| 1 | Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy. | Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou | 2022 | Cell Research2022,32,6: | 10 |
| 2 | Numerical Investigation of the Centrifugal Compressor Stability Improvement by Half Vaned Low Solidity Diffusers显示文摘Centrifugal compressors for the fuel cell vehicles often operate near the surge line compared with the turbocharger compressors.Low solidity and half vaned diffusers are recognized as good ways to improve the stability of the centrifugal compressor.The presented work investigated four diffuser configurations (i.e.,the vaneless diffuser (VLD),full-height low solidity vaned diffuser (LSVD),hub-side half vaned diffuser (HVD) and shroud-side half vaned diffuser (SVD)) through steady-state and unsteady numerical simulations.The results show that the best performance is achieved by the LSVD,HVD and SVD at the design,surge and choke conditions.The flow rate at the surge operating point of the HVD has decreased by 15.53% compared with the LSVD,and 9.21% compared with the VLD.At near surge operating point,a longitudinal suction side passage vortex is formed on the hub of the LSVD and rotates as circumferential stall cells.A hairpin vortex is formed along the leading edge and is dragged by the main flow along the suction side as a local vortex shedding.The mechanism of the stability improvement by half vaned diffusers is that the tip leakage vortex migrates from the clearance side to the vane mounting side and replenishes the low-momentum zone on the mounting side.The best position where the half vaned diffuser should be mounted is based on the impeller outlet flow conditions,namely,the location of the wake region,where the meridional velocity and relative stagnation pressure is low. | CHEN Siyue ZUO Shuguang WEI Kaijun | 2021 | Journal of Thermal Science2021,30,2: | 1 |
| 3 | One‑Step Gas-Solid‑Phase Diffusion‑Induced Elemental Reaction for Bandgap‑Tunable Cu_(a)Agm_(1)Bim_(2)I_(n)/CuI Thin Film Solar Cells显示文摘Lead-free inorganic copper-silver-bismuth-halide materials have attracted more and more attention due to their environmental friendliness,high element abundance,and low cost.Here,we developed a strategy of one-step gas-solid-phase diffusioninduced reaction to fabricate a series of bandgap-tunable Cu_(a)Agm_(1)Bim_(2)I_(n)/CuI bilayer films due to the atomic diffusion effect for the first time.By designing and regulating the sputtered Cu/Ag/Bi metal film thickness,the bandgap of Cu_(a)Agm_(1)Bim_(2)I_(n)/CuI could be reduced from 2.06 to 1.78 eV.Solar cells with the structure of FTO/TiO_(2)/Cu_(a)Agm_(1)Bim_(2)I_(n)/CuI/carbon were constructed,yielding a champion power conversion efficiency of 2.76%,which is the highest reported for this class of materials owing to the bandgap reduction and the peculiar bilayer structure.The current work provides a practical path for developing the next generation of efficient,stable,and environmentally friendly photovoltaic materials. | Erchuang Fan Manying Liu Kangni Yang Siyu Jiang Bingxin Li Dandan Zhao Yanru Guo Yange Zhang Peng Zhang Chuantian Zuo Liming Ding Zhi Zheng | 2023 | Nano-Micro Letters2023,15,4: | 0 |
