upregulation of serum MANCR distinguished MCL patients from controls.MANCR overexpression promoted RUNX2 expression in MCL cells,while RUNX2 overexpression failed to significantly change the expression levels of MANCR.MANCR overexpression promoted the proliferation of MCL cells,while MANCR silencing inhibited the proliferation of MCL cells.In addition,RUNX2 overexpression attenuated the inhibitory effects of MANCR silencing on cell proliferation.However,MANCR overexpression and silencing had no significant effects on cell migration and invasion.Further bioinformatics analysis showed that MANCR may sponge miR-218 to upregulate RUNX2.Therefore,we conclude that downregulation of MANCR may inhibit cancer cell proliferation in MCL possibly by interacting with RUNX2.Shujuan Wen Min Zeng Yan Li Xin Hu Shan Li Xiao Liang Lin Zhu Shune Yang 2019Acta Biochimica et Biophysica Sinica2019,51,11:1

16Asymmetric supercapacitors based on high capacitance Ni6MnO8 and graphene显示文摘Supercapacitors with high energy density and high power density have attracted numerous attentions. Here, we successfully synthesized Ni6MnO8 material by a fast, facile, and cost-effective method, which the area specific capacitance was found to be as high as 1113 mF/cm^2 at a current density of 1 mA/cm^2. Further, the asymmetric supercapacitor was assembled by using Ni6MnO8 as the positive electrode and graphene as the negative electrode with an operation potential from 0 to 1.4 V. It exhibited an area specific capacitance of 69.1m F/cm^2 at 0.5 mA/cm^2 and stable cycling performance which presented about 80% capacitance retention after 5000 cycles at 5 mA/cm^2. 17Engineering the high-entropy phase of Pt-Au-Cu nanowire for electrocatalytic hydrogen evolution显示文摘Hydrogen economy,as the most promising alternative energy system,relies on the hydrogen production through sustainable water splitting which in turn relies on the high efficiency electrocatalysts.PtAuCu A1-phase alloy has been predicted to be a promising electrocatalyst for the hydrogen evolution.As such preferred phase of Pt-Au-Cu is not thermodynamically favored,herein,we stabilize PtAuCu alloy by engineering the high-entropy phase in the form of nanowire.Density functional theory(DFT)calculations indicate that,in comparison with the ordered phase and segregated phases with discrete hydrogen binding energy,the high-entropy phase provides a diverse combination of site composition to continuously tune the hydrogen binding energy,and thus generate a series of highly active sites for the hydrogen evolution.Reflecting the theoretical prediction,electrochemical tests show that the A1-phase PtAuCu nanowire significantly outperforms its nanoparticle counterpart with phase segregation,toward the electrocatalysis of hydrogen evolution,offering one of the best hydrogen evolution electrocatalysts.Yanan Yu Guangdong Liu Shuaihu Jiang Ruya Zhang Huiqiu Deng Eric AStach Shujuan Bao Zhenhua Zeng Yijin Kang 2023Nano Research2023,16,8:0

20Common Postzygotic Mutational Signatures in Healthy Adult Tissues Related to Embryonic Hypoxia显示文摘Postzygotic mutations are acquired in normal tissues throughout an individual’s lifetime and hold clues for identifying mutagenic factors.Here,we investigated postzygotic mutation spectra of healthy individuals using optimized ultra-deep exome sequencing of the time-series samples from the same volunteer as well as the samples from different individuals.In blood,sperm,and muscle cells,we resolved three common types of mutational signatures.Signatures A and B represent clocklike mutational processes,and the polymorphisms of epigenetic regulation genes influence the proportion of signature B in mutation profiles.Notably,signature C,characterized by C>T transitions at GpCpN sites,tends to be a feature of diverse normal tissues.Mutations of this type are likely to occur early during embryonic development,supported by their relatively high allelic frequencies,presence in multiple tissues,and decrease in occurrence with age.Almost none of the public datasets for tumors feature this signature,except for 19.6%of samples of clear cell renal cell carcinoma with increased activation of the hypoxia-inducible factor 1(HIF-1)signaling pathway.Moreover,the accumulation of signature C in the mutation profile was accelerated in a human embryonic stem cell line with drug-induced activation of HIF-1α.Thus,embryonic hypoxia may explain this novel signature across multiple normal tissues.Our study suggests that hypoxic condition in an early stage of embryonic development is a crucial factor inducing C>T transitions at GpCpN sites;and individuals’genetic background may also influence their postzygotic mutation profiles.Yaqiang Hong Dake Zhang Xiangtian Zhou Aili Chen Amir Abliz Jian Bai Liang Wang Qingtao Hu Kenan Gong Xiaonan Guan Mengfei Liu Xinchang Zheng Shujuan Lai Hongzhu Qu Fuxin Zhao Shuang Hao Zhen Wu Hong Cai Shaoyan Hu Yue Ma Junting Zhang Yang Ke Qian-Fei Wang Wei Chen Changqing Zeng 2022Genomics, Proteomics & Bioinformatics2022,20,1:0
