1When worlds collide: Th17 and Treg cells in cancer and autoimmunity显示文摘The balance between Th17 cells and regulatory T cells(Tregs)has emerged as a prominent factor in regulating autoimmunity and cancer.Th17 cells are vital for host defense against pathogens but have also been implicated in causing autoimmune disorders and cancer, though their role in carcinogenesis is less well understood.Tregs are required for self-tolerance and defense against autoimmunity and often correlate with cancer progression. This review addresses the importance of a functional homeostasis between these two subsets in health and the consequences of its disruption when these forces collide in disease.Importantly,we discuss the ability of Th17 cells to mediate cancer regression in immunotherapy, including adoptive transfer and checkpoint blockade therapy, and the therapeutic possibilities of purposefully offsetting the Th17/Treg balance to treat patients with cancer as well as those with autoimmune diseases.Hannah M.Knochelmann Connor J.Dwyer Stefanie R.Bailey Sierra M.Amaya Dirk M.Elston Joni M.Mazza-Mc.Crann Chrystal M.Paulos 2018Cellular & Molecular Immunology2018,15,5:50

3Biometric and ICL-related risk factors associated to sub-optimal vaults in eyes implanted with implantable collamer lenses显示文摘Background:To identify biometric and implantable collamer lens(ICL)-related risk factors associated with suboptimal postoperative vault in eyes implanted with phakic ICL.Methods:This study reports a retrospective case series of the first operated eye in 360 patients implanted with myopic spherical or toric ICL.Preoperatively,white-to-white(WTW),central keratometry(Kc)and central corneal thickness(CCT)were measured using the Pentacam.Anterior-segment optical coherence tomography(AS-OCT,Visante)was applied preoperatively for measuring the horizontal anterior-chamber angle-to-angle distance(ATA),internal anterior chamber depth(ACD),crystalline lens rise(CLR),anterior-chamber angle(ACA)and postoperatively the vault.Eyes were divided into three vault groups:low(LVG:≤250μm),optimal(OVG:>250 and<1000μm)and high(HVG:≥1000μm).Multinomial logistic regression(MLR)was used to find the sub-optimal vault predictors. Results:MLR showed that CLR,ICL size minus the ATA(ICL size-ATA),age,ICL spherical equivalent(ICLSE)and ICL size as contributing factors for sub-optimal vaults(pseudo-R2=0.40).Increased CLR(OR:1.01,CI:1.00-1.01)and less myopic ICLSE(OR:1.22,CI:1.07-1.40)were risk factors for low vaults.Larger ICL size-ATA(OR:41.29,CI:10.57-161.22)and the 13.7mm ICL(OR:7.08,CI:3.16-15.89)were risk factors for high vaults, whereas less myopic ICLSE(OR:0.85,CI:0.76-0.95)and older age(OR:0.92,CI:0.88-0.98)were protective factors. Conclusion:High CLR and low ICLSE were the major risk factors in eyes presenting low vaults. In the opposite direction,ICL size-ATA was the major contributor for high vaults. This relationship was more critical in higher myopic ICLSE,younger eyes and when 13.7mm ICL were used. The findings show that factors influencing the vault have differentiated weight of influence depending on the type of vault(low, optimal or high).Santiago Cerpa Manito Angel Sánchez Trancón Oscar Torrado Sierra António Manuel Baptista Pedro Miguel Serra 2021Eye and Vision2021,8,1:5
4Efficiency of white lupin in the removal of mercury from contaminated soils: Soil and hydroponic experiments显示文摘This study examined the ability of the white lupin to remove mercury (Hg) from a hydroponic system (Hg concentrations 0, 1.25, 2.5, 5 and 10 μmol/L) and from soil in pots and lysimeters (total Hg concentration (19.2 ± 1.9) mg/kg availability 0.07%, and (28.9 ± 0.4) mg/kg availability 0.09%, respectively), and investigated the accumulation and distribution of Hg in different parts of the plant. White lupin roots efficiently took up Hg, but its translocation to the harvestable parts of the plant was low. The Hg concentration in the seeds posed no risk to human health according to the recommendations of the World Health Organization, but the shoots should not be used as fodder for livestock, at least when unmixed with other fodder crops. The accumulation of Hg in the hydroponically-grown plants was linear over the concentration range tested. The amount of Hg retained in the roots, relative to the shoots, was almost constant irrespective of Hg dose (90%). In the soil experiments, Hg accumulation increased with exposure time and was the greater in the lysimeter than in the pot experiments. Although Hg removal was the greater in the hydroponic system, revealing the potential of the white lupin to extract Hg, bioaccumulation was the greatest in the lysimeter-grown plants; the latter system more likely reflects the true behaviour of white lupin in the field when Hg availability is a factor that limits Hg removal. The present results suggest that the white lupin could be used in long-term soil reclamation strategies that include the goal of profitable land use in Hg-polluted areas.Pilar Zornoza Rocío Milln M. 9Actinobacteria-enhanced plant growth, nutrient acquisition, and crop protection:Advances in soil, plant, and microbial multifactorial interactions显示文摘Agricultural areas of land are deteriorating every day owing to population increase, rapid urbanization, and industrialization. To feed today’s huge populations, increased crop production is required from smaller areas, which warrants the continuous application of high doses of inorganic fertilizers to agricultural land. These cause damage to soil health and, therefore, nutrient imbalance conditions in arable soils. Under these conditions, the benefits of microbial inoculants (such as Actinobacteria) as replacements for harmful chemicals and promoting ecofriendly sustainable farming practices have been made clear through recent technological advances. There are multifunctional traits involved in the production of different types of bioactive compounds responsible for plant growth promotion, and the biocontrol of phytopathogens has reduced the use of chemical fertilizers and pesticides. There are some well-known groups of nitrogen-fixing Actinobacteria, such as Frankia, which undergo mutualism with plants and offer enhanced symbiotic trade-offs.In addition to nitrogen fixation, increasing availability of major plant nutrients in soil due to the solubilization of immobilized forms of phosphorus and potassium compounds, production of phytohormones, such as indole-3-acetic acid, indole-3-pyruvic acid, gibberellins, and cytokinins, improving organic matter decomposition by releasing cellulases, xylanase, glucanases, lipases, and proteases, and suppression of soil-borne pathogens by the production of siderophores, ammonia, hydrogen cyanide, and chitinase are important features of Actinobacteria useful for combating biotic and abiotic stresses in plants. The positive influence of Actinobacteria on soil fertility and plant health has motivated us to compile this review of important findings associated with sustaining plant productivity in the long run.Debasis MITRA Rittick MONDAL Bahman KHOSHRU Ansuman SENAPATI T.K.RADHA Bhaswatimayee MAHAKUR Navendra UNIYAL Ei Mon MYO Hanane BOUTAJ Beatriz Elena GUERRA SIERRA Periyasamy PANNEERSELVAM Arakalagud Nanjundaiah GANESHAMURTHY Snežana ANÐELKOVIĆ Tanja VASIĆ Anju RANI Subhadeep DUTTA Pradeep K.DAS MOHAPATRA 2022Pedosphere2022,32,1:3
10H pylori receptor MHC classⅡcontributes to the dynamic gastric epithelial apoptotic response显示文摘AIM: To investigate the role of MHC classⅡin the modulation of gastric epithelial cell apoptosis induced by H pylon infection. METHODS: After stimulating a human gastric epithelial cell line with bacteria or agonist antibodies specific for MHC classⅡand CD95, the quantitation of apoptotic and anti-apoptotic events, including caspase activation, BCL-2 activation, and FADD recruitment, was performed with a fluorometric assay, a cytometric bead array, and confocal microscopy, respectively. RESULTS: Pretreatment of N87 cells with the anti-MHC classⅡIgM antibody RFD1 resulted in a reduction in global caspase activation at 24 h of H pylori infection. When caspase 3 activation was specifically measured, crosslinking of MHC class n resulted in markedly reduced caspase activation, while simple ligation of MHC classⅡdid not. Crosslinking of MHC class n also resulted in an increased activation of the anti-apoptosis molecule BCL-2 compared to simple ligation. Confocal microscope analysis demonstrated that the pretreatment of gastric epithelial cells with a crosslinking anti-MHC classⅡIgM blocked the recruitment of FADD to the cell surface. CONCLUSION: The ability of MHC class n to modulate gastric epithelial apoptosis is at least partially dependent on its crosslinking. The crosslinking of this molecule has anti-apoptotic effects during the earlier time points of H pylori infection. This effect is possibly mediated by the ability of MHC classⅡto modulate the activation of the pro-apoptotic receptor Fas by blocking the recruitment of the accessory molecule FADD, and this delay in apoptosis induction could allow for prolonged cytokine secretion by H pylori-infected gastric epithelial cells.David A Bland Giovanni Suarez Ellen J Beswick Johanna C Sierra Victor E Reyes 2006World Journal of Gastroenterology2006,12,29:3
11Acknowledgment of Libraries in the Journal Literature: An Exploratory Study显示文摘Purpose: This study examines acknowledgments to libraries in the journal literature, as well as the efficacy of using Web of Science(WoS) to locate general acknowledgment text.Design/methodology/approach: This mixed-methods approach quantifies and characterizes acknowledgments to libraries in the journal literature. Using WoS's Funding Text field, the acknowledgments for six peer universities were identified and then characterized. The efficacy of using WoS to locate library acknowledgments was assessed by comparing the WoS Funding Text search results to the actual acknowledgment text found in the articles.Findings: Acknowledgments to libraries were found in articles at all six peer universities, though the absolute and relative numbers were quite low(< 0.5%). Most of the library acknowledgments were for resources(collections, funding, etc.), and many were concentrated in natural history(e.g. zoology). Examination of Texas A&M University zoology articles found that 91.7% of the funding information came from 'acknowledgments' and not specifically a funding acknowledgment section. The WoS Funding Text search found 56% of the library acknowledgments compared to a search of the actual acknowledgment text in the articles.Research limitations: Limiting publications to journals, using a single truncated search term, and including only six research universities in the United States.Practical implications: This study examined library acknowledgments, but the same approach could be applied to searches of other keywords, institutions/organizations, individuals, etc. While not specifically designed to search general acknowledgments, WoS's Funding Text field can be used as an exploratory tool to search acknowledgments beyond funding. Originality/value: There are a few studies that have examined library acknowledgments in the scholarly literature, though to date none of those studies have examined the efficacy of using the WoS Funding Text field to locate those library acknowledgments within the journal literature.David E.Hubbard Sierra Laddusaw 2020Journal of Data and Information Science2020,5,3:2

13Relation of atrophic gastritis with Helicobacter pylori-CagA^+ and interleukin-1 gene polymorphisms显示文摘AIM: To determine the association of Helicobacter pylori (H pylori) CagA+ infection and pro-inflammatory poly-morphisms of the genes interleukin (IL)-1RN and IL-1B with the risk of gastric atrophy and peptic ulcers in a dyspeptic population in Costa Rica, a country with high incidence and mortality of gastric cancer. METHODS: Seven biopsy specimens, a fasting blood sample and a questionnaire concerning nutritional and sociodemographic factors were obtained from 501 con-secutive patients who had undergone endoscopy for dyspeptic symptoms. A histopathological diagnosis was made. Pepsinogen concentrations were analyzed by enzyme linked immunosorbent assay (ELISA). Infection with H pylori CagA+ was determined by serology and polymerase chain reaction (PCR). IL-1B and IL-1RN polymorphisms genotyping was performed by PCR-restriction fragment length polymorphism (PCR-RFLP)and PCR respectively. RESULTS: In this dyspeptic population, 86% were H pylori positive and of these, 67.8% were positive for CagA. Atrophic antral gastritis (AAG) was associated with CagA+ status [odd ratio (OR) = 4.1; P < 0.000] and fruit consumption (OR = 0.3; P < 0.00). Atrophic body gastritis (ABG) was associated with pepsinogen PGI/PGII < 3.4 (OR = 4.9; P < 0.04) and alcohol consumption (OR = 7.3; P < 0.02). Duodenal ulcer was associated with CagA+ (OR = 2.9; P < 0.04) and smoking (OR = 2.4; P < 0.04). PGI < 60 μg/L as well as PGI/PGII < 3.4 were associated with CagA+. CONCLUSION: In a dyspeptic population in Costa Rica, H pylori CagA+ is not associated with ABG, but it is a risk factor for AAG. The pro-inflammatory cytokine poly-morphisms IL-1B + 3945 and IL-1RN are not associated with the atrophic lesions of this dyspeptic population.Rafaela Sierra Clas Une Vanessa Ramírez Warner Alpízar-Alpízar María I González José A Ramírez Antoine de Mascarel Patricia Cuenca Guillermo Pérez-Pérez Francis Mégraud 2008World Journal of Gastroenterology2008,14,42:2

20Clearing the corpses: regulatory mechanisms, novel tools, and therapeutic potential of harnessing microglial phagocytosis in the diseased brain显示文摘Apoptosis is a widespread phenomenon that occurs in the brain in both physiological and pathological conditions. Dead cells must be quickly removed to avoid the further toxic effects they exert in the parenchyma, a process executed by microglia, the brain professional phagocytes. Although phagocytosis is critical to maintain tissue homeostasis, it has long been either overlooked or indirectly assessed based on microglial morphology, expression of classical activation markers, or engulfment of artificial phagocytic targets in vitro. Nevertheless, these indirect methods present several limitations and, thus, direct observation and quantification of microglial phagocytosis is still necessary to fully grasp its relevance in the diseased brain. To overcome these caveats and obtain a comprehensive, quantitative picture of microglial phagocytosis we have developed a novel set of parameters. These parameters have allowed us to identify the different strategies utilized by microglia to cope with apoptotic challenges induced by excitotoxicity or inflammation. In contrast, we discovered that in mouse and human epilepsy microglia failed to find and engulf apoptotic cells, resulting in accumulation of debris and inflammation. Herein, we advocate that the efficiency of microglial phagocytosis should be routinely tested in neurodegenerative and neurological disorders, in order to determine the extent to which it contributes to apoptosis and inflammation found in these conditions. Finally, our findings point towards enhancing microglial phagocytosis as a novel therapeutic strategy to control tissue damage and inflammation, and accelerate recovery in brain diseases.Irune Diaz-Aparicio Sol Beccari Oihane Abiega Amanda Sierra 2016Neural Regeneration Research2016,11,10:2
