1Translational Regulation of Plant Response to High Temperature by a Dual-Function tRNAHls Guanylyltransferase in Rice显示文摘As sessile organisms,plants have evolved numerous strategies to acclimate to changes in environmental temperature.However,the molecular basis of this acclimation remains largely unclear.In this study we identified a tRNAHis guanylyltransferase,AET1,which contributes to the modification of pre-tRNAH,s and is required for normal growth under high-temperature conditions in rice.Interestingly,AET1 possibly interacts with both RACK1A and elF3h in the endoplasmic reticulum.Notably,AET1 can directly bind to OsARF mRNAs including the uORFs of OsARF19 and OsARF23,indicating that AET1 is associated with translation regulation.Furthermore,polysome profiling assays suggest that the translational status remains unaffected in the aet1 mutant,but that the translational efficiency of OsARF19 and OsARF23 is reduced;moreover,OsARF23 protein levels are obviously decreased in the aet1 mutant under high temperature,implying that AET1 regulates auxin signaling in response to high temperature.Ourfindings provide new insights into the molecular mechanisms whereby AET1 regulates the environmental temperature response in rice by playing a dual role in tRNA modification and translational control.Ke Chen Tao Guo Xin-Min Li Yi-Min Zhang Yi-Bing Yang Wang-Wei Ye Nai-Qian Dong Chuan-Lin Shi Yi Kan You-Huang Xiang Hai Zhang Ya-Chao Li Ji-Ping Gao Xuehui Huang Qiang Zhao Bin Han Jun-Xiang Shan Hong-Xuan Lin 2019Molecular Plant2019,12,8:4
2Cytoplasmic and nuclear genome variations of rice hybrids and their parents inform the trajectory and strategy of hybrid rice breeding显示文摘The male sterility(MS)line is a prerequisite for efficient production of hybrid seeds in rice,a self-pollinating species.MS line breeding is pivotal for hybrid rice improvement.Understanding the historical breeding tra-jectory will help to improve hybrid rice breeding strategies.Maternally inherited cytoplasm is an appro-priate tool for phylogenetic reconstruction and pedigree tracing in rice hybrids.In this study,we analyzed the cytoplasmic genomes of 1495 elite hybrid rice varieties and identified five major types of cytoplasm,which correspond to different hybrid production systems.As the cytoplasm donors for hybrids,461 MS lines were also divided into five major types based on cytoplasmic and nuclear genomic architecture.Spe-cific core accessions cooperating with different fertility-associated genes drove the sequence divergence of MS lines.Dozens to hundreds of convergent and divergent selective sweeps spanning several agro-nomic trait-associated genes were identified among different types of MS lines.We further analyzed the cross patterns between different types of MS lines and their corresponding restorers.This study system-atically analyzed the cytoplasmic genomes of rice hybrids revealed their relationships with nuclear ge-nomes of MS lines,and illustrated the trajectory of hybrid rice breeding and the strategies for breeding different types of MS lines providing new insights for future improvement of hybrid rice.Zhoulin Gu Zhou Zhu Zhen Li Qilin Zhan Qi Feng Congcong Zhou Qiang Zhao Yan Zhao Xiaojian Peng Bingxin Dai Rongrong Sun Yan Li Hengyun Lu Lei Zhang Tao Huang Junyi Gong Danfeng Lv Xuehui Huang Bin Han 2021Molecular Plant2021,14,12:2

13Role of social organization engagement in high-risk groups intervention against HIV/AIDS:a case study from 176 cities of China显示文摘Background: A high-risk prevention strategy is an effective way to fight against human immunodeficiency virus (HIV) and acquired immunodeficiency syndrome (AIDS). The China AIDS Fund for Non-Governmental Organizations (CAFNGO) was established in 2015 to help social organizations intervene to protect high-risk populations in 176 cities. This study aimed to evaluate the role of social organizations in high-risk population interventions against HIV/AIDS.Methods: This study was based on the CAFNGO program from 2016 to 2020. The collected data included the number and types of social organizations participating in high-risk group interventions and the amount of funds obtained by these organizations each year. We explored the factors influencing the number of newly diagnosed AIDS cases using a spatial econometric model. Furthermore, we evaluated the effectiveness of intervention activities by comparing the percentages of the individuals who initially tested positive, and the individuals who took the confirmatory test, as well as those who retested positive and underwent the treatment.Results: Overall, from 2016 to 2020, the number of social organizations involved in interventions to protect HIV/AIDS high-risk populations increased from 441 to 532, and the invested fund increased from $3.98 to $10.58 million. The number of newly diagnosed cases decreased from 9128 to 8546 during the same period. Although the number of cities with overall spatial correlations decreased, the spatial agglomeration effect persisted in the large cities. Citywise, the number of social organizations (direct effect 19.13), the permanent resident population (direct effect 0.12), GDP per capita (direct effect 17.58;indirect effect -15.38), and passenger turnover volume (direct effect 5.50;indirect effect -8.64) were the major factors influencing new positive cases confirmed through the testing interventions performed by the social organizations. The initial positive test rates among high-risk populations were below 5.5%, the retesting rates among those who initially tested positive were above 60%, and the treatment rates among diagnosed cases were above 70%.Conclusions: The spatial effect of social organizations participating in interventions targeting high-risk populations funded by CAFNGO is statistically significant. Nevertheless, despite the achievements of these social organizations in tracking new cases and encouraging treatment, a series of measures should be taken to further optimize the use of CAFNGO. Working data should be updated from social organizations to CAFNGO more frequently by establishing a data monitoring system to help better track newly diagnosed AIDS cases. 14Aberrant nuclear lamina contributes to the malignancy of human gliomas显示文摘Glioma is the most common type of tumor in the central nervous system, accounting for around 80% of all malignant brain tumors. Previous studies showed a significant association between nuclear morphology and the malignant progress of gliomas. By virtue of integrated proteomics and genomics analyses as well as experimental validations, we identify three nuclear lamin genes(LMNA, LMNB1, and LMNB2) that are significantly upregulated in glioma tissues compared with normal brain tissues. We show that elevated expressions of LMNB1, LMNB2, and LMNA in glioma cells are highly associated with the rapid progression of the disease and the knockdown of LMNB1, LMNB2, and LMNA dramatically suppresses glioma progression in both in vitro and in vivo mouse models. Moreover, the repression of glioma cell growth by lamin knockdown is mediated by the p Rb-mediated G1-S inhibition. On the contrary, overexpression of lamins in normal human astrocytes dramatically induced nuclear morphological aberrations and accelerated cell growth. Together, our multi-omics-based analysis has revealed a previously unrecognized role of lamin genes in gliomagenesis, providing a strong support for the key link between aberrant tumor nuclear shape and the survival of glioma patients.
