1CMTM family proteins 1–8:roles in cancer biological processes and potential clinical value显示文摘The CKLF-like MARVEL transmembrane domain containing(CMTM)family of genes comprises CKLF and CMTM1–8(previously called chemokine-like factor superfamily 1–8,CKLFSF1–8).The CMTM family proteins contain a structurally conserved MAL and related proteins for vesicle trafficking and membrane linking(MARVEL)domain.Dysregulated expression of multiple CMTM family members is a common feature in many human cancer types.CMTM proteins control critical biological processes in cancer development,including growth factor receptor activation and recycling,cell proliferation,apoptosis,metastasis,and immune evasion.Emergingin vivo andin vitro evidence indicates that the mechanisms of action of most CMTM proteins are complex and multifactorial.This review highlights new findings regarding the roles of CMTM1–8 in cancer,particularly in tumor growth,metastasis,and immune evasion.Additionally,the potential clinical value of CMTMs as novel drug targets or biomarkers is discussed.Jie Wu Lan Li Siyi Wu Bin Xu 2020Cancer Biology & Medicine2020,17,3:12
2MiR156 regulates anthocyanin biosynthesis through SPL targets and other microRNAs in poplar显示文摘Anthocyanins biosynthesized from the flavonoid pathway are types of pigments that are involved in the protection of poplar from biotic and abiotic stresses.Previous researchers studying anthocyanin-related transcription factors and structural genes in poplar have made significant discoveries.However,little is known about the regulatory role of microRNAs in anthocyanin biosynthesis in poplar.Here,we overexpressed miR156 in poplar to study the comprehensive effects of the miR156-SPL module on the biosynthesis of anthocyanins.Small RNA sequencing analysis revealed 228 microRNAs differentially expressed in transgenic poplar plants with dramatically increased miR156 levels.Furthermore,integrated microRNAomic and transcriptomic analysis suggested that two microRNAs,miR160h,and miR858,have the potential to affect anthocyanin accumulation in poplar by regulating auxin response factors and MYB transcription factors,respectively.Additionally,the accumulation of miR160h and miR858 displayed a positive correlation with miR156 levels,suggesting a possible interaction between the miR156-SPL module and these microRNAs in poplar.Last,metabolomics analysis revealed that the levels of anthocyanins,flavones,and flavonols were substantially elevated in transgenic poplar plants overexpressing miR156 compared with the wild type,whereas the total lignin content was reduced in the transgenic plants.Taken together,our results indicate that miR156 can fine tune the anthocyanin biosynthetic pathway via multiple factors,including microRNAs,transcription factors,and the levels of structural genes,in poplar.This provides additional clues for understanding the complex regulatory network of anthocyanin biosynthesis in woody plants.Yamei Wang Wenwen Liu Xinwei Wang Ruijuan Yang Zhenying Wu HanWang LeiWang Zhubing Hu Siyi Guo Hailing Zhang Jinxing Lin Chunxiang Fu 2020Horticulture Research2020,7,1:9
3Colorectal cancer burden,trends and risk factors in China:A review and comparison with the United States显示文摘Objective:China and the United States(the U.S.)have the heaviest colorectal cancer(CRC)burden with considerable variations in temporal trends.This study aims to analyze the temporal patterns of CRC burden and its risk factors in China and the U.S.across the past three decades.Methods:Data were extracted from the Global Burden of Disease(GBD)Study in 2019,including cases,deaths,disability-adjusted life-years(DALYs),age-standardized rate(ASR),and summary exposure value(SEV)of CRC in China and the U.S.between 1990 and 2019.Annual average percentage changes(AAPCs)of CRC burden were calculated using the Joinpoint regression model.The mortality in CRC attributable to potential risk factors was characterized by countries,gender,and age groups.Results:In 2019,there were 607,900 and 227,241 CRC cases,and 261,777 and 84,026 CRC deaths in China and the U.S.,respectively.The age-standardized incidence rate(ASIR)was 30.55 per 100,000 in China and 41.86 per100,000 in the U.S.,and the age-standardized mortality rate(ASMR)was 13.86 per 100,000 in China and 14.77 per100,000 in the U.S.CRC incidence,mortality,and DALY rate in the U.S.showed downward trends in the past three decades(AAPC=-0.47,-1.06,and-0.88,respectively),while upward trends were observed in China(AAPC=3.11,1.05,and 0.91,respectively).Among the cause of CRC,the leading risk factor contributing to CRC death was low milk in China and smoking in the U.S.,respectively.Conclusions:From 1990 to 2019,the burden of CRC in China increased dramatically,particularly for males and middle-aged and elderly people.The management of the major risk factors associated with the high burden of CRC should be enhanced.Qianru Li Hongliang Wu Maomao Cao He Li Siyi He Fan Yang Xinxin Yan Shaoli Zhang Yi Teng Changfa Xia Ji Peng Wanqing Chen 2022Chinese Journal of Cancer Research2022,34,5:6
4The RING E3 ligase CLG1 targets GS3 for degradation via the endosome pathway to determine grain size in rice显示文摘G-protein signaling and ubiquitin-dependent degradation are both involved in grain development in rice,but how these pathways are coordinated in regulating this process is unknown.Here,we show that Chang Li Geng 1(CLG1),which encodes an E3 ligase,regulates grain size by targeting the Gγprotein GS3,a negative regulator of grain length,for degradation.Overexpression of CLG1 led to increased grain length,while overexpression of mutated CLG1 with changes in three conserved amino acids decreased grain length.We found that CLG1 physically interacts with and ubiquitinats GS3which is subsequently degraded through the endosome degradation pathway,leading to increased grain size.Furthermore,we identified a critical SNP in the exon 3 of CLG1 that is significantly associated with grain size variation in a core collection of cultivated rice.This SNP results in an amino acid substitution from Arg to Ser at position 163 of CLG1 that enhances the E3 ligase activity of CLG1 and thus increases rice grain size.Both the expression level of CLG1 and the SNP CLG1163S may be useful variations for manipulating grain size in rice.Wensi Yang Kun Wu Bo Wang Huanhuan Liu Siyi Guo Xiaoyu Guo Wei Luo Shengyuan Sun Yidan Ouyang Xiangdong Fu Kang Chong Qifa Zhang Yunyuan Xu 2021Molecular Plant2021,14,10:6
5全球尺度上氮添加影响植物生物量分配但不影响不同器官间的异速生长关系显示文摘生物量在不同器官间的分配是植物对环境变化响应的一个关键生态生理学过程。然而,在全球尺度上有关不同陆地生态系统植物生物量分配对氮沉降响应的认识还比较欠缺。本文通过整合分析333篇己发表文章的5474个观测值,基于“最优分配假说”和“异速分配假说”,评估了全球尺度上氮添加对植物生物量及其在不同器官间分配的影响。结果表明:(1)氮添加显著增加了整株植物或不同器官的生物量,降低了根冠比和根质量分数,但对叶质量分数和茎质量分数无显著影响;(2)氮添加对不同器官质量分数的影响受实验条件、植物功能性状、纬度、氮添加率等因子单独或交互作用的调控;(3)氮添加对生物量在不同器官间的异速分配率无显著影响,表明氮添加导致的根冠比和根质量分数减少是在异速分配模式下由整株植物生物量增加而致。虽然氮添加改变了地上和地下部分的质量分数,但不同器官生物量间异速分配模式的稳定性表明“异速分配假说”能更好地描述植物生物量分配对氮添加的响应规律。该研究结果将有助于深入认识氮沉降环境下植物生物量的分配规律,同时表明将生物量分配纳入有关氮沉降对陆地生态系统影响的预测模型中的重要性。Kai Yue Dario A.Fomara Wang Li Xiangyin Ni Yan Peng Shu Liao Siyi Tan Dingyi Wang Fuzhong Wu Yusheng Yang 2021Journal of Plant Ecology2021,14,3:5

7Resorption Efficiency of Four Cations in Different Tree Species in a Subtropical Common Garden显示文摘High rainfall in subtropical regions can leach cation elements from ecosystems,which may limit plant growth.Plants often develop efficient resorption patterns to recycle elements,but there is relatively little available information on this topic.In February 2012,a common garden was established in a subtropical forest by planting dominant trees from the area.Green and senescent leaves were sampled from 11 tree species.The concentrations of potassium(K),calcium(Ca),sodium(Na)and magnesium(Mg)were determined,and the resorption efficiencies were calculated.The results showed significant K,Na and Mg resorption in most of the investigated tree species,while Ca mainly displayed accumulation.Evergreen coniferous and evergreen broad-leaved trees(such as Cunninghamia lanceolata,Pinus massoniana,Cinnamomum camphora,and Michelia macclurei)exhibited relatively higher resorption efficiencies of K(39.0%-87.5%)and Na(18.3%-50.2%)than deciduous broad-leaved trees.Higher Mg resorption efficiencies(>50%)were detected in Liriodendron chinense,C.lanceolata and P.massoniana than in other trees.Overall,evergreen coniferous and evergreen broad-leaved trees could show higher cation resorption than deciduous broad-leaved trees.K and Mg resorption efficiencies and Ca accumulation decrease with increasing nutrient concentrations in green leaves.Our results emphasize that nutrient resorption patterns largely depend on elements and plant functions,which provides new insights into the nutrient use strategies of subtropical plants and a reference for the selection of suitable tree species in this region.Yaoyi Zhang Xiangyin Ni Jing Yang Siyi Tan Shu Liao Dingyi Wang Kai Yue Fuzhong Wu 2022Phyton-International Journal of Experimental Botany2022,91,1:2

12Taurocholic acid inhibits the response to interferon-αtherapy in patients with HBeAg-positive chronic hepatitis B by impairing CD8^(+)T and NK cell function显示文摘Pegylated interferon-alpha (PegIFNα) therapy has limited effectiveness in hepatitis B e-antigen (HBeAg)-positive chronic hepatitis B (CHB) patients. However, the mechanism underlying this failure is poorly understood. We aimed to investigate the influence of bile acids (BAs), especially taurocholic acid (TCA), on the response to PegIFNα therapy in CHB patients. Here, we used mass spectrometry to determine serum BA profiles in 110 patients with chronic HBV infection and 20 healthy controls (HCs). We found that serum BAs, especially TCA, were significantly elevated in HBeAg-positive CHB patients compared with those in HCs and patients in other phases of chronic HBV infection. Moreover, serum BAs, particularly TCA, inhibited the response to PegIFNα therapy in HBeAg-positive CHB patients. Mechanistically, the expression levels of IFN-γ, TNF-α, granzyme B, and perforin were measured using flow cytometry to assess the effector functions of immune cells in patients with low or high BA levels. We found that BAs reduced the number and proportion and impaired the effector functions of CD3^(+)CD8^(+) T cells and natural killer (NK) cells in HBeAg-positive CHB patients. TCA in particular reduced the frequency and impaired the effector functions of CD3^(+)CD8^(+) T and NK cells in vitro and in vivo and inhibited the immunoregulatory activity of IFN-α in vitro. Thus, our results show that BAs, especially TCA, inhibit the response to PegIFNα therapy by impairing the effector functions of CD3^(+)CD8^(+) T and NK cells in HBeAg-positive CHB patients. 15Gap edge canopy buffering of throughfall deposition in a subalpine natural forest显示文摘Base cation loads are rarely considered in forest gap edge canopies,but they can play critical roles in capturing or buffering atmospheric deposition in forests with frequent gap disturbances,such as subalpine forests.We selected an expanded gap edge canopy and a closed canopy in a subalpine natural forest on the eastern Tibetan Plateau.The throughfall deposition and canopy exchange processes of common base cations(K^(+),Ca^(2+),Na^(+),and Mg^(2+))were continuously studied over two years.The results showed that the enrichment ratio and fluxes had lower levels of base cations in the gap-edge canopy than in the closed canopy,which indicated that base cations were concentrated more in the closed canopy than in the gap-edge canopy.Although Ca^(2+)in the gap-edge canopy showed a higher net throughfall flux,the annual net throughfall fluxes of K^(+),Na^(+) and Mg^(2+) within the gap-edge canopy were 1.83,6.75 and 2.95 times lower than those in the closed canopy,respectively.Moreover,dry deposition fluxes of base cations significantly decreased in the gap edge canopy compared to those in the closed canopy,and the decreasing tendency was more significant during the snowy season than during the rainy season.Overall,these results suggest that the amount of base cations in subalpine natural forest ecosystems may be overestimated when the throughfall deposition of ions in gap edge canopies is ignored.Siyi Tan Qing Dong Xiangyin Ni Kai Yue Shu Liao Fuzhong Wu 2022Forest Ecosystems2022,9,4:0
16Increased microbial sequestration of soil organic carbon under nitrogen deposition over China’s terrestrial ecosystems显示文摘Background:China’s terrestrial ecosystems have been receiving increasing amounts of reactive nitrogen(N)over recent decades.External N inputs profoundly change microbially mediated soil carbon(C)dynamics,but how elevated N affects the soil organic C that is derived from microbial residues is not fully understood.Here,we evaluated the changes in soil microbial necromass C under N addition at 11 forest,grassland,and cropland sites over China’s terrestrial ecosystems through a meta-analysis based on available data from published articles.Results:Microbial necromass C accounted for an average of 49.5%of the total soil organic C across the studied sites,with higher values observed in croplands(53.0%)and lower values in forests(38.6%).Microbial necromass C was significantly increased by 9.5%after N addition,regardless of N forms,with greater stimulation observed for fungal(+11.2%)than bacterial(+4.5%)necromass C.This increase in microbial necromass C under elevated N was greater under longer experimental periods but showed little variation among different N application rates.The stimulation of soil microbial necromass C under elevated N was proportional to the change in soil organic C.Conclusions:The stimulation of microbial residues after biomass turnover is an important pathway for the observed increase in soil organic C under N deposition across China’s terrestrial ecosystems.Shu Liao Siyi Tan Yan Peng Dingyi Wang Xiangyin Ni Kai Yue Fuzhong Wu Yusheng Yang 2020Ecological Processes2020,9,1:0

18Development of an accurate lateral flow immunoassay for PEDV detection in swine fecal samples with a filter pad design显示文摘Porcine epidemic diarrhea virus(PEDV),as the main causative pathogen of viral diarrhea in pigs,has been reported to result in high morbidity and mortality in neonatal piglets and cause significant economic losses to the swine industry.Rapid diagnosis methods are essential for preventing outbreaks and transmission of this disease.In this study,a paper-based lateral flow immunoassay for the rapid diagnosis of PEDV in swine fecal samples was developed using stable color-rich latex beads as the label.Under optimal conditions,the newly developed latex bead-based lateral flow immunoassay(LBs-LFIA)attained a limit of detection(LOD)as low as 10^(3.60) TCID_(50)/mL and no cross-reactivity with other related swine viruses.To solve swine feces impurity interference,by adding a filtration unit design of LFIA without an additional pretreatment procedure,the LBs-LFIA gave good agreement(92.59%)with RT-PCR results in the analysis of clinical swine fecal samples{n=108),which was more accurate than previously reported colloidal gold LFIA(74.07%)and fluorescent LFIA(86.67%).Moreover,LBs-LFIA showed sufficient accuracy(coefficient of variance[CV]<15%)and stable(room temperature storage life>56 days)performance for PEDV detection,which is promising for on-site analysis and user-driven testing in pig production system.Siyi Zou Lei Wu Gan Li Juan Wang Dongni Cao Tao Xu Aiqing Jia Yong Tang 2021Animal Diseases2021,1,3:0
